Euchrenone A10 attenuates septic lung injury though S100A8/A9-dependent TLR4/MyD88/NF-κB signaling.

BACKGROUND: Euchrenone A10 (A10), an isoprenylated flavanone isolated from Glycyrrhiza (licorice), exhibits significant bioactivities, including anti-inflammatory and antioxidant effects. However, the effects and mechanisms underlying A10's protective role in sepsis-associated acute lung injury (SALI) remain incompletely understood. OBJECTIVE: This study aims to elucidate the pharmacological effects of A10 and the underlying mechanisms by which it protects against SALI, using both in vitro and in vivo experiments. METHODS: To evaluate the effects of A10 against SALI, mice were pretreated wit

Extracellular matrix metabolism-related gene expression in bile duct-ligated rats.

Bile duct-ligated rats have been widely used as a model of cholestatic liver fibrosis. The aim of this study was to analyze the sequential expression of extracellular matrix (ECM) metabolism-related genes in these rats. We analyzed the intrahepatic messenger RNA (mRNA) expression of several ECM metabolism-related genes: transforming growth factor β1 (TGF-β1), connective tissue growth factor (CTGF), procollagen-α1 (collagen-I), matrix metalloproteinase (MMP)-2, MMP-13 and tissue inhibitor of metalloproteinases (TIMP)-1 on days 10, 21 and 42 after bile duct ligation (BDL). A DNA microarray was u

Neutrophil-derived S100A8/A9 impairs megakaryocyte maturation in immune thrombocytopenia.

Immune thrombocytopenia (ITP) features autoantibody-mediated platelet clearance, but how inflammatory mediators impair thrombopoiesis remains less defined. Using single-cell and spatial transcriptomics, we identify a neutrophil-megakaryocyte axis in ITP marrow in which neutrophil-derived S100A8/A9 engages TLR4 and activates JNK/c-Jun signaling. This pathway represses the megakaryocyte master regulator GATA1 and constrains maturation. In primary CD34+-derived megakaryocyte cultures, ITP plasma reduces polyploidization, maturation-marker expression, and platelet-like particle release; tasquinimo

Cytosolic proliferating cell nuclear antigen (PCNA) orchestrates neutrophil hyperactivation in COVID-19.

Neutrophils are central mediators of the hyperinflammatory response in severe SARS-CoV-2 infection. We report elevated cytosolic levels of proliferating cell nuclear antigen (PCNA) in neutrophils from patients with severe and critical COVID-19, correlating with enhanced NADPH oxidase-dependent reactive oxygen species (ROS) generation and neutrophil extracellular trap (NET) formation. Using T2AA, a small-molecule inhibitor of the PCNA scaffold, we demonstrate potent suppression of NADPH oxidase activation and NET release, particularly in response to SARS-CoV-2 RNA. Mechanistically, we identify

CD300lf Regulates Neutrophil Aging and Periodontal Immune Homeostasis.

Immune alterations, such as neutrophil dysfunction, significantly affect the progression and outcome of periodontitis, a prevalent inflammatory disease. Despite this, the molecular mechanisms driving neutrophil dysregulation in periodontitis remain poorly understood. In this study, we demonstrate that CD300lf, a critical immune regulator, is markedly downregulated in neutrophils from a periodontitis mouse model and human patients. The loss of CD300lf accelerates neutrophil aging, as evidenced by increased reactive oxygen species production, the senescence-associated secretory phenotype with el

Post-Aire Medullary Thymic Epithelial Cells and Hassall's Corpuscles as Inducers of Tonic Pro-Inflammatory Microenvironment.

While there is convincing evidence on the role of Aire-positive medullary thymic epithelial cells (mTEC) in the induction of central tolerance, the nature and function of post-Aire mTECs and Hassall's corpuscles have remained enigmatic. Here we summarize the existing data on these late stages of mTEC differentiation with special focus on their potential to contribute to central tolerance induction by triggering the unique pro-inflammatory microenvironment in the thymus. In order to complement the existing evidence that has been obtained from mouse models, we performed proteomic analysis on mic

Dietary titanium dioxide particles (E171) promote colitis-associated colorectal cancer development in mice through macrophage-derived S100A8/S100A9secretion mediated by NLRP3/Caspase 1/GSDMD pathway.

Colitis-associated colorectal cancer (CAC) is a major contributor to cancer-related mortality worldwide. Titanium dioxide (TiO2, E171), a widely used food additive, has been insufficiently studied regarding its effects on macrophages within colon tumors during CAC development. In this study, CAC mouse models were used to investigate the biological impact of dietary E171 on macrophages in vivo, while lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cell lines were employed to elucidate the underlying mechanisms in vitro. We found that dietary E171 intake accelerated CAC development, exac

Demonstration of extracellular traps in tuberculous pleural effusion from adult patients.

