Reproductive and developmental toxicity of degradation products of refrigerants in experimental animals.

The present paper summarizes the results of animal studies on the reproductive and developmental toxicity of the degradation products of refrigerants, including trifluoroacetic acid (TFA), carbon dioxide (CO(2)), carbon monoxide (CO), carbonyl fluoride (CF), hydrogen fluoride (HF) and formic acid (FA). Excessive CO(2) in the atmosphere is testicular and reproductive toxic, embryolethal, developmentally neurotoxic and teratogenic in experimental animals. As for CO, maternal exposure causes prenatal and postnatal lethality and growth retardation, skeletal variations, cardiomegaly, blood biochemi

Studies on the glycemic and lipidemic effect of Murraya koenigii in experimental animals.

Diabetes is often accompanied by lipid abnormalities, which contribute significantly to cardiovascular morbidity and mortality in diabetic patients. Previously, we have demonstrated potent hypoglycemic activity of lyophilized aqueous extract of Murraya koenigii leaves in normal and alloxan induced diabetic rabbits for short duration of 6 h. In this study, we examined the effect of 1 month oral administration of Murraya koenigii aqueous leaves extract in normal and STZ induced severe diabetic rats, at the dose of 300 mg/kg bw, on various biochemical parameters, viz., fasting blood glucose (FBG)

Evaluation of antipyretic activity of Calotropis gigantea (Asclepiadaceae) in experimental animals.

The roots of Calotropis gigantea have been used in leprosy, eczema, syphilis, elephantiasis, ulceration and cough in the Indian system of traditional medicine. The present communication evaluated its antipyretic activity by using yeast-induced and TAB (Typhoid) vaccine-induced pyrexia in rats and rabbits. In both yeast-induced and TAB vaccine-induced fever, the fever was significantly reduced and the body temperature was normalized by administration of 200 and 400 mg/kg dose intraperitoneally. Based on the results of the present study it can be concluded that the extract of C. gigantea has pot

Insights into the pathogenesis of hydrocephalus from transgenic and experimental animal models.

Hydrocephalus is a progressive brain disorder characterized by abnormalities in the flow of cerebrospinal fluid (CSF) and ventricular dilatation that leads to cerebral atrophy, and if left untreated, can be fatal. Genetic mutations, congenital malformations, infectious diseases, intracerebral hemorrhages and tumors are common conditions resulting in hydrocephalus. Although the causes of obstructive hydrocephalus are better understood, the mechanisms resulting in chronic, progressive communicating congenital and acquired hydrocephalus are less well understood. In this regard, recent studies in

Anxiolytic and anticonvulsive activity of Sesbania grandiflora leaves in experimental animals.

Various parts of Sesbania grandiflora have been used in the Indian system of medicine, in particular, the leaves of S. grandiflora are used in Ayurveda for the treatment of epileptic fits. In the present study we have evaluated the anticonvulsive activity of S. grandiflora leaves using a variety of animal models of convulsions. Bioassay guided separation was also carried out to identify the fraction possessing anticonvulsant activity. The benzene:ethyl acetate fraction (BE) of the acetone soluble part of a petroleum ether extract significantly delayed the onset of convulsions in pentylenetetra

Role of cysteinyl leukotrienes in nociceptive and inflammatory conditions in experimental animals.

The leukotrienes are potent inflammatory mediators, which may have a role in inflammatory diseases such as allergic rhinitis, inflammatory bowl disease and asthma. Zafirlukast, a cysteinyl leukotriene receptor antagonist, is claimed to be effective in asthma. However, it is not known whether these leukotrienes are involved in nociceptive and peripheral inflammation. The present study aimed to assess the role of cysteinyl leukotrienes in nociceptive and inflammatory conditions in experimental animals. Central nociception was assessed with tail-flick and hot-plate methods and peripheral nocicept

Animal experimental implantation of an atrial septal defect occluder system.

OBJECTIVE: To establish the implantation technique for the atrial septal defect occluder system (ASDOS) device in an experimental animal model and to determine long term mechanical stability of the device and its in vivo properties in terms of biocompatibility and tissue reaction. MATERIALS AND METHODS: An atrial septal defect was created and the device implanted in 17 pigs (mean weight 30 kg). The implantation technique was refined and modified because of initial technical and anatomical complications during nine acute pilot studies. The technique proved to be feasible in eight subsequent su

Interaction of air pollutants and pulmonary allergic responses in experimental animals.

Asthma, which is primarily an allergic type of respiratory disease, has increased in the U.S. and Europe by 30% over the last decade. Air pollution may play a role in this rise, since during episodes of smog, hospital admissions due to asthma increase. Ambient air quality has generally improved since the Clean Air Act was implemented in 1971 however, and has led some investigators to suggest that the increased risk of asthma is associated with a deterioration of indoor air quality through the introduction of closed ventilation systems and constant climate control. Thus, although the direct hea

The trypanocidal effect of Cannabis sativa constituents in experimental animal trypanosomiasis.

