Chromosomal and gene mapping of uterine fibroids: A systematic review.

Genetic mutations and their phenotypic manifestation have been recognized as critical factors in tumorigenesis. However, the relationship between these mutations and the pathogenesis of uterine leiomyomas (UL) remains inadequately characterized. There is compelling evidence to suggest a genetic underpinning in UL development, alongside influences from epigenetics, environmental stimuli, growth hormones, and growth factors. A plethora of studies have tried to elucidate the genetic and epigenetic etiologies associated with UL, but the definitive implications of these findings remain unclear. An

Mechanisms of transcriptional regulation of CCR5, which is the co-receptor for R5-tropic HIV and is also involved in other disease processes.

C-C chemokine receptor type 5 (CCR5) is the R5-tropic human immunodeficiency virus type 1 or HIV co-receptor. Lower CCR5 levels can reduce T cell and macrophage susceptibility and suppress HIV infection. Moreover, CCR5Δ32 homozygous stem cell transplantation is central to HIV cure. Other studies have shown that CCR5 plays a vital role in cancer development and cell migration, and it was considered a potential therapeutic target for several types of malignancy. In addition to HIV and cancer, CCR5 also participates in immune response and plays a role in graft-versus-host disease in bone marrow t

Boosting Neurogenesis as a Strategy in Treating Alzheimer's Disease.

Alzheimer's disease (AD) causes progressive cognitive decline and neuronal loss, partly due to the buildup of amyloid-β (Aβ) plaques and tau tangles. Despite years of research, treatments targeting these hallmark pathologies have yielded only modest clinical success, prompting interest in regenerative approaches to restore the brain's ability to repair itself. One such approach focuses on adult hippocampal neurogenesis, the process by which neural stem cells (NSCs) produce new neurons throughout life. In AD, this process is impaired, worsening cognitive deficits. In this review, we examine the

Decoding hybridization barriers: the molecular and genetic orchestration of the triploid block in Arabidopsis thaliana.

Plant evolution has been greatly influenced by polyploidization phenomena. Polyploid plants yield more and are more resistant to unfavorable environments than their diploid relatives. The triploid block, a postzygotic barrier that causes failure of endosperm development and thus seed arrest, often prevents polyploid breeding. Alterations in the parental dose in interploidy crosses alter endosperm development by changing the correct maternal: paternal ratio (2m:1p) that this tissue requires to properly fulfill its proliferation and cellularization. After many years of research, the study of epi

Genetic, Epigenetic, and Non-Genetic Factors in Testicular Dysgenesis Syndrome: A Narrative Review.

BACKGROUND: Testicular dysgenesis syndrome (TDS) is a complex disorder of the male reproductive system related to disfunction of the fetal testis. The clinical features of TDS may be evident at birth or infancy (cryptorchidism, hypospadias and/or reduced anogenital distance) or occur later in adulthood (testis cancer, infertility). Genetic background seems to be important for genetic predisposition, with new genes being associated with components of the syndrome in last years. Interestingly, the incidence of clinical manifestations of TDS has been increasing in many countries in recent decades

Genetic Susceptibility and Pathogenesis of Hypospadias.

This study examines the genetic susceptibility and pathogenesis of hypospadias. Hypospadias is a birth defect of the urethra where the urinary opening is on the underside of the penis rather than at the tip. This study aims to identify the genetic basis of this defect and, by extension, improve our understanding of the biological pathways involved in its pathogenesis. Novel genetic loci and variants associated with hypospadias were identified through case-control studies in Europe and North America and, more recently, through large-scale genome-wide association studies (GWAS). The identified g

Epigenetics in Glaucoma.

Primary open angle glaucoma (POAG) is defined as a "genetically complex trait", where modifying factors act on a genetic predisposing background. For the majority of glaucomatous conditions, DNA variants are not sufficient to explain pathogenesis. Some genes are clearly underlying the more "Mendelian" forms, while a growing number of related polymorphisms in other genes have been identified in recent years. Environmental, dietary, or biological factors are known to influence the development of the condition, but interactions between these factors and the genetic background are poorly understoo

Sjogren's syndrome: Everything you always wanted to know about genetic and epigenetic factors.

