Plasmodium knowlesi provides a rapid in vitro and in vivo transfection system that enables double-crossover gene knockout studies.

Transfection technology for malaria parasites provides a valuable tool for analyzing gene function and correlating genotype with phenotype. Transfection models are even more valuable when appropriate animal models are available in addition to complete in vitro systems to be able to fully analyze parasite-host interactions. Here we describe the development of such a model by using the nonhuman primate malaria Plasmodium knowlesi. Blood-stage parasites were adapted to long-term in vitro culture. In vitro-adapted parasites could re-adapt to in vivo growth and regain wild-type characteristics afte

Exploring the dynamics of adaptability in the translation of mental health education material: the perspectives from Chinese tertiary students studying in Australia.

INTRODUCTION: With technological advances, health communication studies have increasingly focused on multimodality. In contrast, as adaptation has been merged with cross-cultural translation, the dynamics of adaptability have been discussed primarily in theoretical research and other areas of translation, and only a few medical and health translation studies have emphasized this point. METHODS: Using the dynamic dimension in Verschueren's Adaptation Theory, the current study used a questionnaire survey and semi-structured interviews to explore the dynamics of adaptability in the Chinese trans

An expeditious and facile method of amyloid beta (1-42) purification.

For the study of amyloid beta (Aβ) associated toxicity which is supposed to be the main pathological agent in Alzheimer's disease (AD), it is important to secure Aβ peptide with appropriate biological activity. However, commercial and synthetic Aβ often have some pitfalls like less cell toxicity, prompt aggregation and excess price, using recombinant technology, these issues can be resolved though the method also suffered from some problems such as low yield, aggregation and prolong time to purify. Thus, we previously developed an easy, economic and convenient method for Aβ42 purification usin

[Progress in studies of seminal microbiome based on next-generation sequencing].

Seminal microbiome plays an important role in male reproductive health, and its composition may be associated with sperm quality, development of oligoasthenospermia, and pregnancy outcomes of assisted reproductive technology. Seminal microbiome may be transmitted to the partner and offspring, and consequently affect female reproduction and the health of offspring. The complete structure of seminal microbiome is difficult to find by conventional bacterial culture due to the length of time taken and restriction of condition. Next-generation sequencing (NGS) can reveal the complete composition of

Cortical Organoids to Model Microcephaly.

How the brain develops and achieves its final size is a fascinating issue that questions cortical evolution across species and man's place in the animal kingdom. Although animal models have so far been highly valuable in understanding the key steps of cortical development, many human specificities call for appropriate models. In particular, microcephaly, a neurodevelopmental disorder that is characterized by a smaller head circumference has been challenging to model in mice, which often do not fully recapitulate the human phenotype. The relatively recent development of brain organoid technolog

How can HIV/STI testing services be more accessible and acceptable for gender and sexually diverse young people? A brief report exploring young people's perspectives in Queensland.

ISSUE ADDRESSED: Gender and sexually diverse young people (GSDYP) are an important target group for HIV/sexually transmitted infection (STI) prevention and there is an immediate need to explore ways to make testing interventions accessible and appropriate for this group. METHODS: We used a modified World Café workshop with 14 GSDYP in Brisbane Australia, to inform the development of a pilot community-based testing intervention. RESULTS: The workshop identified the key features of an ideal service, which would include multiple, accessible sites that offer holistic, affordable services and con

Mesenchymal stem cells in bone development, bone repair, and skeletal regeneration therapy.

Bone formation in the embryo, and during adult fracture repair and remodeling, involves the progeny of a small number of cells called mesenchymal stem cells (MSCs). These cells continuously replicate themselves, while a portion become committed to mesenchymal cell lineages such as bone, cartilage, tendon, ligament, and muscle. The differentiation of these cells, within each lineage, is a complex multistep pathway involving discrete cellular transitions much like that which occurs during hematopoiesis. Progression from one stage to the next depends on the presence of specific bioactive factors,
