Enhancing selective esterification of conjugated linoleic acid isomers by immobilized Candida rugosa lipase via hydrophobic acyl acceptors.

An immobilized Candida rugosa lipase on mesoporous silica functionalized with C18 alkyl chains (MS-C18) was employed to construct a non-aqueous selective esterification system for the resolution of conjugated linoleic acid (CLA) isomers. The effects of acyl acceptors with different hydrophobicities on esterification performance and isomer selectivity were investigated in isooctane. Under controlled reaction conditions (CLA-to-alcohol molar ratio 1:1, 40 °C), increasing acyl acceptor hydrophobicity enhanced both the esterification rate and the differentiation of reaction rates between c9,t11-CL

Effects of Dietary Conjugated Linoleic Acid on the Growth Performance of Fattening Broiler Duck and the Properties of Its Dry-Cured Products.

The effects of dietary supplementation with conjugated linoleic acid (CLA) on broiler duck growth performance and the quality of dry-cured products were investigated. Ten days preslaughter with the supplementation of 2 mL/d CLA was more economical and effective. This feeding protocol did not affect growth performance, with CLA accumulation amounting to 3.61 g/100 g. Histopathological observation of tissue morphology revealed that CLA alleviated intestinal damage with no adverse morphological changes in the heart, spleen, liver, or pancreas. Gut microbiota analysis demonstrated that the benefic

L-Theanine Mitigates Chronic Alcoholic Intestinal Injury by Regulating Intestinal Alcohol and Linoleic-Arachidonic Acid Metabolism in Rats.

Background: Chronic alcohol intake impairs intestinal function, while L-theanine (LTA) may support intestinal health. However, the protective effects of LTA to chronic alcoholic intestinal injuries remain unclear. Methods: SD rats were administered LTA for 8 weeks and then co-administered Lieber-DeCarli liquid alcohol feed and LTA for 4 weeks to establish a chronic alcoholic intestinal injury model and investigate the mitigating influence of LTA on chronic alcoholic intestinal injury. Results: LTA alleviated duodenal pathology and intestinal permeability injury and reduced intestinal oxidative

Mycobacterium tuberculosis-derived linoleic acid increases regulatory T cell function to promote bacterial survival within macrophages.

Regulatory T (Treg) cells expand during Mycobacterium tuberculosis (Mtb) infection and suppress T cell-mediated control. Whether Mtb actively contributes to this process is unclear. Here, using a genome-wide mutant library, we show that the expression of Mtb Rv1272c, an ATP-binding cassette transporter, increased under hypoxic conditions and promotes Mtb survival in vivo by increasing lecithin import, followed by the production and release of linoleic acid. Linoleic acid released by infected macrophages promoted surface trafficking of the immune checkpoint molecule cytotoxic T lymphocyte antig

Multi-omics analysis reveals that linoleic acid metabolism is associated with variations of trained immunity induced by distinct BCG strains.

Trained immunity is one of the mechanisms by which BCG vaccination confers persistent nonspecific protection against diverse diseases. Genomic differences between the different BCG vaccine strains that are in global use could result in variable protection against tuberculosis and therapeutic effects on bladder cancer. In this study, we found that four representative BCG strains (BCG-Russia, BCG-Sweden, BCG-China, and BCG-Pasteur) covering all four genetic clusters differed in their ability to induce trained immunity and nonspecific protection. The trained immunity induced by BCG was associated

Linoleic Acid-Derived Oxylipins Differentiate Early Stage Alcoholic Hepatitis From Mild Alcohol-Associated Liver Injury.

Alcohol-associated liver disease (ALD) is a spectrum of liver disorders ranging from steatosis to steatohepatitis, fibrosis, and cirrhosis. Alcohol-associated hepatitis (AH) is an acute and often severe form of ALD with substantial morbidity and mortality. The mechanisms and mediators of ALD progression and severity are not well understood, and effective therapeutic options are limited. Various bioactive lipid mediators have recently emerged as important factors in ALD pathogenesis. The current study aimed to examine alterations in linoleic acid (LA)-derived lipid metabolites in the plasma of

High conjugated linoleic acid enriched ghee (clarified butter) increases the antioxidant and antiatherogenic potency in female Wistar rats.

