Here we describe the construction of an artificial enzyme bearing a biomimetic PLP-like catalytic center within an apo-myoglobin scaffold through covalent incorporation of an abiological salicylaldehyde moiety. By mimicking the substrate activation strategy employed by PLP-dependent enzymes, the engineered catalyst promotes the formation of reactive glycine ester nucleophiles and enables direct α-functionalization in C-C bond-forming reactions. This work establishes a biocatalytic platform for the synthesis of amino acid derivatives and illustrates how the integration of non-natural catalytic

Aldolases are powerful biocatalysts for the stereoselective formation of carbon-carbon bonds and are widely used in the synthesis of chiral intermediates for pharmaceutical applications. Among them, 2-deoxyribose-5-phosphate aldolase (DERA) has been extensively exploited for the preparation of the conserved side chain of statins. In this work, we report a novel chemoenzymatic approach for the synthesis of nucleobase-substituted lactol products as potential precursors of new statin analogues. A C49M variant of DERA from Pectobacterium atrosepticum (PaDERA C49M) was employed to catalyze sequenti

Vector-borne diseases pose a rising global health challenge, necessitating the development of safe and effective pest protection agents. Here, we report a highly selective biocatalytic direct Prins cyclohydration for the synthesis of (1R)-cis-p-menthane-3,8-diol (PMD), a natural insect repellent with high efficacy. By strategically engineering squalene-hopene cyclases (SHCs), we achieved >96% diastereomeric excess, surpassing previous synthetic methods. Structural and mechanistic analyses suggest direct Prins cyclohydration and a precisely positioned water molecule within the enzyme's active p

Vibrios have sustained various types of ocean ecosystems, being key players in marine mineral cycles and essential partners in specific groups of marine life. Observed genome plasticity and metabolic versatility are some of the unique biological features of vibrios, and these traits could contribute in expanding their ecological niche in marine environments. Vibrios are now recognized as ecophysiologically essential microbial species for our planet. At the time to "Sustainable Development Goals" (SDGs), their genome plasticity and metabolic versatility have also been studied with the aim of so

Pyridoxal 5'-phosphate (PLP)-dependent enzymes are among the most versatile biocatalysts, yet transformations involving the Cγ-nucleophilic vinylglycine quinonoid (VGQ) intermediate remain exceptionally rare. Understanding the untapped reactivity of VGQ could open new avenues for developing PLP-dependent biocatalysts. Here, we establish a biocatalytic platform that artificially accesses and exploits the reactivity of this high-energy intermediate. By reprogramming SphA, a PLP-dependent enzyme that natively catalyzes decarboxylative Claisen condensation, to generate VGQ in situ through facile d

The semipinacol rearrangement comprises a valuable class of synthetic organic transformations that provide access to useful molecular scaffolds via C-C bond cleavage and formation. Naturally occurring semipinacolases have thus far been limited to examples in alkaloid or polyketide biosynthesis, and the only new-to-nature semipinacolase was reported based on Brønsted acid catalysis with limited substrate scope. Herein, we report that flavin-dependent halogenases (FDHs) can catalyze enantioselective halogenative semipinacol rearrangement of prochiral allylic alcohols. This biocatalytic platform

Halogenation reactions are essential for producing many fine and bulk chemicals. Vanadium-dependent chloroperoxidases (VCPOs) are promising biocatalysts for a more sustainable production of halogenated products because they are cofactor independent, use hydrogen peroxide (H2O2) as oxygen donor, tolerate high H2O2 concentrations, and catalyze all halogenations except fluorination. Despite their outstanding catalytic potential, few VCPOs have been identified yet, and detailed information on their catalytic characteristics is lacking. Here, we report the heterologous expression of three previousl

Acrylic acid is a bulk chemical predominantly produced via fossil-based processes. Here, we repurpose and engineer a thiamine diphosphate (ThDP)-dependent enzyme to catalyze direct H 2 PO 4 - elimination from dihydroxyacetone phosphate (DHAP) for acrylic acid synthesis. Quantum chemical calculations reveal the catalytic mechanism involving isomerization, enol-ThDP intermediate formation, H 2 PO 4 - elimination, and hydrolysis. Using an automated high-throughput screening platform, we perform directed evolution and obtain a 13.6-fold improved variant, designated acrylic acid synthase (A

The α,β-desaturation of esters is one of the most challenging transformations in organic synthesis. While transition-metal-catalyzed methods have significantly advanced in the last decades, they often require high catalyst loading and nongreen solvents and rarely enable stereoselective transformations. Here we show that whole cells of Rhodococcus erythropolis enable a dynamic enantioconvergent desaturation of 4,5-disubstituted γ-butyrolactones (Quercus-like lactones), funneling all four initial stereoisomers into a single (R)-configured product with excellent enantioselectivity (up to ee 99%).

