Secondary Deinococcus proteolyticus Infection Following Varicella-Zoster Virus Infection in a Child: A Case Report Describing a Rare Medical Condition.

Deinococcus proteolyticus belongs to Deinococcaceae, a family of bacteria characterized by an exceptional ability to withstand the lethal effects of DNA-damaging agents, including ionizing radiation, ultraviolet (UV) light, and desiccation. It was isolated from the Sahara surface sands, an extreme and nutrient-poor environment, regularly exposed to intense UV radiation, cycles of extreme temperatures, and desiccation. D. proteolyticus infection has seldom been implicated in human infections. The child presented with multiple necrotic skin lesions following a varicella infection, later complica

Adaptive peptide dispersions enable drying-induced biomolecule encapsulation.

Peptides are promising building blocks of designer materials with wide-ranging applications. These materials are stabilized by directional hydrogen-bonding patterns, giving rise to one-dimensional or two-dimensional assembly. It remains a challenge to mimic biology's context-adaptive and flexible structures. Here we introduce minimalistic tripeptide sequences that form highly soluble dynamic ensembles through multivalent side-chain interactions. We observe these supramolecular dispersions undergo drying-induced sequential liquid-liquid phase separation followed by solidification, resulting in

Commercially Available Eye Drops Containing Trehalose Protect Against Dry Conditions via Autophagy Induction.

Purpose: Evaluation of marketed eye drops with or without trehalose, a nonreducing natural osmoprotector disaccharide, in autophagy modulation and its role in cell survival during desiccation. Methods: Eye drops containing either sodium hyaluronate (SH) (Hyabak®, Thea, France) or a combination of SH with trehalose (Thealose Duo®, Thea, France) were compared with control conditions to evaluate the ability to modulate autophagy in human epithelial cells in vitro. Autophagy was monitored using LC3, a marker of the autophagic machinery, by fluorescence microscopy and immunoblot analysis. Control a

Glycerol hypersensitivity in a Drosophila model for glycerol kinase deficiency is affected by mutations in eye pigmentation genes.

Glycerol kinase plays a critical role in metabolism by converting glycerol to glycerol 3-phosphate in an ATP dependent reaction. In humans, glycerol kinase deficiency results in a wide range of phenotypic variability; patients can have severe metabolic and CNS abnormalities, while others possess hyperglycerolemia and glyceroluria with no other apparent phenotype. In an effort to help understand the pathogenic mechanisms underlying the phenotypic variation, we have created a Drosophila model for glycerol kinase deficiency by RNAi targeting of dGyk (CG18374) and dGK (CG7995). As expected, RNAi f

Factors affecting survival of bacteriophage on tomato leaf surfaces.

The ability of bacteriophage to persist in the phyllosphere for extended periods is limited by many factors, including sunlight irradiation, especially in the UV zone, temperature, desiccation, and exposure to copper bactericides. The effects of these factors on persistence of phage and formulated phage (phage mixed with skim milk) were evaluated. In field studies, copper caused significant phage reduction if applied on the day of phage application but not if applied 4 or 7 days in advance. Sunlight UV was evaluated for detrimental effects on phage survival on tomato foliage in the field. Phag

Trehalose augments autophagy to mitigate stress induced inflammation in human corneal cells.

PURPOSE: Cornea acts as a structural barrier and protects the eye from environmental stresses. Inflammation in ocular surface causes discomfort and visual distortion. Defective autophagy has been associated with inflammation and ocular surface diseases. Therefore, we explored the protective role of trehalose on inflammation and desiccation-triggered stress in human corneal cells in vitro and in dry eye patients. METHOD: TNF-α and desiccation stress induced human corneal cells (piHCF and HCE-T) with or without trehalose treatment were analyzed for the expression levels of inflammatory and auto

Laparotomy causes loss of peritoneal mesothelium prevented by humidified CO2 insufflation in rats.

