Sodium channel Nav1.6 involved in modulating isoflurane-induced perioperative cognitive disorder of mice.

Perioperative neurocognitive disorder (PND), a major contributor to poor postoperative outcomes and excessive healthcare costs, has been associated with isoflurane inhalation, although the underlying mechanisms remain poorly defined. Voltage-gated sodium channels (VGSCs or Nav) have been implicated in mediating the anesthetic effects of isoflurane. We previously reported that the Nav1.6 subtype modulates neural network activity and cognitive function. Here we investigated whether Nav1.6-mediated network disturbances contribute to isoflurane-induced PND. In the present study, we observed an inc

Cerebral hyperaemia after isoflurane anaesthesia for craniotomy of patients with supratentorial brain tumour.

BACKGROUND: Few studies look into cerebral blood flow (CBF) changes during emergence from general anaesthesia for craniotomy. The purpose of this study was to assess CBF changes during emergence from general anaesthesia for craniotomy, through monitoring blood oxygen saturation of jugular vein bulb (SjvO2 ) and transcranial Doppler (TCD). METHODS: We enrolled 30 patients undergoing selective craniotomy (group C) for supratentorial brain tumour resection and 30 patients undergoing selective abdominal surgery (group A). Mean velocity of middle cerebral artery (Vmca), mean arterial pressure (MAP

Ketamine-xylazine-acepromazine compared with isoflurane for anesthesia during liver transplantation in rodents.

Orthotopic liver transplantation in mice and rats is used to study a wide range of scientific questions. Inhalant anesthesia is widely used in liver transplantation models in rodents, but drawbacks of inhalant anesthetics include issues of cost, safety, and ease of use. The goal here was to find an effective injection anesthesia protocol that would not directly influence metabolic or functional parameters after liver transplantation. We compared intraperitoneal injection of a ketamine-xylazine-acepromazine cocktail (KXA) with isoflurane during 50% liver transplantation in mice and rats. Anesth

Sevoflurane provides better recovery as compared with isoflurane in children undergoing spinal surgery.

Rapid recovery is desirable in pediatric neurosurgical patients to obtain an early neurologic assessment. We compared the recovery characteristics of 2 commonly used anesthetic agents, sevoflurane and isoflurane, under bispectral index-guided anesthesia in children undergoing spinal surgery. Eighty children who underwent surgery for occult spinal dysraphism at the lumbar and sacral level were randomized to anesthesia with sevoflurane or isoflurane in oxygen and nitrous oxide. Anesthesia depth was guided by a bispectral index monitor kept between 40 and 60. In addition to time at emergence, ext

gamma-Aminobutyric acid-A receptors contribute to isoflurane neuroprotection in organotypic hippocampal cultures.

UNLABELLED: The mechanisms by which anesthetics such as isoflurane reduce cell death in rodent models of cerebral ischemia remain incompletely defined. Reduction in glutamate excitotoxicity explains some but not all of isoflurane's neuroprotection. Because isoflurane potentiates gamma-aminobutyric acid (GABA) receptor-mediated ion fluxes and GABA(A) receptor agonists have neuroprotective effects, we hypothesized that GABA(A) receptors contribute to isoflurane neuroprotection. As a model of cerebral ischemia and recovery, we used rat hippocampal slice cultures. Survival of CA1, CA3, and dentate

Isoflurane preconditions myocardium against infarction via activation of inhibitory guanine nucleotide binding proteins.

BACKGROUND: Isoflurane-induced myocardial protection during ischemia is mediated by adenosine triphosphate-regulated potassium (KATP) channels; however, the intracellular signal transduction cascade responsible for this process has been incompletely evaluated. The authors tested the hypothesis that isoflurane reduces myocardial infarct size through a Gi protein-mediated process. METHODS: Forty-eight hours after pretreatment with vehicle (0.9% saline) or the Gi protein inhibitor pertussis toxin (10 microg/kg intravenously), barbiturate-anesthetized dogs (n = 43) were instrumented for measureme

Isoflurane anesthesia blunts cerebral responses to noxious and innocuous stimuli: a fMRI study.

We used functional magnetic resonance imaging to determine how isoflurane affected cerebral neuronal activation resulting from noxious and innocuous stimuli. Five male volunteers were subjected to mild electrical shock and tactile stimuli applied to the hand. During low (0.7%) and moderate (1.3%) isoflurane anesthesia the stimuli were repeated and a supramaximal electrical shock was also applied. Tactile stimulation activated bilateral SI and SII, but resulted in no significant activation at low or moderate anesthesia. Electrical shock activated contralateral SI and bilateral SII; low anesthes

Isoflurane and halothane produce similar alterations in aortic distensibility and characteristic aortic impedance.

