[Chronic motility disorders of the upper gastrointestinal tract in the elderly. Pharmaceutical, endoscopic and operative therapy].

Primary motility disorders of the upper gastrointestinal (GI) tract result from an impairment of the motor function of the esophagus, stomach, and duodenum by malfunction of the enteric nervous system or degeneration of the gastrointestinal muscle layer. Other forms of upper GI motility disorders occur secondary to underlying systemic diseases. The exact pathophysiology of the disturbances within the enteric nervous system of the upper GI tract is not yet clearly understood. For motility disorders resulting from systemic diseases the lack of knowledge with respect to the underlying pathomechan

The gut is not only the target but a source of inflammatory mediators inhibiting gastrointestinal motility during sepsis.

BACKGROUND: Sepsis is a common problem in intensive care patients leading to multi-organ failure and gastrointestinal paralysis. AIM: The aim of the study was to investigate whether the gastrointestinal tract is not only the target but also the source of inflammatory mediators inhibiting gastrointestinal motility. METHODS: Mesenteric lymph was obtained from rats in which a sepsis was induced by lipopolysaccharide (LPS) intraperitoneally. Gastrointestinal motility was recorded following mesenteric lymph- or TNFα infusion into the jugular vein of separate healthy recipient rats using the strai…

Gastrointestinal motility problems in patients with Parkinson's disease. Effects of antiparkinsonian treatment and guidelines for management.

Gastrointestinal (GI) motility disorders are frequent in patients with Parkinson's disease, manifesting mainly as dysphagia, disorders of gastric emptying and constipation. The most likely causes of these disorders are cerebral degeneration and degeneration of the myenteric plexus. Although the effect of antiparkinsonian medication is largely overestimated, it certainly has an influence and should be adapted accordingly in patients with GI motility disorders. In particular, anticholinergic drugs should be avoided, and anamnesis, clinical examination and, if necessary, diagnostic tests performe

Gastrointestinal motility in microgravity: a critical review of multi-level mechanisms and model-dependent effects.

BACKGROUND: Gastrointestinal motility disturbances rank among the most frequently reported medical complications of spaceflight. Astronauts experience delayed gastric emptying, erratic small intestinal transit and reduced colonic propulsion. The underlying mechanisms are multifactorial. Microgravity alters intra-abdominal physical mechanics, disrupts autonomic and enteric neural circuits, shifts gastrointestinal hormone secretion profiles, inflicts oxidative stress upon effector cells, and perturbs gut microbial communities. Cross-model comparisons reveal substantial disagreement, suggesting t…

Effects of optogenetic activation of the enteric nervous system on gastrointestinal motility in mouse small intestine.

BACKGROUND AND AIMS: Recently, it was demonstrated that optogenetics could be used to stimulate enteric calretinin neurons, leading to increased colonic transit in vitro and in vivo. The aim of the current study was to determine if similar approaches could be used to stimulate the isolated mouse small intestine, with the aim of potentially also improving transit in the small bowel. METHODS: Cre-Lox recombination was used to generate transgenic mice expressing the light-sensitive ion channel channelrhodopsin-2 (ChR2) in calretinin neurons. RESULTS: Spontaneous migrating motor complexes were r

Quantified small bowel motility in patients with ulcerative colitis and gastrointestinal symptoms: a pilot study.

BACKGROUND: Gastrointestinal (GI) symptoms are common in patients with ulcerative colitis (UC), even when the disease is in remission, possibly due to abnormalities in GI motility. Small bowel motility can be assessed globally and in specific intestinal regions during magnetic resonance enterography (MRE) using a displacement mapping technique. PURPOSE: To investigate whether small bowel motility in MRE differs between patients with UC and controls, and if altered motility correlates with GI symptoms. MATERIAL AND METHODS: In 2016-2018, patients who were admitted for MRE, regardless of clini

Impairment of Nrf2- and Nitrergic-Mediated Gastrointestinal Motility in an MPTP Mouse Model of Parkinson's Disease.

BACKGROUND: Gastrointestinal (GI) motility dysfunction is the most common non-motor symptom of Parkinson's disease (PD). Studies have indicated that GI motility functions are impaired before the onset of PD. AIMS: To investigate the underlying mechanism of PD-induced GI dysmotility in MPTP (1-methyl 4-phenyl 1,2,3,6-tetrahydropyridine)-induced animal model. METHODS: C57BL/6 mice were administered with or without a selective dopamine neurotoxin, MPTP, to induce parkinsonian symptoms. In addition to in vivo studies, in vitro experiments were also conducted in colon specimens using l-methyl-4-p

Effects of Serotonin and Slow-Release 5-Hydroxytryptophan on Gastrointestinal Motility in a Mouse Model of Depression.

BACKGROUND & AIMS: Mood disorders and constipation are often comorbid, yet their shared etiologies have rarely been explored. The neurotransmitter serotonin (5-HT) regulates central nervous system and enteric nervous system (ENS) development and long-term functions, including gastrointestinal (GI) motility and mood. Therefore, defects in neuron production of 5-HT might result in brain and intestinal dysfunction. Tryptophan hydroxylase 2 (TPH2) is the rate-limiting enzyme in 5-HT biosynthesis. A variant of TPH2 that encodes the R441H substitution (TPH2-R441H) was identified in individuals with

[Effects of Electroacupuncture on Gastrointestinal Motility and Expressions of VIP and CGRP in Functional Dyspepsia Model Rats].

