Impact of coronavirus disease 2019 restrictions on the efficacy of atropine 0.01% eyedrops for myopia control - Findings from the Western Australia Atropine for the Treatment of Myopia study.

This study explored the impact of short-term coronavirus disease 2019 (COVID-19) restrictions on the efficacy of atropine 0.01% eyedrops on myopia control in a multiethnic cohort of Australian children. In the Western Australia Atropine for the Treatment of Myopia study, 104 and 49 children were randomized to receive atropine 0.01% eyedrops and a placebo, respectively. We compared the 1-year myopia progression and axial elongation following the 2-month lockdown in 2020 to the same months in 2019 and 2021, i.e., the 1-year myopia progression up to May 2019-October 2019 (non-COVID-19) versus the…

In vitro activation of the medial septum-diagonal band complex generates atropine-sensitive and atropine-resistant hippocampal theta rhythm: an investigation using a complete septohippocampal preparation.

The medial septum and diagonal band complex (MS-DB) is believed to play a key role in generating theta oscillations in the hippocampus, a phenomenon critical for learning and memory. Although the importance of the MS-DB in hippocampal theta rhythm generation is generally accepted, it remains to be determined whether the MS-DB alone can generate hippocampal oscillations or is only a transducer of rhythmic activity from other brain areas. Secondly, it is known that hippocampal theta rhythm can be separated into an atropine-sensitive and insensitive component. However, it remains to be establishe…

Short-term effects of atropine on the retina and choroid in young adults.

BACKGROUND: Low concentration atropine is widely prescribed to slow myopia progression in children, yet its short-term retinal and choroidal effects remain incompletely understood. This study aimed to evaluate short-term effects of a range of low atropine concentrations on axial length, retinal and choroidal thickness, and microvasculature. METHODS: In this double-masked, randomized study, twenty healthy adults received a single instillation of placebo, 0.01%, 0.025%, 0.05%, or 0.1% atropine in the right eye across five separate sessions. Retinal and choroidal thickness in the central 1.0 mm …

The role of atropine in myopia control: insights into choroidal and scleral mechanisms.

In this study, we investigate the inhibitory effects of atropine on the progression of experimental myopia by targeting the functions of the choroid and sclera and exploring its potential therapeutic mechanisms. Form deprivation myopia (FDM) was induced in C57BL/6 mice, with treatment groups receiving atropine. We assessed the effects on ocular morphology, extracellular matrix (ECM) protein expression, choroidal and scleral thickness, and choroidal vascular index (CVI) through histopathology, immunofluorescence, and quantitative quantitative polymerase chain reaction (qPCR). In vitro, mouse sc…

Comparison of cycloplegic effects of atropine and tropicamide in children with refractive error.

BACKGROUND: Refractive error is the primary cause of vision impairment in children and requires accurate cycloplegic refraction for proper diagnosis and treatment. Various cycloplegic agents are used clinically, but their comparative efficacy remains controversial, particularly across different pediatric age groups and refractive error types. The purpose of this study was to observe the cycloplegic effects of tropicamide and atropine in children of different ages and different refractive errors, and to screen out more suitable drugs for cycloplegia in children with different refractive errors.…

Additively manufactured microplate for the simultaneous colorimetric and electrochemical detection of atropine.

This work presents the development of a dual mode electrochemical and colorimetric sensing platform, produced in a single print through additive manufacturing. The cell design was based on the dimensions of a standard 96-well plate, with the base replaced by a disc electrode made from bespoke conductive polypropylene for the electrochemical testing, with the walls created from transparent non-conductive polypropylene to allow for the colorimetric tests. This new system was employed for the detection of atropine (ATP) in two distinct steps within the same electrochemical cell: (1) colour change…

Accidental foodborne poisoning by atropine and scopolamine: a fatal case report.

Tropane alkaloids such as atropine and scopolamine are naturally occurring compounds found in various plants of the Solanaceae family, notably Datura species. Although widely used in clinical settings, their potent anticholinergic properties can lead to severe toxic effects. Fatalities involving these substances remain rare, particularly in accidental contexts. Here, the authors report the case of a 58-year-old male who died following the accidental consumption of a meal suspected to have been contaminated with plant material containing tropane alkaloids. At autopsy, blackish vomitus containin…

Preservatives and pH in low-dose atropine formulations for clinical trials.

Over a century ago, atropine has been tested to arrest myopia progression with good results. In recent years, many randomized clinical trials have tested different concentrations against placebo. Three recent such studies with low-dose atropine showed that it was less effective than previous studies, even the last one showing no difference in myopia progression between the treated and control group. Previous randomized studies had been performed in Asian populations, and these last three were extended to Western Caucasian populations, based on the initial observation that differences in iris p…

Synergism between cocaine and atropine at the caudal ventrolateral medulla of cats.

We have previously reported that the anticholinergic properties of cocaine may be important in cocaine induced apneusis. We have studied the effects of the cholinergic muscarinic antagonist atropine (ATR) on cocaine induced apneusis at the caudal chemosensitive areas of the ventrolateral medulla oblongata (CVLM). Experiments were performed in urethane anesthetized and tracheotomized cats with the CVLM surgically exposed. Topical application of ATR (44 mM ) to the CVLM produced significant decrements in minute ventilation (V(E)) and mean arterial blood pressure (MABP) (P<0.05) but the effects o…
