Regulation of the L1 cell adhesion molecule by thyroid hormone in the developing brain.

Thyroid hormone is essential for brain maturation, regulating neuronal differentiation and migration, myelination, and synaptogenesis. Mutations in the cell adhesion molecule L1 cause severe neurological abnormalities in humans. We studied the effect of thyroid hormone deprivation and administration on L1 expression. Northern and in situ hybridization studies showed that hypothyroidism induces a marked increase in L1 mRNA levels in the caudate putamen, cerebral cortex, amygdala, and some thalamic nuclei. L1 protein was overexpressed in embryonic and newborn hypothyroid rats in the caudate puta

Effects of diazepam and ketamine administered individually or in combination on regional rates of glucose utilization in rat brain.

The effects of diazepam, which acts at GABAA receptors to enhance the effects of GABA, and ketamine, a non-competitive N-methyl-D-aspartate receptor antagonist, on local rates of cerebral glucose utilization (ICMRglc) were examined in unrestrained rats. Four groups were studied: vehicle-injected controls; and ketamine-treated, diazepam-treated and combined ketamine- and diazepam-treated animals. Ketamine alone produced a heterogeneous pattern of changes in ICMRglc (e.g. significant increases in the corpus callosum, olfactory tubercle and the entire Papez circuit, in addition to other limbic ar

Distribution of substance P-like immunoreactivity in the brain of the newt (Triturus cristatus).

The distribution of immunoreactive substance P (sP)-containing structures in the newt brain and spinal cord was explored with an indirect immunofluorescence method. Five sP-positive elements were detected: perikarya, dots, fibers, pericellular appositions, and pipe-shaped structures. Perikarya were seen at the levels of the spinal ganglia, spinal cord, raphe nucleus, interpeduncular nucleus, mesencephalon, preoptic area, infundibulum, dorsocaudal part of the ventral hypothalamus, habenula, and corpus striatum. Pericellular terminals were observed in periventricular areas, known to be rich in c

Deep brain stimulation mechanisms in comorbid pain and emotional dysregulation from animal models.

Deep brain stimulation (DBS) is a potent neuromodulatory intervention for chronic pain and comorbid affective disorders. This review synthesizes preclinical evidence detailing the neural circuits and molecular cascades engaged by DBS in models of pain and emotional dysregulation. We delineate how DBS-targeting regions like the prefrontal cortex (PFC), thalamus, and nucleus accumbens (NAc) recruit the prefrontal-midbrain-spinal axis to facilitate endogenous opioid release and modulate neurotransmission, thereby attenuating nociception. Concurrently, stimulating the ventromedial PFC (vmPFC) or l

Pituitary adenylate cyclase-activating polypeptide-immunoreactive (PACAP-IR) nerve fibers in the pig pineal gland.

The present study demonstrates the occurrence of PACAP-immunoreactive (PACAP-IR) nerve fibers in different compartments of the pig pineal gland, including glandular capsule (where they form a very dense network) and subependymal tissue close to the pineal recess (moderate to dense meshwork of varicose fibers). Furthermore, several varicose fibers penetrate from the capsule into the connective tissue septa and then into the parenchyma, where they form unequally distributed, fine network and, in some cases, basket-like structures around pinealocytes. Some of the PACAP-IR nerve fibers, observed b

Stepwise engagement of cingulate ensembles underlies motivational decline in chronic pain.

Chronic pain can progress from sensory hypersensitivity to impaired motivation, but the underlying circuit mechanisms remain unclear. Using whole-brain activity-dependent tagging, projection-specific perturbations, in vivo Ca2+ imaging, and single-unit recordings in male mice, we identify stage-dependent engagement of anterior cingulate cortex (ACC) circuits during neuropathic pain. ACC neurons projecting to the basolateral amygdala (BLA) are preferentially engaged early, respond to allodynic stimulation, and contribute to mechanical hypersensitivity, whereas neurons projecting to the lateral

A dual-pathway dysregulation in prefrontal-habenular circuits mediates stress susceptibility.