With the rapid industrial development, massive fossil fuel use has caused excessive carbon dioxide (CO2) emissions, triggering global warming and environmental issues. Thus, CO2 recovery and reuse have become a research focus, among which artificially designed in vitro biocatalytic pathways for converting CO2 into high-value chemicals show promise, with advantages like shorter routes, higher efficiency and lower energy consumption compared to natural pathways. However, challenges remain due to natural enzymes' issues in specificity, affinity, efficiency, stability and oxygen sensitivity. To ta

Efficient and cost-effective conversion of CO2 to biomass holds the potential to address the climate crisis. Light-driven CO2 conversion can be realized by combining inorganic semiconductors with enzymes or cells. However, designing enzyme cascades for converting CO2 to multicarbon compounds is challenging, and inorganic semiconductors often possess cytotoxicity. Therefore, there is a critical need for a straightforward semiconductor biohybrid system for CO2 conversion. Here, we used a visible-light-responsive and biocompatible C3N4 porous nanosheet, decorated with formate dehydrogenase, forma

Ketoreductase (KRED)-catalyzed asymmetric reduction of prochiral ketones is an attractive method to synthesize chiral alcohols. Herein, two KREDs LfSDR1-V186A/E141I and CgKR1-F92I with complementary stereopreference were identified towards reduction of apremilast prochiral ketone intermediate 1a. LfSDR1-V186A/E141I exhibited >99% conversion and 99.2% ee yielding an apremilast chiral alcohol intermediate ((R)-2a) at 50 g L-1 substrate loading. Furthermore, we investigated the substrate scope of β-keto sulfones by using LfSDR1-V186A/E141I and CgKR1-F92I to produce both enantiomers of the corresp

Alcohol dehydrogenases (ADHs; EC 1.1.1.1) have been widely used for the reversible redox reactions of carbonyl compounds (i.e., aldehydes and ketones) and primary or secondary alcohols, often resulting in optically pure hydroxyl products with high added value. In this work, we report a concise chemoenzymatic route toward xanthine-based enantiomerically pure active pharmaceutical ingredients (API) - proxyphylline, xanthinol, and diprophylline employing various recombinant short-chain ADHs with (R)- or (S)-selectivity as key biocatalysts. By choosing the appropriate ADH, the (R)- as well as the

We define paracatalysis as the acceleration of a reaction that appears abnormal or nonphysiological. With the high specificity of enzymes, side reactivity of this kind is typically negligible. However, enzyme paracatalysis can be amplified to levels that are biologically significant through interactions with a special class of small molecule "antagonist", here termed a paracatalytic inducer. Compounds with this unusual mode of action tend to be natural products, identified by chance through phenotypic screens. In this Perspective, we suggest two general types of paracatalytic inducer. The firs

Polyketides form a structurally diverse and pharmaceutically important class of secondary metabolites. Both diversity and biological activity are largely facilitated by post-polyketide synthase tailoring including methylation, oxidation, reduction, glycosylation, and dimerization. Cytochrome P450 enzymes (CYPs), flavin-dependent monooxygenases (FMOs), and laccases are known to catalyze phenol coupling in the biosynthesis of polyketide dimers. Polyketide homodimers resulting from enzyme catalysis are often formed in a highly regio- and stereoselective manner, in contrast to analogous nonenzymat

Herein, we describe the discovery of a new catalytic promiscuity of ene reductases (EREDs), which enables the selective reduction of azoarenes to the corresponding hydroazoarenes using MorB. The role of active site residues is confirmed by generating H186A and N189A variants of MorB, which show considerable loss of activity towards azo bond reduction. This transformation expands the known catalytic repertoire of EREDs and provides a sustainable biocatalytic route to valuable hydroazo compounds.

Redox-controlled rotaxane formation can be employed as a novel strategy for cyclodextrin interconversion via a process of template-directed enzymatic synthesis, mechanical capture and release. Mechanical bonds are shown to protect cyclodextrins from hydolysis by α-amylase and cyclodextrin glucanotransferase, facilitating rotaxane isolation and enabling size-selective separation of cyclodextrins via reversible rotaxane formation.
