Computational designing of a novel subunit vaccine for human cytomegalovirus by employing the immunoinformatics framework.

Human cytomegalovirus (HCMV) is a widespread virus that can cause serious and irreversible neurological damage in newborns and even death in children who do not have the access to much-needed medications. While some vaccines and drugs are found to be effective against HCMV, their extended use has given rise to dose-limiting toxicities and the development of drug-resistant mutants among patients. Despite half a century's worth of research, the lack of a licensed HCMV vaccine heightens the need to develop newer antiviral therapies and vaccine candidates with improved effectiveness and reduced si

Conserved multiepitope vaccine constructs: A potent HIV-1 therapeutic vaccine in clinical trials.

Despite the success of Antiretroviral Therapy (ART) in preventing HIV-1-associated clinical progression to AIDS, it is unable to eliminate the viral reservoirs and eradicate the HIV-1 infection. Therapeutic vaccination is an alternative approach to alter the HIV-1 infection course. It can induce effective HIV-1-specific immunity to control viremia and eliminate the need for lifelong ART. Immunological data from spontaneous HIV-1 controllers have shown that cross-reactive T-cell responses are the key immune mechanism in HIV-1 control. Directing these responses toward preferred HIV-1 epitopes is

Identification of B and T Cell Epitopes to Design an Epitope-Based Peptide Vaccine against the Cell Surface Binding Protein of Monkeypox Virus: An Immunoinformatics Study.

BACKGROUND: Although the monkeypox virus-associated illness was previously confined to Africa, recently, it has started to spread across the globe and become a significant threat to human lives. Hence, this study was designed to identify the B and T cell epitopes and develop an epitope-based peptide vaccine against this virus's cell surface binding protein through an in silico approach to combat monkeypox-associated diseases. RESULTS: The analysis revealed that the cell surface binding protein of the monkeypox virus contains 30 B cell and 19 T cell epitopes within the given parameter. Among t

A novel multi-epitope mRNA vaccine against Escherichia coli using an in silico reverse vaccinology approach.

The adaptable nature of Escherichia coli, shifting from benign to virulent forms, poses a major healthcare challenge due to rising multidrug resistance and hypervirulence. In this study, using an in silico reverse vaccinology approach, we designed a multiepitope mRNA vaccine candidate targeting the Shiga toxin 1 (Stx1) of Shiga toxin-producing E. coli (STEC). Two cytotoxic T-Lymphocyte (CTL), three Helper T-Lymphocyte (HTL), and two Linear B-Lymphocyte (LBL) epitopes were identified and selected through stringent computational filtering based on high antigenicity, non-allergenicity, and non-to

A comprehensive approach to developing a multi-epitope vaccine against Mycobacterium tuberculosis: from in silico design to in vitro immunization evaluation.

INTRODUCTION: The Bacillus Calmette-Guérin (BCG) vaccine, currently used against tuberculosis (TB), exhibits inconsistent efficacy, highlighting the need for more potent TB vaccines. MATERIALS AND METHODS: In this study, we employed reverse vaccinology techniques to develop a promising multi-epitope vaccine (MEV) candidate, called PP13138R, for TB prevention. PP13138R comprises 34 epitopes, including B-cell, cytotoxic T lymphocyte, and helper T lymphocyte epitopes. Using bioinformatics and immunoinformatics tools, we assessed the physicochemical properties, structural features, and immunologi

Assessment of antigen immunogenicity formulated in minigenes transfected into antigen-presenting cells.

Tumor cells exhibit deficient antigen presentation to T cells, significantly contributing to immune evasion and tumor genesis. Peptide pulsed Antigen-presenting cells (APCs) are commonly used in cancer immunotherapy to circumvent the defects of tumor cells in processing and presenting antigens to T lymphocytes. However, peptides do not always represent epitopes naturally processed by tumor cells, which might reduce the identification of actual immunogenic antigens. Minigenes encoding concatenated immunogenic tumor epitope sequences offer a promising alternative to select tumor antigens natural

Engineering a novel multi-epitope mRNA vaccine against major bacterial meningitis pathogens: E. coli K1, group B Streptococcus, Listeria monocytogenes, Neisseria meningitidis, and Streptococcus pneumoniae.

