Temporal propagation of neural state boundaries in naturalistic context.

Our senses receive a continuous stream of complex information, which we segment into discrete events. Previous research has related such events to neural states: temporally and regionally specific stable patterns of brain activity. The aim of this paper was to investigate whether there was evidence for top-down or bottom-up propagation of neural state boundaries. To do so, we used intracranial measurements with high temporal resolution while subjects were watching a movie. As this is the first study of neural states in intracranial data in the context of event segmentation, we also investigate

Auditory prediction and prediction error responses evoked through a novel cascade roving paradigm: a human ECoG study.

Auditory sensory processing is assumed to occur in a hierarchical structure including the primary auditory cortex (A1), superior temporal gy … digm, which incorporated complex structures (cascade sequences) into a roving paradigm. We analyzed electrocorticography data from six patients with refractory epilepsy who passively listened to this novel auditory paradigm and detected responses to deviants mainly in the superior temporal gyrus and inferior frontal gyrus. Notably, the inferior frontal gyrus exhibited …

Depth versus surface: A critical review of subdural and depth electrodes in intracranial electroencephalographic studies.

Intracranial electroencephalographic (IEEG) recording, using subdural electrodes (SDEs) and stereoelectroencephalography (SEEG), plays a pivotal role in localizing the epileptogenic zone (EZ). SDEs, employed for superficial cortical seizure foci localization, provide information on two-dimensional seizure onset and propagation. In contrast, SEEG, with its three-dimensional sampling, allows exploration of deep brain structures, sulcal folds, and bihemispheric networks. SEEG offers the advantages of fewer complications, better tolerability, and coverage of sulci. Although both modalities allow e

MTL-P300 as a marker of the epileptogenic zone and hippocampal functionality in the presurgical evaluation of temporal lobe epilepsy: a systematic review.

BACKGROUND: In the past twenty years, there has been an increasing interest among neuroscientists and physicians in mapping the cortical areas involved in the epileptogenic zone (EZ) through event-related potentials (ERPs) that enable the evaluation of the functional preservation of these areas. The present review is an update on publications on this topic. OBJECTIVE: To investigate the accuracy of the cognitive evoked of the medial temporal lobe P300 (MTL-P300) potential in detecting the EZ in temporal lobe epilepsy (TLE). METHODS: The systematic review of articles on the PubMed, Embase and

Zero-crossing patterns reveal subtle epileptiform discharges in the scalp EEG.

Clinical diagnosis of epilepsy depends heavily on the detection of interictal epileptiform discharges (IEDs) from scalp electroencephalographic (EEG) signals, which by purely visual means is far from straightforward. Here, we introduce a simple signal analysis procedure based on scalp EEG zero-crossing patterns which can extract the spatiotemporal structure of scalp voltage fluctuations. We analyzed simultaneous scalp and intracranial EEG recordings from patients with pharmacoresistant temporal lobe epilepsy. Our data show that a large proportion of intracranial IEDs manifest only as subtle, l

Development of wirelessly-powered, extracranial brain activator (ECBA) in a large animal model for the future non-invasive human neuromodulation.

As transcranial electrical stimulation (tES) is an emerging and promising technique for neuromodulation, we developed a novel device; wirelessly-powered, extracranial brain activator (ECBA), which is mounted subcutaneously, and its neuromodulation effect was investigated. The oscillatory changes in electrocorticography (EcoG) were analyzed from two types of stimulation. Two weeks prior to the recording experiment, we underwent surgery for implantation of subdural strips and ECBA module over centroparietal regions of anesthetized beagles. Low-frequency stimulation (LFS) and subsequent high-freq…

Bimodal coupling of ripples and slower oscillations during sleep in patients with focal epilepsy.

OBJECTIVE: Differentiating pathologic and physiologic high-frequency oscillations (HFOs) is challenging. In patients with focal epilepsy, HFOs occur during the transitional periods between the up and down state of slow waves. The preferred phase angles of this form of phase-event amplitude coupling are bimodally distributed, and the ripples (80-150 Hz) that occur during the up-down transition more often occur in the seizure-onset zone (SOZ). We investigated if bimodal ripple coupling was also evident for faster sleep oscillations, and could identify the SOZ. METHODS: Using an automated ripple

Theta Oscillations in the Human Medial Temporal Lobe during Real-World Ambulatory Movement.

The theta rhythm-a slow (6-12 Hz) oscillatory component of the local field potential-plays a critical role in spatial navigation and memory by coordinating the activity of neuronal ensembles within the medial temporal lobe (MTL). Although theta has been extensively studied in freely moving rodents, its presence in humans has been elusive and primarily investigated in stationary subjects. Here we used a unique clinical opportunity to examine theta within the human MTL during untethered, real-world ambulatory movement. We recorded intracranial electroencephalographic activity from participants c

OpenSpindleNet: An open-source deep learning network for reliable sleep spindle detection in scalp and intracranial EEG.