Chronic stress disrupts emotional homeostasis, leading to resilience or susceptibility through unclear mechanisms. Here, we identify a dual-pathway mechanism in the dorsomedial prefrontal cortex (dmPFC)-lateral habenula (LHb) circuit in male mice, where glutamatergic and GABAergic projections coordinately regulate stress responses. Chronic stress differentially modulates these pathways, inducing susceptibility-specific hyperactivity in dmPFC-innervated LHb neurons by potentiating excitatory and suppressing inhibitory inputs, disrupting long-range excitatory/inhibitory (E/I) balance. Spatial transcriptomics reveals modulation of transcriptional networks across brain regions after auditory threat conditioning.

Prior research has demonstrated genome-wide transcriptional changes related to fear and anxiety across species, often focusing on individual brain regions or cell types. However, the extent of gene expression differences across brain regions and how these changes interact at the level of transcriptional connectivity remains unclear. To address this, we performed spatial transcriptomics RNAseq analyses in an auditory threat conditioning paradigm in mice. We generated a spatial transcriptomic atlas of a coronal mouse brain section covering cortical and subcortical regions, corresponding to histo

Nicotine addiction: More than just dopamine.

Despite decades of research and anti-tobacco messaging, nicotine addiction remains an important public health problem leading to hundreds of thousands of deaths each year. While fundamental studies have identified molecular, circuit-level and behavioral mechanisms important for nicotine reinforcement and withdrawal, recent studies have identified additional pathways that are important for both nicotine seeking and aversion. In particular, although dopaminergic mechanisms are necessary for nicotine-dependent reward and drug-seeking, novel glutamate and GABA signaling mechanisms in the mesolimbi

Circuits regulating pleasure and happiness - focus on potential biomarkers for circuitry including the habenuloid complex.

INTRODUCTION: The multiplicity and complexity of the neuronal connections in the central nervous system make it difficult to disentangle circuits that play an essential role in the development or treatment of (neuro)psychiatric disorders. By choosing the evolutionary development of the forebrain as a starting point, a certain order in the connections can be created. The dorsal diencephalic connection (DDC) system can be applied for the development of biomarkers that can predict treatment response. MATERIALS AND METHODS: After providing a brief introduction to the theory, we examined neuroanat

Comparative immunohistochemical distribution of amylin-like and calcitonin gene related peptide like immunoreactivity in the rat central nervous system.

Using the indirect immunofluorescence method with a polyclonal antiserum raised in rabbits and directed against amylin (AMY), we have investigated the distribution of AMY-like immunoreactivity (-ir) throughout the central nervous system of the rat. The widespread distribution of AMY-ir was much more abundant than that previously reported for calcitonin gene related peptide (CGRP) immunoreactivity. In most brain areas there was no overlap between AMY- and CGRP-ir cell body groupings, with the exception of the motor nuclei of the hindbrain and spinal cord, which were found to contain large numbe

Distribution of choline acetyltransferase immunoreactivity in the pigeon brain.

We have investigated the distribution of cholinergic perikarya and fibers in the brain of the pigeon (Columba livia). With this aim, pigeon brain sections were processed immunohistochemically by using an antiserum specific for chicken choline acetyltransferase. Our results show cholinergic neurons in the pigeon basal telencephalon, the hypothalamus, the habenula, the pretectum, the midbrain tectum, the dorsal isthmus, the isthmic tegmentum, and the cranial nerve motor nuclei. Cholinergic fibers were prominent in the dorsal telencephalon, the striatum, the thalamus, the tectum, and the interpedu

Region-specific neural activity changes and hierarchical instability following social isolation.

Social hierarchy contributes to maintaining group stability and coordination; yet, how social isolation affects hierarchical dynamics remains poorly understood. Although social isolation is a potent psychosocial stressor, its influence on social rank, stability, and the underlying neural mechanisms has not been clearly delineated. We established two complementary tube-test-based behavioral paradigms to investigate how isolation modulates rank status, social stability, and competitive behaviors: a rank change model assessing pre- and post-isolation rank shifts and a daily rank-trace model captu

A lateral hypothalamic region supporting diverse visual processing and modulation of visually-guided behaviour.