Primary Sjögren's syndrome (pSS) is a chronic, systemic autoimmune disease characterized by a wide spectrum of glandular and extra-glandular features. Genetic and epigenetic factors play an important role in the disease susceptibility and phenotype. There are a multitude of genes that have been identified as implicated in the pathogenesis of pSS, both in HLA and extra-HLA regions with a strong contribution given by genes in interferon signalling pathways. Among the HLA alleles, the most consistent associations have been found with DR2 and DR3 alleles at the DRB1 locus. Moreover, several gene v

A Review of CRISPR Tools for Treating Usher Syndrome: Applicability, Safety, Efficiency, and In Vivo Delivery.

This review considers research into the treatment of Usher syndrome, a deaf-blindness syndrome inherited in an autosomal recessive manner. Usher syndrome mutations are markedly heterogeneous, involving many different genes, and research grants are limited due to minimal patient populations. Furthermore, gene augmentation therapies are impossible in all but three Usher syndromes as the cDNA sequence exceeds the 4.7 kb AAV packaging limit. It is, therefore, vital to focus research efforts on alternative tools with the broadest applicability. The CRISPR field took off in recent years following th

Genetic and epigenetic analyses of panic disorder in the post-GWAS era.

Panic disorder (PD) is a common and debilitating neuropsychiatric disorder characterized by panic attacks coupled with excessive anxiety. Both genetic factors and environmental factors play an important role in PD pathogenesis and response to treatment. However, PD is clinically heterogeneous and genetically complex, and the exact genetic or environmental causes of this disorder remain unclear. Various approaches for detecting disease-causing genes have recently been made available. In particular, genome-wide association studies (GWAS) have attracted attention for the identification of disease

The Immunologic Mechanisms of Eosinophilic Esophagitis.

Eosinophilic esophagitis (EoE) is a chronic allergic inflammatory disease that is triggered by food and/or environmental allergens and is characterized by a clinical and pathologic phenotype of progressive esophageal dysfunction due to tissue inflammation and fibrosis. EoE is suspected in patients with painful swallowing, among other symptoms, and is diagnosed by the presence of 15 or more eosinophils per high-power field in one or more of at least four esophageal biopsy specimens. The prevalence of EoE is increasing and has now reached rates similar to those of other chronic gastrointestinal

Neo-enhancers in T-cell acute lymphoblastic Leukaemia (T-ALL) and beyond.

T-cell acute lymphoblastic leukaemia (T-ALL) is a rare aggressive haematological malignancy characterised by the clonal expansion of immature T-cell precursors. It accounts for 15% of paediatric and 25% of adult ALL. T-ALL is associated with the overexpression of major transcription factors (TLX1/3, TAL1, HOXA) that drive specific transcriptional programmes and constitute the molecular classifying subgroups of T-ALL. Although the dysregulation of transcription factor oncogenes is frequently associated with chromosomal translocations in T-ALL, epigenetic dysregulation resulting in changes to po

Epigenetic signature of MAOA and MAOB genes in mental disorders.

Epigenetic processes such as DNA methylation are considered key mechanisms at the crossroads between genetics and environment in the etiology of mental disorders. The monoamine oxidases A and B (MAOA/MAOB) are prime candidates for the investigation into the role of DNA methylation in mental disorders, given their pivotal role in the metabolism of monoamines and as pharmacological targets of potent antidepressant drugs such as tranylcypromine, phenelzine or moclobemide. The present mini-review aims at summarizing and critically discussing the growing body of the literature supporting a role of

The role of genome-wide DNA methylation and polymorphisms in periodontitis etiology: A narrative review.