BACKGROUND: Hypercholesterolemia and oxidative stress are the main stimulating factors responsible for coronary artery disease and progression of atherosclerosis. Dairy food products are rich in conjugated linoleic acid (CLA) which is considered as an important component due to its potential health benefits such as anticarcinogenic, antiatherogenic, antidiabetic and antiadipogenic properties. In the present study, the effect of CLA enriched ghee on the antioxidant enzyme system and antiatherogenic properties in Wistar rats has been studied. METHODS: Female Wistar rats of 21 days were taken fo

Topical clindamycin 1% vs. linoleic acid-rich phosphatidylcholine and nicotinamide 4% in the treatment of acne: a multicentre-randomized trial.

It has been shown that topical linoleic acid rich-phosphatidylcholine seems to be effective in normalization of follicular hyperkeratinization, whereas nicotinamide has an interesting anti-inflammatory effect. However, little is known about their combined effect on acne. A multicentre, double-blind, 12-week randomized vehicle and parallel-active control study was conducted by clinical and biophysical non-invasive measurements to evaluate the efficacy, tolerability and safety of a 4% nicotinamide-phospholipidic (N-PHCL) emulsion vs. 1% topical clindamycin phosphate applied once daily. trans-10,cis-12 conjugated linoleic acid prevents adiposity but not insulin resistance induced by an atherogenic diet in hamsters.

While conjugated linoleic acid (CLA) has received a great deal of attention as a supplement that can favourably modify body composition, its potential impact on insulin sensitivity has not received equal attention. The aim of the present work was to analyse the effects of trans-10,cis-12 CLA isomer on insulin sensitivity in hamsters fed an atherogenic diet. Hamsters were divided into three groups: one group was fed a chow diet (control) and the other two a semipurified atherogenic diet supplemented with 0.5% linoleic acid (LA) or trans-10,cis-12 CLA, respectively. Serum glucose, FFAs, insulin,

Inhibitory effect of linoleic acid on chain elongation and desaturation of 18:2 c,t isomers in lactating and neonatal rats.

The previous studies showed that dietary 18:2 c,t isomers could be chain-elongated and desaturated to produce unusual 20:4 isomers. The present study was undertaken to determine the minimal amount of 18:2n-6 required to suppress the chain elongation and desaturation of 18:2 c,t isomers in the lactating and neonatal rats when animals were fed 15% partially hydrogenated canola oil diet containing 1.72% energy as 18:2 c,t isomers and varying amounts of free 18:2n-6. These diets induced marginal essential fatty acid (EFA) deficiency states (0.56% energy 18:2n-6) to EFA adequacy (2.56% energy 18:2n

Detection of 11 beta-hydroxysteroid dehydrogenase type 1, the glucocorticoid and mineralocorticoid receptor in various adipose tissue depots of dairy cows supplemented with conjugated linoleic acids.

Early lactating cows mobilize adipose tissue (AT) to provide energy for milk yield and maintenance and are susceptible to metabolic disorders and impaired immune response. Conjugated linoleic acids (CLA), mainly the trans-10, cis-12 isomer, reduce milk fat synthesis and may attenuate negative energy balance. Circulating glucocorticoids (GC) are increased during parturition in dairy cows and mediate differentiating and anti-inflammatory effects via glucocorticoid (GR) and mineralocorticoid receptors (MR) in the presence of the enzyme 11β-hydroxysteroid dehydrogenase type 1 (11βHSD1). Activated …

Using mixtures of linoleic acid, a long-chain polyunsaturated fatty acid, and its monoglyceride in lipid-based formulations to increase drug loading and enhance aqueous microemulsification.

Although lipid-based formulations (LBFs) of self-emulsifying drug delivery systems (SEDDS) have been used for poorly water-soluble drugs, they have many limitations, the first being the low solubility of drugs in lipids, which limits drug loading in formulations, and the second being inadequate dispersion of formulations in the aqueous fluids of the human gastrointestinal tract (GIT). Large amounts of organic cosolvents and surfactants, up to 90% of the formulation, have been used to overcome these limitations. We previously demonstrated that the situation could be improved by using lipid dige

Comparison of shock wave therapy and nutraceutical composed of Echinacea angustifolia, alpha lipoic acid, conjugated linoleic acid and quercetin (perinerv) in patients with carpal tunnel syndrome.

