A pH responsive nanocomposite for combination sonodynamic-immunotherapy with ferroptosis and calcium ion overload via SLC7A11/ACSL4/LPCAT3 pathway.

Sonodynamic therapy offers a non-invasive approach to induce the death of tumor cells. By harnessing ultrasound waves in tandem with sonosensitizers, this method produces reactive oxygen species (ROS) that inflict oxidative damage upon tumor cells, subsequently causing their demise. Ferroptosis is a regulatory form of cell death that differs from other forms, characterized by iron accumulation, ROS accumulation, and lipid peroxidation. In the presented research, a nanoparticle formulation, parthenolide/ICG-CaCO3@lipid (PTL/ICG-CaCO3@Lip), has been engineered to amplify ferroptosis in tumor cel

A dual PROTAC nanocarrier amplifies DNA damage and STING activation for cancer immunotherapy.

The cytoplasmic accumulation of damaged DNA to activate the STING pathway has emerged as a promising strategy to enhance tumor immunogenicity and improve the efficacy of immune checkpoint blockade (ICB) therapy. Herein, a stimuli-responsive and dual PROTAC-embedded immunoactivator (denoted as Sd@Lip) is developed to potentiate ICB through amplified DNA damage and robust STING activation. Sd@Lip comprises an acid-sensitive liposomal nanocarrier co-encapsulating two targeted degraders of dBET1 (a BRD4 degrader) and SK-575 (a PARP1 degrader), which enhances drug solubility, stability, and enable

Direct Anthropometry of Repaired Bilateral Complete Cleft Lip: A Long-Term Assessment.

BACKGROUND: Repair of bilateral complete cleft lip requires more than following a design in a textbook. The operative strategy should incorporate knowledge of anticipated nasolabial changes with growth. METHODS: Direct nasolabial anthropometry was recorded in 174 children with bilateral complete cleft lip before and immediately after primary repair. Serial measurements were taken in 66 Caucasian patients between the ages of 6 months and 15 years and compared to Farkas' normal values. Data for upper philtral width were unavailable; therefore, this dimension was measured in 454 nonsyndromic Cau

Secondary Narrow Nostril Deformity After Unilateral Cleft Lip Repair: Etiology, Classification, and Surgical Strategies.

Secondary narrow nostril deformity after unilateral cleft lip repair remains a stubborn clinical challenge, with an incidence of approximately 84%. It compromises both nasal airway function and facial esthetics. The etiology is multifactorial, involving congenital tissue deficiencies, abnormal orbicularis oris insertion, and iatrogenic deformities resulting from primary repair. Current reconstructive approaches can be broadly categorized into tissue augmentation, structural reinforcement, and contour-modifying strategies, with postoperative nasal retainers serving as a key adjunct device. [Peribuccal esthetic surgery].

This paper concerns the procedures of remodelling the peribuccal area for aesthetic purposes; these techniques may be planned in isolation or as a complement to a face-lift. In our hands these interventions are helpful firstly to correct and excessively deep nasolabial fold and sublabial crease using dermis or fascia grafts. Secondly, are designed to correct the contour of the upper lip either by smoothening the fine vertical folds, excising a fine strip of skin along the cupid's bow associated with dermabrasion or by diminishing the labial height resecting a buffalo-horn shaped cutaneous stri

Management of Skeletal Class III Malocclusion in an Adolescent Using Maxillary Skeletal Expander and Facemask - A Case Report.

Skeletal Class III malocclusion in adolescents can be effectively managed using the combination of maxillary skeletal expander (MSE) and facemask (FM). An adolescent presented concerns about her lower front teeth positioned ahead of the upper front teeth, difficulty closing the lips, and dissatisfaction with the facial appearance. The patient exhibited a skeletal Class III profile, reverse overjet, a 2 mm overbite, concave facial profile, midface retrusion, mandibular prognathism, chin deviation to the right, and increased lower facial height (LFH). Management involved the use of MSE and protr

Measuring quality of life in cleft lip and palate patients: currently available patient-reported outcomes measures.

BACKGROUND: Patient-reported outcomes in cleft lip and palate treatment are critical for patient care. Traditional surgical outcomes focused on objective measures, such as photographs, anatomic measurements, morbidity, and mortality. Although these remain important, they leave many questions unanswered. Surveys that include aesthetics, speech, functionality, self-image, and quality of life provide more thorough outcomes assessment. It is vital that reliable, valid, and comprehensive questionnaires are available to craniofacial surgeons. METHODS: The authors performed a literature review to id

Total mandibular subapical osteotomy and Le Fort I osteotomy using piezosurgery and computer-aided designed and manufactured surgical splints: a favorable combination of three techniques in the management of severe mouth asymmetry in Parry-Romberg syndrome.

Although its pathogenesis remains obscure, Parry-Romberg syndrome (PRS) has been associated with the linear scleroderma en coup de sabre. PRS is characterized by unilateral facial atrophy of the skin, subcutaneous tissue, muscles, and bones with at least 1 dermatome supplied by the trigeminal nerve. Facial asymmetry represents the most common sequela and can involve the soft tissues, craniomaxillofacial skeleton, dentoalveolar area, and temporomandibular joint. Although orthognathic procedures have been reported for skeletal reconstruction, treatment of facial asymmetry has been directed to au

Incorporating Cleft Lip Simulation Into a "Bootcamp-Style" Curriculum.

