Evidence of a louse-borne outbreak involving typhus in Douai, 1710-1712 during the war of Spanish succession.

BACKGROUND: The new field of paleomicrobiology allows past outbreaks to be identified by testing dental pulp of human remains with PCR. METHODS: We identified a mass grave in Douai, France dating from the early XVIII(th) century. This city was besieged during the European war of Spanish succession. We tested dental pulp from 1192 teeth (including 40 from Douai) by quantitative PCR (qPCR) for R. prowazekii and B. quintana. We also used ultra-sensitive suicide PCR to detect R. prowazekii and genotyped positive samples. RESULTS AND DISCUSSION: In the Douai remains, we identified one case of B.

The effect of BMP9 on inflammation in the early stage of pulpitis.

BACKGROUND: Bone morphogenetic protein 9 (BMP9) tends to be associated with various inflammatory responses of diseases, but its relationship with pulpitis remains unknown. OBJECTIVE: This study aimed to evaluate the effects and mechanisms of BMP9 in pulpitis. METHODOLOGY: A rat model of pulpitis was used to evaluate the expression of BMP9, which was also analysed in Porphyromonas gingivalis lipopolysaccharide (Pg-LPS)-stimulated human dental pulp cells (hDPCs). The effects and mechanism of BMP9 on the regulation of inflammatory factors and matrix metalloproteinase-2 (MMP2) were evaluated usi

Dental iatrogenesis, Part 2.

To substantiate biocompatibility of dental materials and techniques, and thereby reduce iatrogenesis, tests for pulp and dentinal responses have been developed. These have shown that high-speed tooth cutting techniques are superior to low-speed techniques even when both incorporate air-water coolant sprays; pressure when placing restorative materials intensifies pulp responses induced by the cutting procedure, and that chemically self-cured resin composites requiring the application of a matrix to enhance adaptation intensify pulp response as compared to visible light-cured equivalents applied

Endodontic Management Before, During and After Head and Neck Radiotherapy: Biological, Diagnostic and Clinical Considerations in Head and Neck Cancer Patients.

BACKGROUND: Head and neck radiotherapy creates a high-consequence dental environment in which tooth loss may precipitate osteoradionecrosis, particularly after extraction in high-dose mandibular regions. Tooth preservation through nonsurgical root canal treatment is therefore a risk reduction strategy. However, evidence relevant to endodontic decision-making remains dispersed among clinical series, laboratory research, pulp testing studies and oncology dental guidance. OBJECTIVE: To synthesise contemporary evidence relevant to endodontic management before, during and after head and neck radio

Transient apical breakdown during orthodontic incisor intrusion treated with non-steroidal anti-inflammatory drugs: a case report.

Transient apical breakdown (TAB) may occur during pulp repair after dental traumatic injury, including orthodontic force. Herein, we describe TAB in the maxillary right central incisor of a 24-year-old woman during orthodontic treatment for a severe gummy smile. Eight months after initiating intrusion of the maxillary anterior teeth using temporary anchorage devices, the maxillary right central incisor showed discoloration and was non-responsive to electric pulp tests. The orthodontic force on the maxillary anterior teeth was immediately removed, and loxoprofen sodium, a non-steroidal anti-inf

Refining Pulpal Diagnostics for Predictable Vital Pulp Treatment.

INTRODUCTION: Vital pulp treatment (VPT) comprises a range of biologically based procedures aimed at preserving pulp vitality, reducing the need for invasive interventions, and improving long-term tooth survival. Despite growing evidence supporting these approaches, their implementation in clinical practice remains limited by the lack of reliable methods for accurately assessing the inflammatory status of the dental pulp and by the continued use of diagnostic terminology that does not adequately support pulp-preserving treatment strategies. METHODS: This narrative review examines the limitati

Circadian rhythm disruption impairs time-dependent dentine regeneration and reprograms the late-stage dental proteome in a mouse pulp injury model.

Circadian rhythms coordinate metabolic and regenerative programs, yet their contribution to pulp healing and tertiary dentinogenesis remains unclear. Here, we investigated whether chronic circadian rhythm disruption (CRD) alters time-dependent dentine repair and proteomic remodeling after dental pulp injury (DPI) in male BALB/c mice. Animals were maintained either under a stable 12:12 h light-dark cycle or under a chronic jet-lag model with a daily 2 h phase advance. DPI was induced in the maxillary first molars, followed by immediate direct pulp capping with mineral trioxide aggregate and sea

Comparison of three different materials used for indirect pulp capping in permanent molars: An in vivo study.

BACKGROUND: Pulp capping should always be considered as the primary treatment of choice for teeth without irreversible pulpitis in lesions approaching dental pulp. The predictability of vital pulp therapy has improved with the introduction of newer bioceramic materials. AIM: The aim of this study was to evaluate the clinical success of Biodentine, calcium hydroxide (CH), and mineral trioxide aggregate (MTA) as pulp capping materials for indirect pulp capping in carious permanent teeth. MATERIALS AND METHODS: Indirect pulp capping was done for 36 molars of 36 patients with deep caries lesions

In Vivo Study of the Action of a Topical Anti-Inflammatory Drug In Rat Teeth Submitted To Dental Bleaching.

Bleaching gel containing hydrogen peroxide (H2O2) cause damages in pulp tissue. This study investigated the action of a topical anti-inflammatory, the Otosporin®, in rats' bleached teeth with the null hypothesis of which the Otosporin® is no able to minimize the pulp inflammation that bleaching gel generates. The rat's molars were divided into groups: BLE: bleached (35% H2O2 concentration /single application of 30 min); BLE-O: bleached followed by Otosporin® (10 min); and control: placebo gel. In the second day after dental bleaching, the rats were killed, and the jaws were processed for hemat

A Two-Visit Pulpotomy Approach for the Management of Uncontrolled Bleeding in a Crown-Root Fractured Immature Incisor: A Case Report.

Background/Objective: The present case demonstrates the successful preservation of a severely inflamed dental pulp despite uncontrolled bleeding occurring during pulpotomy. This novel therapeutic approach was developed when a 7-year-old boy presented for endodontic treatment one week after sustaining a crown-root fracture in an immature incisor. The tooth exhibited slight hypersensitivity to cold testing, with no tenderness to percussion or palpation. An exposed pulp was present; however, when he could avoid contact with the exposed pulp tissue, the patient reported no symptoms. Methods: The c

Dysregulation of miR-140-3p is involved in the pathogenesis of pulpitis via the regulation of ACSL1.

Pulpitis is an inflammatory disease affecting the dental pulp tissue, impacting patients' oral health and daily life. MicroRNAs (miRNAs) are involved in the progression of pulpitis. This study aims to elucidate the regulatory role of the miR-140-3p/ACSL1 axis in pulpitis, particularly its molecular mechanisms within dental pulp cells. The levels of miR-140-3p and long-chain acyl CoA synthetase 1 (ACSL1) was measured by RT-qPCR. The receiver operating characteristic (ROC) curve represented the diagnostic capability of miR-140-3p. Lipopolysaccharide (LPS) derived from Escherichia coli was applie
