Transcriptomic dynamics reveals sequential acquisition of complement resistance during prolonged starvation of Trypanosoma cruzi epimastigote.

BACKGROUND: The life cycle of the parasitic protozoan Trypanosoma cruzi, the etiological agent of Chagas disease (CD), includes two well-recognised insect-dwelling stages: the replicative non-infective epimastigotes and the non-replicative infective metacyclic trypomastigotes. Nonetheless, the existence of multiple intermediate forms has been reported. Since nutrient restriction is considered one of the main factors driving metacyclogenesis and is very frequent due to the long-term starvation periods that the insect vectors commonly undergo, we have studied the transcriptomic effects of nutrie

Changes in amounts of total salivary gland proteins of Lutzomyia longipallpis (Diptera: Psychodidae) according to age and diet.

Saliva plays important roles in facilitation of a bloodmeal, lubrication of mouthparts, and parasite transmission for some vector insects. Salivary composition changes during the lifetime of an insect, and differences in the salivary profile may influence its functions. In this report, the amount and profile of salivary gland protein of the American visceral leishmaniasis vector Lutzomyia longipalpis (Lutz & Neiva, 1912) were analyzed at different times of insect development and diet. Protein content from unfed female sand flies increased significantly with age, and a significant difference wa

Characterization of a leucokinin binding protein in Aedes aegypti (Diptera: Culicidae) Malpighian tubule.

The insect myokinin (leucokinin-like) neuropeptide family includes peptides that have different physiological effects such as the induction of hindgut myotropic activity and stimulation of urine production. The C-terminal pentamer of myokinins Phe-X-(Ser/Pro/Ala)-Trp-Gly-amide [X=Phe, His, Asn, Ser or Tyr], had been previously determined as the minimum fragment able to elicit a functional response. The receptor(s) for these insect neuropeptides has not yet been identified. In order to characterize the Malpighian tubule leucokinin-like peptide receptor(s) from the yellow fever mosquito (Aedes a

Effect of Wolbachia Infection and Adult Food on the Sexual Signaling of Males of the Mediterranean Fruit Fly Ceratitis capitata.

Sexual signaling is a fundamental component of sexual behavior of Ceratitis capitata that highly determines males' mating success. Nutritional status and age are dominant factors known to affect males' signaling performance and define the female decision to accept a male as a sexual partner. Wolbachia pipientis, a widespread endosymbiotic bacterium of insects and other arthropods, exerts several biological effects on its hosts. However, the effects of Wolbachia infection on the sexual behavior of medfly and the interaction between Wolbachia infection and adult food remain unexplored. This stud

Drosophila X virus-like particles as delivery carriers for improved oral insecticidal efficacy of scorpion Androctonus australis peptide against the invasive fruit fly, Drosophila suzukii.

Insect-specific neurotoxic peptides derived from the venoms of scorpions and spiders can cause acute paralysis and death when injected into insects, offering a promising insecticidal component for insect pest control. However, effective delivery systems are required to help neurotoxic peptides pass through the gut barrier into the hemolymph, where they can act. Here, we investigated the potential of a novel nanocarrier, Drosophila X virus-like particle (DXV-VLP), for delivering a neurotoxin from the scorpion Androctonus australis Hector (AaIT) against the invasive pest fruit fly, Drosophila su

The whole genome sequence of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann), reveals insights into the biology and adaptive evolution of a highly invasive pest species.

BACKGROUND: The Mediterranean fruit fly (medfly), Ceratitis capitata, is a major destructive insect pest due to its broad host range, which includes hundreds of fruits and vegetables. It exhibits a unique ability to invade and adapt to ecological niches throughout tropical and subtropical regions of the world, though medfly infestations have been prevented and controlled by the sterile insect technique (SIT) as part of integrated pest management programs (IPMs). The genetic analysis and manipulation of medfly has been subject to intensive study in an effort to improve SIT efficacy and other as

Real-time cell analysis and heat shock protein gene expression in the TcA Tribolium castaneum cell line in response to environmental stress conditions.