The effects of volatile anesthetics on arterial mechanical properties in vivo are incompletely understood. The present investigation tested the hypothesis that isoflurane and halothane cause differential actions on the mechanical characteristics of the aorta in barbiturate-anesthetized dogs instrumented for measurement of aortic blood pressure, blood flow, and diameter. Aortic distensibility and characteristic aortic impedance were calculated from aortic diameter measurements. Oscillatory (Wo) and mean (Wm) hydraulic power were determined from aortic pressure and blood flow waveforms. Hemodyna

Humidification reduces coughing and breath-holding during inhalation induction with isoflurane in children.

PURPOSE: Inhalation induction using isoflurane is associated with airway irritability, coughing, breath-holding and laryngospasm. These complications are more common in children. This study was designed to determine if humidification of isoflurane in oxygen/nitrous oxide would reduce respiratory complications and hypoxic episodes at induction. METHODS: Fifty-nine unpremedicated children, aged three months to 12 yr, were enrolled in the study and randomised to receive either humidified isoflurane (n = 27, Group A), or non-humidified isoflurane (n = 32, Group B). Isoflurane inhibits hypoxic pulmonary vasoconstriction. An in vivo fluorescence microscopic study in rabbits.

BACKGROUND: Contradictory results have been reported in previous studies investigating the effect of isoflurane on hypoxic pulmonary vasoconstriction by indirect approaches. The current study measured the effects of one-lung ventilation (1LV) and isoflurane 1.5% by direct visual observation of the pulmonary microcirculation. METHODS: Ten New Zealand White rabbits were anesthetized with intravenous thiopental, alpha-chloralose, and piritramid. Arterial, central venous, pulmonary arterial, left atrial, and airway pressures and cardiac output were recorded continuously. 1LV was facilitated by a

Suspected malignant hyperthermia syndrome in a miniature pot-bellied pig anesthetized with isoflurane.

Hyperthermia developed in a 7-week-old Vietnamese pot-bellied pig after isoflurane anesthesia for routine ovariohysterectomy. Shivering and a sudden increase in heart rate were noticed 90 minutes after anesthetic induction, both of which persisted throughout the remaining anesthetic period. Rectal temperature increased from 37 C immediately after anesthetic recovery to a peak of 41.5 C, and remained high for 18 hours. Treatment included external cooling and treatment with dantrolene sodium. Malignant hyperthermia was suspected to be the cause of the clinical signs in this pig, but specific tes

Central circulation during halothane-diethyl-ether azeotrope and isoflurane anaesthesia in the pig.

The fatal anaesthetic ratio (FAR) has been determined for halothane-diethyl-ether (HE) in pigs, and the central circulation during hypovolaemia has been investigated using a well-documented agent such as isoflurane as a standard. The fatal anaesthetic ratio for HE in pigs was (3.21/1.03) = 3.12. This is high compared to the FAR for halothane of 1.7. The central circulation was investigated in 12 pigs which were randomly allocated to either HE or isoflurance anaesthesia, respectively. Baseline values were recorded when they were stable at 1.3 MAC of the volatile anaesthetic used. The pigs were

Haemodynamic and catecholamine response to isoflurane anaesthesia in patients undergoing coronary artery surgery.

Haemodynamic and plasma catecholamine responses were evaluated during isoflurane anaesthesia in ten patients undergoing coronary artery bypass surgery. Following thiopentone induction the patients were anaesthetized with isoflurane 1.5-2.0 per cent in oxygen. The results show that after 10 minutes of isoflurane anaesthesia there was a significant increase from baseline in heart rate, 68 to 80; cardiac output, 3.75 to 4.61; and plasma epinephrine, 0.80 to 1.33 microgram/l. Conversely, there was a significant reduction in systemic vascular resistance index, 3388 to 2260, and plasma norepinephrin

Metabolic Differences in Neuroimaging with [18F]FDG in Rats Under Isoflurane and Hypnorm-Dormicum.

BACKGROUND: Anesthesia can significantly impact positron emission tomography (PET) neuroimaging in preclinical studies. Therefore, understanding these effects is crucial for accurate interpretation of the results. In this experiment, we investigate the effect of [18F]-labeled glucose analog fluorodeoxyglucose ([18F]FDG) uptake in the brains of rats anesthetized with two commonly used anesthetics for rodents: isoflurane, an inhalation anesthetic, and Hypnorm-Dormicum, a combination injection anesthetic. Brief isoflurane administration as an adjunct treatment to control organophosphate-induced convulsions and neuropathology.

Organophosphate-based chemical agents (OP), including nerve agents and certain pesticides such as paraoxon, are potent acetylcholinesterase inhibitors that cause severe convulsions and seizures, leading to permanent central nervous system (CNS) damage if not treated promptly. The current treatment regimen for OP poisoning is intramuscular injection of atropine sulfate with an oxime such as pralidoxime (2-PAM) to mitigate cholinergic over-activation of the somatic musculature and autonomic nervous system. This treatment does not provide protection against CNS cholinergic overactivation and ther