Even though the initial treatment of carpal tunnel syndrome (CTS) is conservative, knowledge of the clinical effects of supplements and of some methods of physiotherapy is still preliminary. Many biological mechanisms can support the administration of shock wave therapy (ESWT) or of alpha lipoic acid (ALA) based nutraceutical, conjugated linoleic acid (GLA), anti-oxidants and Echinacea angustifolia for CTS. The shock waves reduce the nerve compression, produce an anti-inflammatory action, and accelerate the regeneration of neuropathy. ALA and GLA induce antioxidant protective actions, reduce i

Glucose metabolism and the somatotropic axis in dairy cows after abomasal infusion of essential fatty acids together with conjugated linoleic acid during late gestation and early lactation.

Sufficient glucose availability is crucial for exploiting the genetic potential of milk production during early lactation, and endocrine changes are mainly related to repartitioning of nutrient supplies toward the mammary gland. Long-chain fatty acids, such as essential fatty acids (EFA) and conjugated linoleic acid (CLA), have the potential to improve negative energy balance and modify endocrine changes. In the present study, the hypothesis that combined CLA and EFA treatment supports glucose metabolism around the time of calving and stimulates insulin action and the somatotropic axis in cows

Oxidized metabolites of linoleic acid as biomarkers of pancreatic beta-cell function in the first-onset type 2 diabetic patients with obesity.

OBJECTIVES: The role of oxylipins in type 2 diabetes (T2D) with obesity remains poorly understood. This study aimed to characterize oxylipin profiles in T2D patients with obesity, identify associations with pancreatic beta-cell function, and suggest potential therapeutic targets. METHODS: We recruited first-onset T2D patients with obesity (OB group, n = 15) and normal-weight T2D patients (NW group, n = 15), matched by gender, age, and blood lipid levels. Serum samples were collected in a fasting state before medication. Oxylipin profiling was analyzed using liquid chromatography-tandem mass sp

Resveratrol ameliorates PM2.5-aggravated IBS-D mouse symptoms by restoring the gut bacteria and hypoxanthine/linoleic acid metabolism homeostasis.

Fine particulate matter (PM2.5) is an emerging environmental risk factor for gut disorders. Still, the toxicity of PM2.5 in diarrhea-predominant irritable bowel syndrome (IBS-D) mice and the intervention role of resveratrol remain unclear. This study integrated non-targeted metabolomics and gut microbiota analysis in mice of IBS-D, PM2.5 exposure, and resveratrol + PM2.5 + IBS-D. The colonic histopathology, oxidative stress, colon barrier function, and visceral hypersensitivity were detected. Results showed that PM2.5 significantly exacerbated IBS-D symptoms in mice. Such effects were accompan

Linoleic Acid Enhances Murine Natural Killer Cell Function In Vitro but Compromises Mitochondrial Bioenergetic Function During Retroviral Infection.

Natural killer (NK) cells are innate immune cells that eliminate virus-infected and tumour cells by a plethora of signalling pathways, involving the production of cytokines and cytotoxic molecules. NK cell activation and functions demand substantial energy and fatty acids are energy-rich molecules reported to be important for NK cell functions upon viral infections. However, it remains unclear whether NK cells preferentially take up a specific fatty acid. Here, we show in the acute Friend retrovirus infection of mice that linoleic acid (LA) exerts a dual role in modulating NK cells. Interestin

Synthesis, characterization and evaluation of linoleic acid-loaded nano-carriers: a novel drug delivery system for efficient treatment of hepatocellular carcinoma.

Hepatocellular carcinoma is the main primary liver cancer. Due to the high recurrence rate and potential resistance to treatments, there is an urgent need to find more effective and targeted therapies. Combinational therapies and using advanced drug delivery methods are promising approaches. Niosomes are one of the popular types of nanoparticles in modern drug delivery systems and have recently attracted more attention. They are biodegradable, nonimmunogenic, and more stable carriers than liposomes. They can release anticancer drugs in an efficient and controlled manner. The aim of this study
