Integrative analysis and in vitro assessment of the effects of α-mangostin on high-glucose-induced inflammatory and oxidative stress responses in RAW264.7 macrophages.

BACKGROUND: Persistent macrophage-mediated inflammation under hyperglycemic conditions is an important feature of the inflammatory microenvironment associated with diabetic wounds. α-Mangostin (α-MG), a natural xanthone derived from Garcinia mangostana, has been reported to exert anti-inflammatory activity in various experimental models. However, its effects on high-glucose-induced macrophage inflammation and oxidative stress remain insufficiently characterized. OBJECTIVE: This study aimed to investigate the effects of α-MG on inflammatory and oxidative responses in high-glucose-stimulated ma

Integrated physiological, transcriptomic and widely targeted metabolomic analyses reveal the underlying mechanism of cold stress response in the panicle apical abortion line J876 derived from Oryza rufipogon.

Cold stress is a major environmental factor that significantly limits rice yield. However, the coordinated systemic responses of rice leaves and roots to cold stress remain poorly understood. In this study, we developed Oryza rufipogon introgression lines (ILs) using the recipient parent Dianjingyou 1 (DJY1). Among these lines, J876 exhibits panicle apical abortion and was selected to investigate cold tolerance at the seedling stage. Phenotypic analysis showed that J876 was significantly more sensitive to cold stress than DJY1. To elucidate the underlying mechanisms, we conducted integrated ph

Integrated Multi-Omics Exploration of the Molecular Mechanisms Underlying Differential Responses to Saline-Alkaline Stress in Leaves of Populus simonii × P. nigra.

Populus simonii × P. nigra is a valuable model for studying woody plant responses to saline-alkaline stress, however the molecular mechanisms underlying its adaptation remain poorly understood. Here, integrated transcriptomic and metabolomic analyses were used to examine the responses in leaves to low (50 mmol/L) and high (200 mmol/L) alkaline salt stress over 7 and 14 days. Physiological assessments revealed that high-concentration stress inhibited growth, induced oxidative damage, elevated malondialdehyde (MDA) content, enhanced antioxidant enzyme activity (SOD, POD, CAT), and promoted osmor

Integrated Transcriptomic and Metabolomic Analyses Reveal Physiological and Hepatic Metabolic Responses of Largemouth Bass (Micropterus salmoides) to Subacute Saline-Alkaline Stress.

Freshwater scarcity makes saline-alkaline water essential for sustainable aquaculture. Current research has primarily focused on individual salinity or alkalinity effects on fish, with limited studies addressing their interactive effects. We found significant synergistic toxicity between salinity and alkalinity (r ≈ -0.950/-0.925) in largemouth bass (Micropterus salmoides), demonstrating higher salinity levels corresponding to lower 96-h median lethal concentration (96 h LC50) values for alkalinity, and vice versa. A subsequent 56-day subacute stress trial (salinity: 6‱; alkalinity: 20 mmol/L)

Effects of dietary Bacillus subtilis 14823 on growth performance, gut barrier integrity and inflammatory response of broilers raised in a stressful tropical environment.

Heat stress (HS) has become a major concern for the poultry industry in many countries. HS impacts gut health by causing damaged mucosal microstructures, increased oxidative stress, weakened immunity, and heightened permeability to toxins and poultry pathogens. We investigated the potential benefits to broiler chickens subjected to HS of dietary supplementation with Bacillus subtilis 14823. Growth performance, gut barrier integrity, and expressions of inflammatory cytokines were analyzed. The results indicated that dietary supplementation with B. subtilis spores at concentrations of 1 × 106 CF

Appropriate Regulation of the σE-Dependent Envelope Stress Response Is Necessary To Maintain Cell Envelope Integrity and Stationary-Phase Survival in Escherichia coli.

The alternative sigma factor σE is a key component of the Escherichia coli response to cell envelope stress and is required for viability even in the absence of stress. The activity of σE increases during entry into stationary phase, suggesting an important role for σE when nutrients are limiting. Elevated σE activity has been proposed to activate a pathway leading to the lysis of nonculturable cells that accumulate during early stationary phase. To better understand σE-directed cell lysis and the role of σE in stationary phase, we investigated the effects of elevated σE activity in cultures g

FvBck1, a component of cell wall integrity MAP kinase pathway, is required for virulence and oxidative stress response in sugarcane Pokkah Boeng pathogen.

Fusarium verticillioides (formerly F. moniliforme) is suggested as one of the causal agents of Pokkah Boeng, a serious disease of sugarcane worldwide. Currently, detailed molecular and physiological mechanism of pathogenesis is unknown. In this study, we focused on cell wall integrity MAPK pathway as one of the potential signaling mechanisms associated with Pokkah Boeng pathogenesis. We identified FvBCK1 gene that encodes a MAP kinase kinase kinase homolog and determined that it is not only required for growth, micro- and macro-conidia production, and cell wall integrity but also for response

The development of persistent pain and psychological morbidity after motor vehicle collision: integrating the potential role of stress response systems into a biopsychosocial model.

OBJECTIVES: Persistent pain and psychological sequelae are common after motor vehicle collision (MVC), but their etiology remains poorly understood. Such common sequelae include whiplash-associated disorders (WAD), fibromyalgia, and posttraumatic stress disorder (PTSD). Increasing evidence suggests that these disorders share overlapping epidemiologic and clinical features. A model is proposed in which central neurobiological systems, including physiologic systems and neuroanatomical structures involved in the stress response, are an important substrate for the development of all 3 disorders an

Integrated transcriptomic and metabolomic analyses reveal coordinated molecular responses associated with nano-enabled arbuscular mycorrhizal symbiosis for salt stress tolerance in rice.

Salinity stress is a major environmental constraint limiting rice growth and yield. Seed nanopriming with calcium oxide nanomaterials (CaO NMs) in combination with arbuscular mycorrhizal fungus (AMF) has recently emerged as an effective strategy to enhance resilience, although the underlying responses remain unexplored. In the present study, rice seeds primed with CaO NMs (80 ppm) and inoculated with the AMF Claroideoglomus claroideum at transplanting were evaluated under 175 mM NaCl stress to assess their effects on plant performance and metabolic responses. The combined treatment significant

Integrative analysis of transcriptome and chromatin accessibility reveals promoter-proximal regulation and identifies candidate ABC transporters associated with cold stress responses in maize.

BACKGROUND: Low-temperature stress is a formidable environmental constraint that severely limits the growth and productivity of maize (Zea mays L.), particularly during the highly vulnerable early seedling stage. While cold tolerance is a critical agronomic objective, the integrated transcriptional and epigenetic regulatory mechanisms that govern this trait remain largely elusive. Characterizing these coordinated molecular networks is fundamental to the genetic enhancement of cold resilience in maize. METHODS: Using two maize inbred lines contrasting in chilling response (ZHB12 tolerant, B73

Integrated analysis of structure, physiology, transcriptome and metabolome reveals key metabolic pathway responses of cold-tolerant pear cultivar germplasms under low-temperature stress.

Freezing injury during winter is a critical abiotic stress that severely impacts the growth, development and, fruit quality of deciduous fruit trees. Cold tolerance can be induced through seasonal cold acclimation, which involves coordinated adjustments in tissue structure, physiology, and biochemistry driven by natural low-temperature, with distinct strategies across plant species. However, the cold-tolerant mechanisms of pear trees during cold acclimation remain poorly understood. Here, one-year-old branches of 10 pear cultivar germplasms were evaluated for cold tolerance based on the semi-l
