Gene expression profiling in porocarcinoma indicates heterogeneous tumor development and substantiates poromas as precursor lesions.

BACKGROUND AND OBJECTIVES: Malignant sweat gland tumors are rare, with the most common being eccrine porocarcinoma (EP). Approximately 18% of benign eccrine poroma (EPO) transit to EP. Previous research has provided first insights into the mutational landscape of EP. However, only few studies have performed gene expression analyses. This leaves a gap in the understanding of EP biology and potential drivers of malignant transformation from EPO to EP. METHODS: Transcriptome profiling of 23 samples of primary EP and normal skin (NS). Findings from the EP samples were then tested in 17 samples of

Multiomic analysis of familial adenomatous polyposis reveals molecular pathways associated with early tumorigenesis.

Familial adenomatous polyposis (FAP) is a genetic disease causing hundreds of premalignant polyps in affected persons and is an ideal model to study transitions of early precancer states to colorectal cancer (CRC). We performed deep multiomic profiling of 93 samples, including normal mucosa, benign polyps and dysplastic polyps, from six persons with FAP. Transcriptomic, proteomic, metabolomic and lipidomic analyses revealed a dynamic choreography of thousands of molecular and cellular events that occur during precancerous transitions toward cancer formation. These involve processes such as cel

A BAC transgenic reporter recapitulates in vivo regulation of human telomerase reverse transcriptase in development and tumorigenesis.

Telomerase is tightly regulated in humans relative to mice, owing to the differential regulation of TERT genes. To explore hTERT regulation in vivo, we engineered mice with a 160-kb transgenic bacterial artificial chromosome (BAC) spanning the hTERT locus with a Renilla luciferase (Rluc) cassette downstream of its promoter. Analysis of multiple founder lines revealed that the Rluc expression profile from the transgenic hTERT reporter locus reproduced that of the native hTERT gene in all tissues and organs examined, demonstrating that genetic sequence determined the species-specific development

Topology-Based Biomarkers Accurately Predict Breast Cancer Outcome and Survival.

UNLABELLED: Loss of organized structure is a hallmark of malignant transformation in breast cancer. Traditionally, such morphologic features are captured by descriptive histologic assessments, such as grade, that represent reliable diagnostic and prognostic determinants. Nonetheless, the predictive value of these semiquantitative approaches is limited by their subjective nature and the computational restrictions inherent to discrete integer-based scoring systems. In this study, we describe an application of topological measurements and statistical modeling to derive continuous mathematical sco

Integrative transcriptomic and CRISPR dependency analysis identifies hepatoblastoma-specific essential genes and actionable vulnerabilities.

BACKGROUND: Hepatoblastoma (HB) is the most common primary liver malignancy in childhood, yet its molecular determinants, functional dependencies, and therapeutic vulnerabilities remain incompletely characterized. Integrative analyses combining transcriptomic profiling with functional genomic datasets provide a strategy to identify essential genes, biomarkers predictive of tumor behavior and treatment response. METHODS: Differential expression analysis comparing HB tumors with normal liver was processed on training cohort. These genes were integrated with DepMap CRISPR-Cas9 dependency scores

LINC00265 is significantly upregulated in breast carcinoma: expression, correlation networks, and potential prognostic value.

BACKGROUND: Several signaling pathways and related transcripts have been found to be dysregulated in breast cancer. Methods and Results: In this study, we used an in silico step aiming to find the most relevant biological pathways and related long non-coding RNAs (lncRNAs) in the pathoetiology of breast cancer. This step led to identification of EGFR and its related lncRNAs as contributors to the pathology of this type of cancer. Then, we assessed expression of these transcripts in breast cancer samples and their adjacent non-cancerous tissues (ANCTs). Among the assessed transcripts, LINC00265

Nuclear factor I/B: Duality in action in cancer pathophysiology.

The nuclear factor I (NFI) family of transcription factors plays a decisive role in organ development and maturation. Their deregulation has been linked with various diseases, most notably cancer. NFIB stands apart from the other NFI family members given its unique ability to drive both tumor suppressive and oncogenic programs. Thus, the ultimate impact of deregulated NFIB signaling is cancer-specific and strongly influenced by an intricate network of upstream regulators and downstream effectors. Deciphering the events that drive NFIB's paradoxical roles within these networks will enable us to

A concise review on miRNAs as regulators of colon cancer stem cells and associated signalling pathways.

Despite recent therapy advances and a better understanding of colon cancer biology, it remains one of the major causes of death. The cancer stem cells, associated with the progression, metastasis, and recurrence of colon cancer, play a major role in promoting the development of tumour and are found to be chemo resistant. The stroma of the tumour, which makes up the bulk of the tumour mass, is composed of the tumour microenvironment. With the advent of theranostic and the development of personalised medicine, miRNAs are becoming increasingly important in the context of colon malignancies. A hol

Aberrant Regulation of mRNA m⁶A Modification in Cancer Development.

