Carnivore Attractant or Plant Elicitor? Multifunctional Roles of Methyl Salicylate Lures in Tomato Defense.

Synthetic plant volatile lures attract natural enemies, but may have non-target effects due to the multifunctional nature of volatile signals. For example, methyl salicylate (MeSA) is used to attract predators, yet also serves as a signaling hormone involved in plant pathogen defense. We investigated the consequences of deploying MeSA lures to attract predators for tomato (Solanum lycopersicum) defense against herbivores. To understand the spatial distribution of the lure's effect, we exposed tomatoes in the field to MeSA along a linear distance gradient and induced defenses by simulating feed

Multimodal Insecticidal Activity of α-Humulene Against Chilo suppressalis via Endocrine Disruption and Transcriptional Suppression of Cuticular Proteins.

Striped rice stem borer (Chilo suppressalis, SSB) poses a substantial threat to global rice production, leading to considerable economic loss. In this study, we explored the insecticidal mechanism of α-humulene, a sesquiterpene derived from Humulus lupulus, against SSB larvae. Bioassays indicated that α-humulene markedly elevated larval mortality rates and extended developmental periods. Transcriptomic analysis revealed the upregulation of genes involved in juvenile hormone (JH) biosynthesis and the downregulation of cuticle protein genes. Molecular docking experiments demonstrated that α-humu

Membrane-localized MET engages PVR to mediate extranuclear juvenile hormone signaling in Aedes aegypti.

Although Methoprene-tolerant (MET) is well established as the intracellular receptor mediating the genomic actions of juvenile hormone (JH) in insects, the identity of the receptor responsible for initiating extranuclear JH responses has remained elusive. In the yellow fever mosquito Aedes aegypti, we identify the platelet-derived growth factor- and vascular endothelial growth factor-receptor related (PVR) protein as a key mediator of membrane-initiated JH signaling. JH treatment induces robust PVR phosphorylation in the fat body of adult female mosquitoes. Disruption of PVR function suppresse

Histone deacetylases synergistically regulate juvenile hormone signaling in the yellow fever mosquito, Aedes aegypti.

Controlling Aedes aegypti mosquitoes is crucial for managing mosquito-transmitted diseases like dengue, zika, chikungunya, and yellow fever. One of the efficient methods to control mosquitoes is to block their progression from the larval to the adult stage. Juvenile hormones (JH) maintain the larval stage and ensure proper developmental timing for transitioning from larval-pupal-adult stages. Our previous studies showed that histone deacetylases (HDACs) regulate JH signaling and metamorphosis in the red flour beetle Tribolium castaneum. However, the role of HDACs in regulating JH signaling in

Leucokinin-like immunoreactive neurones in the central nervous system of the spider Cupiennius salei.

Leucokinin is a member of the myokinin peptide family. These myotropic peptides are widely distributed in arthropods. A specific antiserum immunoreactive to the neuroactive octapeptide leucokinin I (LKI) has been used to map neurones within the central nervous system of the central American wandering spider Cupiennius salei. The antiserum labels nine pairs of cell bodies and their axons. The somata are grouped near the joint of the optic lobes in the dorsal part of the supraoesophageal ganglion. The axons descend in a bundle, pass the oesophagus and divide to innervate the suboesophageal gangl

Reduced 20-Hydroxyecdysone Signaling Modulates Antifungal Immune Responses and Growth in Lymantria dispar Against Beauveria bassiana.

20-Hydroxyecdysone (20E) is the active form of the steroid hormone ecdysone and is responsible for regulating insect development, molting, and metamorphosis. Infection with the entomopathogenic Beauveria bassiana enhances innate immunity and reduces 20E titers in Lymantria dispar. To further explore the role of the 20E signaling pathway in antifungal immunity, we combined exogenous hormone assays, proteomic profiling, and RNAi-mediated knockdown. In this study, exogenous hormone assays revealed that 20E accelerated the death of B. bassiana-infected larvae, whereas juvenile hormone (JH) markedl

Developmental bias of body size and caste by the fire ant social supergene.

