Cholinergic activation of phosphoinositide metabolism during soman-induced seizures.

In the present study we investigated the role of the cholinergic pathway in phosphoinositide metabolism activation observed during soman-induced convulsions. We thus studied the effect of atropine sulphate, a muscarinic antagonist (20 mg kg-1, i.p.), on IP3 levels in rat hippocampus. We demonstrated that initially, the increase of IP3 is closely seizure-related. On the other hand, after 10 min of seizures, the IP3 enhancement and the seizure activity are no longer correlated. After 20 min of seizures, atropine failed to inhibit soman-induced IP3 enhancement, suggesting that the activation of a

Chronic hypoxic incubation blunts thermally dependent cholinergic tone on the cardiovascular system in embryonic American alligator (Alligator mississippiensis).

Environmental conditions play a major role in shaping reptilian embryonic development, but studies addressing the impact of interactions between chronic and acute environmental stressors on embryonic systems are lacking. In the present study, we investigated thermal dependence of cholinergic and adrenergic cardiovascular tone in embryonic American alligators (Alligator mississippiensis) and assessed possible phenotypic plasticity in a chronic hypoxic incubation treatment. We compared changes in heart rate (f H) and mean arterial blood pressure (P M) for chronically hypoxic and normoxic-incubat

Increased cholinergic contractions of jejunal smooth muscle caused by a high cholesterol diet are prevented by the 5-HT4 agonist--tegaserod.

BACKGROUND: Excess cholesterol in bile and in blood is a major risk factor for the respective development of gallbladder disease and atherosclerosis. This lipid in excess negatively impacts the functioning of other smooth muscles, including the intestine. Serotonin is an important mediator of the contractile responses of the small intestine. Drugs targeting the serotonin receptor are used as prokinetic agents to manage intestinal motor disorders, in particular irritable bowel syndrome. Thus, tegaserod, acting on 5-HT4 receptor, ideally should obviate detrimental effects of excessive cholestero

A self-amplifying nerve-fibroblast circuit drives colorectal cancer progression.

Nerves and cancer-associated fibroblasts (CAFs) have each been shown to regulate cancer progression directly. However, whether these cells interact to control tumor progression remains largely unknown. We show that in colorectal cancer (CRC), cholinergic stimulation induces CHRM3/Gq-dependent NTN1 secretion from CAFs, which in turn enhances intratumor cholinergic innervation. Within this feedforward loop, cholinergic stimulation promotes CRC growth directly through tumoral CHRM3/Gq-mediated YAP activation, while CAF-derived NTN1 promotes CRC growth and epithelial-to-mesenchymal transition-like

Autonomic nervous system control of multiple myeloma.

The autonomic nervous system (ANS), which consists of antagonistic sympathetic (adrenergic) and parasympathetic (cholinergic) arms, has emerged as an important regulator of neoplastic development, yet little is known about its role in multiple myeloma (MM). Clinical findings that anti-adrenergic β-blocker intake reduces risk of disease-specific death and overall mortality in patients with MM have indicated that adrenergic input may worsen myeloma outcome. However, preclinical studies using β-adrenergic receptor agonists or antagonists produced controversial results as to whether sympathetic pa

Exercise-conditioned plasma ameliorates postoperative cognitive dysfunction by activating hippocampal cholinergic circuit and enhancing BDNF/TrkB signaling.

BACKGROUND: Postoperative cognitive dysfunction (POCD) is a prevalent complication following anesthesia and surgery, particularly in the elderly, leading to increased mortality and reduced quality of life. Despite its prevalence, there are no effective clinical treatments. Exercise has shown cognitive benefits in aging and various diseases, which can be transferred to sedentary animals through plasma. However, it is unclear if exercise-conditioned plasma can replicate these benefits in the context of POCD. METHODS: Sixteen-month-old male C57BL/6J mice underwent 30 days of voluntary running wh

Trospium Chloride in the Treatment of Overactive Bladder Syndrome and Detrusor Overactivity.

