Regulatory T cells as novel cell-based therapy for ischemic stroke.

Ischemic stroke is a leading cause of mortality worldwide, with an excessive immune response playing a major role in exacerbating post-stroke pathogenesis. Regulatory T (Treg) cells, known for their immunosuppressive properties, are believed to be crucial in mitigating post-stroke inflammation. Moreover, emerging evidence suggests that Treg cells contribute to ischemic stroke recovery by suppressing neurotoxic astrogliosis and by promoting myelination regeneration. However, significant challenges remain before the clinical translation of Treg cell-based therapies can be realized. In this revie

Beyond Vision: An Overview of Regenerative Medicine and Its Current Applications in Ophthalmological Care.

Regenerative medicine (RM) has emerged as a promising and revolutionary solution to address a range of unmet needs in healthcare, including ophthalmology. Moreover, RM takes advantage of the body's innate ability to repair and replace pathologically affected tissues. On the other hand, despite its immense promise, RM faces challenges such as ethical concerns, host-related immune responses, and the need for additional scientific validation, among others. The primary aim of this review is to present a high-level overview of current strategies in the domain of RM (cell therapy, exosomes, scaffold

Development of the aganglionic colon following surgical rescue in a cell therapy model of Hirschsprung disease in rat.

Patients with Hirschsprung disease lack enteric ganglia in the distal colon and propulsion of colorectal content is substantially impaired. Proposed stem cell therapies to replace neurons require surgical bypass of the aganglionic bowel during re-colonization, but there is inadequate knowledge of the consequences of bypass. We performed bypass surgery in Ednrb-/- Hirschsprung rat pups. Surgically rescued rats failed to thrive, an outcome reversed by supplying electrolyte- and glucose-enriched drinking water. Histologically, the bypassed colon had normal structure, but grew substantially less i

Cell-based Therapies for Corneal and Retinal Disorders.

Maintenance of the visual function is the desired outcome of ophthalmologic therapies. The shortcomings of the current treatment options, like partial recovery, post-operation failure, rigorous post-operative care, complications, etc., which are usually encountered with the conventional treatment options has warranted newer treatment options that may eliminate the root cause of diseases and minimize the side effects. Cell therapies, a class of regenerative medicines, have emerged as cutting-edge treatment option. The corneal and retinal dystrophies during the ocular disorders are the major cau

RPE based gene and cell therapy for inherited retinal diseases: A review.

The Retinal Pigment Epithelium (RPE) is the supportive layer located beneath the neural retina. Its health is essential for the proper function of photoreceptors. Indeed, any condition involving the RPE has the potential to induce an antegrade degeneration of the photoreceptors and inner retinal layers. Traditionally, degenerative disorders of the neural retina have been considered untreatable. However, the advent of gene and cell replacement therapies brings hope to halt or even cure retinal degenerative diseases. This study aims to review the most recent clinical trials registered on the RPE

Clinical translation of stem cell therapy for spinal cord injury still premature: results from a single-arm meta-analysis based on 62 clinical trials.

BACKGROUND: How much scientific evidence is there to show that stem cell therapy is sufficient in preclinical and clinical studies of spinal cord injury before it is translated into clinical practice? This is a complicated problem. A single, small-sample clinical trial is difficult to answer, and accurate insights into this question can only be given by systematically evaluating all the existing evidence. METHODS: The PubMed, Ovid-Embase, Web of Science, and Cochrane databases were searched from inception to February 10, 2022. Two independent reviewers performed the literature search, identif

Modulation of the tumor microenvironment with an oncolytic adenovirus for effective T-cell therapy and checkpoint inhibition.

Despite exciting proof-of-concept data mediated by adoptive T-cell transfer and checkpoint blockade, major challenges imposed by the tumor microenvironment restrict clinical benefits to a minority of patients with advanced or metastatic solid malignancies. While employment of toxic pre- and postconditioning regimens to circumvent the inefficacy of T-cell transfer presents a fundamental problem for heavily pretreated cancer patients, for checkpoint blockade, the main issue relates to low single-agent response rates. To overcome these hurdles, combination therapy with oncolytic adenovirus is bec

Apoptotic Bodies: Decoding Their Functional Messages in Physiology and Pathology.