Supergenes are clusters of linked genes that drive complex phenotypic divergence. In the fire ant Solenopsis invicta, a supergene determines polymorphic social organization, and its derived (b) haplotype is over-represented among large workers and gynes relative to the ancestral (B) haplotype, acting as a selfish genetic element. We tested whether this caste bias reflects altered juvenile hormone (JH) mediated growth by treating colonies with a JH analogue. We show that BB and Bb pupae reach similar maximal sizes and exhibit a comparable JH-mediated growth response despite consistent Bb over-r

Hormone-dependent miRNA cascades converge on glycogen phosphorylase to regulate wing plasticity in aphids.

Phenotypic plasticity enables organisms to adapt to changing environments, yet how environmental cues generate alternative phenotypes remains largely unclear. Aphid wing plasticity offers a tractable model for understanding at a molecular level how winged and wingless morphs are induced and determined. In aphids, endocrine signals such as juvenile hormone (JH) and insulin, together with miRNAs, have been implicated in regulating wing plasticity, but whether and how these regulatory layers are mechanistically connected remains unclear. Here, we identified two hormone-responsive miRNA cascades t

Self-toxicity and tolerance mechanisms of honeybee venom in honeybees.

Honeybee venom (apitoxin) is a potent mixture of biologically active peptides and enzymes, primarily evolved as a defence against insect predators - including other bees. Recent evidence suggests that honeybees also employ components of apitoxin for both external and internal defence against parasites and microbial infections. Consequently, they are predicted to exhibit a remarkable resistance to their own venom, which they frequently encounter within colonies. To investigate this phenomenon, we examined the physiological responses of honeybee workers and drones to envenomation. Individuals we

A dual-barrier disruption strategy employing lufenuron/dsRNA nanocomplexes overcomes both cuticular and intestinal barriers in late-instar lepidopteran pests.

Managing late-instar lepidopteran pests remains a critical challenge due to their robust physiological and physical barriers, which severely compromise the efficacy of insecticides. Lufenuron, an insect growth regulator, controls lepidopteran pests by inhibiting chitin biosynthesis, but its efficacy against late-instar larvae needs improvement. Here, we developed a dual-barrier disruption strategy by integrating nano-lufenuron with RNA interference targeting the chitin synthase genes PxCHS1 and PxCHS2 in the diamondback moth, Plutella xylostella. The nanocarrier star polycation (SPc)-mediated

Elimination of certain honeybee venom activities by adipokinetic hormone.

The primary aim of this study was to analyse the influence of honeybee venom on various aspects of Drosophila melanogaster physiology and to assess the efficacy of adipokinetic hormone (AKH) in mitigating venom toxicity. We examined the harmful effects of venom on the thoracic muscles and central nervous system of Drosophila, as well as the potential use of AKH to counteract these effects. The results demonstrated that envenomation altered AKH levels in the Drosophila CNS, promoted cell metabolism, as evidenced by an increase in citrate synthase activity in muscles, and improved relative cell

Neuroendocrine modulation of olfactory sensory neuron signal reception via axo-dendritic synapses in the antennae of the mosquito, Aedes aegypti.

An ultrastructural study of the antennae of the yellow fever mosquito, Aedes aegypti, revealed that as in the salt marsh mosquito, Culex salinarius, the first flagellar segment of both sexes of A. aegypti contain neuroendocrine cells. These cells not only extend an axon via the antennal nerve to the antennal lobe of the deutocerebrum, but project collaterals to the periphery of the antennae, where they modulate the antennal sensory neurons by forming synapses with the dendrites of these afferent neurons. To our knowledge, this is the first report in any animal of neurites of neuroendocrine cel

Effects of pyriproxyfen spray, powder, and oral bait treatments on the relative abundance of fleas (Siphonaptera: Ceratophyllidae) in black-tailed prairie dog (Rodentia: Sciuridae) towns.

Separate black-tailed prairie dog, Cynomys ludovicianus (Ord), towns on the Rocky Mountain Arsenal National Wildlife Refuge, Colorado, were treated with technical pyriproxyfen (Nylar) using spray, powder, and oral bait carriers. Direct combing methods (1997 and 1998) and burrow flagging (1998) were used to estimate relative abundance of the plague vector Oropsylla hirsuta (Baker). Pyriproxyfen spray (0.05%) and powder (0.05%) did not significantly reduce (P > 0.05) O. hirsuta abundance. Pyriproxyfen bait, when applied every 4 wk at a concentration of 286 mg/50 g bait, significantly reduced (P

Insulin signaling couples growth and early maturation to cholesterol intake in Drosophila.

