Regulation of calcium signalling in T lymphocytes by the second messenger cyclic ADP-ribose.

Cyclic ADP-ribose (cADPR) is a natural compound that mobilizes calcium ions in several eukaryotic cells. Although it can lead to the release of calcium ions in T lymphocytes, it has not been firmly established as a second messenger in these cells. Here, using high-performance liquid chromatography analysis, we show that stimulation of the T-cell receptor/CD3 (TCR/CD3) complex results in activation of a soluble ADP-ribosyl cyclase and a sustained increase in intracellular levels of cADPR. There is a causal relation between increased cADPR concentrations, sustained calcium signalling and activat

Histomorphological, immunohistochemical, and ultrastructural study on ontogeny of ileocaecal lymphoglandular complexes in prenatal and postnatal Indian buffalo: An innate mucosal immune barrier.

The present study was conducted on prenatal and postnatal development of lymphoglandular complexes (LGCs) in ileocaecal region of buffalo fetuses (n = 15) ranging from 11.5 cm curved crown rump length (CVRL) (80 days) to 100 cm CVRL (299 days) and neonatal buffalo calves (n = 10). The fetuses were categorized into three groups based on their CVRL. LGC formation was not evident in ileocaecal junction up to 32 cm CVRL (145 days). At 35 cm CVRL (152 days), diffuse lymphocytes were scattered around the base of glands that encircled them. At 54 cm CVRL (195 days), lymphoid aggregates were present i

A primary T-cell immunodeficiency associated with defective transmembrane calcium influx.

We investigated a T-cell activation deficiency in a 3-month-old boy with protracted diarrhea, serious cytomegalovirus pneumonia, and a family history (in a brother) of cytomegalovirus infection and toxoplasmosis. In spite of detection of normal number of peripheral lymphocytes, T cells did not proliferate after activation by anti-CD3 and anti-CD2 antibodies, although proliferation induced by antigens was detectable. We sought to determine the origin of this defect as it potentially represented a valuable tool to analyze T-cell physiology. T-cell activation by anti-CD3 antibody or phytohemagglu

Intracellular CD3 and surface CD8β distinguish activated human T cells from natural killer cells after αCD3/αCD28 stimulation.

Many studies of human T cell function involve activating primary T cells in vitro to assess functional outcomes. A common model of human T cell activation is polyclonal stimulation with αCD3/αCD28 antibodies. However, subsequent identification of these stimulated T cells in a mixed population is complicated by surface CD3 loss. In particular, both cytotoxic T cells and natural killer (NK) cells can appear CD3-CD8α+, leading to misidentification of NK cells as cytotoxic T cells. To address this limitation, we investigated incorporating intracellular CD3 and surface CD8β staining following αCD3/

Empowering exhausted T cells of Glioblastoma patients by Neurotransmitters and Neuropeptides: decreasing immune checkpoint inhibitors, and increasing CD3zeta, proliferation and Glioblastoma arrest.

BACKGROUND: Glioblastoma is the most common malignant brain tumor, with extremely poor prognosis, and patient's T cells are exhausted, dysfunctional, and unable to eliminate Glioblastoma. To our knowledge, there is no effective and safe treatment for rejuvenating and improving multiple functions of exhausted T cells. We previously found that specific Neurotransmitters and Neuropeptides induce direct, potent and beneficial effects on human T cells. METHOD: We studied if Dopamine, Glutamate, GnRH-II, Neuropeptide Y or their combinations, can rejuvenate and improve peripheral T cells of four Gli

A bispecific CD3×CD19 antibody for systemic lupus erythematosus: a phase 1 trial.

Early-phase studies of deep B cell depletion with anti-CD19 chimeric antigen receptor (CAR) T have produced prolonged, drug-free remission in refractory autoimmune disease. However, CAR T cell therapy requires lymphodepleting chemotherapy, autologous cell manufacturing and specialized infrastructure, limiting reach to a fraction of patients who might benefit. Bispecific T cell engagers (TCEs) offer a potent, off-the-shelf approach to deep B cell depletion; however, controlled clinical evaluation in rheumatic disease remains limited. Here we report results from the intravenous treatment arm of

Novel case of cytoplasmic CD3 positive, surface CD4 positive high-grade B-cell lymphoma with MYC rearrangement.

