Hematopoietic progenitors in cyclic neutropenia: effect of granulocyte colony-stimulating factor in vivo.

The number and growth factor requirements of committed progenitor cells (colony-forming units-granulocyte/macrophage and burst-forming units-erythroid) in three patients with cyclic neutropenia (two congenital, one acquired) were studied before and during therapy with recombinant human granulocyte colony-stimulating factor (G-CSF; 3 to 10 micrograms/kg/d). When the patients with congenital disease were treated with G-CSF, the cycling of blood cells persisted, but the cycle length was shortened from 21 days to 14 days, and the amplitude of variations in blood counts increased. There was a paral…

Treatment of refractory aplastic anemia with recombinant human granulocyte-macrophage-colony-stimulating factor.

Fifteen patients with refractory aplastic anemia or agranulocytosis received treatment with recombinant human granulocyte-macrophage-colony-stimulating factor (rhGM-CSF) in doses from 4 to 64 micrograms/kg/d by continuous intravenous (IV) infusion. Ten of 11 evaluable patients with aplastic anemia had substantial increments in granulocytes, monocytes, and eosinophils associated with myeloid and eosinophilic hyperplasia in the bone marrow. Patients with pretreatment granulocytes greater than 0.3 x 10(9)/L had greater increments in circulating myeloid cells than patients with more severe granulo

Recombinant granulocyte/macrophage-colony stimulating factor in human and murine bone marrow transplantation.

Recombinant granulocyte/macrophage-colony stimulating factor (rGM-CSF) is a protein that mediates a broad range of biological activities including inducing proliferation of bone marrow progenitor cells and up-regulating the functional capabilities of neutrophils, monocytes, and macrophages. We have administered rGM-CSF in vivo in the context of allogeneic (murine) and autologous (human) bone marrow transplantation (BMT). In a murine allogeneic model of T-cell depletion constructed to favor graft failure/rejection of donor marrow, ex vivo treatment of marrow with rGM-CSF significantly promoted

Use of recombinant granulocyte-macrophage colony stimulating factor in the Brazil radiation accident.

8 patients with bone marrow failure after a caesium-137 radiation accident were treated with recombinant human granulocyte-macrophage colony stimulating factor (rHuGM-CSF). The 7 who were evaluable had prompt increases in granulocytes and bone marrow cellularity. 2 patients died of radiation toxicity and haemorrhage and 2 of bacterial sepsis acquired before the start of rHuGM-CSF treatment. 4 patients survive, including 2 who were treated early and never became infected. This therapeutic approach to radiation-induced granulocytopenia may therefore be useful after radiation and nuclear accident

The effects of ethanol, tumor necrosis factor, and granulocyte colony-stimulating factor on lung antibacterial defenses.

In summary, evidence is emerging indicating that alcohol-abusing hosts are seriously undermined by profound disturbances in cytokine production and activity. These alterations likely play a critical role in the development and clinical sequelae of their immunosuppressed status. Recombinant technology currently provides the clinician with the potential to immunologically reconstitute and restore host defenses in these hosts. While the ultimate role of these agents in patients will require extensive clinical investigations into their multiple biologic effects and interactions, cytokines, when pr

Proinflammatory monocyte-derived granulocyte-macrophage colony-stimulating factor fuels airway inflammation in bronchopulmonary dysplasia.

BACKGROUND: Bronchopulmonary dysplasia (BPD) remains a leading cause of morbidity and long-term respiratory complications in preterm neonates. Effective strategies for moderate-severe BPD are currently lacking. METHODS: Tracheal aspirate (TA) samples were collected from 62 preterm neonates, and the immune cell composition was analyzed using multicolor flow cytometry. Subsequently, CD14+ monocytes were isolated and purified from the TA samples for transcriptomic sequencing analysis. Cytokine arrays and enzyme-linked immunosorbent assays were utilized in the cytokine level detection from TA sam

Protective effect of granulocyte colony stimulating factor on cyclophosphamide-induced prostate injury in rats.

Cyclophosphamide (CPA) is a commonly used chemotherapeutic agent whose metabolites readily accumulate in male reproductive organs such as the testes and epididymis, exhibiting reproductive toxicity. Granulocyte colony-stimulating factor (G-CSF) possesses anti-inflammatory and antioxidant effects, but its protective mechanism against CPA-induced prostate injury remains unclear. This study aimed to investigate the protective effect of G-CSF against CPA-induced prostate injury in rats. Thirty SPF male Sprague-Dawley (SD) rats were randomly divided into a control group (C group), a CPA model group…

Granulocyte colony-stimulating factor (G-CSF)-induced aortitis in a woman with breast cancer.

