In vitro extracellular replication of Wolbachia endobacteria.

Obligate intracellular endobacteria of the genus Wolbachia are widespread in arthropods and several filarial nematodes. Control programs for vector-borne diseases (dengue, Zika, malaria) and anti-filarial therapy with antibiotics are based on this important endosymbiont. Investigating Wolbachia, however, is impeded by the need for host cells. In this study, the requirements for Wolbachia wAlbB growth in a host cell-free in vitro culture system were characterized via qPCRs. A cell lysate fraction from Aedes albopictus C6/36 insect cells containing cell membranes and medium with fetal bovine ser

The whole genome sequence of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann), reveals insights into the biology and adaptive evolution of a highly invasive pest species.

BACKGROUND: The Mediterranean fruit fly (medfly), Ceratitis capitata, is a major destructive insect pest due to its broad host range, which includes hundreds of fruits and vegetables. It exhibits a unique ability to invade and adapt to ecological niches throughout tropical and subtropical regions of the world, though medfly infestations have been prevented and controlled by the sterile insect technique (SIT) as part of integrated pest management programs (IPMs). The genetic analysis and manipulation of medfly has been subject to intensive study in an effort to improve SIT efficacy and other as

Optimization of the spraying equipment and technology used in ornamental crops.

In 2006, a research project concerning the optimization of the spraying equipment and technology used in ornamental crops was started. First, several greenhouse growers were surveyed on the spray equipment and technology they were using for their plant protection. Later on, different parts of their equipment were evaluated. It this way, we could advice the connected growers on how to improve their own techniques and equipment. Additionally, the survey showed that growers predominantly use knapsack sprayers and lances for crop protection purposes. These techniques are often proven to be less ef

Transgenic Metarhizium rapidly kills mosquitoes in a malaria-endemic region of Burkina Faso.

Malaria control efforts require implementation of new technologies that manage insecticide resistance. Metarhizium pingshaense provides an effective, mosquito-specific delivery system for potent insect-selective toxins. A semifield trial in a MosquitoSphere (a contained, near-natural environment) in Soumousso, a region of Burkina Faso where malaria is endemic, confirmed that the expression of an insect-specific toxin (Hybrid) increased fungal lethality and the likelihood that insecticide-resistant mosquitoes would be eliminated from a site. Also, as Hybrid-expressing M. pingshaense is effectiv

Morphological and Chemical Changes in the Hemolymph of the Wax Moth Galleria mellonella Infected by the Entomopathogenic Fungus Conidiobolus coronatus.

Hemolymph enables communication between organs in insects and ensures necessary coordination and homeostasis. Its composition can provide important information about the physiological state of an insect and can have diagnostic significance, which might be particularly important in the case of harmful insects subjected to biological control. Galleria mellonella Linnaeus 1758 (Lepidoptera: Pyralidae) is a global pest to honey bee colonies. The hemolymph of its larvae was examined after infection with the soil fungus Conidiobolus coronatus (Constantin) Batko 1964 (Entomophthorales). It was found

Pesticide resistance: can we make it a renewable resource?

Negative cross-resistance (NCR) occurs when a mutant allele confers (i) resistance to one toxic chemical and (ii) hyper-susceptibility to another. Sequential deployment of NCR toxins is useful for insect control in few situations (Pittendrigh et al., 2000). Using Monte Carlo simulations, we investigated the concurrent use of a pair of NCR toxins to control a hypothetical insect pest population. When the toxins killed more heterozygotes than homozygotes, the resistance allele became either extremely common or rare depending on starting allelic frequency. If the NCR toxins did not kill the two h…

The deployment of semiochemical-based pest management tools for woody plants in managed residential and commercial landscapes: current status, challenges, and future directions.

Managed residential and commercial landscapes are primarily managed for aesthetic purposes or to provide social benefits (e.g. shade). These landscapes, and the woody plants that comprise them, exist in environments that pose unique challenges for the management of pests, such as the discontinuous nature and patchiness of habitat and hosts, abiotic conditions that result in plant stress, and the existence of ecologically arbitrary but legally meaningful boundaries. Due to challenges like these and other factors, plant health care managers have heavily relied upon traditional pesticides for dec

Estimating the efficacy and plume reach of semiochemical-baited traps on the capture of Trogoderma granarium everts.

BACKGROUND: The khapra beetle, Trogoderma granarium, is a serious pest of stored grains that infests more than 100 different commodities. The current wall trap used for monitoring is optimized for the behavior of khapra beetles. In this study, we tested existing and modified trap designs for Trogoderma after release of T. granarium exclusively. The work aimed to evaluate trap efficacy, while providing a template for further attract-and-kill research. Three bioassays were conducted in nine rooms. Four traps were evaluated: dome, wall, modified wall, and the Wilkins interception trap. RESULTS:

Reassessing ecdysteroidogenic cells from the cell membrane receptors' perspective.

Ecdysteroids secreted by the prothoracic gland (PG) cells of insects control the developmental timing of their immature life stages. These cells have been historically considered as carrying out a single function in insects, namely the biochemical conversion of cholesterol to ecdysteroids and their secretion. A growing body of evidence shows that PG cells receive multiple cues during insect development so we tested the hypothesis that they carry out more than just one function in insects. We characterised the molecular nature and developmental profiles of cell membrane receptors in PG cells of

Immune tolerance of vector beetle to its partner plant parasitic nematode modulated by its insect parasitic nematode.