INTRODUCTION: Avoiding tissue desiccation is a common recommendation to reduce postoperative complications following open abdominal surgery, although difficult to achieve delicately without damaging the peritoneal mesothelium. Insufflation of humidified-warm CO2 into the abdomen during open abdominal surgery is proposed as an invisible, effortless way to prevent desiccation. We hypothesized that desiccation during open abdominal surgery would cause loss of peritoneal mesothelium that would be prevented by insufflation of humidified-warm CO2. METHODS: Nine Wistar rats were assigned to 1 h of a

Species-specific vulnerability of benthic marine embryos of congeneric snails (Haminoea spp.) to ultraviolet radiation and other intertidal stressors.

We used field surveys and multi-factorial experiments to examine synergistic effects of ultraviolet radiation (UVR) and low tide conditions on the embryonic mortality of two bubble-shell snail species that deposit gelatinous egg masses in intertidal mudflats: Haminoea zelandiae from New Zealand, and Haminoea vesicula from Washington, USA. Egg masses of both species were predominantly found in shallow pools at low tide, and a substantial proportion of both were found in sunny as well as shaded microhabitats. Both exposure to sun and desiccation led to increased embryonic mortality for naturally

AvaKa, a protein of unknown function, contributes to iron homeostasis in the cyanobacterium Anabaena sp. strain PCC 7120.

Water is indispensable to life, yet some cyanobacteria inhabit hyper-arid deserts and withstand complete desiccation. However, the molecular mechanisms enabling such acclimation remain incompletely understood. In Anabaena sp. strain PCC 7120, the gene avaKa, which encodes a protein of unknown function, has been shown to be required for desiccation tolerance. Here, we characterized the avaKa disruptant DRavaKa under dehydration-related stress conditions. DRavaKa displayed hypersensitivity to EDTA, and transcripts of iron-deficiency-inducible genes (isiA1, isiB, furA, and sufB) were elevated eve

Simplified field preservation of tissues for subsequent DNA analyses.

Successful DNA-based identification of mass disaster victims depends on acquiring tissues that are not highly degraded. In this study, multiple protocols for field preservation of tissues for later DNA analysis were tested. Skin and muscle samples were collected from decaying pig carcasses. Tissues were preserved using cold storage, desiccation, or room temperature storage in preservative solutions for up to 6 months. DNA quality was assessed through amplification of successively larger segments of nuclear DNA. Solution-based storage, including a DMSO/NaCl/EDTA mixture, alcohols, and RNAlater

Effects of Thermal Acclimation on the Tolerance of Bactrocera zonata (Diptera: Tephritidae) to Hydric Stress.

Insects, similarly to other small terrestrial invertebrates, are particularly susceptible to climatic stress. Physiological adjustments to cope with the environment (i.e., acclimation) together with genetic makeup eventually determine the tolerance of a species to climatic extremes, and constrain its distribution. Temperature and desiccation resistance in insects are both conditioned by acclimation and may be interconnected, particularly for species inhabiting xeric environments. We determined the effect of temperature acclimation on desiccation resistance of the peach fruit fly (Bactrocera zo

Clusterin in the eye: An old dog with new tricks at the ocular surface.

The multifunctional protein clusterin (CLU) was first described in 1983 as a secreted glycoprotein present in ram rete testis fluid that enhanced aggregation ('clustering') of a variety of cells in vitro. It was also independently discovered in a number of other systems. By the early 1990s, CLU was known under many names and its expression had been demonstrated throughout the body, including in the eye. Its homeostatic activities in proteostasis, cytoprotection, and anti-inflammation have been well documented, however its roles in health and disease are still not well understood. CLU is promin

Impact of Nanostructured Lipid Carriers as an Artificial Tear Film in a Rabbit Evaporative Dry Eye Model.

PURPOSE: To characterize formulations of nanostructured lipid carriers (NLCs) as an artificial tear film and evaluate their efficacy in protecting the ocular surface epithelial cells from desiccating stress in vivo. METHODS: The physicochemical properties of NLCs, produced with components similar to the tear film such as phosphatidylcholine and squalene, were determined. In vitro cytotoxicity of NLCs was evaluated by a short-time exposure test in porcine corneal epithelial cells using a methyl thiazol diphenyl-tetrazolium bromide assay. The residence time of NLCs in rabbit eyes and the effica

Tardigrades as a potential model organism in space research.

