Effect of papaverine on the longitudinal and circular smooth muscle of the guinea-pig caecum.

Electrical and mechanical activity of smooth muscle of the guinea-pig caecum was recorded by means of the sucrose-gap technique. The responses of longitudinal and circular smooth muscle to acetycholine were differently affected by changes of extracellular calcium concentration (0.8, 2.5 and 7.5 mM). The contractions of both preparations were depressed at high Ca++ concentrations, whereas at low Ca++ concentrations only contractions of the circular smooth muscle were augmented. The stimulatory effect of acetylcholine was decreased by papaverine in both preparations at all three concentrations o

Pressure effects on the distribution of extracellular materials in the rhesus monkey outflow apparatus.

An electron-microscopic investigation was conducted to determine the distribution of extracellular materials in the rhesus monkey outflow ap … 5 mm Hg), heavy concentrations of stain were detected within the cores of the trabeculae and in the extracellular spaces of the endothelial meshwork. With pressure elevation there was a progressive decrease in the surface staining on the meshwork cells, an increase in staining intensity within the trabecular cores, and a 'washout' of extracellular materials from the …

Comparative ribosome profiling uncovers a dominant role for translational control in Toxoplasma gondii.

BACKGROUND: The lytic cycle of the protozoan parasite Toxoplasma gondii, which involves a brief sojourn in the extracellular space, is characterized by defined transcriptional profiles. For an obligate intracellular parasite that is shielded from the cytosolic host immune factors by a parasitophorous vacuole, the brief entry into the extracellular space is likely to exert enormous stress. Due to its role in cellular stress response, we hypothesize that translational control plays an important role in regulating gene expression in Toxoplasma during the lytic cycle. Unlike transcriptional profil…

Carbonic anhydrase inhibitor coated gold nanoparticles selectively inhibit the tumor-associated isoform IX over the cytosolic isozymes I and II.

An approach for the synthesis of carbonic anhydrase (CA, EC 4.2.1.1) inhibitor coated gold nanoparticles is reported. This nanomaterial selectively inhibited the tumor-associated isoform CA IX overexpressed in hypoxic cancers over the ubiquitous, cytosolic housekeeping isozymes CA I and II and was membrane impermeant. As CA IX has an extracellular active site, the new nanomaterial which is confined to the extracellular space may be useful for imaging and treatment of hypoxic tumors.

Extracellular DNA traps in inflammation, injury and healing.

Following strong activation signals, several types of immune cells reportedly release chromatin and granular proteins into the extracellular space, forming DNA traps. This process is especially prominent in neutrophils but also occurs in other innate immune cells such as macrophages, eosinophils, basophils and mast cells. Initial reports demonstrated that extracellular traps belong to the bactericidal and anti-fungal armamentarium of leukocytes, but subsequent studies also linked trap formation to a variety of human diseases. These pathological roles of extracellular DNA traps are now the focu…

Enhanced phototoxicity of photodynamic treatment by Cx26-composed GJIC via ROS-, calcium- and lipid peroxide-mediated pathways.

In spite of the promising initial treatment responses presented by photodynamic therapy (PDT), 5-year recurrence rates remain high level. Therefore, improvement in the efficacy of PDT is needed. There are reports showing that connexin(Cx) 26-composed gap junctional intercellular communication (GJIC) enhances the intercellular propagation of "death signal", thereby increasing chemotherapeutic cytotoxicity. However, it is unclear whether Cx26-formed GJIC has an effect on PDT phototoxicity. The results in the present study showed that Cx26-composed GJ formation at high density enhances the photot

[Anatomical structures, physiology and pathophysiology of cerebrospinal fluid metabolism; a review for an understanding of cerebrospinal fluid findings].

The very different macroscopic and microscopic details of the structures involved in production and reabsorption of cerebrospinal fluid in the different parts of the central nervous system are causing confusing laboratory findings in cerebrospinal fluids, the ingredients of which change even under normal condition in these different parts. Moreover the chemical ingredients of cerebrospinal fluid differ especially from the normal situation during some diseases of the central nervous system. Although we can be sure that there is much more diagnostic information in cerebrospinal fluid, we mainly

A sensitive in vivo model for quantifying interstitial convective transport of injected macromolecules and nanoparticles.

Effective interstitial transport of particles is necessary for injected drug/diagnostic agents to reach the intended target; however, quantitative methods to estimate such transport parameters are lacking. In this study, we develop an in vivo model for evaluating interstitial convection of injected macromolecules and nanoparticles. Fluorescently labeled macromolecules and particles are coinfused with a reference solute at constant infusion pressure intradermally into the mouse tail tip, and their relative convection coefficients are determined from spatial and temporal interstitial concentrati

The Role of Extracellular Histones in Disease Progression: A Potential Therapeutic Target for Disease Modulation.