The rust red flour beetle, Tribolium castaneum (Herbst, 1797) (Coleoptera: Tenebrionidae), is a pest of stored grain and one of the most studied insect model species. Some of the previous studies involved heat response studies in terms of survival and heat shock protein expression, which are regulated to protect other proteins against environmental stress conditions. In the present study, we characterize the impedance profile with the xCELLigence Real-Time Cell Analyzer and study the effect of increased temperature in cell growth and viability in the cell line BCIRL-TcA-CLG1 (TcA) of T. castan

Review on Structures of Pesticide Targets.

Molecular targets play important roles in agrochemical discovery. Numerous pesticides target the key proteins in pathogens, insect, or plants. Investigating ligand-binding pockets and/or active sites in the proteins' structures is usually the first step in designing new green pesticides. Thus, molecular target structures are extremely important for the discovery and development of such pesticides. In this manuscript, we present a review of the molecular target structures, including those of antiviral, fungicidal, bactericidal, insecticidal, herbicidal, and plant growth-regulator targets, curre

Oxidative stress from environmental pollutants.

Recently progress has been made on O2 toxicity and pathology related to numerous environmental contaminants in insects. The pro-oxidants studied included: dioxin, paraquat, and an assorted array of quinones, 8-methoxypsorlen, arsenic, and mercury. The responses to these oxidants are diverse, but they arise from the reactive oxygen species. These pro-oxidants in insects cause lipid peroxidation, protein and enzyme oxidation, and GSH depletion. Potentially, they may also cause DNA oxidation, and form DNA adducts. Oxidative challenge is alleviated by antioxidant compounds, but more importantly by

Antisense peptide nucleic acids targeting ribosomal protein genes: A novel strategy against carbapenem-resistant Acinetobacter baumannii.

Carbapenem-resistant Acinetobacter baumannii (CRAB) is a nosocomial pathogen associated with substantial morbidity and mortality globally. Due to its high resistance to most antimicrobial agents, including carbapenems, novel treatment strategies are urgently required for CRAB infection. We explored peptide nucleic acids (PNAs) targeting essential ribosomal proteins against CRAB. Potential targets were identified using the Database of Essential Genes, and a cell-penetrating peptide (CPP)-PNA conjugate library was synthesized. The PNAs were screened using colony-forming unit counts and minimum i

Harnessing next-generation molecular technologies for precision control of Tuta absoluta and Helicoverpa armigera in tomato.

Tomato (Solanum lycopersicum) is one of the most significant vegetable crops cultivated around the world. Among biotic stresses, production of tomato is severely affected by insect pests mainly by tomato fruit borer (Helicoverpa armigera) and tomato leaf miner (Tuta absoluta). Traditionally to manage the destruction caused by insect pests, farmers mostly used chemical insecticides and by different cultural practices. Use of chemical pesticides is of great concern as it is harmful for the environment and other living beings and with the passage of time, insects develop resistance against pestic

Lipophorin levels in the yellow fever mosquito, Aedes aegypti, and the effect of feeding.

High density lipophorin (HDLp) is the major lipid transport vehicle in insect hemolymph. Using an indirect ELISA, levels of HDLp were measured in the yellow fever mosquito, Aedes aegypti. The level of lipophorin, when normalized to the total weight of the insect, was similar in the different developmental stages. Starvation (access to water only) of adult females did not affect the level of HDLp nor its density when compared to sugar-fed females. On the other hand, blood feeding (of normally sugar-fed females) resulted in a three-fold increase of the HDLp level at 40 h after feeding. This incr

Participation of 14-3-3ε and 14-3-3ζ proteins in the phagocytosis, component of cellular immune response, in Aedes mosquito cell lines.

BACKGROUND: Better knowledge of the innate immune system of insects will improve our understanding of mosquitoes as potential vectors of diverse pathogens. The ubiquitously expressed 14-3-3 protein family is evolutionarily conserved from yeast to mammals, and at least two isoforms of 14-3-3, the ε and ζ, have been identified in insects. These proteins have been shown to participate in both humoral and cellular immune responses in Drosophila. As mosquitoes of the genus Aedes are the primary vectors for arboviruses, causing several diseases such as dengue fever, yellow fever, Zika and chikunguny

Identification and characterization of two chitin synthase genes in African malaria mosquito, Anopheles gambiae.

