Therapeutic hypothermia by exogenous cooling induces potent neuroprotection. Post-stroke, therapeutic hypothermia so far did not translate into clinically applicable therapies due to hypothermia-associated side-effects compromising patient outcome. The hypothalamus contains two major thermoregulatory centers in the ventrolateral preoptic area (vlPOA) and dorsomedial hypothalamus (DMH), which are connected via gamma-aminobutyric acid (GABA)-ergic fibers. Using chemogenetic and optogenetic approaches, we explored the role of this GABAergic projection in regulating body temperature responses, cer

OBJECTIVE: The melanocortin receptor 4 (MC4R) plays a key role in the CNS regulation of metabolism. In addition to its role within the hypothalamus, other brain areas, including the dorsal raphe nucleus (DRN), express MC4R. However, the identity and role of these neurons in metabolism regulation are not fully understood. We performed studies to address these questions.
METHODS: We generated Mc4r-cre;Vgat-FlpO and Mc4r-cre;Vglut2-FlpO mice to determine the contribution of these MC4R neuronal populations in DRN. We then chemogenetically activated or inhibited the GABAergic and glutamatergic pop

Designer receptors exclusively activated by designer drugs (DREADDs) are a powerful chemogenetic tool for controlling targeted cell populations and manipulating brain functions. As a typical chemogenetic actuator in DREADD systems, clozapine-N-oxide (CNO) is widely utilized in neurobiological and pharmacological research. However, the acute off-target effects of CNO on somatosensory (e.g., itch, pain) stimulation-induced and anxiety-related behaviors in rodents have not been systematically investigated. Through classical behavioral paradigms, including evoked itch-scratching, noxious thermal p

BACKGROUND: Clinical and translational research has identified deficits in the dopaminergic neurotransmission in the striatum in Alzheimer's disease (AD) and this could be related to the pathophysiology of psychiatric symptoms appearing even at early stages of the pathology.
HYPOTHESIS: We hypothesized that AD pathology in the hippocampus may influence dopaminergic neurotransmission even in the absence of AD-related lesion in the mesostriatal circuit.
METHODS: We chemogenetically manipulated the activity of hippocampal neurons and astrocytes in wild-type and hemizygous TgF344-AD (Tg) rats, a

BACKGROUND: Previous studies indicated the involvement of cholinergic neurons in seizure; however, the specific role of the medial septum (MS)-hippocampus cholinergic circuit in temporal lobe epilepsy (TLE) has not yet been completely elucidated.
METHODS: In the current study, we used magnetic resonance imaging and diffusion tensor imaging to characterize the pathological change of the MS-hippocampus circuit in 42 patients with TLE compared with 22 healthy volunteers. Using optogenetics and chemogenetics, combined with in vivo or in vitro electrophysiology and retrograde rabies virus tracing,…

BACKGROUND AND HYPOTHESIS: Although the 5-hydroxytr-yptamine 2C receptor (5-HT2C) receptor agonists have demonstrated efficacy in mitigating sensorimotor gating deficits in the schizophrenic animal model of glutamatergic dysfunction, the precise neural mechanisms underlying these effects remain incompletely understood. This study investigates how 5-HT2C receptor activation improves sensorimotor gating and cognitive function by modulating Excitation/Inhibition balance in the medial prefrontal cortex (mPFC).

Social defeat stress (SDS) is a rodent model used to assess the effect of chronic stress on depressive-like behavior and alcohol consumption. Our previous studies have indicated that the neurokinin-1 receptor (NK1R) mediates the behavioral responses to this stressor, especially through its actions in the nucleus accumbens (NAc). The NK1R is the high affinity, endogenous target of the neuropeptide substance P (SP). In the experiments presented here, we first infused a cre-dependent, retrogradely transported virus into the NAc of Tac1-cre mice (Tac1 is the gene for SP) to identify brain regions

BACKGROUND: Mental disorders frequently co-occur with pain, yet pain mechanisms in non-peripheral etiologies (e.g., chronic psychological stress) remain underexplored. The ventral anterior-lateral thalamic complex (VAL) is implicated in emotional processing, but its role in chronic stress-induced pain hypersensitivity is unclear. Electroacupuncture (EA) is clinically used for pain management, but its efficacy and mechanisms in chronic stress-driven pain hypersensitivity require validation.
METHODS: A chronic restraint stress (CRS) model was established in male mice. Behavioral assessments wer

Irritable bowel syndrome (IBS), which is characterized by chronic abdominal pain, has a high global prevalence. The anterior cingulate cortex (ACC), which is a pivotal region involved in pain processing, should be further investigated regarding its role in the regulation of visceral sensitivity and mental disorders. A C57BL/6J mouse model for IBS was established using chronic acute combining stress (CACS). IBS-like symptoms were assessed using behavioral tests, intestinal motility measurements, and abdominal withdrawal reflex scores. Fluoro-Gold retrograde tracing and immunohistochemistry tech

