Therapies for Prevention and Treatment of Alzheimer's Disease.

Alzheimer's disease (AD) is the most common cause of dementia associated with a progressive neurodegenerative disorder, with a prevalence of 44 million people throughout the world in 2015, and this figure is estimated to double by 2050. This disease is characterized by blood-brain barrier disruption, oxidative stress, mitochondrial impairment, neuroinflammation, and hypometabolism; it is related to amyloid-β peptide accumulation and tau hyperphosphorylation as well as a decrease in acetylcholine levels and a reduction of cerebral blood flow. Obesity is a major risk factor for AD, because it in

Thromboxane may be important in the organ damage and hypotension of malaria.

Generalized circulatory changes, manifesting as pulmonary oedema, acute renal failure, liver damage and severe hypotension, are well recognized aspects of acute falciparum malaria. The organ pathology is thought to be associated with a restricted local blood flow. These aspects of falciparum malaria are strikingly analogous to the shock syndrome which follows trauma, the injection of indotoxin, or some bacterial infections. Two of the cyclooxygenase products of arachidonic acid, thromboxane A2 and prostacyclin, acting through their opposing effects on vasoactivity and platelet aggregation, are

Accidental inhalation of mercury vapour: respiratory and toxicologic consequences.

Four adults, including a pregnant woman, and three children were admitted to hospital following accidental exposure to mercury vapour produced by heating mercury-gold amalgam. Initial symptoms and signs included a paroxysmal cough, dyspnea, chest pain, tachypnea, nausea, vomiting, fever and leukocytosis. Pulmonary function testing performed on the second day after exposure revealed air-flow obstruction and minor restrictive defects in three patients. The diffusing capacity of the lung for carbon monoxide was reduced in two of these patients. The mean initial blood mercury level (+/- one standa

Preoperative HAIC enlarges future liver remnant in initial unresectable HCC patients with Vp3 tumor thrombus.

BACKGROUND: Conversion strategy is the optimal treatment for patients with unresectable hepatocellular carcinoma (uHCC). However, a severe lack of future liver remnant (FLR) may result in early withdrawal. In this small sample retrospective study, we investigated FLR changes after conversion therapies. Notably, patients with ipsilateral Vp3 portal vein tumor thrombus (PVTT) showed a significant FLR increase after treatment and provided an advantage for carrying out radical surgery. METHODS: 53 uHCC patients with Vp0 to Vp3 tumor thrombus were enrolled and divided into two groups (Vp0-2 and Vp

Hepatorenal Syndrome in Focus: Emerging Diagnostic Criteria and Current Therapeutic Approaches.

Hepatorenal syndrome (HRS) is a phenotype of kidney injury specific to patients with liver cirrhosis and ascites, characterized by impaired kidney function due to reduced blood flow and reduced glomerular filtration rate. HRS can occur quickly with acute kidney injury (HRS-AKI). In those not meeting HRS-AKI criteria, this can develop as HRS-acute kidney disease (HRS-AKD) or HRS-chronic kidney disease (HRS-CKD). This pathophysiology occurs along a continuum, dependent on timing of the disease course, and the presence of functional and structural abnormalities. Traditionally, pharmacologic treat

The role of placental hormones and metabolites in modulating uterine circulation in physiological and pathological pregnancies.

The adaptation of the uterine circulation during pregnancy is fundamental to ensure an adequate supply of oxygen and nutrients to the fetus, and this process is largely orchestrated by placental hormones/metabolites. In this review, we comprehensively examine the role of placental hormones, growth factors, and proteins in mediating vascular remodeling, vasodilation, and angiogenesis within the uterine circulation under both physiological and pathological conditions. Key molecules such as estrogens, progesterone, relaxin, VEGF, PlGF, and PTHrP, among others, promote structural and functional ad

A Novel Mutation in Insulin-Like Growth Factor 1 Receptor (c.641-2A>G) Is Associated with Impaired Growth, Hypoglycemia, and Modified Immune Phenotypes.

INTRODUCTION: Insulin-like growth factor 1 receptor (IGF1R) mutations lead to systemic disturbances in growth and glucose homeostasis due to widespread IGF1R expression throughout the body. IGF1R is expressed by innate and adaptive immune cells, facilitating their development and exerting immunomodulatory roles in the periphery. CASE PRESENTATION: We report on a family presenting with a novel heterozygous IGF1R mutation with characterization of the mutation, IGF1R expression, and immune phenotyping. Twin probands presented clinically with short stature and hypoglycemia. Variable phenotypic ex

Circulating CD22+/CD19-/CD24- progenitors and CD22+/CD19+/CD24- mature B cells: Diagnostic pitfalls for minimal residual disease detection in B-lymphoblastic leukemia.

BACKGROUND: Multiparametric flow cytometry (MFC) has become a powerful tool in minimal residual disease (MRD) detection in B-lymphoblastic leukemia/lymphoma (B-ALL). In the setting of targeted immunotherapy, B-ALL MRD detection often relies on alterative gating strategies, such as the utilization of CD22 and CD24. It is important to depict the full diversity of normal cell populations included in the alternative B-cell gating methods to avoid false-positive results. We describe two CD22-positive non-neoplastic cell populations in the peripheral blood (PB), including one progenitor population o

Trends in hydrogel-based encapsulation technologies for advanced cell therapies applied to limb ischemia.

