Synthesis of the pluramycins 1: two designed anthrones as enabling platforms for flexible bis-C-glycosylation.

Two effective tricyclic platforms are reported for the installation of the two constituent sugars, L-vancosamine and D-angolosamine, in a regio- and stereoselective manner for the synthesis of the pluramycin class of bis-C-glycoside antitumor antibiotics. Two complementary protocols are now available that differ in the order in which the two sugar moieties are installed. Sc(OTf)3 was effective as the Lewis acid.

Studies on protein-polysaccharides from pig laryngeal cartilage. Extraction and purification.

1. Protein-polysaccharides of chondroitin sulphate were extracted from fresh laryngeal cartilage at pH6.8 by two procedures. Procedure I consisted of brief low-speed homogenization in 0.15m (iso-osmotic) sodium acetate and procedure II consisted of longer homogenization followed by prolonged extraction in 10% calcium chloride solution. 2. The protein-polysaccharides in both extracts were isolated and purified by precipitation with 9-aminoacridine hydrochloride. They were free from serum proteins, collagen and nucleic acids and also of degradative enzymes. The absence of such enzymes was shown

ManNAc attenuates Man5 glycoform abundance through GNE-mediated metabolic channeling of UDP-GlcNAc to N-glycosylation modifications via CMP-Neu5Ac biosynthesis.

N-glycosylation, a critical quality attribute of monoclonal antibodies, plays a pivotal role in regulating pharmacokinetics and pharmacodynamics through high-mannose (Man5) glycoform modulation. While our previous work demonstrated that N-acetyl-D-mannosamine (ManNAc) supplementation effectively reduces Man5 levels without compromising antibody yield or other critical quality attributes, the mechanistic basis remained unclear. This study systematically investigates ManNAc's regulatory mechanism through a multi-parametric analysis. Cellular uptake studies revealed a 3-day latency period precedi

N-acetylmannosamine (ManNAc) supports the growth of Borrelia burgdorferi in the absence of N-acetylglucosamine (GlcNAc).

Borrelia burgdorferi, the causative agent of Lyme disease, lacks the ability to biosynthesize many essential nutrients de novo, including N-acetylglucosamine (GlcNAc). This amino sugar is required for cell wall synthesis, and is a component of the complex growth medium used for in vitro propagation. When cultured without free GlcNAc, B. burgdorferi cells exhibit a unique biphasic growth pattern. We hypothesized that genes involved in the GlcNAc starvation response would be differentially expressed when compared to cells cultured in complete medium, and investigated this using transcriptomics.

Guanidinylated 2,5-dideoxystreptamine derivatives as anthrax lethal factor inhibitors.

Anthrax lethal factor is a Zn(2+)-dependent metalloprotease and the key virulence factor of tripartite anthrax toxin secreted by Bacillus anthracis, the causative agent of anthrax. A series of guanidinylated 2,5-dideoxystreptamine derivatives were designed and synthesized as inhibitors of lethal factor, some of which show strong inhibitory activity against lethal factor in an in vitro FRET assay. Preparation and structure-activity relationships of these compounds are presented.

Long-term fate and effects of exercise on sternal cartilage autografts used for repair of large osteochondral defects in horses.

Bilateral osteochondral defects (10 mm2 x 3 mm deep) were created on the distal articular surface of the radial carpal bone of ten, 2- to 3-year-old horses. One defect of each horse was repaired, using a sternal cartilage autograft (treated), and the other was left untreated (control). The horses were exercised on a high-speed treadmill at incrementally increased speed and duration over the course of 12 months. Horses were evaluated arthroscopically at 6 to 7 weeks, and clinical examinations were conducted weekly at exercise. Twelve months after surgery, carpuses of each horse were radiographe

Glucosamine as a regulator of O-GlcNAc signaling: linking metabolism to disease pathogenesis.

BACKGROUND: Glucosamine, a naturally occurring amino sugar abundant in cartilage, has long been utilized as a dietary supplement to alleviate osteoarthritis (OA) and joint pain. Beyond its structural role in maintaining joint integrity, glucosamine is metabolized through the hexosamine biosynthetic pathway (HBP) to generate UDP-N-acetylglucosamine (UDP-GlcNAc), the indispensable donor substrate for protein O-linked N-acetylglucosamine (O-GlcNAc) modification. FINDINGS: Accumulating evidence indicates that glucosamine-driven modulation of HBP flux and the consequent alteration of O-GlcNAcylati

Glucosamine Promotes Autophagy and Attenuates Hepatic Steatosis Via O-GlcNAcylation-Mediated Mechanisms.