Bacterial meningitis poses a persistent global health threat, underscoring the need for innovative preventive strategies. This study details the computational design of a novel multi-epitope mRNA vaccine targeting outer membrane porins (OMPs), crucial virulence factors, from five primary causative pathogens: Escherichia coli K1, Group B Streptococcus, Listeria monocytogenes, Neisseria meningitidis, and Streptococcus pneumoniae. Employing a comprehensive in silico approach, we identified promising cytotoxic T-lymphocyte (CTL), B-cell, and helper T-lymphocyte (HTL) epitopes derived from these OM

A blind spot of human T cell immunology: epitope specificity in secondary lymphoid organs.

The fate of T cells is determined through the expression of a unique T cell receptor (TCR). TCR epitope specificity drives spatial localization, phenotypic and metabolic plasticity, effector function, as well as the evolution of complex repertoires of T cell clones. However, human immunology is often restricted to the peripheral blood compartment and neglects 'professional' lymphoid organs. Furthermore, despite the generation of single-cell 'atlases', our knowledge of the epitopes that are recognized by human T cells is largely limited to only a handful of antigenic specificities from conventi

Employing computational tools to design a multi-epitope vaccine targeting human immunodeficiency virus-1 (HIV-1).

BACKGROUND: Despite being in the 21st century, the world has still not been able to vanquish the global AIDS epidemic, and the only foreseeable solution seems to be a safe and effective vaccine. Unfortunately, vaccine trials so far have returned unfruitful results, possibly due to their inability to induce effective cellular, humoral and innate immune responses. The current study aims to tackle these limitations and propose the desired vaccine utilizing immunoinformatic approaches that have returned promising results in designing vaccines against various rapidly mutating organisms. For this, a

Demonstration of epitope spreading in bullous pemphigoid: results of a prospective multicenter study.

Di Zenzo and colleagues have undertaken a multicenter prospective study to clarify the epitope profile for IgG anti-BP180 and BP230 antibodies in 35 patients with bullous pemphigoid (BP). Both intra- and intermolecular epitope spreading events were observed, in which epitopes shifted exclusively from extracellular to intracellular domains. The presence of IgG antibodies to the BP180 C-terminal domain and BP230, in addition to the BP180-NC16A domain, correlated with disease severity and activity, suggesting specific pathogenic relevance for anti-BP230 antibodies. Epitope spreading was found in

Designing a broad-spectrum multi-epitope subunit vaccine against leptospirosis using immunoinformatics and structural approaches.

INTRODUCTION: Leptospirosis, caused by Leptospira interrogans, is a neglected zoonotic disease that poses a significant global health risk to both humans and animals. The rise of antimicrobial resistance and the inefficacy of existing vaccines highlight the urgent need for new preventive strategies. METHODS: An immunoinformatics approach was employed to design a multi-epitope subunit vaccine (MESV) against leptospirosis. B-cell, cytotoxic T lymphocyte (CTL), and helper T lymphocyte (HTL) epitopes were selected from five key Leptospira proteins. These epitopes were fused with a heparin-binding

Computational design of a multi-epitope mRNA vaccine against orthopoxviruses: A path toward comprehensive poxvirus protection.

Along with the recent outbreak of monkeypox (MPOX), there are worries about other outbreaks of the poxvirus genus that threaten global public health. Until now, although there are no specific vaccines for MPOXV, there are no comprehensive vaccines for all dangerous poxviruses. This research utilizes immunoinformatics and structural vaccinology methodologies to develop a multi-epitope mRNA vaccine targeting variola virus (VARV), vaccinia virus (VACV), monkeypox virus (MPOXV), and cowpox virus (CPXV), which are four pathogenic orthopoxviruses (OPV). Accordingly, A29L, M1R, A35R, B6R, and F8L ant

A novel DNA vaccine containing multiple TB-specific epitopes cast in a natural structure elicits enhanced Th1 immunity compared with BCG.