Trospium chloride (TCL) is a quaternary amine derived from atropine, classified as an oral anticholinergic, the class of drugs most commonly used in the treatment of overactive bladder syndrome (OAB) and detrusor overactivity (DO). It is a large and hydrophilic molecule, with a low water-oil partition coefficient, which hinders the passage through lipid membranes and has demonstrated poor penetration into the central nervous system (CNS), generally without deleterious effects on the cognitive functions of healthy patients or those already with dementia. This molecule is excreted practically un

A control experiment revealed no change in horizontal activity and rearing when only the cholinergic antagonists (atropine; mecamylamine) were administered. This is the first study that shows a substantial role of muscarinic receptors for the development of behavioral sensitization to MDMA.

Blockade of GABA, type A, receptors in the rat pontine reticular formation induces rapid eye movement sleep that is dependent upon the cholinergic system.

The brainstem reticular formation is an area important to the control of rapid eye movement (REM) sleep. The antagonist of GABA-type A (GABA(A)) receptors, bicuculline methiodide (BMI), injected into the rat nucleus pontis oralis (PnO) of the reticular formation resulted in a long-lasting increase in REM sleep. Thus, one factor controlling REM sleep appears to be the number of functional GABA(A) receptors in the PnO. The long-lasting effect produced by BMI may result from secondary influences on other neurotransmitter systems known to have long-lasting effects. To study this question, rats wer

Cholinergic dopamine release from the in vitro rabbit carotid body.

The aim of this study was to test whether cholinergic mechanisms regulate dopamine (DA) release from the carotid body (CB) and interact with DA D(2) autoreceptors. One hundred forty-two CBs from adult rabbits were infused in vitro in a surviving medium bubbled with O(2) (Bairam A, Marchal F, Cottet-Emard JM, Basson H, Pequignot JM, Hascoet JM, and Lahiri S. J Appl Physiol 80: 20-24, 1996). CB DA content and release were measured after 1 h of exposure to various treatments: control, cholinergic agonist (0.1-50 microM carbachol), full muscarinic antagonist (1 and 10 microM atropine), antagonists

Behavioral and neurochemical alterations following thiamine deficiency in rodents: relationship to functions of cholinergic neurons.

Memory deficits are induced during the late stage (20-25 days) of thiamine-deficient (TD) feeding. In this review, the role of cholinergic neurons on the memory deficit induced by TD feeding are summarized. Although memory deficit cannot be suppressed by an injection of thiamine once it appears, such impairment was found to be protected by early treatment with thiamine during TD feeding. Administration of muscarinic M(1) agonist McN-A-343 reversed the memory deficit observed in TD mice, although the muscarinic M(2) antagonist methoctramine did not. The "kampo" (traditional herbal) medicine, "k

Role of dentate gyrus cells in retention of a radial arm maze task and sensitivity of rats to cholinergic drugs.

Male adult Fischer-344 rats that received bilateral injections of colchicine into two rostrocaudal sites showed relatively long-lasting alterations in the performance of a previously acquired radial arm maze task and specific destruction of dentate granule cells. Results of subsequent experiments with cholinergic drugs indicated that physostigmine or nicotine had no effect on the number of errors made in the maze, although other signs of cholinergic or pharmacological activity were present. RS-86, an analog of the muscarinic agonist arecoline, decreased errors in colchicine-treated rats, but t

Muscarinic cholinergic receptors in rat heart. Effects of thyroidectomy.

The effects of thyroid status on muscarinic cholinergic receptors in the rat myocardium were investigated. The potent muscarinic antagonist [3H]quinuclidinyl benzilate was used to identify muscarinic cholinergic receptors in rat heart particulate fractions from control, hypothyroid, and hyperthyroid rats. Thyroidectomy increased specific [3H]quinuclidinyl benzilate binding to heart particulate fractions by about 60% as compared to euthyroid rat cardiac preparations. Administration of triiodothyronine to euthyroid rats decreased specific binding to muscarinic cholinergic receptors by about 20%.