High-grade B-cell lymphoma, not otherwise specified (HGBCL-NOS), is an aggressive mature B-cell neoplasm whose diagnosis remains challenging due to significant morphologic overlap with diffuse large B-cell lymphoma and Burkitt lymphoma, and the absence of defining molecular alterations. Aberrant antigen expression can further complicate diagnoses and may result in lineage ambiguity. We report a unique case of HGBCL-NOS with MYC rearrangement demonstrating aberrant cytoplasmic CD3 and surface CD4 expression, leading to an initial misdiagnosis as a T-cell lymphoma at an outside hospital. TCR signal-enhancing mutation alters lipid metabolism of thymocytes and impairs antitumor immunity of mature T cells.

The T cell receptor (TCR), a master regulator of adaptive immunity, serves as a molecular transducer that converts antigen recognition into precisely modulated intracellular signals, orchestrating both T cell development and effector functions. In this study, we leveraged a germline CD3εI173A mutation, a previously characterized alteration that amplifies TCR signaling through the disruption of inhibitory lipid interactions, to dissect how thymocyte-intrinsic TCR signaling amplification influences the fate of mature T cells. Remarkably, thymic double-positive cells in CD3εI173A mice with altere

Case Report: Flow cytometric differential diagnosis of a peripheral T-cell lymphoma, NOS with complete loss of CD45 and dim expression of CD3.

Peripheral T-cell lymphomas (PTCLs) are a group of non-Hodgkin lymphomas originating from mature T-lymphocytes. Despite encompassing several well-defined entities, about 25% of the PTCLs do not fulfill the requirements of any of the subcategories. These diseases are classified as PTCL, not otherwise specified (PTCL, NOS), and often associated with poor prognosis. Hereby we present a case of a female patient, diagnosed with PTCL, NOS from her skin biopsy specimen. Besides histology and immunohistochemistry, flow cytometry was used for phenotyping and staging (peripheral blood, bone marrow). Pat

Emerging biomarkers for CD3×CD20 bispecific antibodies in lymphoma.

Novel CD3×CD20 bispecific antibody (BsAb) immunotherapies have entered the armamentarium for follicular lymphoma and diffuse large B-cell lymphoma based on accelerated approvals. The primary challenge in utilizing BsAbs lies in patient selection due to variable responses, unique toxicity, and health economics. To date, no validated biomarkers of therapy response exist, however data demonstrating potential clinical, imaging, and biological markers relating to BsAbs are growing. This review examines current prognostic and potentially predictive biomarkers and explores future directions for nuanc

Integrated bulk RNA sequencing and mass cytometry analysis reveal the circulating immune landscape in ischemic and hemorrhagic Moyamoya disease.

BACKGROUND: Moyamoya disease (MMD) is increasingly recognized as being influenced by chronic inflammation, with circulating immune cells playing a role in its progression. However, research on the immune characteristics of different MMD subtypes is limited. This study aims to compare the peripheral immune profiles of ischemic and hemorrhagic MMD patients. METHODS: Peripheral immune profiles were analyzed using transcriptome sequencing and mass cytometry. Data preprocessing was followed by functional and gene set enrichment analyses, as well as the construction of immune-related gene sets and

Comparison of Three CD3-Specific Separation Methods Leading to Labeled and Label-Free T Cells.

T cells are an essential part of the immune system. They determine the specificity of the immune response to foreign substances and, thus, help to protect the body from infections and cancer. Recently, T cells have gained much attention as promising tools in adoptive T cell transfer for cancer treatment. However, it is crucial not only for medical purposes but also for research to obtain T cells in large quantities, of high purity and functionality. To fulfill these criteria, efficient and robust isolation methods are needed. We used three different isolation methods to separate CD3-specific T

Validation of simple prediction algorithms to consistently achieve CD3+ and postselection CD34+ targets with leukapheresis.