N⁶-methyladenosine (m⁶A) is the most prevalent internal modification of eukaryotic messenger RNAs (mRNAs). The m⁶A modification in RNA can be catalyzed by methyltransferases, or removed by demethylases, which are termed m⁶A writers and erasers, respectively. Selective recognition and binding by distinct m⁶A reader proteins lead mRNA to divergent destinies. m⁶A has been reported to influence almost every stage of mRNA metabolism and to regulate multiple biological processes. Accumulating evidence strongly supports the correlation between aberrant cellular m⁶A level and cancer. We summarize here

Upregulated CircRNAs With Efficacy in Preclinical In Vivo Models Reveal New Targets for Therapeutic Intervention in Glioma.

There is a significant need to improve treatment efficacy for patients with glioma. Therefore, we have analyzed the literature for the role of circular RNAs (circRNAs) with respect to selected pathology-related topics of glioma. We have focused on upregulated circRNAs with efficacy in preclinical in vivo models. The identified circRNAs cover the following topics: Temozolomide (TMZ) resistance, transmembrane and secreted proteins, wingless-related integration site (WNT) signaling, cytoskeleton dynamics, cell-cell and cell-extracellular matrix interactions, as well as circRNAs with additional fu

The role of the WD40-repeat protein family in cancer.

WD40-repeat (WDR) proteins constitute one of the largest and most functionally diverse scaffold families in eukaryotes. By folding tandem WD repeats into β-propeller domains, they provide modular interaction surfaces that assemble multi-protein complexes governing transcription and epigenetic control, ubiquitin-dependent proteostasis, RNA metabolism, and cell-cycle progression. Despite their pervasive involvement in oncogenic signaling and hallmark cancer phenotypes, the cancer field still lacks an integrated framework that connects WDR structural logic to context-dependent mechanisms and, cri

miR‑324‑5p in oncology: Molecular mechanisms and clinical outlook (Review).

BACKGROUND: Batracylin is a heterocyclic arylamine topoisomerase inhibitor with preclinical anticancer activity. Marked species differences in sensitivity to the toxicity of batracylin were observed and attributed to differential formation of N-acetylbatracylin by N-acetyltransferase. A Phase I trial of batracylin in cancer patients with slow acetylator genotypes identified a dose-limiting toxicity of hemorrhagic cystitis. To further explore the metabolism of batracylin and N-acetylbatracylin across species, detailed studies using human, rat, and dog liver microsomal and hepatocyte preparation

The role and mechanism of METTL3 in cancer: emerging insights into m6A methylation and therapeutic potential.

N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, plays a crucial role in regulating RNA metabolism and gene expression. Among m6A regulators, METTL3 functions as the catalytic core of the m6A methyltransferase complex and has emerged as a critical epitranscriptomic modulator in cancer. An expanding body of evidence demonstrates that METTL3 is aberrantly expressed in various malignancies, contributing to tumor progression, metastasis, stemness, immune evasion, and resistance to chemotherapy and targeted therapies. Nonetheless, its role remains highly context

Molecular Alterations and Pathways in Intrahepatic Cholangiocarcinoma: Available Evidence and New Perspectives.

Intrahepatic cholangiocarcinoma (iCCA) is an aggressive cancer arising within the liver from the bile ducts, and it is characterized by limited therapeutic options and a poor prognosis. This neoplasm exhibits both high intra-tumor and inter-tumor heterogeneity and many oncogenic and tumor suppressor genes are involved in its development and progression. Here, we summarize the major pathways and driver genes involved in the genesis and progression of iCCA, with a special look at their potential therapeutic values. We approach not only the well-known FGFR, IDH and HER2 alterations but also delve

"Friends or foes": a new perspective of tumour metabolic transcriptional modification.

Energy metabolism plays a pivotal role in cancer clinical treatment and has become an important means of clinical diagnosis of tumour progression. However, current research mostly focuses on changes in metabolic products and neglects the deeper mechanisms of transcriptional regulation. This paper proposes a new perspective, establishing a comprehensive network that reveals the interaction between metabolism and transcription, which explores how tumour metabolism affects tumour progression through transcriptional modifications, and provides a novel approach for optimizing tumour treatment strat

Gas1-Mediated Suppression of Hepatoblastoma Tumorigenesis.

Hepatoblastoma (HB), the most common pediatric liver cancer, is associated with dysregulated Wnt/β-catenin, Hippo, and/or nuclear factor erythroid 2 ligand 2/nuclear respiratory factor 2 (NFE2L2/NRF2) pathways. In mice, pairwise combinations of oncogenically active forms of the terminal transcription factors of these pathways, namely, β-catenin (B), Yes-associated protein (YAP; Y), and Nrf2 (N), generate HBs, with the triple combination (B + Y + N) being particularly potent. Each tumor group alters the expression of thousands of B-, Y-, and N-driven unique and common target genes. The identifi

Aberrant DNA Methylation as a Biomarker and a Therapeutic Target of Cholangiocarcinoma.

Cholangiocarcinoma is an epithelial malignancy arising in the region between the intrahepatic bile ducts and the ampulla of Vater at the distal end of the common bile duct. The effect of current chemotherapy regimens against cholangiocarcinoma is limited, and the prognosis of patients with cholangiocarcinoma is poor. Aberrant DNA methylation and histone modification induce silencing of tumor suppressor genes and chromosomal instability during carcinogenesis. Studies have shown that the tumor suppressor genes and microRNAs (miRNAs) including MLH1, p14, p16, death-associated protein kinase (DAPK
