OBJECTIVE: Polycystic ovary syndrome (PCOS) is a condition of androgen excess and chronic anovulation frequently associated with insulin resistance. We combined a nontargeted and targeted metabolomics approach to identify pathways and metabolites that distinguished PCOS from metabolic syndrome (MetS).
METHODS: Twenty obese women with PCOS were compared with 18 obese women without PCOS. Both groups met criteria for MetS but could not have diabetes mellitus or take medications that treat PCOS or affect lipids or insulin sensitivity. Insulin sensitivity was derived from the frequently sampled in

Realizing fluorescence emission from non-aromatic amino acids is a difficult challenge in the development of optical materials in the absence of polymeric backbones and covalent cross-linkers. Herein, a non-covalent approach is used to fabricate a series of bulk glasses exhibiting strong fluorescence from non-aromatic amino acids (ʟ-arginine, ʟ-lysine, ʟ-proline, and ʟ-histidine) by co-assembling them with tetrafluorophthalic acid (FA). FA, amino acids, and non-covalent bonding between them play essential roles in both glass formation and fluorescent properties. The high viscosity and rapid an

Lipolysis is an essential metabolic process that releases unesterified fatty acids from neutral lipid stores to maintain energy homeostasis in living organisms. Adipose triglyceride lipase (ATGL) plays a key role in intracellular lipolysis and can be coactivated upon interaction with the protein comparative gene identification-58 (CGI-58). The underlying molecular mechanism of ATGL stimulation by CGI-58 is incompletely understood. Based on analysis of evolutionary conservation, we used site directed mutagenesis to study a C-terminally truncated variant and full-length mouse ATGL providing insi

Feline dysautonomia (FD) is a multiple system neuropathy of unknown aetiology. An apparently identical disease occurs in horses (equine grass sickness, EGS), dogs, rabbits, hares, sheep, alpacas and llamas. Horses with acute EGS have a marked reduction in plasma concentrations of the sulphur amino acids (SAA) cyst(e)ine and methionine, which may reflect exposure to a neurotoxic xenobiotic. The aim of this study was to determine whether FD cats have alterations in amino acid profiles similar to those of EGS horses. Amino acids were quantified in plasma/serum from 14 FD cats, 5 healthy in-contac

Chaerophyllum macrospermum is consumed in parts of Iran for its nutritional and antioxidant properties, but rapid postharvest yellowing limits its shelf-life. This study assessed its nutritional composition, bioactive compounds, and postharvest preservation strategies. Edible parts (leaf-blades, buds, petioles) were analyzed for nutrients, antioxidants, essential oils, fatty acids, amino acids, and phenolics. Buds contained the highest levels of nitrogen, zinc, and protein, while leaves were richest in iron and manganese. Fresh essential oil was dominated by α-pinene (26.35%) and α-ocimene, wh

An infant with a prenatal diagnosis of citrullinemia, who started standard treatment at birth (L-arginine; sodium benzoate and a personalized diet characterized by a low protein intake and supplementation of essential nutrients and amino acids), presented at 4 months of age with extended, progressive, and severe skin lesions consistent with acrodermatitis dysmetabolica. Guidelines for the diagnosis and management of urea cycle disorders underline that a low-protein diet places patients at risk of essential fatty acids, trace elements, and vitamin deficiency. At hospital admission, our patient

Element composition of whole canine heartworms (Dirofilaria immitis) was determined using inductively coupled argon plasma emission spectroscopy. The elements evaluated included Na, K, Ca, Mg, P, Al, B, Ba, Cu, Fe, Mn, Sr, Zn, As, and Pb. The only significant sex difference was calcium concentration which was higher in females. Amino acid analyses of body fluid revealed presence of nine essential and eight non-essential amino acids. Concentrations for male and female heartworms were not significantly different. The spectrum of amino acids was similar to that previously reported for canine plas

Amino acids and proteins in whole saliva of normal and malnourished Egyptian infants and young children were investigated. 16 amino acids, 7 of them are essential, could be detected in saliva of normal infants. Electrophoretic analysis revealed the existence of 10 protein components in saliva of either normal or malnourished infants. In oedematous cases, salivary aminoacids, particularly the essential ones, were increased as well as most of the protein components. In non-oedematous cases, the pattern was more or less normal, but in 3rd grade marasmus, amino acid and protein components of saliv

