Inactivation of the adenosine A2A receptor protects apolipoprotein E-deficient mice from atherosclerosis.

BACKGROUND: Atherosclerosis is a chronic inflammatory disease of the arterial vessel wall. The A(2A) receptor (A(2A)R) plays a central role in many antiinflammatory effects of adenosine. However, the role of A(2A)R in atherosclerosis is not clear. METHODS AND RESULTS: The knockout of A(2A)R in apolipoprotein E-deficient (Apoe(-/-)/A(2A)R(-/-)) mice led to an increase in body weight and levels of blood cholesterol and proinflammatory cytokines, as well as the inflammation status of atherosclerotic lesions. Unexpectedly, Apoe(-/-)/A(2A)R(-/-) mice developed smaller lesions, as did chimeric Apoe

Nephrotic syndrome and pulmonary hypertension as complications of intravenous drug abuse.

This case report discusses a patient with nephrotic syndrome, pulmonary hypertension, and repeated episodes of infections. He had a history of intravenous drug abuse. Kidney biopsy revealed the rare finding of numerous foam cells, mainly in glomeruli. The solvent used for the drugs is thought to be responsible for the foam cells. In line with previous reports, we suspect that the pulmonary hypertension is consistent with foam cells in pulmonary capillaries or fat embolism syndrome due to the intravenous administered drugs. Our case demonstrates that the use of intravenous drugs can lead to wid

A Bioorthogonal Ratiometric Fluorescence Nanoprobe for Imaging the Regulation of O-GlcNAcylation in Atherosclerosis.

Protein post-translational modification (PTM) plays a significant role in the progression of atherosclerosis (AS). However, the role of O-GlcNAcylation, as an important O-glycosylation modification, in the formation of foam cells remains unclear, limiting early diagnosis and mechanistic studies. Here, utilizing two bioorthogonal reactions, we developed a ratiometric fluorescent nanoprobe-based tracker for O-GlcNAcylation. This tracker (BORFI) enables dynamic tracing, relative quantification, and multi-scale in situ visualization of protein O-GlcNAcylation during cellular events, and images the

Processing-Microstructure-Performance Relations in Thermoformed Auxetic Hyperelastic Foams with Enhanced Energy Absorption Capacity.

Auxetic (negative Poisson's ratio) foams with reentrant cell structures exhibit enhanced mechanical properties such as superior strength, energy absorption, and fracture resistance, compared to their nonauxetic counterparts. A well-established method for inducing auxeticity in cellular solids involves permanently changing the cell ribs that are buckled under compressive loads. This permanent change can be achieved by heating a deformed foam for a specific duration. In this study, a thermoforming process is developed to convert closed-cell hyperelastic polyurea foams into auxetic structures. Th

Cutaneous verruciform xanthoma: a report of five cases investigating the etiology and nature of xanthomatous cells.

Verruciform xanthoma is a rare clinicopathologic entity of uncertain etiology that occurs primarily in the oral mucosa. Aggregates of foam cells in the submucosal stroma or papillary dermis in association with verrucous epithelial hyperplasia are the hallmark of this lesion. Extraoral (cutaneous) occurrence of verruciform xanthoma is much rarer and has been reported mostly in the genital skin. Five cases of extraoral cutaneous verruciform xanthoma (three from the scrotum, one from the penis, and one from the nose) and one histologic "simulant" (from skin of the nose) were studied. The lesions

Compressive Mechanical Behavior and Corresponding Failure Mechanism of Polymethacrylimide Foam Induced by Thermo-Mechanical Coupling.

Thermal-mechanical coupling during the molding process can cause compressive yield in the polymer foam core and then affect the molding quality of the sandwich structure. This work investigates the compressive mechanical properties and failure mechanism of polymethacrylimide (PMI) foam in the molding temperature range of 20-120 °C. First, the DMA result indicates that PMI foam has minimal mechanical loss in the 20~120 °C range and can be regarded as an elastoplastic material, and the TGA curve further proves that the PMI foam is thermally stable within 120 °C. Then, the compression results sho

Atherosclerosis in mice lacking apo E. Evaluation of lesional development and progression.