Nutrition is one of the most important influences on growth and the timing of maturational transitions including mammalian puberty and insect metamorphosis. Childhood obesity is associated with precocious puberty, but the assessment mechanism that links body fat to early maturation is unknown. During development, the intake of nutrients promotes signaling through insulin-like systems that govern the growth of cells and tissues and also regulates the timely production of the steroid hormones that initiate the juvenile-adult transition. We show here that the dietary lipid cholesterol, which is r

AaCAT1 of the yellow fever mosquito, Aedes aegypti: a novel histidine-specific amino acid transporter from the SLC7 family.

Insect yolk protein precursor gene expression is regulated by nutritional and endocrine signals. A surge of amino acids in the hemolymph of blood-fed female mosquitoes activates a nutrient signaling system in the fat bodies, which subsequently derepresses yolk protein precursor genes and makes them responsive to activation by steroid hormones. Orphan transporters of the SLC7 family were identified as essential upstream components of the nutrient signaling system in the fat body of fruit flies and the yellow fever mosquito, Aedes aegypti. However, the transport function of these proteins was un

The somatic sex determines the requirement for ovarian tumor gene activity in the proliferation of the Drosophila germline.

Gametogenesis in Drosophila requires sex-specific interactions between the soma and germline to control germ cell viability, proliferation, and differentiation. To determine what genetic components are involved in this interaction, we examined whether changes in the sexual identity of the soma affected the function of the ovarian tumor (otu) and ovo genes. These genes are required cell autonomously in the female germline for germ cell proliferation and differentiation. Mutations in otu and ovo cause a range of ovarian defects, including agametic ovaries and tumorous egg cysts, but do not affec

Inhibition of the Ca(2+)-activated K(+)-channel by sapecin B, an insect antibacterial protein.

Sapecin is an antibacterial protein of the flesh fly and sapecin B is its homologue structurally similar to charybdotoxin of scorpion venom, which is known to be a K+ channel inhibitor. We found that, like charybdotoxin, sapecin B inhibits part of the voltage pulse-induced K+ currents of rat cerebellar Purkinje cells. We suggest that this effect is due to inhibition of the Ca(+)-activated K+ channel. Probably, sapecin B is a naturally occurring K+ channel inhibitor as well as an antibacterial protein.

[Factors affecting the efficacy of 2 growth regulator compounds, OMS 3010 (phenoxycarb) and OMS 3019 (ethoxypyridine), on larvae of the Simulium damnosum complex].

Two insect growth regulators (IGR'S) acting as juvenile hormone analogs (JHA'S), OMS 3010 (phenoxycarb) and OMS 3019 (ethoxypyridine) were selected after a screening of several compounds proposed by industry on black fly larvae of the Simulium damnosum complex (Diptera: Simuliidae) the vector of onchocerciasis in West Africa. Bioassays were designed to study two parameters: larval age and exposure time. The results showed that these compounds are more efficient on old larvae (6th and 7th instars) than young larvae (3rd, 4th and 5th instars). However, exposure time seems essential. The black fl

Stimulation of JH biosynthesis by the corpora allata of adult female Aedes aegypti in vitro: effect of farnesoic acid and Aedes allatotropin.

Previous studies have demonstrated that the synthesis of juvenile hormone (JH) by the isolated corpora allata (CA) complex in vitro as well as the JH titer in the yellow fever mosquito Aedes aegypti are elevated before feeding and low after a blood meal. In the present study, we used an in vitro radiochemical assay to analyze the effect of farnesoic acid (FA) and Aedes allatotropin (Aedes-AT) on the biosynthesis of JH and methyl farnesoate (MF) by the isolated CA complex of A. aegypti adult female. CA complex from day-0 females (0-1 h after emergence) exhibited a low basal juvenile hormone III

Delayed diagnosis of T3 supplementation in a bodybuilder presenting with tachycardia and features of sepsis.

A 25-year-old man presented generally unwell to the emergency department. Initial assessment identified systemic inflammatory response syndrome markers with an insect bite as a potential source of infection and he was treated for presumed sepsis. Tachycardia persisted and baseline thyroid function testing showed undetectable free thyroxine and thyroid-stimulating hormone (TSH), prompting further endocrine investigation. Triiodothyronine (T3) was markedly raised with normal TSH receptor antibodies, and the patient later confessed to supplementary testosterone and T3 use as part of bodybuilding
