A host-restricted viral vector for antigen-specific immunization against Lyme disease pathogen.

Newcastle disease virus (NDV) is an avian virus that is attenuated in primates and is a potential vaccine vector for human use. We evaluated NDV as a vector for expressing selected antigens of the Lyme disease pathogen Borrelia burgdorferi. A series of recombinant NDVs were generated that expressed intracellular or extracellular forms of two B. burgdorferi antigens: namely, the basic membrane protein A (BmpA) and the outer surface protein C (OspC). Expression of the intracellular and extracellular forms of these antigens was confirmed in cultured chicken cells. C3H or Balb/C mice that were imm

The Trw type IV secretion system of Bartonella mediates host-specific adhesion to erythrocytes.

Bacterial pathogens typically infect only a limited range of hosts; however, the genetic mechanisms governing host-specificity are poorly understood. The alpha-proteobacterial genus Bartonella comprises 21 species that cause host-specific intraerythrocytic bacteremia as hallmark of infection in their respective mammalian reservoirs, including the human-specific pathogens Bartonella quintana and Bartonella bacilliformis that cause trench fever and Oroya fever, respectively. Here, we have identified bacterial factors that mediate host-specific erythrocyte colonization in the mammalian reservoirs

Identification of genetic determinants of a tick-borne flavivirus associated with host-specific adaptation and pathogenicity.

Tick-borne flaviviruses are maintained in nature in an enzootic cycle involving a tick vector and a vertebrate host. Thus, the virus replicates in two disparate hosts, each providing selective pressures that can influence virus replication and pathogenicity. To identify viral determinants associated with replication in the individual hosts, plaque purified Langat virus (TP21pp) was adapted to growth in mouse or tick cell lines to generate two virus variants, MNBp20 and ISEp20, respectively. Virus adaptation to mouse cells resulted in four amino acid changes in MNBp20 relative to TP21pp, occurr

Genome sequence of a human tumorigenic poxvirus: prediction of specific host response-evasion genes.

Molluscum contagiosum virus (MCV) commonly causes asymptomatic cutaneous neoplasms in children and sexually active adults as well as persistent opportunistic acquired immunodeficiency syndrome (AIDS)-associated disease. Sequencing the 190-kilobase pair genome of MCV has now revealed that the virus potentially encodes 163 proteins, of which 103 have homologs in the smallpox virus. MCV lacks counterparts to 83 genes of the smallpox virus, including those important in suppression of host responses to infection, nucleotide biosynthesis, and cell proliferation. MCV possesses 59 genes that are predi

Donor-derived cytomegalovirus-specific CD8+ T cells restricted to shared, donor-specific, or host-specific HLA after HLA mismatched hematopoietic stem cell transplantation.

Immune reconstitution after human leukocyte antigen (HLA)-mismatched (haploidentical) hematopoietic stem cell transplantation (haplo-HCT) can significantly influence long-term outcomes. The three possible HLA haplotypes after transplantation are: one carried by both the patient and the donor (shared HLA), one by donor only (donor-specific HLA), and one by patient only (host-specific HLA), and the donor T cells remain restricted to one of these three haplotypes. Understanding the presence of donor T cells restricted to each haplotype may provide more detailed insights into post-transplant immun

Two known and one new species of Proctoeces from Australian teleosts: Variable host-specificity for closely related species identified through multi-locus molecular data.

Species of Proctoeces Odhner, 1911 (Trematoda: Fellodistomidae) have been reported from a wide range of marine animals globally. Members of the genus tend to lack strongly distinguishing morphological features for diagnosis, making identification difficult and the true number of species in the genus contentious. Combined morphological and molecular analyses were used to characterise three species of Proctoeces from Moreton Bay and the southern Great Barrier Reef. Data for two ribosomal regions and one mitochondrial region were generated for specimens collected from Australia. Three unique 18S-

Redescription of Babesia capreoli (Enigk and Friedhoff, 1962) from roe deer (Capreolus capreolus): isolation, cultivation, host specificity, molecular characterisation and differentiation from Babesia divergens.

The recent use of the sole molecular identification of Babesia infecting European cervids has led to confusion between the closely related Babesia divergens and Babesia capreoli, and to their grouping together as "B. divergens-like". In order to clarify this taxonomic confusion, Babesia from roe deer, cattle and human blood were isolated, cultured and their biological as well as molecular characteristics compared. On this basis, we conclude that: (i) the parasites isolated from roe deer blood are B. capreoli; (ii) there are no intraspecific variations in the 18S rDNA within B. capreoli and B.

Diversity of bartonellae in mites (Acari: Mesostigmata: Macronyssidae and Spinturnicidae) of boreal forest bats: Association of host specificity of mites and habitat selection of hosts with vector potential.

Research into various bacterial pathogens that can be transmitted between different animals and may have zoonotic potential has led to the discovery of different strains of Bartonella sp. in bats and their associated ectoparasites. Despite their enormous species diversity, only a few studies have focussed on the detection of bacterial pathogens in insectivorous bats of boreal forests and their associated Macronyssidae and Spinturnicidae mites. We collected and molecularly analysed mite samples from forest-dwelling bat species distributed all along the boreal belt of the Palearctic, from Centra

Host Tau Genotype Specifically Designs and Regulates Tau Seeding and Spreading and Host Tau Transformation Following Intrahippocampal Injection of Identical Tau AD Inoculum.