Ischemia occurs when blood flow is reduced or restricted, leading to a lack of oxygen and nutrient supply and removal of metabolites in a body part. Critical limb ischemia (CLI) is a severe clinical manifestation of peripheral arterial disease. Atherosclerosis serves as the main cause of CLI, which arises from the deposition of lipids in the artery wall, forming atheroma and causing inflammation. Although several therapies exist for the treatment of CLI, pharmacotherapy still has low efficacy, and vascular surgery often cannot be performed due to the pathophysiological heterogeneity of each pa

The Efficacy of Blood Flow Restriction Training to Improve Quadriceps Muscle Function after Anterior Cruciate Ligament Reconstruction.

BACKGROUND: Blood flow restriction training (BFRT) is a popular rehabilitation intervention after anterior cruciate ligament reconstruction (ACLR). However, there are a lack of clinical trials establishing the efficacy of using BFRT during rehabilitation to improve quadriceps muscle function. PURPOSE: The purpose of this study is to evaluate the efficacy of blood flow restriction training to improve quadriceps muscle strength, morphology, and physiology, and knee biomechanics in individuals after ACLR in a double-blind, randomized, placebo-controlled clinical trial (NCT03364647). METHODS: Fo

The clinical trial of Women On the Move through Activity and Nutrition (WOMAN) study.

The Women On the Move through Activity and Nutrition (WOMAN) study is the first randomized clinical trial of nonpharmacological intervention designed to modify lipoproteins, weight loss and exercise among postmenopausal women using noninvasive measures of atherosclerosis as the primary endpoint. The trial was initially designed to test whether intervention as compared to health education would be more effective in slowing progression of subclinical atherosclerosis among women on hormone therapy (HT), estrogen or estrogen+progestin. It was designed and implemented prior to the results of the Wo

A review on the alterations in the brain of persistent postural-perceptual dizziness patients and non-pharmacological interventions for its management.

Persistent postural perceptual dizziness (PPPD) is a relatively newer term, and this term is included in the International Classification of Diseases in its 11th revision. The typical features of PPPD include the presence of persistent dizziness, non-spinning vertigo, and unsteadiness, and these symptoms are exacerbated during upright posture, movement, or visual stimuli. Moreover, the structural changes have also been identified in the brains of PPPD patients, particularly in visual, vestibular, and limbic areas. These include a decrease in the volume and gyration of gray matter, a decrease i

Functional dystonia.

Although currently lacking a sensitive and specific electrophysiologic battery test, functional (psychogenic) dystonia can sometimes be diagnosed with clinically definite certainty using available criteria. Certain regional phenotypes have been recognized as distinctive, such as unilateral lip and jaw deviation, laterocollis with ipsilateral shoulder elevation and contralateral shoulder depression, fixed wrist and finger flexion with relative sparing of the thumb and index fingers, and fixed foot plantar flexion and inversion. The pathophysiologic abnormalities in functional dystonia overlap s

Pulmonary functions in Egyptian children with transfusion-dependent β-thalassemia.

BACKGROUND: In β-thalassemia, there are varying degrees of ineffective haematopoiesis, intermittent haemolysis and iron overload. Excess iron is deposited in organs such as the heart, the liver, the endocrine glands and the lungs. OBJECTIVES: To evaluate the pulmonary functions in asymptomatic beta thalassemic children on regular transfusion therapy and their relation to iron overload. METHODS: The study included 50 transfusion-dependent β-thalassemic children and 50 apparently healthy children as control. All children had undergone pulmonary function tests (spirometry, lung volumes and diff

Hypoxia and anemia: effects on tumor biology and treatment resistance.

In locally advanced solid tumors, oxygen (O2) delivery is frequently reduced or even abolished. This is due to abnormalities of the tumor microvasculature, adverse diffusion geometries, and tumor-associated and/or therapy-induced anemia. Up to 50-60% of locally advanced solid tumors may exhibit hypoxic and/or anoxic tissue areas that are heterogeneously distributed within the tumor mass. In approximately 30% of pretreatment patients, a decreased O2 transport capacity of the blood as a result of tumor-associated anemia can greatly contribute to the development of tumor hypoxia. While normal tis

Personalized haemodynamic modelling for non-invasive quantification of arterial stiffness in the human vasculature.

Arterial stiffness is a pivotal predictor of cardiovascular events, yet its in vivo characterization remains elusive because arterial wall mechanics cannot be directly measured. Conventional inverse approaches require high-resolution imaging to resolve wall motion, restricting their clinical applicability. Here, we present a non-invasive computational framework that infers regional arterial stiffness solely from routinely acquired pressure and blood flow velocity waveforms by leveraging personalized one-dimensional haemodynamic models of the human arterial network. Patient-specific models were

[Arterial thyroid embolization in thyroid diseases].

Traditional forms of treatment of the thyroid diseases: pharmacotherapy, radioiodine therapy and surgery can not always be applied. Intolerance, side effects of antithyroid drugs, low iodine uptake, high risk of surgery or disagreement with the proposed treatment was the reason for seeking alternative healing methods. With the development of interventional radiology, and gained experience in the use of arterial embolization, this method has become possible to use in treatment of thyroid diseases. The essence of this treatment is to shut down blood flow in major arteries of the thyroid by direc

Biomimetic Silica Nanomotors Loaded with Food-Derived Antithrombotic Peptides for NIR Photothermal Efficient Clot Thrombolysis-Anticoagulation.

Here we report a fuel-free, all-optical platform that overcomes key limitations of conventional thrombolysis and offers a promising strategy for targeted, combined thrombolytic-anticoagulant vascular therapy based on food-derived bioactive substances. Thrombotic diseases remain difficult to treat because mature fibrin-rich clots restrict drug penetration, while systemic thrombolytics carry substantial bleeding risks. To address these challenges, we developed a near-infrared (NIR)-propelled biomimetic nanomotor (NM) loaded with a food-derived antithrombotic peptide to achieve integrated thrombo