We have tested the effect of substitution of an amino acid at position 195 of the F protein of mumps virus on cell-to-cell fusion caused by the virus. Introduction of amino acids with aromatic side chains into this position resulted in reduction of fusion induction. Furthermore the F protein was not cleaved when the amino acid at this position was substituted by several amino acids, suggesting that the amino acid at this position was essential to keep the tertiary structure of the protein that might be required not only for proper folding of the protein but also for induction of membrane fusio

Malaria remains a significant global health burden, particularly in regions like Nigeria. This study investigated the impact of malaria infection on plasma amino acid and micronutrient levels in a Nigerian population. A cross-sectional study was conducted on 200 participants, including malarious and healthy controls. Plasma levels of amino acids and selected micronutrients (Ca, Mg, Na, K, Zn, Fe, Co) were measured using HPLC and ICP-MS, respectively. Results revealed significant alterations (p < 0.01) in plasma amino acid profiles (except histidine and proline) with increasing malaria severity

In suckling hyperphenylalaninemic (hyper-Phe) rats, all essential amino acids including tryptophan are depleted in the blood. The inadequate supply of Trp to the developing brain leads to a decline of Trp, of serotonin, and of 5-hydroxyindoleacetic acid. The exhaustion of amino acids in both blood and brain can be restored by administration of Lys. Even though Phe is still elevated in blood and brain, Trp, serotonin and 5-hydroxyindoleacetic acid, are no longer depleted in the brain. This observation contradicts the idea that the serotonin deficit in the developing hyper-Phe brain is caused by

Trans-placental transport of amino acids is vital for the developing fetus. Using the BeWo cell line as a placental model, we investigated the effect of restricting amino acid availability on amino acid transport system type A. BeWo cells were cultured either in amino acid-depleted (without non-essential amino acids) or control media for 1, 3, 5 or 6 h. System A function was analysed using alpha(methyl-amino)isobutyric acid (MeAIB) transcellular transport studies. Transporter (sodium coupled neutral amino acid transporter (SNAT1/2)) expression was analysed at mRNA and protein level by Northern

Amino acid requirements of a Chlamydia trachomatis were defined in McCoy cell monolayer cultures, depleted of their nutrient reserves by maintenance in Earle balanced salt solution for at least 2 days, and then treated at infection with 1 microgram of cycloheximide per ml, an inhibitor of host cell protein and deoxyribonucleic acid synthesis. Serum-free Waymouth MB 752/1 medium lacking one amino acid or a group of related amino acids was added to depleted cultures before infection. Eight of the amino acids normally present in MB 752/1 were essential for chlamydial growth, six were advantageous

Amino acids are the building blocks for producing proteins. They are essential to the efficient operation of body functions and the structure of cells. They supply the energy needed for normal cell division and growth. An amino acid that is classified as semi-essential or conditionally essential is arginine. Arginine is essential for maintaining the immune system and for treating several disorders. Numerous studies have demonstrated the effectiveness of using dietary supplements containing arginine and other amino acids in improving a range of medical conditions. This review will cover the cla

Apart from cancer, metabolic reprogramming is also prevalent in other diseases, such as bacterial infections. Bacterial infections can affect a variety of cells, tissues, organs, and bodies, leading to a series of clinical diseases. Common Pathogenic bacteria include Helicobacter pylori, Salmonella enterica, Mycobacterium tuberculosis, Staphylococcus aureus, and so on. Amino acids are important and essential nutrients in bacterial physiology and support not only their proliferation but also their evasion of host immune defenses. Many pathogenic bacteria or opportunistic pathogens infect the ho

The biosynthesis of structural and signaling molecules depends on intracellular concentrations of essential amino acids, which are maintained by a specific system of plasma membrane transporters. We identify a unique population of nutrient amino acid transporters (NATs) within the sodium-neurotransmitter symporter family and have characterized a member of the NAT subfamily from the larval midgut of the Yellow Fever vector mosquito, Aedes aegypti (aeAAT1, AAR08269), which primarily supplies phenylalanine, an essential substrate for the synthesis of neuronal and cuticular catecholamines. Further