Apolipoprotein E-deficient mice have spontaneous elevations of total plasma cholesterol and triglycerides and reduced high-density lipoprotein. The mice develop arterial lesions in a time-dependent manner. Lesional distribution was centered in the aortic sinus in young mice, and the lesions were widely distributed throughout the arterial tree in mice at 8 to 9 months of age. In young mice, subendothelial foam cell deposits were present in the aortic sinus adjacent to valve-attachment sites. By 5 months of age, foam cell deposits, free cholesterol, and admixed smooth muscle cells composed the d

Peripheral nerve regeneration using bioresorbable macroporous polylactide scaffolds.

The ability of DRG-derived neurons to survive and attach onto macroporous polylactide (PLA) foams was assessed in vitro. The foams were fabricated using a thermally induced polymer-solvent phase separation. Two types of pore structures, namely oriented or interconnected pores, can be produced, depending on the mechanism of phase separation, which in turn can be predicted by the thermodynamics of the polymer-solvent pair. Coating of the porous foams with polyvinylalcohol (PVA) considerably improved the wettability of the foams and allowed for cell culture. The in vitro biocompatibility of the P

Study on the preparation and properties of lignin-based flexible polyurethane foams with NaCl as a medium.

Currently, most polyols used in polyurethane foam (PUF) synthesis are derived from petrochemicals. However, lignin as the most abundant aromatic biopolymer rich in hydroxyl groups, is a suitable filler for synthesizing polyurethane foam. Therefore, in this study, Kraft lignin (KL) was utilized as a partial substitute for flexible polyethylene glycol 400 (PEG400). After adding 15 % KL, NaCl was introduced as a medium for synthesizing LFPUF and Polydimethylsiloxane (PDMS) was used as a foam stabilizer. Lignin-based flexible polyurethane foam (LFPUF) with high elasticity was synthesized. The arom

Comprehensive Multimodal Profiling of Atherosclerosis Reveals Bhlhe40 as a Potential Regulator of Vascular Smooth Muscle Cell Phenotypic Modulation.

BACKGROUND: Vascular smooth muscle cells (VSMCs) play a central role in atherosclerosis by undergoing phenotypic modulation from a quiescent, contractile state to a range of synthetic phenotypes, including fibroblast-like, macrophage-like, and lipid-laden foam cell-like states. However, a comprehensive multimodal characterization and understanding of the transcriptional programs driving these transitions remain incomplete. METHODS: To comprehensively define the phenotypic diversity of VSMCs during atherosclerosis progression, we performed in-depth profiling using cellular indexing of transcri

Macropinocytosis enhances foamy macrophage formation and cholesterol crystallization to activate NLRP3 inflammasome after spinal cord injury.

After spinal cord injury (SCI), phagocytes endocytose myelin debris to form foam cells, exacerbating the inflammatory response. It has been previously shown that macrophages become foam cells through the phagocytosis of myelin debris via receptor-dependent mechanisms after SCI. Blocking receptor-mediated endocytosis did not completely prevent foam cell formation, so we investigated receptor-independent endocytosis. Here, we revealed that foam cells formed after myelin debris internalization were predominantly macrophages rather than microglia. Receptor-independent macropinocytosis has an impor

A novel aluminum foam structure for combined excellent wave attenuation and ventilation performance.

In this paper, a novel aluminum foam structure with wave attenuation and ventilation performance suitable for underground space is designed and prepared. It focuses on dynamic response of aluminum foam structure under explosion impact load and ventilation resistance at different wind speeds. Failure modes of each component are analyzed and attenuation mechanism of explosive shock wave are revealed. The results indicate that: under the synergistic action of the crushing behavior of the aluminum foam cells, the rough wall structure of the ventilation holes and the special diagonal square honeyco

Lipid droplet-associated proteins in atherosclerosis (Review).

Accumulation of atherosclerotic plaques in arterial walls leads to major cardiovascular diseases and stroke. Macrophages/foam cells are central components of atherosclerotic plaques, which populate the arterial wall in order to remove harmful modified low‑density lipoprotein (LDL) particles, resulting in the accumulation of lipids, mostly LDL‑derived cholesterol ester, in cytosolic lipid droplets (LDs). At present, LDs are recognized as dynamic organelles that govern cellular metabolic processes. LDs consist of an inner core of neutral lipids surrounded by a monolayer of phospholipids and free

Degradation of h-acid by free and immobilized cells of Alcaligenes latus.