Hypothalamic retinal input is traditionally considered distinct from the subcortical pathways supporting vision, specialised to adjust physiology and behaviour alongside variations in ambient illumination. Investigations of retinohypothalamic function have overwhelming focussed on the suprachiasmatic nucleus circadian clock, however. Here we employ multielectrode recording, viral tracing and chemogenetic manipulation in mice to show that another retinohypothalamic target, the anterior lateral hypothalamic area (LHA), displays diverse visual processing capabilities, supporting regulation of mor

Chemogenetic inhibition of prefrontal cortex inputs to dorsal raphe reduces anxiety behaviors in male rat model of fetal alcohol spectrum disorder.

Prenatal ethanol exposure (PE) causes Fetal Alcohol Spectrum Disorders (FASD), characterized by cognitive, behavioral, and emotional deficits, including anxiety and depression. PE-induced alteration in the function of dorsal raphe nucleus (DRN) serotonin (5-HT) neurons is thought to be major contributing factor for increased anxiety. However, the precise neuronal circuits involved are unknown. Using electrophysiology, optogenetics, chemogenetics, and behavioral approaches, we find that PE preferentially potentiates medial prefrontal cortex (mPFC) glutamatergic inputs, but not lateral habenula

Ventral pallidum DRD3 potentiates a pallido-habenular circuit driving accumbal dopamine release and cocaine seeking.

Drugs of abuse induce persistent remodeling of reward circuit function, a process thought to underlie the emergence of drug craving and relapse to drug use. However, how circuit-specific, drug-induced molecular and cellular plasticity can have distributed effects on the mesolimbic dopamine reward system to facilitate relapse to drug use is not fully elucidated. Here, we demonstrate that dopamine receptor D3 (DRD3)-dependent plasticity in the ventral pallidum (VP) drives potentiation of dopamine release in the nucleus accumbens during relapse to cocaine seeking after abstinence. We show that tw

The rostromedial tegmental nucleus (RMTg), a GABAergic afferent to midbrain dopamine neurons, encodes aversive stimuli and inhibits motor responses.

Separate studies have implicated the lateral habenula (LHb) or amygdala-related regions in processing aversive stimuli, but their relationships to each other and to appetitive motivational systems are poorly understood. We show that neurons in the recently identified GABAergic rostromedial tegmental nucleus (RMTg), which receive a major LHb input, project heavily to midbrain dopamine neurons, and show phasic activations and/or Fos induction after aversive stimuli (footshocks, shock-predictive cues, food deprivation, or reward omission) and inhibitions after rewards or reward-predictive stimuli

Bsx controls pineal complex development.

Neuroendocrine cells in the pineal gland release melatonin during the night and, in teleosts, are directly photoreceptive. During development of the pineal complex, a small number of cells migrate leftward away from the pineal anlage to form the parapineal cell cluster, a process that is crucial for asymmetrical development of the bilateral habenular nuclei. Here, we show that, throughout zebrafish embryonic development, the brain-specific homeobox (bsx) gene is expressed in all cell types of the pineal complex. We identified Bmp and Noto/Flh as major regulators of bsx expression in the pineal

Immunohistochemical localization of the P2Y(1) purinergic receptor in neurons and glial cells of the central nervous system.

This study reports the characterization of a polyclonal antiserum to a carboxy-terminal epitope of the P2Y(1) receptor and its use in immunolocalization studies of this receptor in the CNS. The antibody recognized a major band of 42 kDa in Western blot of tissue homogenates from rat and bovine brain. Immunohistochemical studies confirmed early reports about the presence of the P2Y(1) receptor in the corpus callosum, habenula and ductal cells of the salivary gland. In addition, we found that the P2Y(1) receptor is intensely expressed in Purkinje cells, in deep layers of the cerebral cortex and

Recruitment of inhibitory neuronal pathways regulating dopaminergic activity for the control of cocaine seeking.

Drug seeking is associated with the ventral tegmental area (VTA) dopaminergic (DA) activity. Previously, we have shown that brief optogenetic inhibition of VTA DA neurons with 1 s pulses delivered every 9 s attenuates cocaine seeking under extinction conditions in rats without producing overt signs of dysphoria or locomotor sedation. Whether recruitment of neuronal pathways inhibiting VTA neuronal activity would suppress drug seeking remains unknown. Here, we asked if optogenetic stimulation of the lateral habenula (LHb) efferents in the rostromedial tegmental nucleus (RMTg) as well as RMTg ef