BACKGROUND: The thalamus system plays critical roles in the regulation of reversible unconsciousness induced by general anesthetics, especially the arousal stage of general anesthesia (GA). But the function of thalamus in GA-induced loss of consciousness (LOC) is little known. The thalamic reticular nucleus (TRN) is the only GABAergic neurons-composed nucleus in the thalamus, which is composed of parvalbumin (PV) and somatostatin (SST)-expressing GABAergic neurons. The anterior sector of TRN (aTRN) is indicated to participate in the induction of anesthesia, but the roles remain unclear. This s

RATIONALE: Clinical and preclinical studies have demonstrated that estradiol withdrawal after delivery is one of important factors involved in the pathogenesis of postpartum depression (PPD). The infralimbic cortex (IL) is related to anxiety and mood disorders. Whether IL neurons mediate PPD is still unclear.
OBJECTIVES: This study was to observe the antidepressant effect and expression of BDNF and β-catenin in IL by allopregnanolone (ALLO) treatment or the selective activation or inhibition of IL neurons using a chemogenetic approach in a pseudopregnancy model of PPD.
METHODS: Administratio

BACKGROUND: Anxiety disorders are associated with an altered perception of the body's internal state. Therefore, understanding the neuronal basis of interoception can foster novel anxiety therapies. In rodents, the feeding status bidirectionally modulates anxiety-like behavior but how the sensing of gastrointestinal state affects anxiety remains unclear.
METHODS: We combined chemogenetics, neuropharmacology, and behavioral approaches in male and female rats to test whether vagal afferents terminating in the gastrointestinal tract mediate feeding-induced tuning of anxiety. Using saporin-based …

A growing body of literature implicates noradrenergic (NE) signaling in the modulation of ethanol consumption. However, relatively few studies have detailed specific brain pathways that mediate NE-associated binge-like ethanol consumption. To begin to fill this gap in the literature, male and female C57BL6/J and TH-ires-cre mice underwent pharmacological and chemogenetic testing, respectively, in combination with "drinking in the dark" procedures to model binge-like consumption of ethanol or sucrose solutions. First, we showed that intraperitoneal administration of the NE reuptake inhibitor, r

Stress undermines extinction learning and hinders exposure-based clinical therapies for a variety of neuropsychiatric disorders. In both animals and humans, dysfunction in the ventromedial prefrontal cortex (vmPFC) contributes to stress-impaired extinction, but the neural circuit by which stress modulates vmPFC function is not known. We hypothesize that locus coeruleus (LC) norepinephrine undermines extinction learning by recruiting projections from the basolateral amygdala (BLA) to vmPFC. Using a combination of circuit-specific chemogenetics and calcium imaging, we find that activation of LC …

Rapid eye movement (REM) sleep is thought to be a cortical state closer to wakefulness than non-REM sleep, yet a few brain regions regulate both states. This study identifies the glutamatergic neurons in the lateral periaqueductal gray (LPAG) that are specifically activated during wakefulness and REM sleep, as demonstrated by fiber photometry and optic tetrode recordings. Chemogenetic inhibition of LPAG vesicular glutamate transporter 2 (Vglut2) neurons reduced REM sleep and wakefulness while boosting non-REM sleep; their activation induced wakefulness, with high theta power and immobility. Chronic stress influences hematopoietic stem cells (HSCs). However, how psychological stress regulates HSC function remains incompletely understood. Here, we show that psychological stress impairs HSC self-renewal and lymphoid differentiation, inducing aging-like phenotypes. Stress suppresses neuronal activity in the medial prefrontal cortex (mPFC) and periaqueductal gray (PAG), leading to HSC dysfunction, whereas chemogenetic activation of these regions restores HSC function. Psychological stress or chemogenetic inhibition of the mPFC and PAG reduces the abundance of L. reuteri in the gut mic

Binge drinking substantially elevates the risk of developing alcohol use disorder, with pronounced sex-specific differences observed. However, there is a limited understanding of the underlying sex differences in the neural mechanisms that underpin binge drinking. The present study utilized the Drinking in the Dark (DID) paradigm, in combination with electrophysiology and chemogenetics, to reveal the critical role of the locus coeruleus (LC) in regulating sexually dimorphic binge-like drinking behavior. Electrophysiological analysis revealed that after three weeks of DID modeling, female mice …

In the cerebellum, climbing fibers (CFs) provide instructive signals for supervised learning at parallel fiber to Purkinje cell synapses. It has not been tested so far whether CF signaling may also influence plasticity in other brain areas. Here, we show that optogenetic CF activation suppresses potentiation of whisker responses in layer 2/3 pyramidal cells in the primary somatosensory (S1) cortex of awake mice that is observed after repeated whisker stimulation. Using two-photon imaging and chemogenetics, we find that CFs control plasticity by modulating SST- and VIP-positive interneurons in …
