Effects on the liver lipidome of rat offspring prenatally exposed to bisphenol A.

Bisphenol A (BPA) is a well-known endocrine disruptor that has obesogenic properties. We have previously reported sex- and age-dependent changes in hepatic transcriptome and proteome of several lipid homeostasis-related genes in rat offspring prenatally exposed to BPA. To further understand the impacts of prenatal BPA exposure, we analyzed lipidomic profiles in the postnatal day (PND) 21 and 60 rats using a high-resolution QTOF mass spectrometer coupled with a HPLC system. We found that the total lipid content was significantly decreased in PND21 females prenatally exposed to 5000 μg/kg bw/day

Comprehensive Lipidome Profiling of the Kidney in Early-Stage Diabetic Nephropathy.

Metabolic changes associated with diabetes are reported to lead to the onset of early-stage diabetic nephropathy (DN). Furthermore, lipotoxicity is implicated in renal dysfunction. Most studies of DN have focused on a single or limited number of lipids, and the lipidome of the kidney during early-stage DN remains to be elucidated. In the present study, we aimed to comprehensively identify lipid abnormalities during early-stage DN; to this end, we established an early-stage DN rat model by feeding a high-sucrose and high-fat diet combined with administration of low-dose streptozotocin. Using a

The Role of Clinical Proteomics, Lipidomics, and Genomics in the Diagnosis of Alzheimer's Disease.

The early diagnosis of Alzheimer's disease (AD) has become important to the reversal and treatment of neurodegeneration, which may be relevant to premature brain aging that is associated with chronic disease progression. Clinical proteomics allows the detection of various proteins in fluids such as the urine, plasma, and cerebrospinal fluid for the diagnosis of AD. Interest in lipidomics has accelerated with plasma testing for various lipid biomarkers that may with clinical proteomics provide a more reproducible diagnosis for early brain aging that is connected to other chronic diseases. The c

Clinical chorioamnionitis at term: the amniotic fluid fatty acyl lipidome.

Clinical chorioamnionitis at term (TCC) is the most common obstetrical infliction diagnosed in labor and delivery units worldwide and is associated with a substantial increase in maternal and neonatal morbidity and mortality. This obstetrical complication is a heterogeneous condition, as only half of patients have detectable microorganisms in the amniotic cavity. Because bioactive lipids play a key role in the initiation and resolution of an inflammatory response, we aimed to characterize the amniotic fluid lipidome in patients with TCC. We studied the amniotic fluid of patients in the followi

Brain lipidomes of subcortical ischemic vascular dementia and mixed dementia.

Despite its importance as the leading cause of vascular dementia, the primary pathogenic mechanisms in subcortical ischemic vascular dementia (SIVD) have remained elusive. Because of the lack of approved therapeutic agents for SIVD, there is a pressing need to identify novel therapeutic targets. Comparative lipidomic analyses of SIVD and mixed dementia (i.e., SIVD and Alzheimer's disease, MixD) may also confer new insights pertaining to the possible interaction between neurodegenerative and vascular mechanisms in the pathogenesis of dementia. Liquid chromatography coupled to mass spectrometry

Comparative lipidomic analysis of mouse and human brain with Alzheimer disease.

Lipids are key regulators of brain function and have been increasingly implicated in neurodegenerative disorders including Alzheimer disease (AD). Here, a systems-based approach was employed to determine the lipidome of brain tissues affected by AD. Specifically, we used liquid chromatography-mass spectrometry to profile extracts from the prefrontal cortex, entorhinal cortex, and cerebellum of late-onset AD (LOAD) patients, as well as the forebrain of three transgenic familial AD (FAD) mouse models. Although the cerebellum lacked major alterations in lipid composition, we found an elevation of

Spatial Lipidomics and Transcriptomics Reveal Nicotine-Mediated Restoration of Lipid Homeostasis in Parkinson's Disease.

Parkinson's disease (PD) is characterized by progressive dopaminergic neurodegeneration, and increasing evidence implicates disrupted lipid metabolism as a critical contributor to disease pathogenesis. Although nicotine has been associated with a reduced risk of PD, the mechanisms underlying its neuroprotective effects remain incompletely understood. Here, we investigated whether nicotine protects against PD by modulating brain lipid metabolism. Using 1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced mouse model of PD, we combined behavioral assessments, transcriptomic profiling, and

Artificial Intelligence in Mass Spectrometry-Based Metabolomics and Lipidomics for Drug Discovery and Development.