Granulocyte colony-stimulating factor (G-CSF) is used alongside cytotoxic chemotherapy regimens to reduce the severity and duration of neutropenia and decrease infection risk. Here, we add to the growing body of evidence that aortitis can be considered a recognised side effect of G-CSF therapy.A woman in her 60s with breast carcinoma treated with G-CSF cover alongside adjuvant chemotherapy presented with intermittent fever and epigastric pain. There were no localising signs of infection. Results demonstrated raised inflammatory markers. The patient did not respond to antibiotics, and imaging s…

Sarcopenia attenuates the efficacy of PEGylated granulocyte colony-stimulating factor in preventing febrile neutropenia.

PURPOSE: The aim of this study was to develop a predictive model integrating sarcopenia, PEGylated granulocyte colony-stimulating factor (PEG-G-CSF) administration, and conventional risk factors to predict febrile neutropenia (FN) and grade 4 neutropenia (G4 NP) in patients with diffuse large B-cell lymphoma (DLBCL) treated with R-CHOP. METHODS: A retrospective cohort of 305 patients was analyzed. FN and G4 NP incidence rates after the first cycle were assessed along with predictors of these toxicities using multivariable logistic regression. Nomograms were developed and internally validated …

[A Case of Granulocyte-Colony Stimulating Factor-Producing Undifferentiated Sarcoma of the Transverse Colon].

We report a case of granulocyte-colony stimulating factor(G-CSF)-producing undifferentiated sarcoma of the transverse colon. A 80-year-old woman was referred to our hematology department after visiting her primary physician with chief complaints of decreased appetite and fatigue. An elevated white blood cell count was observed during examination. Further laboratory tests at our hospital revealed an elevated white blood cell count of 95,800/μL and a plasma G-CSF level of 895 pg/mL. The patient was referred to our department after a tumor lesion was found in the transverse colon during computed

Granulocyte colony-stimulating factor with or without immunosuppression reduction in neutropenic kidney transplant recipients.

INTRODUCTION: Neutropenia post-kidney transplantation is associated with adverse graft and patient outcomes. We aimed to analyze the effect of granulocyte colony-stimulating factor (G-CSF) use with and without immunosuppression reduction on graft outcomes in neutropenic recipients. METHODS: In this retrospective cohort study, we identified 120 recipients with neutropenia, within the first-year post-transplant. RESULTS: Of these, 45.0% underwent no intervention, 17.5% had immunosuppression reduced, 18.3% were only given G-CSF, and 19.2% had both interventions. Overall, 61 patients experienced…

Granulocyte Macrophage Colony-Stimulating Factor-Specific Autoantibodies and Cerebral Nocardia With Pulmonary Alveolar Proteinosis.

In this study, we report the history of a 40-year-old man with a primary cerebral abscess caused by Nocardia abscessus that led to the discovery of autoimmune pulmonary alveolar lipoproteinosis (anti-granulocyte-macrophage colony-stimulating factor [GM-CSF] autoantibodies). Anti-GM-CSF autoantibodies promote immunodeficiency and should be monitored to prevent opportunistic and disseminated infections and to diagnose asymptomatic pulmonary alveolar lipoproteinosis.

Safety of recombinant human granulocyte-macrophage colony-stimulating factor in healing pediatric severe burns.

We explored the safety of recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF) for healing burns in children. Subjects were randomly assigned to two groups: the experimental group received external rhGM-CSF gel, and the control group received rhGM-CSF gel matrix components, applied to the burn surface. Neither group was given any other drugs that promote wound healing. Each day we recorded the pulse, body temperature, and respiration status in the two groups. We detected the blood routine, urine routine, and hepatic and renal function before the patients received drug

Osteoclasts are dispensable for hematopoietic progenitor mobilization by granulocyte colony-stimulating factor in mice.

The contribution of osteoclasts to hematopoietic stem/progenitor cell (HSPC) retention in the bone marrow is controversial. Studies of HSPC trafficking in osteoclast-deficient mice are limited by osteopetrosis. Here, we employed two non-osteopetrotic mouse models to assess the contribution of osteoclasts to basal and granulocyte colony-stimulating factor (G-CSF)-induced HSPC mobilization. We generated Rank(-/-) fetal liver chimeras using Csf3r(-/-) recipients to produce mice lacking G-CSF receptor expression in osteoclasts. Basal and G-CSF-induced HSPC mobilization was normal in these chimeras…
