BACKGROUND: Fusobacterium nucleatum (Fn) drives chemotherapy resistance in colorectal cancer (CRC) by activating hypoxia-mimicking signaling pathways, yet the molecular mechanisms linking Fn to 5-fluorouracil (5-FU) resistance remain unclear. We investigated how Fn-induced hypoxia-related gene expression contributes to 5-FU resistance and identified a pharmacological strategy to overcome it.
METHODS: DEGs from Fn-infected CRC cells (GSE90944) were intersected with GSEA hypoxia-related genes, and Cox regression on TCGA-CRC data prioritized the top candidate, DDIT3. Its role in 5-FU resistance …

Untargeted metabolite profiling, aka metabolomics, offers a powerful technology to potentially reveal fundamental molecular mechanisms whereby a perturbation in cell metabolism leads to an alteration in cell growth or physiological maintenance. Notably, this discovery approach does not require any preconceived notion of underlying molecular mechanisms, affording an opportunity to effectively shine a molecular mechanistic spotlight in a direction that an informed investigator may not have thought to consider. Fusobacterium nucleatum colonization has been linked to the initiation and promotion o…

BACKGROUND: Immune disorders and impaired intestinal barrier function resulting from dysbiosis of the intestinal flora play pivotal roles in the pathogenesis of ulcerative colitis (UC). Fusobacterium nucleatum (F. nucleatum, Fn) can secrete extracellular vesicles (EVs) that surmount the mucus layer, thereby contributing to UC progression. However, the underlying mechanisms and their clinical significance remain elusive. METHODS: Bioinformatics and clinical sample analysis were employed to explore the relationship between F. nucleatum and the absent in melanoma 2 (AIM2) inflammasome. Both in vi…

The ubiquitous inflammophilic oral pathobiont Fusobacterium nucleatum (Fn) is widely recognized for its strong association with inflammatory dysbiotic diseases and cancer. Fn is subdivided into four subspecies, which are historically considered functionally interchangeable in the oral cavity. To test this assumption, we analyzed patient-matched dental plaque and odontogenic abscess clinical specimens and examined whether an inflammatory environment selects for/against particular Fn subspecies. Dental plaque harbored a greater diversity of fusobacteria, with Fn. polymorphum dominating, whereas …

Immune checkpoint blockade (ICB) therapy has been approved for breast cancer (BC), but clinical response rates are limited. Recent studies have shown that commensal microbes colonize a variety of tumors and are closely related to the host immune system response. Here, we demonstrated that Fusobacterium nucleatum (F.n), which is prevalent in BC, creates an immunosuppressive tumor microenvironment (ITME) characterized by a high-influx of myeloid cells that hinders ICB therapy. Administering the antibiotic metronidazole in BC can deplete F.n and remodel the ITME. To prevent an imbalance in the sy…

The lipopolysaccharide (LPS) of Fusobacterium nucleatum JCM 8532 was isolated by hot-phenol water extraction. Most of the LPS was extracted in the phenolic phase and shown to be the smooth-type, whereas the aqueous phase contained mainly rough-type LPS. The chemical composition of the LPS was similar to that reported in other studies, but D-quinovosamine, which may be a major component of O-antigenic polysaccharide, and 3-deoxy-D-manno-2-octulosonic acid (Kdoi) were detected for the first time by gas chromatography-mass spectrometry. The biological activities of smooth-type LPS, including limu…

Intracellular Fe3+ amount is one of the critical determinants of human health. The development of simple and effective probes for the quantitative detection of Fe3+in vivo is of great significance for the early diagnosis of disease or disorder associated with iron deficiency or overload. In this study, remarkable carbon dots, which can serve as a biosensor for efficient intracellular Fe3+ detection, were synthesized by hydrothermal carbonization of Fusobacterium nucleatum, an anaerobic bacterium. The achieved F. nucleatum-carbon dots (Fn-CDs) possessed the features of strong fluorescence, high…