INTRODUCTION: The traditions of surgical education have changed little over the years. However, the increasing focus on patient safety and duty hour restrictions mandates that residents start developing complex skill sets earlier to ensure they graduate with procedural competency. Surgical training is poised to exploit high-fidelity simulation technology to mitigate these pressures. METHODS: By revisiting principles of adult learning theory, the authors created a "bootcamp-style" cleft lip curriculum that sought to (1) maximize educational impact and (2) pilot a high-fidelity procedural train

Folate-modified liposomes for co-delivery of schisandrin A and oxaliplatin to enhance colorectal cancer cell death.

The incidence of colorectal cancer (CRC) poses a significant threat to human life. Immunotherapy has emerged as an efficacious treatment against CRC. However, it is hindered by the challenge of inadequate immunogenicity. In addition to exerting inhibitory effects on cancer cell activity, the chemotherapeutic agent oxaliplatin (OXP) possesses the advantage of inducing immunogenic cell death (ICD) through reactive oxygen species (ROS). Schisandrin A (SCHA) can augment ROS production stimulated by OXP in cancer cells and enhance the efficacy of ICD. Consequently, we developed a liposome formulati

Co-encapsulated Ce6 + CpG and biopeptide-modified liposomes for enhanced transdermal photo-immunotherapy of superficial tumors.

Cancer immunotherapy encounters challenges of a low treatment response rate due to the tumor immunosuppressive microenvironment and immune-related adverse events caused by off-target immunotherapy agents delivered through systemic administration in clinical practice. Photodynamic therapy (PDT) offers a viable approach to improve the immunotherapy efficacy through inducing immunogenic tumor cell death and is particularly advantageous in superficial tumor treatment. Therefore, leveraging integrated nanomaterials for photo-immunotherapy appears to be an ideal strategy to improve therapeutic outco

Management of the alveolar cleft.

Orthopedic and orthodontic management of patients born with clefts of the lip, alveolus and palate is based on the application of basic biomechanical principles adapted to the individualized cleft anatomy. This article focuses on orthopedic and orthodontic preparation for 2 stages of interdisciplinary orthodontic/surgical cleft care: presurgical infant orthopedics (nasoalveolar molding) for lip/alveolus/nasal surgical repair and maxillary arch preparation for secondary alveolar bone grafting. These preparatory stages of orthopedic/orthodontic therapy are undertaken with the goal of restoring n

Combined skeletal and soft tissue reconstruction for severe Parry-Romberg syndrome.

Parry-Romberg syndrome is an acquired facial deformity that manifests as progressive hemifacial atrophy that has unknown cause. Many surgical techniques to address it have been reported, such as fat transplantation and use of free flaps. Undoubtedly, the results of such techniques can be satisfactory for mild hemifacial atrophy after soft tissue restoration. But in severe cases, except for large-scale soft tissue atrophy, the osseous framework is involved, rendering their management difficult, and the results are often inadequate. On the basis of the severity of facial soft tissue atrophy and

Naso-alveolar Asymmetry in Unilateral Cleft Lip and Palate Patients-CT Analysis of the Paranasal Region.

Secondary deformities to the midfacial area in adolescence and adulthood are common sequelae after primary cleft lip and palate repair and lead to significant functional, aesthetic and psychosocial challenges. Treatment for patients with normal occlusion or treated malocclusion and remaining midfacial hypoplasia includes autologous and alloplastic augmentation of the maxilla. Because of the great variation of deformities, accurate evaluation is essential for appropriate treatment planning. To provide insights in the altered midfacial anatomy in cleft patients, a retrospective computed tomograp

The clinical application of rhBMP-7 for the reconstruction of alveolar cleft.

In this study, radiographic assessment was performed to find out the effectiveness of bone regeneration following the application of recombinant human bone morphogenetic protein 7 (rhBMP-7) for the reconstruction of alveolar cleft defects in 11 cases: nine unilateral and two bilateral alveolar clefs. Reconstruction of the alveolar cleft was performed by using 3.5 mg of rhBMP-7 (Osigraft OP1) on a type I collagen carrier. Radiographs were taken 6 months post operation using a Gendex Intraoral Unit with Agfa Dentus M2 Comfort occlusal film. The amount of bony infill was graded on a Kindelan four

Nasolabial aesthetics.

Orthognathic surgery, as it relates to the maxilla, attempts to correct underlying skeletal deformities as well as improve function. In addition, it has the potential to significantly alter the central aesthetic unit of the face, the nasolabial region. A key to achieving a good functional as well as aesthetic result involves both comprehensive surgical planning and an understanding of the effects that orthognathic surgery of the maxilla will have on the soft tissues within the nasolabial region. The LeFort I osteotomy is one of the most commonly performed techniques to correct maxillary dentof

Selective Neurectomy of the Facial Nerve with Cross-Face Nerve Graft for Treating Postparalytic Facial Nerve Syndrome.

Although postparalytic facial nerve syndrome (PFS) is a frequent sequela of partial facial palsy, no effective treatment is currently available. Herein, we report a case of a cross-face nerve graft (CFNG) technique with selective neurectomy of the facial nerve in a 52-year-old female with moderate PFS (especially oral-ocular synkinesis and facial contracture) and a House-Brackmann score grade III. Selective neurectomy resulted in the release of the synkinesis and contractures. Furthermore, we reinnervated the levator muscles of the upper lip and oral commissure by connecting the contralateral