Autophagy is a key cellular process regulating lipid turnover and maintaining hepatic homeostasis, and its impairment is closely associated with the pathogenesis of nonalcoholic fatty liver disease (NAFLD). In this study, we examined the effects of glucosamine (GlcN), a hexosamine biosynthetic pathway intermediate, on autophagy and lipid accumulation using both human hepatocellular carcinoma (HepG2) cells and a high-fat diet (HFD)-induced NAFLD mouse model. GlcN treatment led to a dose- and time-dependent increase in the expression of autophagy-related markers LC3 and p62 at both mRNA and prot

Human anti-glycan reactivity emerges from B cells utilizing private gene rearrangements that are affinity maturated in germinal centers.

The human antibody repertoire includes reactivity toward a broad array of carbohydrate antigens associated with commensal microflora and pathogenic organisms. Here, we examined the ontogeny and diversity of the human anti-glycan repertoire. Antibodies reactive with group A Streptococcus cell wall carbohydrate (GAC) were absent at birth but reached adult frequencies in childhood, concomitant with the emergence of circulating GAC-reactive memory B cells. GAC-binding B cells expressing germinal center (GC) markers were abundant in tonsil tissue from children. Recombinant antibodies derived from t

Early Cardiomyopathy in Prediabetic NDPK-B-Deficient Mice Is Associated with Remodeling of the Mitochondrial O-GlcNAc Proteome.

Diabetic cardiomyopathy (DCM) is characterized by myocardial remodeling that may already be evident during prediabetes, yet the molecular alterations accompanying these early changes remain poorly understood. The present study examined mouse models of Nucleoside diphosphate kinase B (NDPK-B)-deficient prediabetes and streptozotocin-induced diabetes using O-GlcNAc-associated proteomic profiling to define stage-specific molecular alterations during the progression from prediabetic to diabetic cardiomyopathy. Both models exhibited increased left ventricular extracellular matrix deposition and imp

Natural Biologics Accelerate Healing of Diabetic Foot Ulcers by Regulating Oxidative Stress.

Difficult or even non-healing diabetic foot ulcers (DFU) are a global medical challenge. Although current treatments such as debridement, offloading, and infection control have resulted in partial improvement in DFU, the incidence, amputation, and mortality rates of DFU remain high. Therefore, there is an urgent need to find new or more effective drugs. Numerous studies have shown that oxidative stress plays an important role in the pathophysiology of DFU. The nuclear factor erythroid 2-related factor (Nrf2) signaling pathway and the advanced glycated end products (AGEs)-receptor for advanced

Glucose-6 Phosphate, A Central Hub for Liver Carbohydrate Metabolism.

: Cells efficiently adjust their metabolism according to the abundance of nutrients and energy. The ability to switch cellular metabolism between anabolic and catabolic processes is critical for cell growth. Glucose-6 phosphate is the first intermediate of glucose metabolism and plays a central role in the energy metabolism of the liver. It acts as a hub to metabolically connect glycolysis, the pentose phosphate pathway, glycogen synthesis, de novo lipogenesis, and the hexosamine pathway. In this review, we describe the metabolic fate of glucose-6 phosphate in a healthy liver and the metabolic

Cardiac LXRα protects against pathological cardiac hypertrophy and dysfunction by enhancing glucose uptake and utilization.

Pathological cardiac hypertrophy is characterized by a shift in metabolic substrate utilization from fatty acids to glucose, but the molecular events underlying the metabolic remodeling remain poorly understood. Here, we investigated the role of liver X receptors (LXRs), which are key regulators of glucose and lipid metabolism, in cardiac hypertrophic pathogenesis. Using a transgenic approach in mice, we show that overexpression of LXRα acts to protect the heart against hypertrophy, fibrosis, and dysfunction. Gene expression profiling studies revealed that genes regulating metabolic pathways w

Cytokines and proteoglycans: an introductory overview.

The defining characteristic of the glycoproteins known as proteoglycans is the presence of O-linked acidic polysaccharides known as GAGs (glycosaminoglycans). The backbone of these linear polysaccharides is a repeating disaccharide, comprising N-acetyl hexosamine alternating with beta-D-glucuronic acid, alpha-L-iduronic acid, or galactose. For some GAGs, partial deacetylation, epimerization of glucuronic acid, and substitution with N- and O-sulphates result in highly complex, heterogeneous structures. The interactions with proteins through which GAGs exert their biological effects depend on th

Effects of a polysaccharide fraction from the roots of Bupleurum falcatum L. on experimental gastric ulcer models in rats and mice.

Effects of an acidic polysaccharide fraction, BR-2, from the roots of Bupleurum falcatum L., on HCl-ethanol, ethanol and water immersion stress-induced gastric lesions in mice and pylorus-ligated ulcers in rats have been studied. Oral administration of BR-2 at doses of 50 to 200 mg kg-1 inhibited the formation of the gastric lesions induced by necrotizing agents such as HCl-ethanol and ethanol, in a dose dependent manner. This protective effect was observed after oral, intraperitoneal, and subcutaneous administration of BR-2 (25-100 mg kg-1). BR-2 also inhibited the formation of gastric ulcers
