Subcortical gray matter N-acetylaspartate reduction in two cases of vascular dementia.

Many of the previous studies of vascular dementia using proton magnetic resonance (MR) spectroscopy had been carried out on white matter. However, no proton spectroscopic data of the subcortical gray matter are available in such disease. We report two cases suffering from vascular dementia, with an unilateral N-acetylaspartate (NAA) decrease on subcortical gray matter. This significant reduction in NAA ratios was associated with an increase of choline on the ipsilateral centrum semiovale. We discuss the pathophysiology of these cases.

Reduced cortical gray matter in narcolepsy: preliminary findings with voxel-based morphometry.

There is a selective loss of hypocretin/orexin-containing hypothalamic neurons in patients with narcolepsy. The authors compared MRI-derived gray matter maps of 12 patients with narcolepsy with matched controls using voxel-based morphometry to ascertain whether there are other structural brain abnormalities. Patients with narcolepsy showed bilateral cortical gray matter reductions predominantly in inferior temporal and inferior frontal brain regions. Relative global gray matter loss was independent of disease duration or medication history. No significant subcortical gray matter alterations we…

Morphological alteration of temporal lobe gray matter in dyslexia: an MRI study.

Functional imaging studies of developmental dyslexia have reported reduced task-related neural activity in the temporal and inferior parieta … analysis of tissue composition revealed that this reduction was primarily attributable to decreased gray matter within the left temporal lobe (p < .002). Further segmentation of the temporal lobe showed that this reduction was not confined to the superior temporal gyrus, the primary location of primary auditory cortex. Reductions of temporal lobe gray matter …

Association of heterotopic gray matter with seizures: MR imaging. Work in progress.

Heterotopic gray matter, which previously had been associated with severe congenital malformations of the brain and developmental delay, was found without these associated conditions. The authors found ten cases of heterotopic gray matter on magnetic resonance (MR) images. The lesions had a signal intensity that was isointense compared with that of gray matter on T1, spin-density, and T2-weighted images. Nine of the ten cases were associated with a seizure disorder. The tenth case, discovered during a workup for metastatic lung disease, was confirmed with pathologic studies. Heterotopic gray m…

Structural abnormalities in gray matter, white matter, and cerebrospinal fluid in parkinson's disease: a voxel-based morphometry study.

OBJECTIVE: Parkinson's disease (PD) is a neurodegenerative disorder affecting multiple brain systems, yet whole-brain structural changes remain incompletely characterized. We used voxel-based morphometry (VBM) to systematically quantify gray matter (GM), white matter (WM), and cerebrospinal fluid (CSF) volume alterations in PD and explore their potential clinical significance for deep brain stimulation (DBS). METHODS: High-resolution T1-weighted MRI scans were acquired from 200 PD patients and 176 age- and sex-matched healthy controls. VBM analysis was performed using SPM12/CAT12, with age, s…

Gray matter and white matter functional connectivity changes induced by rTMS concurrent with cognitive training in Alzheimer's disease.

BACKGROUND AND PURPOSE: Primarily by targeting the gray matter (GM), repetitive transcranial magnetic stimulation (rTMS) has shown promise in improving cognitive function in individuals with Alzheimer's disease (AD). However, the impact of rTMS on white matter (WM) remains poorly understood. This study aimed to investigate the functional connectivity (FC) changes in both GM and WM induced by rTMS, and explore their relationship with the clinical manifestation of the disease. METHODS: Sixteen patients with mild to moderate AD were enrolled and randomly assigned to either the real rTMS group (n…

Menstrual Cycle Variations in Gray Matter Volume, White Matter Volume and Functional Connectivity: Critical Impact on Parietal Lobe.

The role of gonadal hormones in neural plasticity remains unclear. This study aimed to examine the effects of naturally fluctuating hormone levels over the menstrual cycle in healthy females. Gray matter, functional connectivity (FC) and white matter changes over the cycle were assessed by using functional magnetic resonance imaging (fMRI), resting state fMRI, and structural MRIs, respectively, and associated with serum gonadal hormone levels. Moreover, electrocutaneous sensitivity was evaluated in 14 women in four phases of their menstrual cycle (menstrual, follicular, ovulatory, and luteal).…

Network-based mapping and neurotransmitter architecture of gray matter correlates of neuroticism.