Vaccination is expected to make a major contribution to the goal of eliminating tuberculosis worldwide by 2050. Because the protection afforded by the currently available tuberculosis vaccine, BCG, is insufficient, new vaccine strategies are urgently needed. Protective immunity against MTB depends on generation of a Th1-type cellular immune response characterized by secretion of IFN-gamma from antigen-specific T cells. Epitope-driven vaccines are created from sub-sequences of proteins (epitopes) derived by scanning the protein sequences of pathogens and selecting epitopes with patterns of amin

Immunogenicity and Efficacy of a Novel Multi-Antigenic Peptide Vaccine Based on Cross-Reactivity between Feline and Human Immunodeficiency Viruses.

For the development of an effective HIV-1 vaccine, evolutionarily conserved epitopes between feline and human immunodeficiency viruses (FIV and HIV-1) were determined by analyzing overlapping peptides from retroviral genomes that induced both anti-FIV/HIV T cell-immunity in the peripheral blood mononuclear cells from the FIV-vaccinated cats and the HIV-infected humans. The conserved T-cell epitopes on p24 and reverse transcriptase were selected based on their robust FIV/HIV-specific CD8⁺ cytotoxic T lymphocyte (CTL), CD4⁺ CTL, and polyfunctional T-cell activities. Four such evolutionarily cons

T-h-2 immunity and CD3+CD45RBlow-activated T cells in mice immunized with recombinant bacillus Calmette-Guérin expressing HIV-1 principal neutralizing determinant epitope.

The genetic engineering of Mycobacterium bovis-bacillus Calmette-Guérin to express foreign epitopes is an attractive strategy in the field of epitope vaccines. We constructed an 'epitope-trap vector' with Mycobacterium tuberculosis chaperonin-10 as a carrier antigen and used it to express the HIV-1 principal neutralizing determinant epitope. We also identified a new chaperonin-10 promoter that was hyperexpressive compared with the heat shock protein-65 promoter. Splenocytes from recombinant bacillus Calmette-Guérin-immunized mice showed enhanced lymphocyte proliferation and interleukin-4 (but

Designing B- and T-cell multi-epitope based subunit vaccine using immunoinformatics approach to control Zika virus infection.

The Zika virus is a rapidly spreading Aedes mosquito-borne sickness, which creates an unanticipated linkage birth deformity and neurological turmoil. This study represents the use of the combinatorial immunoinformatics approach to develop a multiepitope subunit vaccine using the structural and nonstructural proteins of the Zika virus. The designed subunit vaccine consists of cytotoxic T-lymphocyte and helper T-lymphocyte epitopes accompanied by suitable adjuvant and linkers. The presence of humoral immune response specific B-cell epitopes was also confirmed by B-cell epitope mapping among vacc

MHC basis of T cell-dependent heterologous immunity to arenaviruses.

Having a history of infection with one pathogen may sometimes provide a level of T cell-dependent protective heterologous immunity to another pathogen. This immunity was initially thought due to cross-reactive T cell epitopes, but recent work has suggested that such protective immunity can be initiated nonspecifically by the action of cytokines on memory T cells. We retested this concept using two small and well-defined arenaviruses, lymphocytic choriomeningitis virus (LCMV) and Pichinde virus (PV), and found that heterologous immunity in these systems was indeed linked to T cell epitopes and

Proteomics-based vaccine targets annotation and design of multi-epitope vaccine against antibiotic-resistant Streptococcus gallolyticus.

Streptococcus gallolyticus is a non-motile, gram-positive bacterium that causes infective endocarditis. S. gallolyticus has developed resistance to existing antibiotics, and no vaccine is currently available. Therefore, it is essential to develop an effective S. gallolyticus vaccine. Core proteomics was used in this study together with subtractive proteomics and reverse vaccinology approach to find antigenic proteins that could be utilized for the design of the S. gallolyticus multi-epitope vaccine. The pipeline identified two antigenic proteins as potential vaccine targets: penicillin-binding
