Effect of human cord and postoperative serum on experimental inflammation in the rat.

Human cord and postoperative serum depressed the oedema provoked by mediators of the inflammatory reaction such as bradykinin, histamine, serotonin and prostaglandin E2 and also the experimental inflammation caused by carrageenin. Normal human and pregnancy serum did not have such an effect. In two cases of open neural tube defect, one of anencephaly and another of spina bifida, human amniotic fluid also had a strongly depressing effect on the experimental oedema provoked by serotonin. Human amniotic fluid from normal pregnancies did not inhibit this experimental inflammation. A protein-fracti

Experimental transient bacteraemias in human subjects with clinically healthy gingivae.

Thirty-six subjects with no evidence of clinical gingival inflammation underwent a standardized toothbrushing procedure. Blood specimens, obtained from a vein in the antecubital fossa during the last 30 seconds of brushing, were cultured under aerobic and stringent anaerobic conditions. Three subjects exhibited detectable bacteraemias, Propionibacterium sp. being isolated from two of the subjects, while Actinomyces sp., Streptococcus sanguis and Streptococcus mitis were isolated from the third. The implications of these results are discussed.

TRANSMISSION AND CULTIVATION EXPERIMENTS WITH HUMAN TRACHOMA AND THE EXPERIMENTAL DISEASE IN MONKEYS.

1. Conjunctival tissue derived from alien and native American white persons in New York City, having trachoma in an advanced stage, has been used successfully to induce in Macacus rhesus monkeys characteristic granular conjunctivitis. The transfer of infection was effected either by a single subconjunctival injection, or by repeated swabbing with conjunctival secretions. 2. Pathogenic strains of Bacterium granulosis have been recovered from the trachomatous tissues of six out of eleven patients. In addition, the organisms have been isolated from the monkeys infected with human material. 3. Rep

Concordance between sites of tumor development in humans and in experimental animals for 111 agents that are carcinogenic to humans.

Since the inception of the IARC Monographs Programme in the early 1970s, this Programme has developed 119 Monograph Volumes on more than 1000 agents for which there exists some evidence of cancer risk to humans. Of these, 120 agents were found to meet the criteria for classification as carcinogenic to humans (Group 1). Volume 100 of the IARC Monographs, compiled in 2008-2009 and published in 2012, provided a review and update of the 107 Group 1 agents identified as of 2009. These agents were divided into six broad categories: (I) pharmaceuticals; (II) biological agents; (III) arsenic, metals,

New experimental data on the human dermal absorption of Simazine and Carbendazim help to refine the assessment of human exposure.

Due to their widespread usage, people are exposed to pesticides on a daily basis. Although these compounds may have adverse effects on their health, there is a gap in the data and the methodology needed to reliably quantify the risks of non-occupational human dermal exposure to pesticides. We used Franz cells and human skin in order to measure the dermal absorption kinetics (steady-state flux, lag time and permeability coefficient) of Carbendazim and Simazine. These parameters were then used to refine the dermal exposure model and a probabilistic simulation was used to quantify risks resulting

Human anti-laminin 5 autoantibodies induce subepidermal blisters in an experimental human skin graft model.

Patients with one form of cicatricial pemphigoid have IgG antibasement membrane autoantibodies against laminin 5 (alpha3beta3gamma2). Although passive transfer of rabbit anti-laminin 5 IgG to neonatal mice has been shown to induce subepidermal blisters that mimic those in patients, it has not been possible to directly assess the pathogenic activity of human autoantibodies in this animal model because the latter do not bind murine skin. To address this question, a disease model in adult mice as well as SCID mice bearing human skin grafts was developed. Adult BALB/C mice challenged with rabbit a

Are PrP(C)s involved in some human myelin diseases? Relating experimental studies to human pathology.

We have experimentally demonstrated that cobalamin (Cbl) deficiency increases normal cellular prion (PrP(C)) levels in rat spinal cord (SC) and cerebrospinal fluid (CSF), and decreases PrP(C)-mRNA levels in rat SC. Repeated intracerebroventricular administrations of anti-octapeptide repeat-PrP(C)-region antibodies to Cbl-deficient (Cbl-D) rats prevent SC myelin lesions, and the administrations of PrP(C)s to otherwise normal rats cause SC white matter lesions similar to those induced by Cbl deficiency. Cbl positively regulates SC PrP(C) synthesis in rat by stimulating the local synthesis of epi

A Practical Experimental Protocol for Identification and Validation of UFMylation Substrate in Human Cells.

UFMylation is an evolutionarily conserved ubiquitin-like modification that covalently conjugates UFM1 to lysine residues of substrates via a sequential E1-E2-E3 enzymatic cascade. UFMylation plays a pivotal role in maintaining cellular homeostasis, and its dysregulation is closely linked to multiple major diseases, including malignant tumors, hematopoietic defects, neurodegenerative disorders, and congenital developmental defects, highlighting its important biological significance. However, few substrates of UFMylation have been reported to date, limiting our deep understanding of the mechanis

Amplification of seeding-competent tau aggregates by PMCA in human and experimental tauopathies.

Tau assemblies, associated with tauopathies, are believed to self-propagate through prion-like mechanisms in the central nervous system, driving neurodegeneration. Recently, protein seed amplification assays have emerged as highly sensitive methods for detecting trace amounts of misfolded protein assemblies across various neurodegenerative diseases. In this study, we utilized protein misfolding cyclic amplification (PMCA) to demonstrate that tau assemblies from the brains of transgenic mice or human patients with tauopathies can be efficiently amplified. Amplification was achieved using comple

Systematic review of experimental studies in humans on transcranial temporal interference stimulation.

Transcranial temporal interference stimulation (tTIS) has recently emerged as a non-invasive neuromodulation method aimed at reaching deeper brain regions than conventional techniques. However, many questions about its effects remain, requiring further experimental studies. This review consolidates the experimental literature on tTIS's effects in the human brain, clarifies existing evidence, identifies knowledge gaps, and proposes future research directions to evaluate its potential. A systematic literature search was performed in PubMed, Web of Science, and Scopus for studies published up to

Reproductive toxicity of micro- and nanoplastics: Insights from experimental and human studies.

The exponential rise in plastic production has driven widespread contamination by micro- and nanoplastics (MNPs) in the environment. These plastic particles and their chemical additives have been detected in water sources, human bodily fluids, and reproductive tissues. With global fertility rates declining, their role as potential contributors is under investigation. This scoping review compares findings from in vitro experiments, in vivo studies across animal models, and epidemiological data to assess potential reproductive hazards associated with MNP exposure. Forty original studies publishe

Experimental Investigation and Safety Classification Evaluation of Small Drone Collision with Humans.

The safety of small drones in collision with humans has become a key focus in engineering and research fields. This study presents a vertical drop test platform for collision tests involving three representative drones (Air, Mavic 2, and M200) impacting the head of a Hybrid III dummy from different heights and orientations. The deformation and damage of the drones during various collision scenarios, as well as the dynamic responses of the dummy head and neck, are analyzed. The head injury criterion (HIC), together with 3 ms cumulative acceleration (head acc 3 ms), are used to evaluate head inj