Exposure of living organisms to open space requires a high level of tolerance to desiccation, cold, and radiation. Among animals, only anhydrobiotic species can fulfill these requirements. The invertebrate phylum Tardigrada includes many anhydrobiotic species, which are adapted to survive in very dry or cold environmental conditions. As a likely by-product of the adaptations for desiccation and freezing, tardigrades also show a very high tolerance to a number of other, unnatural conditions, including exposure to ionizing radiation. This makes tardigrades an interesting candidate for experiment

Cuticular differences associated with aridity acclimation in African malaria vectors carrying alternative arrangements of inversion 2La.

BACKGROUND: Principal malaria vectors in Africa, An. gambiae and An. coluzzii, share an inversion polymorphism on the left arm of chromosome 2 (2La/2L+a) that is distributed non-randomly in the environment. Genomic sequencing studies support the role of strong natural selection in maintaining steep clines in 2La inversion frequency along environmental gradients of aridity, and physiological studies have directly implicated 2La in heat and desiccation tolerance, but the precise genetic basis and the underlying behavioral and physiological mechanisms remain unknown. As the insect cuticle is the

Apolipoprotein D3 is required for long-chain cuticular hydrocarbon accumulation and water retention in Acyrthosiphon pisum.

BACKGROUND: The pea aphid, Acyrthosiphon pisum, is a damaging phloem-feeding pest of legume crops, and RNA interference (RNAi)-based targeting of stress-adaptation genes offers a promising route for target-specific pest management. Apolipoprotein D (ApoD) proteins are putative lipid carriers in insects, but the function of the chromosome-3 paralog ApApoD3 in aphid cuticular waterproofing remains unknown. We investigated whether ApApoD3 regulates cuticular hydrocarbon (CHC) accumulation and desiccation tolerance in A. pisum using RNAi, GC-MS-based CHC profiling, water-balance and survival assay

Survival of Cryptosporidium parvum oocysts under various environmental pressures.

The survival of various isolates of Cryptosporidium parvum oocysts under a range of environmental pressures including freezing, desiccation, and water treatment processes and in physical environments commonly associated with oocysts such as feces and various water types was monitored. Oocyst viability was assessed by in vitro excystation and by a viability assay based on the exclusion or inclusion of two fluorogenic vital dyes. Although desiccation was found to be lethal, a small proportion of oocysts were able to withstand exposure to temperatures as low as -22 degrees C. The water treatment

Chemical profiling of the street cocktail drug 'nyaope' in South Africa using GC-MS II: Stability studies of the cannabinoid, opiate and antiretroviral components during sample storage.

Nyaope is a mixture of low grade heroin, cannabis products, antiretroviral drugs and other materials added as bulking agents. It is a highly physically additive mixture which is smoked by users. As part of the development of a method for the analysis and profiling of nyaope this study evaluates the stability of the cannabinoid, opiate and antiretroviral components of nyaope during storage following seizure. Conditions used were those typically used for storage of drug seizures: in a desiccator in a refrigerator, in a desiccator in the dark at room temperature, in a desiccator in daylight at ro

Crepuscular vocal partitioning and thermal limits on calling behavior in the Bubbling Kassina (Kassina senegalensis) in a sub-equatorial savanna.

Acoustic communication in anurans is energetically costly and constrained by environmental factors, especially in open savanna ecosystems where extreme temperature fluctuations, desiccation risks, and wind-induced noise pose significant challenges to signal propagation and caller physiology. We analyzed the acoustic ecology of the widespread bubbling Kassina (Kassina senegalensis) in Mozambique, and compared it with that of other locations. We examined the influence of temperature, photoperiod and moonlight on calling phenology. Calling phenology was strongly seasonal driven, while daily patte

Heat stress and amphibian immunity in a time of climate change.

As a class of vertebrates, amphibians, are at greater risk for declines or extinctions than any other vertebrate group, including birds and mammals. There are many threats, including habitat destruction, invasive species, overuse by humans, toxic chemicals and emerging diseases. Climate change which brings unpredictable temperature changes and rainfall constitutes an additional threat. Survival of amphibians depends on immune defences functioning well under these combined threats. Here, we review the current state of knowledge of how amphibians respond to some natural stressors, including heat
