Blood-brain barrier-restricted translocation of Toxoplasma gondii from cortical capillaries.

The cellular barriers of the central nervous system proficiently protect the brain parenchyma from infectious insults. Yet, the single-celled parasite Toxoplasma gondii commonly causes latent cerebral infection in humans and other vertebrates. Here, we addressed the role of the cerebral vasculature in the passage of T. gondii to the brain parenchyma. Shortly after inoculation in mice, parasites mainly localized to cortical capillaries, in preference over post-capillary venules, cortical arterioles or meningeal and choroidal vessels. Early invasion to the parenchyma (days 1-5) occurred in absen

Structure of intercellular junctions in the endothelium.

Endothelial cell junctions are complex structures formed by transmembrane adhesive molecules linked to a network of cytoplasmic/cytoskeletal proteins. At least three different types of endothelial junctions have been described: tight junctions, gap junctions and adherens junctions. These structures have some features and components in common with epithelium but also some which are specific for endothelium. We still know very little about the pathologic consequences of alterations in the functional behaviour or synthesis of endothelial cell junction proteins. It is possible that pathologies lin

[Vascular permeability of fluorescent substance in cranial nerve roots].

Recently, it has been reported that the facial nerve in the facial canal shows enhancement on gadolinium-enhanced magnetic resonance imaging (Gd-MRI) in patients with a clinically normal facial nerve. Enhancement is also observed in the Gasserian ganglion and in the second and third divisions of the trigeminal nerve at the base of the skull. The aim of the present investigation was to study the vascular permeability of cranial nerve roots using the tracer technique. Vascular permeability in the cranial nerve root was examined after intravenous injection of sodium fluorescein (Fluorecite) as a

Influence of surfactant-facilitated interfacial tension reduction on chlorinated solvent migration in porous media: observations and numerical simulation.

The ability of a multiphase flow model to capture the migration behavior of chlorinated solvents under conditions of surfactant-facilitated interfacial tension (IFT) reduction is assessed through comparison of model predictions with observations from controlled laboratory experiments. Tetrachloroethene (PCE) was released in two-dimensional saturated systems, packed with sandy media that incorporated rectangular lenses of capillary contrast. Spatially uniform interfacial tension conditions were created in the tanks by pre-flushing the porous medium with either Milli Q water or an aqueous surfac

Modeling the impact of the revised starling hypothesis on vascular refilling during ultrafiltration in humans.

The physical attributes of continuous capillaries, the glycocalyx, and endothelial tight junctions, influence the distribution of forces that affect transcapillary solvent and solute transport. The traditional Starling hypothesis does not account for the effects of transcapillary solvent flux on the protein content of the pericapillary space, whereas the "Revised" Starling model (RSM) incorporates the impact of the glycocalyx and endothelial tight junctions on transcapillary solute flux, which results in an inhomogeneous distribution of oncotic pressures across the capillary wall. We compared

Pulpal vascular changes in inflammation.

Changes in pulpal vessels in experimentally induced acute and chronic pulpitis in dog tooth were investigated using corrosive resin casts and scanning electron microscopic examination. Following a cavity preparation without water spray, increased permeability of blood vessels occurred in the primary stage of acute pulpitis. This was evidenced by the extravasation of resin from the vessel. This phenomenon was found initially in the venular network as well as in the capillary network located under the dentin. The morphological change was minimal in the vascular network underneath the cavity. Thi

Claudication induces systemic capillary endothelial swelling.

An in vivo model of intermittent claudication has been developed to investigate systemic reperfusion injury associated with transient muscle ischaemia. Rats were subjected to unilateral common iliac artery ligation and two weeks of intermittent hind limb muscle stimulation. Electron microscopy demonstrated a significantly increased percentage of swollen capillary endothelial cells both locally and systemically in these "claudicant" rats, compared with controls or those undergoing muscle stimulation or artery ligation alone. These results support human data suggesting that claudication induces

[Ultrastructure of the air-blood-intestinal respiration barrier of the American sheatfish Hoplosternum thoracatum].

The fish, Hoplosternum thoracatum, lives in water with a permanently reduced content of oxygen. The respiratory gut as an accessory respiratory organ makes it possible to breath atmospheric oxygen and facilitates gaseous interchange. It forms a large, dilated tube containing a voluminous bubble of air. The wall of the respiratory gut is extremely thin. Below the peritoneum there is situated a branching plexus of blood vessels. The tributaries extent perpendicularly and reach the epithelium. They form a rich anastomosing capillary network. The capillaries are covered with attenuated processes o

CRISPR/Cas13a-Responsive and RNA-Bridged DNA Hydrogel Capillary Sensor for Point-of-Care Detection of RNA.