Chitin synthase (CHS) represents an attractive target site for combating insect pests as insect growth and development are strictly dependent on precisely tuned chitin biosynthesis and this pathway is absent in humans and other vertebrates. Current knowledge on CHS in insects, especially their structures, functions, and regulations is still very limited. We report the identification and characterization of two chitin synthase genes, AgCHS1 and AgCHS2, in African malaria mosquito, Anopheles gambiae. AgCHS1 and AgCHS2 were predicted to encode proteins of 1,578 and 1,586 amino acid residues, resp

Immunochemical studies of stinging insect venom allergens.

Several major venom allergens from different insects of the Hymenoptera order have been cloned and sequenced by different laboratories. These insects include fire ants, honey bees, hornets, yellowjackets and wasps. These venom allergens have different biochemical functions, but have one feature in common, their varying extents of sequence identity with other proteins in our environment. Our studies in mice suggest that recombinant fragments containing regions of sequence identity of venom allergen(s) and host protein(s) show antigenic cross-reactivity. These studies lead to the hypothesis that

Evidence for an early immunosuppressive role for related Campoletis sonorensis venom and ovarian proteins in Heliothis virescens.

Shared epitopes among venom, ovarian, and viral proteins may indicate that related proteins have similar functional roles during parasitization of Heliothis virescens by Campoletis sonorensis. Venom and ovarian proteins are introduced directly into the hemolymph during parasitization where they may target hemocytes or other components of the immune system. Polydnavirus expression has been detected in hemocytes, fat body, and other tissues but has not been detected earlier than 4 h after parasitization. Therefore, effects on hemocytes at times earlier than 4 h may not be caused by polydnavirus

Genome sequences of the human body louse and its primary endosymbiont provide insights into the permanent parasitic lifestyle.

As an obligatory parasite of humans, the body louse (Pediculus humanus humanus) is an important vector for human diseases, including epidemic typhus, relapsing fever, and trench fever. Here, we present genome sequences of the body louse and its primary bacterial endosymbiont Candidatus Riesia pediculicola. The body louse has the smallest known insect genome, spanning 108 Mb. Despite its status as an obligate parasite, it retains a remarkably complete basal insect repertoire of 10,773 protein-coding genes and 57 microRNAs. Representing hemimetabolous insects, the genome of the body louse thus p

Consumers' Perceptions about Edible Insects' Nutritional Value and Health Effects: Study Involving 14 Countries.

Insects have been consumed for time immemorial in many regions of the globe. However, in other parts, they are not traditionally eaten. Because they are a more sustainable source of animal protein and provide valuable nutrients as well as bioactive compounds with beneficial effects on the human body, their consumption is encouraged. Knowledge can serve as a tool for better acceptance of insects as food. In this context, the present work investigated the knowledge about the nutritional value and health effects of edible insects in different countries. Data were collected by employing a question

Use of lactic acid bacteria to reduce the IgE-reactivity of the arginine kinase allergen from the edible insect Hermetia illucens.

One of the main concerns associated to the consumption of edible insects is the allergenic risk. Arginine kinase is known to cause allergic reactions to mites and crustaceans, and recent studies have supported its role as an allergenic protein in edible insects. In this work, we explore the use of proliferating and non-proliferating lactic acid bacteria from two different species (Lacticaseibacillus rhamnosus and Lacticaseibacillus paracasei) to induce modification of the structure of arginine kinase, thereby reducing its IgE-reactivity. Incubation with proliferating bacteria leads to the inso

Molecular cloning, characterization and functional analysis of GluCl from the oriental armyworm, Mythimna separata Walker.

Glutamate-gated chloride channels (GluCls) mediate inhibitory synaptic transmission in invertebrate nervous systems, and only one GluCl gene has been found in insects. Therefore, insect GluCls are one of the major targets of insecticides including avermectins. In the present study, a 1347 bp full-length cDNA encoding a 449-amino acid protein (named MsGluCl, GenBank ID: MK336885) was cloned from the oriental armyworm, Mythimna separata, and characterized two alternative splicing variants of MsGluCl. The protein shares 76.9-98.6% identity with other insect GluCl isoforms. Spatial and temporal ex