Alcaligenes latus, isolated from industrial effluent, was able to grow in mineral salts medium with 50 ppm (0.15 mM) of H-acid as a sole source of carbon. Immobilization of Alcaligenes latus in Ca-alginate and polyurethane foam resulted in cells embedded in the matrices. When free cells and immobilized cells were used for biodegradation studies at concentration ranging from 100 ppm (0.3 mM) to 500 ppm (1.15 mM) degradation rate was enhanced with immobilized cells. Cells immobilized in polyurethane foam showed 100% degradation up to 350 ppm (1.05 mM) and 57% degradation at 500 ppm (1.5 mM). Deg

Gadolinium Chloride Rescues Niemann⁻Pick Type C Liver Damage.

Niemann⁻Pick type C (NPC) disease is a rare neurovisceral cholesterol storage disorder that arises from loss of function mutations in the NPC1 or NPC2 genes. Soon after birth, some patients present with an aggressive hepatosplenomegaly and cholestatic signs. Histopathologically, the liver presents with large numbers of foam cells; however, their role in disease pathogenesis has not been explored in depth. Here, we studied the consequences of gadolinium chloride (GdCl₃) treatment, a well-known Kupffer/foam cell inhibitor, at late stages of NPC liver disease and compared it with NPC1 genetic res

Preparation of anti-shrinkage branched poly (butylene succinate-co-butylene terephthalate)/cellulose nanocrystal foam with excellent degradability and thermal insulation.

Branched poly (butylene succinate-co-butylene terephthalate) (BPBST) was synthesized by in-situ polycondensation to enhance the foamability of poly (butylene succinate-co-butylene terephthalate) (PBST) and was blended with cellulose nanocrystals (CNC) to address foam shrinkage. The introduction of 2 wt% CNC increased the crystallization temperature of BPBST from 66.6 °C to 87.7 °C, accelerating its crystallization rate and improving both the mechanical properties and complex viscosity of the polymer matrix. Microcellular foams were produced using supercritical CO2 as a blowing agent, with CNC

Reciprocal Regulation Between miR-148a/152 and DNA Methyltransferase 1 Is Associated with Hyperhomocysteinemia-Accelerated Atherosclerosis.

DNA methyltransferase 1 (DNMT1) and miRNAs are both important regulators of gene expression that have been implicated in the pathogenesis of atherosclerosis. This study was designed to elucidate the potential interaction between DNMT1 and miRNAs in the context of hyperhomocysteinemia (HHcy)-related atherosclerosis. In the aorta of ApoE-/- mice fed a high methionine diet, increased expression of miR-148a/152, with decreased DNMT1 mRNA and protein levels, was detected. Similar changes were observed in cultured foam cells stimulated with homocysteine. When miR-148a/152 was overexpressed using vir

[Mesangial lipidosis associated with cholesterol polycoria].

Accumulation of glomerular foam cells were seen in a kidney biopsy of a 59-years old patient with persistent proteinuria and mild renal insufficiency. Further investigations revealed hypercholesterolemia, hypertriglyceridemia, normal activity of LCAT, absence of lipoprotein x. Foam cells which were shown by histochemical methods to be loaded with intracytoplasmic cholesterol were found in bone marrow. These findings strongly suggest cholesterol-ester-storage disease with glomerular involvement.

Ultra-High-Expansion-Ratio Nanocellular Semicrystalline Polymer Foams: Preparation of Chain-Extender-Mediated PLA/PPC Foams via Supercritical CO2.

The inherent brittleness, retarded crystallization kinetics and insufficient melt strength of polylactic acid (PLA) severely limit its applications in foaming. This study proposed a reactive chain extender and compatibilizer strategy, which involved introducing the multifunctional epoxy-based chain extender KL-E4370 (0.6 wt%) into a PLA/polypropylene carbonate (PPC) blend system to simultaneously enhance melt strength and interfacial compatibility, with the aim of preparing high-expansion-ratio nano-cellular foams by means of the physical foaming assisted by supercritical CO2.