BACKGROUND: Cellular therapies using engineered T cells, haploidentical transplants, and autologous gene therapy are increasing. Specified CD3+ or high CD34+ doses are typically required for subsequent manufacturing, manipulation, or CD34+ selection. Simple, practical, and reliable lymphocyte and hematopoietic progenitor cell (HPC) collection algorithms accounting for subsequent CD34+ selection have not been published. STUDY DESIGN AND METHODS: In this analysis of 15 haploidentical donors undergoing tandem lymphocyte and HPC collections, we validated one-step, practical prediction algorithms

Colonal monomorphic epitheliotropic intestinal T-cell lymphoma with novel phenotype of cytoplasmic CD3 expression.

Monomorphic epitheliotropic intestinal T-cell lymphoma (MEITL) is a new clinical entity that was reclassified from enteropathy-associated T-cell lymphoma in the 2016 WHO classification. An 83-year-old man with fever and diarrhea was referred to our hospital because of free air in the abdominal cavity and wall thickening of the large intestine on CT. Colonofiberscopic examination revealed mucosal edema and multiple ulcers at the sigmoid colon, splenic flexure, and transverse colon. Histopathological examination of the mucosal biopsy specimen demonstrated dense infiltration of small lymphocytes

Immunodeficiency and immune dysregulation associated with proximal defects of T cell receptor signaling.

Engagement of the TCR/CD3 complex triggers a cascade of events that result in T lymphocyte activation and promote positive and negative selection of thymocytes, T lymphocyte migration and effector functions, development and activation of regulatory T cells. Gene mutations that abrogate early TCR signaling are associated with profound abnormalities of T lymphocyte development and function both in humans and in mice, causing susceptibility to severe infections since early in life. In recent years, a growing number of genetic defects have been discovered that reduce, but do not completely abrogat…

A possible role for CD8+ T lymphocytes in the cell-mediated pathogenesis of pemphigus vulgaris.

Pemphigus vulgaris (PV) is an autoimmune blistering disease whose pathogenesis involves both humoral and cell-mediated immune response. Though the pathogenetic role of autoantibodies directed against desmoglein 3 is certain, a number of other factors have been suggested to determine acantholysis in PV. In this study we examined the possible role of CD8+ T cells in the development of acantholysis by a passive transfer of PV autoantibodies using CD8 deficient mice, and we also studied the inflammatory infiltrate of PV skin lesions by immunohistochemical staining. The results of the immunohistoch

Severe combined immunodeficiency caused by a splicing abnormality of the CD3delta gene.

UNLABELLED: CD3delta deficiency is a recently identified rare form of severe combined immunodeficiency. We analysed the CD3delta gene in a Japanese family with severe combined immunodeficiency. The patients lacked T-cells with normal numbers of B-cells and natural killer cells in peripheral blood. We found a novel homozygous mutation in the splicing acceptor site of intron 2 (IVS2-2A --> G) in these patients. Analysis of patients' mononuclear cells revealed the CD3delta splicing abnormality. Chest X-ray films and computed tomography revealed small sized thymuses in these patients. CONCLUSION:

Exposure of cynomolgus monkey embryos to retinoic acid causes thymic defects: effects on peripheral lymphoid organ development.

We have previously reported that exposure of monkey embryos to 13-cis-retinoic acid (cRA) results in thymic defects. In this study, we analyzed lymphocyte and antigen-presenting cell populations at gestational days (GDs) 80-100 in the thymus, spleen, mesenteric lymph nodes, and gut-associated lymphoid tissue following a teratogenic dosing regimen of cRA (2.5 and 5 mg/kg) at GD14-27. Tissue sections were immunostained for T-cells (anti-CD3), B-cells (anti-CD20), dendritic cells (p55), and major histocompatibility class II (anti-HLA-DR). Digital images of spleen sections were analyzed to obtain