Histones were once thought to be exclusive to the nucleus, but were recently discovered in the extracellular space, where they play important roles in disease pathogenesis. In addition to their traditional functions in chromatin organization and gene regulation, extracellular histones also serve as damage-associated molecular patterns (DAMPs), drive inflammation and immune responses, and are responsible for the progression of diseases such as sepsis, autoimmune diseases, and inflammatory diseases. To effectively target extracellular histones and improve disease progression, this review begins …

Morphologic features of the aqueous humor drainage pathways in horses.

The iridocorneal angle of the horse was anteriorly lined by a pectinate ligament, consisting of several layers of vertically oriented collagenous beams, that were interrupted by a network of extracellular spaces. The anterior chamber angle consisted mostly of a large uveal trabecular meshwork that was composed of widely-spaced, thick trabeculae. These trabeculae were attached in part posteriorly to extensions of the ciliary body musculature and anteriorly inserted into the prominent pectinate ligament. The corneoscleral trabecular meshwork was shaped hemiobovately and formed the external bound…

Ultrastructure of the human spinal arachnoid mater and dura mater.

Human spinal dura and arachnoid, obtained during neurosurgical operations, were studied by transmission electron microscopy. The ultrastructure of spinal meninges largely conformed to the morphology of the cranial meninges, but some minor differences were detected. The dura was composed of an outermost loosely arranged fibroelastic layer, a middle basically fibrous portion and an innermost cellular layer (dural border cell layer). The dural border cell layer was characterised by multiple interdigitating cell processes, no extracellular collagen, significant extracellular spaces and few cell ju…

Extracellular alveolar proteasome: possible role in lung injury and repair.

The ubiquitin-proteasome system is the major intracellular pathway for protein degradation in eukaryotes, and it also generates oligopeptides for antigen presentation. However, the 20S proteasome is also associated with the cell's outer membrane, and observations indicate its physiologic presence and biological activity in the extracellular alveolar space (i.e., in the epithelial lining fluid). Furthermore, its concentration is increased in the adult respiratory distress syndrome, acute lung injury, and other inflammatory lung disease. While its cellular origin, potential extracellular biologi

Disordered purinergic signaling inhibits pathological angiogenesis in cd39/Entpd1-null mice.

CD39/ecto-nucleoside triphosphate diphosphohydrolase-type-1 (ENTPD1) is the dominant vascular ecto-nucleotidase that catalyzes the phosphohydrolysis of extracellular nucleotides in the blood and extracellular space. This ecto-enzymatic process modulates endothelial cell, leukocyte, and platelet purinergic receptor-mediated responses to extracellular nucleotides in the setting of thrombosis and vascular inflammation. We show here that deletion of Cd39/Entpd1 results in abrogation of angiogenesis, causing decreased growth of implanted tumors and inhibiting development of pulmonary metastases. Qu…

An electron microscopic study of microfibrils of bone marrow.

After fixation of bone marrow with glutaraldehyde, tannic acid and saponin, a delicate network of microfibrils (10 nm) was observed in the extracellular space. Masses of microfibrils were most frequently observed between the endothelial cells of the sinusoidal wall and adventitial reticular cells, but were also observed at other sites throughout the marrow stroma. Microfibrils are an important component of the extracellular material of bone marrow and appear to provide an anchoring substrate for the endothelium.

Development of a three-dimensional extracellular matrix synthesized by human diploid fibroblasts in vitro.

Development and maturation of an extracellular matrix, synthesized by human gingival fibroblasts, have been studied microscopically. Pairs of demineralized, fibronectin-coated slices of human tooth root, 300 micron thick, were placed on confluent cell layers, defining a 0.5 mm wide space. The cultures were grown under standard conditions with ascorbic acid (50 micrograms ml-1) added daily. At various times up to 13 weeks, the cultures were fixed and the samples prepared for light and electron microscopy. Cells from the monolayer became attached to, and migrated up, the vertical root surface an

Vesicle inhibition reduces Candida biofilm resistance.

Candida biofilm matrix components are delivered to the extracellular space by vesicles where they deposit and confer biofilm-associated drug resistance. Here, we present evidence that drugs designed to inhibit mammalian exosome production exhibit similar effects on C. albicans extracellular vesicles, ultimately eliminating biofilm matrix assembly. We find that vesicle reduction renders biofilm communities susceptible to the antifungal fluconazole. Our findings argue that vesicle trafficking pathways represent a promising target to optimize for recalcitrant fungal biofilms.

Extracellular Circular RNAs Act as Novel First Messengers Mediating Cell Cross-Talk in Ischemic Cardiac Injury and Myocardial Remodeling.

Myocardial infarction (MI) causes most of the mortality worldwide. Coronary obstruction-caused myocardial ischemic injury leads to permanent … ury and myocardial remodeling. circRNAs can be loaded into extracellular vesicles and released into extracellular space. More recently, it was uncovered that the extracellular circRNAs can regulate intercellular communications, similar to "first messengers." The cross-talk mediated by extracellular circRNAs had been demonstrated to play important roles in pathologica …