The unusual adrenergic-like excitatory action of acetylcholine on the ventricular cardiac muscle of the horned shark, Heterodontus portusjacksoni.

The atypical excitatory effect of acetylcholine on cardiac ventricular muscle was investigated in the horned shark, Heterodontus portusjacksoni. Electrically paced ventricular strips produced a massive 391.45% (+/-26.39%) increase in basal force of contraction in response to exogenously applied acetylcholine. The response was similar in nature to that produced by applied adrenaline, which caused a 382.52% (+/-72.47%) increase. The response to acetylcholine was blocked by the muscarinic cholinoceptor antagonist atropine and the competitive beta-adrenoceptor antagonist propranolol and was reduce

Role of brain histamine H1- and H2-receptors in neostigmine-induced hyperglycemia in rats.

We previously reported that when neostigmine, an inhibitor of acetylcholine esterase, was injected into the third cerebral ventricle, the concentration of hepatic venous plasma glucose was increased via central muscarinic receptors in anesthetized rats. To determine whether brain histamine receptors are involved in cholinergic system transmission with regard to central nervous system (CNS)-mediated glucoregulation, we examined the effects of the H1 receptor antagonist pyrilamine and the H2 receptor antagonist ranitidine on neostigmine-induced hyperglycemia in anesthetized rats. The injection o

Systemic blockade of nicotinic and purinergic receptors inhibits ventilation and increases apnoea frequency in newborn rats.

We hypothesized that the combined blockade of peripheral cholinergic and purinergic receptors alters the baseline breathing pattern and respiratory responses to carotid body stimuli (hypoxia, hyperoxia and hypercapnia). Rat pups at 4 (P4) and 12 days of postnatal age (P12) received an intraperitoneal injection of either saline vehicle or hexamethonium + suramin (Hex, 1 mg kg(-1), nicotinic receptor antagonist; Sur, 40 mg kg(-1), P2X receptor antagonist; both of which act mainly on peripheral receptors). Compared with the control animals (saline-injected rats), the Hex + Sur-treated rats demons

Amygdala beta-noradrenergic influences on memory storage involve cholinergic activation.

These experiments examined the involvement of the amygdaloid complex as a site of interaction of adrenergic and muscarinic cholinergic influences on memory storage. Male Sprague-Dawley rats (60 days old; 250-300 g) were given a single training trial in an inhibitory avoidance task and a retention test trial 48 h later. Immediately after training buffer control or drug solutions (0.5 microliter) were infused into the amygdala and, in the first experiment only, other drugs were administered intraperitoneally (ip). The first experiment examined the effects of post-training systemic injections of

Characterization of muscarinic cholinergic receptor binding in the vas deferens, bladder, prostate and penis of the rabbit.

Radioligand receptor binding techniques were used to characterize the muscarinic cholinergic receptor in the vas deferens, bladder, prostate and penis of the rabbit. This study represents the first comparative investigation of a neurotransmitter receptor in the genitourinary tract using radioligand receptor binding methods. A single high affinity muscarinic binding site was identified in the vas deferens (Kd = 0.16 nM), bladder (Kd = 0.15 nM) and prostate (Kd = 0.17 nM), using [3H]N-methylscopolamine ([3H]NMS), a muscarinic antagonist. Two high affinity [3H]NMS binding sites (Kd1 = 0.08 nM; Kd

Optogenetic stimulation of the brainstem dorsal motor nucleus ameliorates acute pancreatitis.

INTRODUCTION: Inflammation is an inherently self-amplifying process, resulting in progressive tissue damage when unresolved. A brake on this positive feedback system is provided by the nervous system which has evolved to detect inflammatory signals and respond by activating anti-inflammatory processes, including the cholinergic anti-inflammatory pathway mediated by the vagus nerve. Acute pancreatitis, a common and serious condition without effective therapy, develops when acinar cell injury activates intrapancreatic inflammation. Prior study has shown that electrical stimulation of the carotid