Immune response of insect vectors to transmitted pathogens or insect hosts against parasites are well studied, whereas the mechanism of tripartite interactions remains elusive. In this study, we investigated the immune interactions of the vector beetle Monochamus alternatus ( Ma) to the devastating plant parasitic nematode Bursaphelenchus xylophilus ( Bx) and the insect parasitic nematode Howardula phyllotretae ( Hp). We report the unique immune mechanism by which the vector beetle tolerates many devastating Bx in its trachea, yet that immune tolerance is compromised by the parasitic nematode

Modulation of insect Ca(v) channels by peptidic spider toxins.

Insects have a much smaller repertoire of voltage-gated calcium (Ca(V)) channels than vertebrates. Drosophila melanogaster harbors only a single ortholog of each of the vertebrate Ca(V)1, Ca(V)2, and Ca(V)3 subtypes, although its basal inventory is expanded by alternative splicing and editing of Ca(V) channel transcripts. Nevertheless, there appears to be little functional plasticity within this limited panel of insect Ca(V) channels, since severe loss-of-function mutations in genes encoding the pore-forming alpha1 subunits in Drosophila are embryonic lethal. Since the primary role of spider v

High efficacy of integrated preventive measures against zoonotic cutaneous leishmaniasis in northern Afghanistan, as revealed by Quantified Infection Rates.

Following an outbreak of more than 200 cases of zoonotic cutaneous leishmaniasis (ZCL) during 2004 and 2005 among International Security Assistance Force (ISAF) troops stationed in the Mazar-e Sharif (MeS) airport area, epidemiological investigations indicated the presence of a local high-density ZCL focus. Based on ZCL's specific transmission modes, density and seasonality, integrated preventive measures were continuously implemented from February 2006 at a German military camp constructed at MeS. Preventive techniques included such synergistic measures as skin repellents and insecticide-impr

Edward F. Knipling: titan and driving force in ecologically selective area-wide pest management.

Few entomologists have impacted insect pest management as profoundly as Edward F Knipling (1909-2000). During WWII, Knipling and his colleagues developed highly effective measures to protect both military personnel and civilian populations from major arthropod-borne diseases. The sterile insect technique was Knipling's conception, and he successfully guided its development and use against the screwworm, Cochliomyia hominivorax Coquerel, and against various other pests. He inspired and guided the development of a wide range of ecologically selective methods of insect detection and suppression.

The effect of anti-sandfly saliva antibodies on Phlebotomus argentipes and Leishmania donovani.

A study was undertaken to find the effect of repeated bites of the sandfly, Phlebotomus argentipes, on its host as well as on the vector itself. The study also aimed to find the effect of the immune serum on the parasite, Leishmania donovani, naturally transmitted by the vector. The hamster which was exposed to sandfly feeding showed good antibody titre against the sandfly salivary-gland secretion, which indicates that the salivary-gland secretion is immunogenic in nature. The result also revealed that the feeding attraction of the females, which has been expressed as the percentage of engorge

Protective netting as a barrier against the X-disease phytoplasma vector Colladonus reductus (Hemiptera: Cicadellidae) in cherry orchards.

X-disease, caused by "Candidatus Phytoplasma pruni," poses a significant economic threat to the stone fruit industry, causing the removal of vast cherry acreage in Washington State. The most abundant vector in these orchards is the leafhopper Colladonus reductus (Van Duzee). Growers need season-long preventative alternatives to insecticides for a robust integrated pest management program. Thus, this study evaluated the efficacy of protective netting as a barrier to C. reductus. Between 2024 and 2025, 3 field trials were conducted testing 4 types of netting (20% shade, 30% shade, Mesh 25, and M

Bacteriocyte-specific antimicrobial peptides regulate the Rhynchophorus ferrugineus-Nardonella symbiosis and represent novel targets for symbiosis-based pest control.

BACKGROUND: Many beetles require tyrosine supplementation from endosymbiotic bacteria for exoskeleton synthesis. Weevils harbor the ancient endosymbiont Nardonella within specialized bacteriocytes, a mutualistic association maintained for >125 million years in which the bacterium exclusively preserves tyrosine biosynthetic capability. The red palm weevil, one of the world's most destructive invasive pests causing widespread devastation to palm industries across continents, depends on its Nardonella endosymbiont for survival. Disrupting this obligate symbiosis represents a promising pest contro

Novel methyl salicylate derivatives containing nerol and citronellol moieties for improved Acyrthosiphon pisum management.

BACKGROUND: Aphids have primarily been controlled by traditional chemical insecticides over the past decades, leading to health risks for honey bees. By contrast, plant volatiles such as methyl salicylate (MeSA) can not only repel aphids, but also attract their natural enemies, while being safe for bees, thus showing a promising prospect in aphid management. Our research group has previously discovered more active compounds for aphid management based on MeSA. Therefore, further structural optimization with MeSA as the lead compound is worth investigating. RESULTS: Through the structural combi