Objective To observe the effects of electroacupuncture (EA) on vasoactive intesti- nal peptide (VIP) , calcitonin gene-related peptide (CGRP … tinal tract, accelerate gastric emptying rate and small intestinal transit rate. EA's improving the gastrointestinal motility might be related to decreasing mRNA expressions of VIP and CGRP in gastrointestinal tract, indicating that abnormal secretion braingut peptide might be one of important mechanisms for FD.

Motility of the gastrointestinal tract and gallbladder during long-term total parenteral nutrition in dogs.

BACKGROUND: The motility of the gastrointestinal tract during total parenteral nutrition (TPN) remains poorly understood. The objective of this study was to determine the motility pattern not only in the gastrointestinal tract but also in the gallbladders of dogs maintained by TPN. METHODS: Central venous catheters were inserted through the external jugular vein of 5 dogs and 6 strain gauge force transducers were sewn to the stomach, small intestine, and gallbladder. Two weeks later, oral food was discontinued and motility was recorded for 24 hours after the first migrating motor complex (MMC

Management of gastrointestinal motility disorders. A practical guide to drug selection and appropriate ancillary measures.

The focus of management of gastrointestinal motility disorders should be to improve survival and quality of life. Some motor disorders are annoying, but are compatible with virtually normal activity and carry no significant life risk. Conversely, other motor disorders are highly incapacitating and may shorten life expectancy because of complications and nutritional impairment. Management is based first on establishing the correct diagnosis and prognosis; secondly, on adjusting therapy to the severity of illness; and thirdly, on preventing significant complications. Simple recommendations on ap…

[Effects of smoking on motility of the upper gastrointestinal tract in healthy persons and patients with peptic ulcer].

59 male volunteers (16 healthy and 43 ulcer subjects) entered the study on effects of smoking on gastrointestinal motility. The latter was assessed within 12-50 hours by means of probes fitted with tensiometers. Smokers, especially heavy smokers, regardless of the disease presence, had more regular phase alterations in the cycles of the motility and more regular rhythm of phase III, more frequent involvement of the stomach in frontal motor activity. A direct effect of the cigarette smoked on gastric and intestinal motility was associated with condition of the patient and time after meal.

The gastrointestinal tract of the rock hyrax (Procavia habessinica). 1. Morphology and motility patterns of the tract.

The stomach of the rock hyrax (Procavia habessinica) is divided into a non glandular part with very slow movements, and a glandular part which rapidly mix the digesta. The large intestine has two fermentation chambers, the caecum, which rapidly mixes the digesta, and the colonic sac, which efficiently, but slowly, mixes digesta. Between these chambers runs the connecting colon. No retrograde transport is observed in any part of the large intestine.

Does disordered upper gastrointestinal motility predispose to Helicobacter pylori colonization of the stomach in children?

Helicobacter pylori is an important factor in the pathogenesis of chronic gastritis and gastroduodenal ulcer disease. However, the basic causal mechanisms of H pylori colonization on the gastric mucosa are still unclear. The authors evaluated the prevalence of H pylori colonization in 266 children who underwent upper gastrointestinal endoscopy during a 12-month period. The indications for endoscopy were follow-up of esophagitis related to gastroesophageal reflux (n = 17), suspicion of gastroesophageal reflux (n = 51), abdominal pain (n = 28), vomiting (n = 30), follow-up of esophageal atresia

Effects of corticotropin-releasing factor on plasma motilin and somatostatin levels and gastrointestinal motility in dogs.

The effects of intracerebroventricular and intravenous administration of corticotropin-releasing factor (CRF) on gastrointestinal motility, motilin-induced gastric motor response, and plasma motilin and somatostatin levels were investigated in fasted dogs chronically prepared with strain gauge transducers on the antrum and proximal jejunum. Administered intracerebroventricularly at doses of 20 and 100 ng/kg in fasted dogs, CRF suppressed for 4-5 h the gastric cyclic migrating motor complex. A similar dose (100 ng/kg) administered intravenously was inactive. Corticotropin-releasing factor admin…

Does salmon calcitonin influence the motility of the human gastrointestinal tract? An electromanometric and endoscopic study.

The effect of salmon calcitonin--as a relatively high dose of 25 mug by single iv. injection--upon the basal and metoclopramide-stimulated motility phenomena of the stomach and proximal duodenum as well as upon the unstimulated colon and rectosigmoid was evaluated in 12 subjects without gastrointestinal disease by quantitative electromanometric measurements performed for one hour with the aid of fluid-filled micro-balloon-catheter-systems. Additionally, in 9 subjects the reaction of the cardia, stomach, pylorus and proximal duodenum upon the same dose of the hormone was observed endoscopically

Dup15q model mice exhibit impaired gastrointestinal motility and a constipation-like phenotype, alleviated by prucalopride.

Chromosome 15q duplication syndrome (Dup15q) is a neurodevelopmental disorder linked to autism spectrum disorder (ASD), involving increased copies of the 15q11.2-q13 region. About 80% of individuals with Dup15q experience gastrointestinal (GI) dysfunction, including constipation. The duplicated region encodes GABA receptor A subunits, affecting GABAergic signalling, while reduced serotonin (5-HT) levels impair neuronal activity and social behaviour in a mouse model of Dup15q (15q dup). Given that both GABA and serotonin are expressed in the enteric nervous system (ENS), this study investigates