In pleural tuberculosis (TB), pleural effusion is typically characterized by a lymphocytic-rich exudate; however, neutrophils and macrophages predominate during the initial phase of infection. These immune cells are known to combat pathogens via phagocytosis, degranulation, and formation of extracellular traps (ETs)-a recently recognized defence mechanism. Although in-vitro and in-vivo studies have demonstrated ETs formation during Mycobacterium tuberculosis infection, their presence in clinical samples of patients with pleural TB has not been previously demonstrated. In this study, we employe

Clinical parameters and biological markers associated with acute severe ocular complications in Stevens-Johnson syndrome and toxic epidermal necrolysis.

Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) are severe cutaneous adverse drug reactions with high mortality rates. Its sequelae, such as blindness, persist even after recovery. Patients with SJS/TEN should be accurately diagnosed and receive appropriate treatment as soon as possible. Therefore, identifying the factors for severity prediction is necessary. We aimed to clarify the clinical parameters and biological markers that can predict acute severe ocular complications (SOCs) in SJS/TEN. This retrospective cross-sectional study enrolled 47 patients with SJS/TEN who we

Regulation of host and viral promoters during human cytomegalovirus latency via US28 and CTCF.

Viral latency is an active process during which the host cell environment is optimized for latent carriage and reactivation. This requires control of both viral and host gene promoters and enhancers often at the level of chromatin, and several viruses co-opt the chromatin organiser CTCF to control gene expression during latency. While CTCF has a role in the latencies of alpha- and gamma-herpesviruses, it was not known whether CTCF played a role in the latency of the beta-herpesvirus human cytomegalovirus (HCMV). Here, we show that HCMV latency is associated with increased CTCF expression and C

Methylprednisolone alleviates multiple sclerosis by expanding myeloid-derived suppressor cells via glucocorticoid receptor β and S100A8/9 up-regulation.

Methylprednisolone is an effective drug in the treatment of autoimmune disease, such as multiple sclerosis (MS), due to long-acting anti-inflammatory, antiallergic and immunosuppressant. Previous studies have noted the importance of myeloid-derived suppressor cells (MDSC) in MS progression. However, it is still not known whether methylprednisolone could influence the ratio and function of MDSC during MS treatment. In the current study, we found an increased ratio of MDSC at the onset of EAE in mice model; but methylprednisolone pulse therapy (MPPT) did not alter the percentage and suppressive

Elevated levels of serum MRP8/14 in ankylosing spondylitis: associated with peripheral arthritis and active disease.

Monocytes of patients with ankylosing spondylitis (AS) have toll-like receptor 4 (TLR4) overexpression on their monocytes. Myeloid-related protein (MRP) 8/14 protein complexes are calcium-binding proteins, which act as endogenous ligands to TLR4. Thus, we studied the levels of MRP8/14 in adult AS patients. MRP8/14 levels were assessed in 99 adult AS patients satisfying Assessments in Ankylosing Spondylitis International Society 2010 criteria and 71 healthy controls by ELISA. Patient disease parameters like patient and physician global assessment, Bath Ankylosing Spondylitis Disease Activity In

Identification of the mouse calcium-binding proteins, MRP 8 and MRP 14, in Schistosoma mansoni-induced granulomas: biochemical and functional characterization.

Granuloma initiation factor (GIF), which elicits a granulomatous reaction in naive murine skin, contains low-molecular-weight proteins partially purified from organized granulomas developed in livers of mice with schistosomiasis. In this study, we found that 10- and 14-kDa proteins in the GIF are highly homologous to mouse migration inhibitory factor-related protein (MRP) 8 and MRP 14. Compared with the N-terminal amino acid sequence deduced from each corresponding cDNA, the 10-kDa protein from the granuloma lacks the first methionine, whereas the 14 kDa misses methionine, alanine, and asparag

Development of Paquinimod-Loaded Implants to Modulate the Foreign Body Response.

The foreign body response (FBR) is an immune reaction from the host to any foreign material that makes it beyond barrier tissues, resulting in the recruitment of immune cells and fibroblasts. The FBR to medical implants can lead to degradation and encapsulation, affecting their efficacy, lifespan, and safety. A key protein involved in myriad inflammatory responses is S100A9, inducing inflammatory cytokine secretion and leukocyte chemotaxis. Inspired by recent studies indicating that inhibiting systemic S100A9 in mice results in a diminished FBR, we have identified S100A9 as a potential locally