Disease diagnostics and surveillance increasingly highlight the importance of portable, cost-effective, and sensitive point-of-care (POC) detection of nucleic acids. Here, we report a CRISPR/Cas13a-responsive and RNA-bridged DNA hydrogel capillary sensor for the direct and visual detection of specific RNA with high sensitivity. The capillary sensor was simply prepared by loading RNA-cross-linking DNA hydrogel film (∼0.2 mm ± 0.02 mm) at the end of a capillary. When CRISPR/Cas13a specifically recognizes the target RNA, the RNA bridge in the hydrogel film is cleaved by the trans-cleavage activit

[Pathogenesis of the venous leg ulcer].

Based on different experimental findings, the following sequence of pathogenetically important factors for the development of venous ulcers is outlined: Valvular incompetence, reflux----chronic venous ambulatory hypertension----capillary dilatation and deformation----hyperemia----increased transendothelial passage (protein), microlymphangiopathy----protein deposition in the tissue. Deficient fibrinolysis----pericapillary fibrin cuffs----diffusion block----local hypoxia, tissue death. Although other mechanisms and complex factors may also play important additional roles, for the first time a re

An electron microscope study on permeability in cerebral venules in the rats with hypertensive encephalopathy.

Hypertensive encephalopathy was induced in the rat by clipping one renal artery and contralateral nephrectomy. The possible changes of vascular permeability of the cerebral blood capillaries and venules were investigated by using ferritin as a tracer. The uninephrectomized rats served as controls. In controls, ferritin was never seen in the basement membranes, within plasmalemmal vesicles on the basal surface of the endothelium, or in endothelial cell junctions of cerebral capillaries and venules up to 180 min after the injection. In venules of the brain in rats with hypertensive encephalopath

Endocytosis of serum albumin-gold conjugates by microvascular endothelial cells in rat adrenal gland: regional differences between cortex and medulla.

We investigated the distribution of serum albumin (bovine) modified by adsorption to 15 nm diameter colloidal gold (Au15BSA) in rat adrenal vasculature after perfusion of Au15BSA in situ for 6-20 min. A striking difference in permeability to and uptake of Au15BSA by adrenal cortical and medullary endothelial cells was found. The fenestrated capillary endothelial cells in the cortex were permeable to Au15BSA particles, but showed no significant endocytosis of these conjugates. Endothelial cells of the medullary capillaries and venous tributaries of the central vein were markedly less fenestrate

A mechanical modeling framework to study endothelial permeability.

The inner lining of blood vessels, the endothelium, is made up of endothelial cells. Vascular endothelial (VE)-cadherin protein forms a bond with VE-cadherin from neighboring cells to determine the size of gaps between the cells and thereby regulate the size of particles that can cross the endothelium. Chemical cues such as thrombin, along with mechanical properties of the cell and extracellular matrix are known to affect the permeability of endothelial cells. Abnormal permeability is found in patients suffering from diseases including cardiovascular diseases, cancer, and COVID-19. Even though

MicroRNA-451 increases vascular permeability and suppresses angiogenesis in pulmonary burn injury in a rat model.

BACKGROUND: Burns are common traumas that cause systemic symptoms by increasing vascular permeability. OBJECTIVES: To investigate the role of miRNA-451 and to clarify the underlying mechanism of the burn process. MATERIAL AND METHODS: We established a heat-induced third-degree burn with acute lung injury (ALI) model in rats. Hematoxylin and eosin (H&E) staining and in situ hybridization were performed. Overexpressed miRNA-451 in human umbilical vascular endothelial cells (HUVEC) were carried out. The migration and proliferation of HUVEC cells were examined. RESULTS: The H&E staining showed

Leptin receptor deficiency induces early, transient and hyperglycaemia-independent blood-brain barrier dysfunction.

Diabetes mellitus (DM) significantly increases susceptibility to central nervous system (CNS) pathologies, including stroke, vascular dementia, cognitive deficits and Alzheimer's disease. Previous studies (mostly using the streptozotocin model) suggested that blood-brain barrier (BBB) disruption is involved in these conditions. Here, we examined the integrity of brain capillaries and BBB permeability in Leprdb/db obesity-related diabetic mice. Surprisingly, significant BBB leakage was observed only in young mice at the pre-hyperglycemic stage. Thorough examination of barrier permeability at la

[A change in glomerular permeability in renovascular hypertension. Participation of angiotensin and structural lesions].

To characterize the decrease in glomerular permeability that occurs in contralateral kidney of renovascular hypertension, glomerular hemodynamics were studied in Goldblatt hypertensive and normotensive control rats. The effects of converting enzyme inhibition (captopril) and renal vasodilatation induced with hyperoncotic plasma were evaluated: in addition, glomerular morphometry was performed. In hypertension, glomerular capillary pressure was increased, ultrafiltration coefficient was decreased, single-nephron filtration rate was normal and afferent resistance was elevated. Captopril rose glo