Periodontitis is a multifactorial inflammatory disease influenced by genetic, epigenetic, and environmental factors. Recent advancements in genomic and epigenomic research have highlighted the role of genetic polymorphisms and genome-wide DNA methylation in its pathogenesis. DNA methylation regulates gene expression, affecting immune responses and inflammatory pathways, while genetic polymorphisms may predispose individuals to altered host-microbial interactions and increased susceptibility to periodontal destruction. Recent studies have identified promising periodontal biomarkers, including s

Genetic and epigenetic factors shape phenotypes and outcomes in systemic lupus erythematosus - focus on juvenile-onset systemic lupus erythematosus.

PURPOSE OF REVIEW: Systemic lupus erythematosus (SLE) is a severe autoimmune/inflammatory disease. Patients with juvenile disease-onset and those of non-European ancestry are most severely affected. While the exact pathophysiology remains unknown, common and rare gene variants in the context of environmental exposure and epigenetic alterations are involved. This manuscript summarizes the current understanding of genetic and epigenetic contributors to SLE risk, manifestations and outcomes. RECENT FINDINGS: Though SLE is a mechanistically complex disease, we are beginning to understand the impa

From engineered embryos to animal models: the structure-regulation gap in synthetic mammalian reproduction.

Alternative approaches to mammalian reproduction have made remarkable progress with the combination of genome engineering, stem cell technologies and synthetic embryos, allowing the study of molecular and cellular processes during germline development and early development. These models offer insightful platforms to investigate in vivo inaccessible molecular and cellular processes. Yet, while these advances have been made, the current models are still limited to partial reconstitution of development. A major limitation of these approaches is their inability to orchestrate diverse regulatory pa

Gene set mediation analysis in high-dimensional epigenetic studies.

Mediation analysis has significantly enhanced the elucidation of underlying epigenetic mechanisms by identifying mediators that influence the effect of an exposure on an outcome. In epigenomics research, the exponential increase in the dimensionality of mediators necessitates the development of advanced high-dimensional mediation models. Here we introduce a novel Gene Set High-Dimensional Mediation Analysis (gHDMA) model to understand the gene-level mediation role of DNA methylations, building upon our previous HDMA method. We begin our analysis by applying functional principal component analy

Nonlinearities and Switch-Like Behavior in Gene Expression: From Genetics to Biochemistry and Back.

Many developmental and physiological outcomes depend on transcriptional thresholds or switches. The transcriptional output, ultimately measured as mature RNA, is determined by a series of molecular steps that can introduce thresholds, bottlenecks, or amplification mechanisms. Their combination can convert quantitative changes in regulator abundance into more-than-proportional changes in mature RNA production. Such nonlinear, S-shaped/sigmoidal input-output relationships may account, at least in part, for the haploinsufficiency (HI) of transcription factors (TFs) and chromatin regulators. Here,

Comparing the predictors of mutability among healthy human tissues inferred from mutations in single-cell genome data.

Studying mutation in healthy somatic tissues is the key for understanding the genesis of cancer and other genetic diseases. Mutation rate varies from site to site in the human genome by up to 100-fold and is influenced by numerous epigenetic and genetic factors including GC content, trinucleotide sequence context, and DNAse accessibility. These factors influence mutation at both local and regional scales and are often interrelated with one another, meaning that predicting mutability or uncovering its drivers requires modelling multiple factors and scales simultaneously. Historically, most inve

Genetic and Epigenetic Approaches to Opioid Use Disorder.

BACKGROUND: Opioid use disorder (OUD) is a major global-scale social issue affecting public health. The high potential for addiction and dependence makes opioid use a significant concern, contributing to substance-related disorders. Both genetic and environmental factors contribute to the predisposition to OUD, with the opioidergic, dopaminergic, and GABAergic systems playing primary roles in itsonset. METHODS: This narrative review documents the association between genes and their variants related to these three systems, along with current evidence on epigenetic interventions in OUD. Relevan