OBJECTIVES: Although neuroticism is a major risk factor for adverse health outcomes, its neural basis is obscured by inconsistent findings from studies of regional gray matter volume (GMV) correlates. This study sought to identify convergent functional brain networks underlying these heterogeneous GMV correlates using functional connectivity network mapping (FCNM), and to explore their neurochemical basis. METHODS: We systematically identified 10 voxel-based morphometry (VBM) studies (N = 1,595) reporting neuroticism-associated GMV coordinates. Using resting-state fMRI data from 1,093 healthy…

Transcriptomic signatures of gray matter volume loss in cerebral small vessel disease.

BACKGROUND: White matter hyperintensities (WMHs) are a hallmark imaging sign of cerebral small vessel disease (CSVD) and are closely linked to motor and cognitive deficits. Using voxel‑based morphometry (VBM) together with spatial transcriptomics, this study explored the altered molecular pathways underlying the reduction in gray matter volume (GMV) associated with WMHs. METHODS: We included 25 patients with WMHs and 26 healthy controls. The VBM analysis was used to identify brain regions with reduced GMV. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment…

Estradiol, Emotion Regulation, and the Limbic System: Effects on Gray Matter Volume.

BACKGROUND: Mastering emotion regulation is crucial for social skills and mental health. Hormonal fluctuations, particularly in estradiol (E2) levels across the menstrual cycle, significantly impact emotion processing. E2 is known to influence emotion regulation, mental health, and the plasticity of limbic and striatal regions, which are involved in emotion processing and are rich in E2 receptors. Although research indicates that E2 levels may impact gray matter volume (GMV) of limbic and striatal areas, sufficient causal evidence is missing so far. Furthermore, because of the additional fluct…

Convergent imaging and genetic signatures of gray matter atrophy in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by widespread structural brain alterations, yet the specific patterns of brain atrophy and their underlying genetic mechanisms remain incompletely understood. Here, we integrated large-scale neuroimaging meta-analysis with population-scale imaging genetics to systematically characterize the genetic architecture linking gray matter volume (GMV) abnormalities to PD. We first performed a meta-analysis of structural MRI studies comprising 3212 patients with PD and 2056 controls, identifying robust patterns of GMV re…

Gray matter abnormalities and memory impairment in left mesial temporal lobe epilepsy.

OBJECTIVES: Mesial temporal lobe epilepsy (MTLE) is a common neurological disorder, with memory impairment being one of its typical symptoms. Most previous studies have focused on assessing declarative memory directly related to hippocampal functions, but emerging data suggest a decline in the efficiency of other types of memory as well. The aim of this study was to comprehensively assess various types of memory and analyze the relationship between memory performance and the volume of selected gray matter structures. METHODS: In total, 21 patients with left-MTLE and 28 age- and education-matc…

Long-term evolution of white and gray matter structural properties in migraine.

OBJECTIVE: To elucidate the specific brain changes linked to clinical diagnoses and distinct temporal progression in migraine. BACKGROUND: Gray (GM) and white matter (WM) differences were previously identified in chronic migraine (CM) compared to episodic migraine (EM). Regarding GM, patients with CM showed increased cortical thickness in the inferior temporal gyrus, and reduced surface area in the precuneus cortex, superior frontal and temporal gyri, and supramarginal gyrus. In the WM, widespread reduced axial and mean diffusivity have been observed in patients with CM in tracts such as the

Individualized gray matter morphological abnormalities unveil two neuroanatomical internet gaming disorder subtypes.

BACKGROUND: High heterogeneity among individuals with internet gaming disorder (IGDs) hampers the discovery of neuroimaging findings. To address this heterogeneity, this study adopted the newly proposed approach based on individualized gray matter morphological differences to objectively characterize neuroanatomical subtypes of IGDs. METHODS: 92 IGDs and 56 demographically matched healthy controls (HCs) were recruited for structural imaging scans and clinical tests. Heterogeneity through discriminative analysis identifies IGD subtypes through individual local GMV aberrances. Voxel-wise volume…

Disentangling False Memories: Gray Matter Correlates of Memory Sensitivity and Decision Bias.

Human memory is inherently susceptible to errors, including the formation of false memories-instances where individuals mistakenly recall information they were never exposed to. While prior research has largely focused on neural activity associated with false memory, the structural brain correlates of this phenomenon remain relatively unexplored. This study bridges that gap by investigating gray matter structure as it relates to individual differences in false memory performance. Using publicly available magnetic resonance imaging datasets, we analyzed cortical thickness (CT) in neural regions…