Artificial intelligence applied to mass spectrometry-based metabolomics and lipidomics has advanced through three technological generations: chemometric and classical machine learning, deep learning, and self-supervised foundation models, with large language models so far entering mainly at the edges. The metabolome and lipidome lie closest to drug action, yet they have been among the slowest omics layers to adopt modern artificial intelligence. This paradox arises from their downstream position, which leaves them without a closed molecular reference and without the standardized, readily model

Histology-guided spatial lipidomics and proteomics of the trisynaptic circuit in the human hippocampus.

The human hippocampal trisynaptic circuit activity is essential for learning and memory. This canonical circuit has spatially distinct populations of neurons, but their unique contributions to neurodevelopment, as well as to dysfunction in neurodegenerative disorders, are missed when analyzing bulk tissue homogenates. Using matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MSI) to guide laser capture microdissection (LCMD) of regions of interest for spatial multimodal analyses is a relatively new approach to study topographically distinct neuronal cell populations

Circulating phospholipid species and internet addiction severity in Japanese adolescents: a pilot lipidomics study.

Internet addiction is increasing among adolescents, yet biological markers remain scarce. Lipid metabolism influences neurobehavior, but its association with internet addiction is unclear. Plasma concentrations of 150 phospholipid species from eight classes-phosphatidylinositol (PI), PI3P, PI4P, PI(3,5)P2, PI(4,5)P2, phosphatidylethanolamine (PE), phosphatidylcholine (PC), and PIP3-were quantified in 34 Japanese adolescents (mean ± SD age = 16.4 ± 0.5 y) using targeted LC-MS/MS with isotopic internal standards. Internet addiction severity was assessed by the 20-item Internet Addiction Test (IA

Lipidomics reveals divergent root and shoot responses to combined nitrogen deficiency and heat stress.

INTRODUCTION: Soybean productivity is highly sensitive to heat and insufficient nitrogen supply, stresses that can occur simultaneously. However, organ-specific lipid responses to this stress combination remain poorly resolved. METHODS: We exposed hydroponically grown soybean plants to nitrogen deficiency, heat stress (HS) and their combination and compared physiological, lipidomic and gene-expression responses in roots and leaves. RESULTS: Combined stress caused the largest reductions in biomass and photosynthetic performance and the highest H₂O₂ and MDA accumulation and relative total abun

Translational lipidomics reveals BMP and its precursor LPG as biomarkers for CLN5 Batten disease.

CLN5 Batten disease, caused by biallelic mutations in CLN5, is a rare, early-onset neurodegenerative lysosomal storage disorder that has no cure and lacks validated biomarkers, hindering accurate diagnosis and assessment of therapeutic response. We recently identified CLN5 as the synthase of bis(monoacylglycero)phosphate (BMP), an endolysosomal phospholipid crucial for lysosome function and lipid catabolism. This suggested BMP and its precursor lysophosphatidylglycerol (LPG) as clinically relevant biomarkers. It also prompted in vivo confirmation of CLN5 as the biologically relevant lysosomal

Integrated lipidomics and fatty acid positional distribution for nutritional assessment of nine mammalian milks.

The nutritional value of milk fat depends on lipid and fatty acid composition. This study characterized the lipidomics and fatty acid positional distribution of milk from nine mammalian species: cow, buffalo, yak, goat, sheep, camel, mare, donkey, and human. Lipid molecular species were profiled by liquid chromatography-tandem mass spectrometry. Fatty acid positional distribution was determined using NH₂ and Si solid-phase extraction, selective sn-1/3 hydrolysis by Candida antarctica lipase B, and sn-2 monoacylglycerol quantification by gas chromatography-mass spectrometry. Ruminant milks show

Current perspectives for metabolomics and lipidomics in dyslipidemia of acne vulgaris: a mini review.

Acne vulgaris (AV) is a common inflammatory disorder involving the pilosebaceous unit. Many studies have reported that people with AV have higher levels of total cholesterol (TC), triglycerides (TG), and low-density lipoprotein cholesterol (LDL-c) compared to healthy controls. Hence, they concluded that an unhealthy lipid profile is an independent risk factor for AV. Recent research in metabolomics and lipidomics has been propelled by rapid advancements in technologies including computational methods and mass spectrometry. Using metabolomics and lipidomics approach, a broad range of structural

Omics to Unveil Diabetes Mellitus Pathogenesis and Biomarkers: Focus on Proteomics, Lipidomics, and Metabolomics.

Diabetes mellitus (DM) is a chronic metabolic disorder characterized by elevated blood sugar levels, resulting from either body's inability to produce or effectively utilize insulin. There are several types of DM, but the most common are type 1 diabetes (T1D), type 2 diabetes (T2D), and gestational diabetes mellitus (GDM). DM is a complex disease and a global health concern, and the current clinical markers, such as fasting glucose, are helpful in the diagnosis of DM, but are not specific and sensitive, especially when measured on the beginning of the pathogenesis. Therefore, there is a pressi