Sleep spindles, an oscillatory brain activity occurring during light non-rapid eye movement (NREM) sleep, are important for memory consolidation and cognitive functions. Accurate detection is important for understanding the role of spindles in sleep state physiology and brain health and for better understanding sleep and neurological disorders. However, manual spindle labeling of electroencephalography (EEG) data is time-consuming and impractical for most clinical and research settings and intracranial EEG (iEEG) presents additional challenges for spindle identification due to its unique signa

A neural computational framework for face processing in the human temporal lobe.

A key question in cognitive neuroscience is how unified identity representations emerge from visual inputs. Here, we recorded intracranial electroencephalography (iEEG) from the human ventral temporal cortex (VTC) and medial temporal lobe (MTL), as well as single-neuron activity in the MTL, to demonstrate how dense feature-based representations in the VTC are translated into sparse identity-based representations in the MTL. First, we characterized the spatiotemporal neural dynamics of face coding in the VTC and MTL. The VTC, particularly the fusiform gyrus, exhibits robust axis-based feature c

An electronic neuromorphic system for real-time detection of high frequency oscillations (HFO) in intracranial EEG.

The analysis of biomedical signals for clinical studies and therapeutic applications can benefit from embedded devices that can process these signals locally and in real-time. An example is the analysis of intracranial EEG (iEEG) from epilepsy patients for the detection of High Frequency Oscillations (HFO), which are a biomarker for epileptogenic brain tissue. Mixed-signal neuromorphic circuits offer the possibility of building compact and low-power neural network processing systems that can analyze data on-line in real-time. Here we present a neuromorphic system that combines a neural recordi

Refractory epilepsy secondary to anti-GAD encephalitis treated with DBS post SEEG evaluation: a novel case report based on stimulation findings.

We report a case of medically refractory anti-GAD encephalitis which was treated with deep brain stimulation (DBS) after seizure termination was achieved using cortical stimulation during stereo-electroencephalography (SEEG) evaluation. The patient underwent bilateral SEEG implantation and cortical stimulation. Upon stimulation, mimicking the intrinsic seizures (at 1 Hz), it was possible to induce seizures with typical semiology, on multiple attempts. Stimulation during these seizures with high frequency (50 Hz) resulted in complete termination of the seizure. DBS was inserted after the SEEG e

Electrocortical spectral analysis and fractal methods for assessing the effects of unilateral brain injury on rat cerebellum.

We used electrocortical spectral analysis and fractal methods for assessing the effects of unilateral, single brain injury on cerebellum. Cerebellar electrocortical activity was recorded in control state (before the injury) and after a single brain injury of the cerebellar cortex in anesthetized rats. We noticed that the mean power in gamma high-frequency domain (32-128 Hz) of the cerebellum, was increased after the first brain injury, while after a two-week recovery, it was larger than before the injury. The unilateral brain injury induced a permanent increase of the mild gamma activity in bo

Arousal transitions in sleep, wakefulness, and anesthesia are characterized by an orderly sequence of cortical events.

Many aspects of brain function are influenced by modulatory processes, including arousal. The most abrupt changes in arousal occur at the wake-sleep transition and at the induction of anesthetic conditions. They are accompanied by major electrophysiological changes, including an emergence of low-frequency (sleep-like) activity and a loss of mid-frequency (wake-like) activity that has been linked to feedback processes of the brain. Nevertheless, the causal relationship between these two types of electrophysiological changes, as well as the cortical mechanisms underlying changes in arousal and c

Frontal cortex organization supporting audiovisual processing during naturalistic viewing.

Our brains dynamically adapt to a multisensory world by orchestrating diverse inputs across sensory streams. This process engages multiple brain regions, but it remains unclear how audiovisual stimuli are represented and evolve over time, especially in naturalistic scenarios. Here, we employed a movie-viewing paradigm to explore this question. We recorded intracranial electrocorticography (iEEG) to measure brain activity in 19 participants watching a short multilingual movie. Using unsupervised clustering and supervised encoding models, we identified a robust modality-specific gradient in the …

Association between motor cortex grey matter loss and inability to control an ECoG-based implanted Brain-Computer Interface in ALS.

BACKGROUND: The field of implantable Brain-Computer Interfaces (iBCIs) is rapidly advancing, with individuals with amyotrophic lateral sclerosis (ALS) as key beneficiaries. However, ALS-related cortical degeneration may impair iBCI effectiveness. This study investigated whether structural magnetic resonance imaging (MRI) and functional MRI (fMRI) metrics are associated with the quality of electrocorticography (ECoG) signals critical for iBCI use. METHODS: Six late-stage ALS participants and 76 controls underwent T1-weighted structural MRI and task-based fMRI during right-hand movement or atte…

Purkinje cell dysfunction causes disrupted sleep in ataxic mice.

Purkinje cell dysfunction disrupts movement and causes disorders such as ataxia. Recent evidence suggests that Purkinje cell dysfunction may also alter sleep regulation. Here, we used an ataxic mouse model generated by silencing Purkinje cell neurotransmission (L7Cre;Vgatfx/fx) to better understand how cerebellar dysfunction impacts sleep physiology. We focused our analysis on sleep architecture and electrocorticography (ECoG) patterns based on their relevance to extracting physiological measurements during sleep. We found that circadian activity was unaltered in the mutant mice, although thei…