The effect of Cannabis sativa on trypanosome-infected rats was examined. An aqueous extract of the seeds administered at a dose of 50 mg/kg/d cured animals infected with Trypanosome brucei brucei of blood stream parasites. Six fractions eluted from the crude extract by column chromatography were assessed for trypanocidal properties. Of these, only 2 fractions retained trypanocidal activity by curing mice infected with T brucei brucei.

Allergenic potential of gonadotrophic preparations in experimental animals: relevance of purity.

Local reactions have been frequently reported following repeated injections of human menopausal gonadotrophins (HMG) for the treatment of infertility. Also immunoglobulin (Ig) E-mediated systemic reactions have sporadically been observed. Since most HMG preparations contain significant amounts of non-hormonal urine-derived proteins, it was suggested that these contaminating proteins are responsible for the various allergic reactions. In order to verify this hypothesis, different human follicle stimulating hormone (HFSH) and HMG preparations (Metrodin and Pergonal from Ares-Serono, and Humegon

Pharmacokinetics of antibiotics in natural and experimental superficial compartments in animals and humans.

Transcapillary exchange of antibiotics and other small molecules is diffusion driven and occurs in the capillary beds of the tissues. Small polar molecules are ionized at physiological pH and diffusion is pore-mediated. More lipophilic substances can also leave capillaries by the transcellular route. Splanchnic tissues have fenestrated capillary walls while somatic tissues have mainly micropores in their capillary walls. Under normal physiological conditions the ratio of capillary surface area to volume of fluid present (SA/V) is very large (> 100) and the rate of exchange of substances betwee

Valvular heart disease: medical therapy and experimental and animal models.

Vasodilators have been advocated for the treatment of regurgitant valvular heart diseases for more than 12 years, and new information on their mechanisms of action is available. Anticoagulants continue as a mainstay of therapy for patients with prosthetic heart valves, and the most desirable levels of anticoagulation for different conditions have been examined. Clinical and animal research studies of cardiac adaptation to mitral and aortic valve diseases are discussed.

[Effectiveness of combined use of verapamil and adenosine triphosphate in experimental animals].

A wide application of ATP to abolish reciprocal paroxysmal tachycardia is limited by frequent and adverse effects of the drug. Experiments on 49 rats have revealed that successive intravenous injections of verapamil and ATP have a more pronounced and prolonged action than the drugs alone. It is assumed that decreased pacemaker activity of sinus nodal cells and reduced atrioventricular conduction ("binodal dysfunction") are as a result of drug calcium channel block in the cellular membranes of myocytes. Potentiation of the pharmacological action of the drugs in question may be used in the clini

Cancer cachexia, malnutrition, and tissue protein turnover in experimental animals.

The role of malnutrition in the development of cachexia in rats bearing the Yoshida ascites hepatoma AH-130 was investigated by comparing the changes in tissue protein turnover in these animals with those observed in pair-fed controls. The tumor elicited in rats an early and conspicuous loss of body weight and tissue waste. Protein loss was particularly prominent for the gastrocnemius muscle and the heart and less pronounced for the soleus, while the diaphragm was little affected. Liver, kidneys, and spleen transiently increased in weight then regressed and eventually atrophied, while adrenals

Pathogenesis of toxicity with human-derived interleukin-2 in experimental animals.

It should be pointed out that although repeated systemic administration of exogenous rIL-2 in nonphysiological doses to mice, rats, and monkeys produced various toxicity-related exaggerated pharmacodynamic effects in many experimental parameters, pathological findings were substantially similar among all species. Furthermore, because toxicities observed in our studies were identical to those reported in humans dosed with rIL-2, preclinical toxicity using animal models was fully predictive of the pathogenesis of rIL-2 in human. With continuous infusion, which maintained high blood concentration

Experimental infection of Thailand Taenia (Chiengmai strain) in domestic animals.

Twenty-five gravid proglottides of a Thailand Taenia were obtained from a patient in Chiengmai, Thailand, and brought to our laboratory. The tapeworm was determined to be T. saginata-like by counting uterine branches (mean number 16, range 12-19 on each side). The eggs from these proglottides remained infective under storage at room temperature for 14 days followed by refrigeration (4-8 degrees C) for 131 days. Eight Small-Ear-Miniature pigs and two Holstein calves were each fed with 3000 eggs and sacrificed 12-76 days afterwards. Six pigs became infected and 16 cysticerci were recovered from

A modification of the rat kidney transplantation model: reduction of experimental animals.

Inbred rats display several abnormalities of the renal arteries, which are relevant for the technique used in the kidney transplantation model. After analysis of anatomical variations of renal vascular supply in inbred Lewis rats we describe an improved standardized technique of bilateral kidney grafting. After harvesting both kidneys from the donor animal they can be transplanted into 2 recipient animals. That allows to save up to 50% of donor animals.