Apoptosis, as the predominant form of programmed cell death, plays a crucial role in maintaining tissue homeostasis. Apoptotic bodies (ABs), which are extracellular vesicles released during the terminal phase of apoptosis, were initially characterized as functionless cellular debris. However, they are now recognized for mediating intercellular communication via biomolecule delivery in both physiological and pathological states. Owing to their diverse biological properties, ABs have emerged as promising therapeutic candidates for treating a wide range of diseases, including cancer, cardiovascul

Cell therapy in multiple sclerosis: An overview.

Despite major advances in therapy for multiple sclerosis (MS) patients, substantial unmet needs remain, particularly regarding the prevention of disability progression and the treatment of progressive and aggressive forms of the disease. While early use of high-efficacy therapies has improved inflammatory disease control, their impact on long-term neurodegeneration is limited, and therapeutic options for progressive MS remain scarce. Autologous hematopoietic stem cell transplantation (AHSCT) has emerged as a highly effective escalation strategy for selected patients with aggressive, inflammato

Barriers to Effective Cryopreservation of Cell Therapies: Challenges and Emerging Solution.

Many cellular therapies require a cryopreservation step during their lifecycle. Cryopreservation offers many advantages such as extending product shelf life, decoupling manufacture from "just-in-time" delivery, and allowing pre-conditioning and other treatment interventions before administration. Nevertheless, it can also pose significant risks to the product and by extension the patient, if the process is non-optimized or misapplied. A clear understanding of the principles and their relationship to cell viability and functionality is therefore a pre-requisite for effective cryopreservation. T

Proposed Mechanisms of Cell Therapy for Alzheimer's Disease.

Alzheimer's disease is a progressive neurodegenerative disorder characterized by mitochondria dysfunction, accumulation of beta-amyloid plaques, and hyperphosphorylated tau tangles in the brain leading to memory loss and cognitive deficits. There is currently no cure for this condition, but the potential of stem cells for the therapy of neurodegenerative pathologies is actively being researched. This review discusses preclinical and clinical studies that have used mouse models and human patients to investigate the use of novel types of stem cell treatment approaches. The findings provide valua

A comprehensive review on the role of T cell subsets and CAR-T cell therapy in Aspergillus fumigatus infection.

Understanding the immune response to Aspergillus fumigatus, a common cause of invasive fungal infections (IFIs) in immunocompromised individuals, is critical for developing effective treatments. Tcells play a critical role in the immune response to A. fumigatus, with different subsets having distinct functions. Th1 cells are important for controlling fungal growth, while Th2 cells can exacerbate infection. Th17 cells promote the clearance of fungi indirectly by stimulating the production of various antimicrobial peptides from epithelial cells and directly by recruiting and activating neutrophi

Cell therapy in Sjögren's syndrome: opportunities and challenges.

Sjögren's syndrome (SS) is a chronic autoimmune disease caused by immune system disorders. The main clinical manifestations of SS are dry mouth and eyes caused by the destruction of exocrine glands, such as the salivary and lacrimal glands, and systemic manifestations, such as interstitial pneumonia, interstitial nephritis and vasculitis. The pathogenesis of this condition is complex. However, this has not been fully elucidated. Treatment mainly consists of glucocorticoids, disease-modifying antirheumatic drugs and biological agents, which can only control inflammation but not repair the tissu

Neural stem cell-derived extracellular vesicles: The light of central nervous system diseases.

Central nervous system (CNS) diseases are the leading cause of death worldwide. By performing compensatory functions and improving the inflammatory microenvironment, the transplantation of neural stem cells (NSCs) can promote functional recovery from brain injury, aging, brain tumours, and other diseases. However, the ability of NSCs to differentiate into neurons is limited, and they are associated with a risk of tumourigenicity. NSC-derived extracellular vesicles (NSC-EVs) can modulate the local microenvironment of the nervous system as well as distant neuronal functions. Thus, cell-free ther