Several studies have demonstrated the different characteristics of tau seeding and spreading following intracerebral inoculation in murine models of tau-enriched fractions of brain homogenates from AD and other tauopathies. The present study is centered on the importance of host tau in tau seeding and the molecular changes associated with the transformation of host tau into abnormal tau. The brains of three adult murine genotypes expressing different forms of tau-WT (murine 4Rtau), hTau (homozygous transgenic mice knock-out for murine tau protein and heterozygous expressing human forms of 3Rta

Evaluating the Biological and Ecological Factors Influencing Transmission of Larval Digenetic Trematodes: A Test of Second Intermediate Host Specificity of Two North American Halipegus Species.

Host specificity of parasites is a basic principle in parasitology; however, it is not easily measured. Previously, host specificity was calculated as the number of species that a parasite infected, but this is not an accurate description of host usage because some species are capable of being infected but do not contribute to the completion of the life cycle. Instead, measures of host specificity should take into consideration interactions between a parasite and a potential host species as well as interactions between current and subsequent hosts in the life cycle. The objectives of this stud…

Host diversity and landscape structure drive genospecies-specific Lyme disease risk across Eurasia.

Lyme disease poses an escalating global health challenge partly driven by climate change and land-use transformation, while mechanistic understanding of transmission remains limited. We analyzed prevalence records from 43 Eurasian countries to map pathogen biogeography and applied theory-driven structural equation modeling (SEM) to evaluate hypothesized direct, indirect, and cascading pathways linking climate, land cover, landscape structure, and host biodiversity to pathogen prevalence. Genospecies exhibited distinct biogeographic patterns shaped by vector-host specificity, with Ixodes ricinu…

Host-Dependent Endophytes as a Resilient Biological Reservoir: Niche-Specific Expansion and Holobiont Reassembly in Grapevine.

Plants and their endophytic microbiota form a functional holobiont, wherein host-dependent endophytes (HDEs) are proposed to serve as persistent microbial components associated with host continuity. Using long-term in vitro grapevine plantlets, we investigated HDE redistribution and reassembly across successive agar-based subcultures and during the transition to soil-based cultivation. Results showed that the initial tissue niche (shoot-tip versus root-base) imprinted a persistent compositional signature on HDE communities, with distinct cultivar-dependent effects. Numerous microbial amplicon

Helminth diversity, prevalence, and host-specific patterns in wild and domestic ruminants of the Bukhara region, Uzbekistan.

Objectives: Helminth infections impose major health burdens on livestock and threaten wildlife at the livestock-wildlife interface in Central Asian drylands. Despite rising concern over parasite transmission between domestic and wild ungulates, integrated surveys remain limited. This study characterized helminth species richness, prevalence, and infection patterns in domestic ruminants (sheep, goats, and cattle) and critically endangered wild ungulates (Ovis ammon bocharensis, Gazella subgutturosa, and Cervus hanglu yarkandensis) in the Bukhara region of Uzbekistan. Overcoming the curse of knowledge: guidelines for reporting system-specific details of host infection and migration dynamics.

Literature syntheses link knowledge across study systems and regions. However, specific details on the biological systems of the included publications may not always be explicitly outlined because of communication gaps (a.k.a. The Curse of Knowledge) and/or knowledge gaps. These gaps can lead to inappropriate comparisons across studies if researchers unfamiliar with the systems make inaccurate assumptions about aspects of study design or natural history. In the context of movement ecology, a growing number of publications explore how host infection and host migration intersect, work that is cr

Site-specific microRNA responses in lungs of pigs depend on the host-adaptation of H1N1 influenza A virus.

Cross-species transmission of influenza A virus (IAV) represents a major risk for emergence of new pandemics, however, the host molecular responses invoked by cross-species IAV infection remain incompletely defined. MicroRNAs (miRNAs) are important post-transcriptional regulators of innate immunity, but their role during infection with IAV strains of the same subtype but of different host origin has not been investigated. Here, we investigated pulmonary miRNA responses in upper and lower lung sites from pigs inoculated with either a swine-adapted (swH1N1) or a human-adapted (huH1N1) IAV origin

Spatial transcriptomic atlas of murine neurotoxocariasis reveals region-specific host responses and dysfunction in the brain.

Toxocara canis is a globally distributed parasite that infects dogs and other canids, shedding eggs into the environment. Humans become accidental hosts by ingesting infective eggs, leading to syndromes such as visceral, ocular and neurotoxocariasis - the latter being poorly understood. Neurotoxocariasis has been proposed to be associated with epilepsy, cognitive impairment and schizophrenia, but its pathomechanism remains unclear. Here, we combine spatial transcriptomics, single-nucleus RNA sequencing, lipidomics and behavioural assays in a mouse model to characterise the host-parasite interf

Multi-platform profiling reveals host- and cell -type-specific pseudorabies virus gene expression.

Pseudorabies virus (PRV) is an alphaherpesvirus that follows a conserved immediate-early → early → late transcriptional cascade, yet how this program adapts to diverse cell types is unclear. We profiled PRV transcription in four permissive cell lines-two epithelial (porcine kidney PK-15 and rat kidney NRK), one glial (rat glioma C6), and one neuron-like (rat PC-12)-at six time points (1-12 h post-infection). Host-dependent differences peaked early in infection, particularly for the regulators ie180, ep0, and us1. ie180 showed strong species bias, with high expression in PK-15 but minimal in ro

Cognac distillation wines host specific Oenococcus oeni strains representing a novel genetic lineage.

Oenococcus oeni is the lactic acid bacteria species best adapted to wine. It proliferates during alcoholic fermentation and typically becomes the most abundant species during malolactic fermentation. A large diversity of O. oeni strains is present in all wine-producing regions. These strains group into several genetic lineages, which are more product-specific rather than linked to geography. I. e. strains of one genetic lineage are often detected in the same wine type produced in distant regions. This study aimed to investigate the biodiversity of O. oeni involved in malolactic fermentations o