Fusobacterium nucleatum bacteremia is critical and not well defined. To identify the clinical characteristics and outcomes, we conducted a retrospective review of hospitalized patients from January 2004 to December 2009 at a tertiary center in northern Taiwan. Fifty-seven patients were enrolled. The mean age was 58.1 years, and the mean Pitt bacteremia score was 4.7. Males predominated (59.6%), and the overall 30-day mortality rate was up to 47.4%. Malignancy was the major comorbidity (26/57, 45.6%), especially oropharyngeal and gastrointestinal cancers (19/26, 73.1%). Pneumonia (17/57, 29.8%)…

Patients with metastasis have an extremely poor prognosis in head and neck squamous cell carcinoma (HNSCC). Emerging studies have illuminated the impact of intratumor microbiota on cancer metastasis, though the specific role of Fusobacterium nucleatum (F. nucleatum) in HNSCC metastasis remains unresolved. This research found that intratumoral F. nucleatum abundance was elevated and correlated to diminished disease-free survival in HNSCC patients exhibiting lymph node metastasis. F. nucleatum invasion into primary and metastatic tumor tissues was observed using fluorescence in situ hybridizatio…

The oral-gut microbiota axis is increasingly recognized as a critical bridge linking local infection to systemic diseases. Growing evidence demonstrates that oral microbes not only enter the gut via saliva, bloodstream, and direct migration but also successfully colonize under specific microenvironmental conditions, reshaping the local microbial ecosystem. Periodontal pathogens such as P. gingivalis and F. nucleatum drive gastrointestinal tumorigenesis through chronic inflammation, metabolite production, and signaling pathway reprogramming. Moreover, oral-gut microbial interactions reveal canc

OBJECTIVE: The red-complex bacteria Porphyromonas gingivalis and Tannerella forsythia together with Fusobacterium nucleatum are essential players in periodontitis. This study investigated the bacterial interplay with human periodontal ligament mesenchymal stromal cells (hPDL-MSCs) which act in the acute phase of periodontal infection.
DESIGN: The capability of the bacteria to induce an inflammatory response as well as their viability, cellular adhesion and invasion were analyzed upon mono- and co-infections of hPDL-MSCs to delineate potential synergistic or antagonistic effects. The expressio…

Intratumoral pathogens can contribute to cancer progression and affect therapeutic response. Fusobacterium nucleatum, a core pathogen of colorectal cancer (CRC), is an important cause of low therapeutic efficacy and metastasis. Thus, the modulation of intratumoral pathogens may provide a target for cancer therapy and metastasis inhibition. Herein, we propose an intratumoral F. nucleatum-modulating strategy for enhancing the therapeutic efficacy of CRC and inhibiting lung metastasis by designing an antibacterial nanoplatform (Au@BSA-CuPpIX), which produced reactive oxygen species (ROS) under ul…

BACKGROUND/AIM: Despite evidence of an association between pulmonary diseases and periodontopathic bacteria, the molecular mechanisms remain unknown. Matrix metalloproteinase-9 (MMP9) plays important roles in pneumonia, chronic obstructive pulmonary disease, and asthma; therefore, we assessed the effects of Fusobacterium nucleatum on MMP9 expression in mouse lung and A549 human alveolar epithelial cells.
MATERIALS AND METHODS: Heat-killed F. nucleatum was administered to the trachea of mice or added to A549 cell cultures. MMP9 expression was determined using real-time PCR and western blotting…

There have been few studies on the detection of direct nitric oxide (NO) production and interferon-gamma (IFN-gamma) in vivo without using animal cell culture. We questioned whether NO and IFN-gamma could be produced at the site of infection. The peritoneal cavity of mice was used as the local infection model. NO and IFN-gamma in abdominal washings from these mice were measured directly at various times after injection of Fusobacterium nucleatum, a gram-negative rod periodontal pathogen. The mice were divided into three groups: those treated with live bacteria (LB), those treated with heat-kil…
