Translocation of Bioactive Molecules through Carbon Nanotubes Embedded in the Lipid Membrane.

One of the major challenges of nanomedicine and gene therapy is the effective translocation of drugs and genes across cell membranes. In this study, we describe a systematic procedure that could be useful for efficient drug and gene delivery into the cell. Using fully atomistic molecular dynamics (MD) simulations, we show that molecules of various shapes, sizes, and chemistries can be spontaneously encapsulated in a single-walled carbon nanotube (SWCNT) embedded in a 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) lipid bilayer, as we have exemplified with dendrimers, asiRNA, ssDNA, an

3D structure of a Brucella melitensis porin: molecular modelling in lipid membranes.

Brucella melitensis is a pathogenic bacterium responsible for brucellosis in mammals and humans. Its outer membrane proteins (Omp) control the diffusion of solutes through the membrane, and they consequently have a crucial role in the design of diagnostics and vaccines. Moreover, such proteins have recently revealed their potential for protein-based biomaterials. In the present contribution, the structure of the B. melitensis porin Omp2a is built using the RaptorX threading method. This is a 16-stranded β-barrel with an α-helix on the third loop folding inside the barrel and forming the constr

Influence of a curcumin derivative on hIAPP aggregation in the absence and presence of lipid membranes.

The deposition of aggregates of human islet amyloid polypeptide (hIAPP) has been correlated with the death of β-cells in type II diabetes mellitus. The actual molecular mechanism of cell death remains largely unknown; however, it has been postulated that the process of aggregation from monomeric hIAPP is closely involved. A possible cause of cellular toxicity may be through the disruption of structural integrity of the cell membrane by IAPP. Herein, a water-soluble curcumin derivative, CurDAc, is used to investigate the mitigation of hIAPP aggregation in the absence and presence of lipid membr

Proteins and lipids of glycosomal membranes from Leishmania tarentolae and Trypanosoma brucei.

In kinetoplastid protists, several metabolic pathways, including glycolysis and purine salvage, are located in glycosomes, which are microbodies that are evolutionarily related to peroxisomes. With the exception of some potential transporters for fatty acids, and one member of the mitochondrial carrier protein family, proteins that transport metabolites across the glycosomal membrane have yet to be identified. We show here that the phosphatidylcholine species composition of Trypanosoma brucei glycosomal membranes resembles that of other cellular membranes, which means that glycosomal membranes

Influence of chloroform in liquid-ordered and liquid-disordered phases in lipid membranes.

Molecular dynamics simulations are used to study the influence of chloroform in two different lipid membranes: one representative of a liquid-disordered phase and another one mixed with cholesterol and representative of a liquid-ordered phase. When chloroform is added to the cholesterol-containing membrane, a strong chain disordering is induced. In both cases, chloroform laterally disorganizes the membranes. The analysis of the main structural and dynamical membrane properties reveals that the interaction with cholesterol is the main factor to explain the strong disordering effect of chlorofor

Lipid rafts are primary mediators of amyloid oxidative attack on plasma membrane.

Increasing evidence indicates that cell surfaces are early interaction sites for Abeta-derived diffusible ligands (ADDLs) and neurons in Alzheimer's disease (AD) pathogenesis. Our previous data showed significant oxidative damage at the plasma membrane in fibroblasts from familial AD patients with enhanced Abeta production. Here, we report that lipid rafts, ordered membrane microdomains, are chronic mediators of Abeta-induced lipid peroxidation in SH-SY5Y human neuroblastoma cells overexpressing amyloid precursor protein (APPwt) and APPV717G genes and in fibroblasts bearing the APPV717I gene m

[Impact of HBV infection and alcohol surrogates on the lipid composition of lymphocytic membranes].

Our study was undertaken to reveal the specific features of changes in the content of different fractions of total and lymphocytic membrane phospholipids under the influence of HBV infection and alcohol substitutes. The lipid spectrum of lymphocytic membranes was determined in 50 healthy individuals, 50 patients with acute viral hepatitis, and 62 patients with toxic hepatitis. Once HBV infection occurs, there is a reduction in the content of total phospholipids, free cholesterol, phosphatidylcholine, and phosphatidylethanolamine and an increase in the level of triglycerides, free fatty acids,

Actin polymerization serves as a membrane domain switch in model lipid bilayers.

The ability of cells to mount localized responses to external or internal stimuli is critically dependent on organization of lipids and proteins in the plasma membrane. Involvement of the actin cytoskeleton in membrane organization has been documented, but an active role for actin networks that directly links internal organization of the cytoskeleton with membrane organization has not yet been identified. Here we show that branched actin networks formed on model lipid membranes enriched with the lipid second messenger PIP(2) trigger both temporal and spatial rearrangement of membrane component

The effect of binding of spider-derived antimicrobial peptides, oxyopinins, on lipid membranes.

Oxyopinins (Oxki1 and Oxki2) are antimicrobial peptides isolated from the crude venom of the wolf spider Oxyopes kitabensis. The effect of oxyopinins on lipid bilayers was investigated using high-sensitivity titration calorimetry and (31)P solid-state NMR spectroscopy. High-sensitivity titration calorimetry experiments showed that the binding of oxyopinins was exothermic, and the binding enthalpies (DeltaH) to 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphatidylcholine (POPC) small unilamellar vesicles (SUVs) were -18.1 kcal/mol and -15.0 kcal/mol for Oxki1 and Oxki2, respectively, and peptide parti

[Lipids content in renal tissue membranes in patients with urolithiasis and secondary pyelonephritis].

We studied cytomembranes of the interstitial tissue of renal medullary layer obtained by lifetime biopsy of renal tissue in the course of pyelolithotomy in 46 patients with urolithiasis and secondary pyelonephritis. Biopsy structure was controlled with histological methods using standard staining. Basic phospholipids and cholesterol fractions in renal tissue cytomembranes were assayed at thin layer chromatography. The findings show significant changes in a nephron cytomembrane lipid phase in patients with urolithiasis and secondary pyelonephritis. There was a structural rearrangement of lipid

Hereditary spherocytosis--defects in proteins that connect the membrane skeleton to the lipid bilayer.

The molecular causes of hereditary spherocytosis (HS) have been unraveled in the past decade. No frequent defect is found, and nearly every family has a unique mutation. In dominant HS, nonsense and frameshift mutations of ankyrin, band 3, and beta-spectrin predominate. Recessive HS is most often due to compound heterozygosity of defects in ankyrin, alpha-spectrin, or protein 4.2. Common combinations include a defect in the promoter or 5'-untranslated region of ankyrin paired with a missense mutation, a low expression allele of alpha-spectrin plus a missense mutation, and various mutations in

The Omp85 protein of Neisseria meningitidis is required for lipid export to the outer membrane.

In Gram-negative bacteria, lipopolysaccharide and phospholipid biosynthesis takes place at the inner membrane. How the completed lipid molecules are subsequently transported to the outer membrane remains unknown. Omp85 of Neisseria meningitidis is representative for a family of outer membrane proteins conserved among Gram-negative bacteria. We first demonstrated that the omp85 gene is co-transcribed with genes involved in lipid biosynthesis, suggesting an involvement in lipid assembly. A meningococcal strain was constructed in which Omp85 expression could be switched on or off through a tac pr

[Correction of the lipid composition of the liver plasma membranes during heliotrine intoxication].

Antioxidant properties of yantocine, a preparation derived from Alhagi pseudalhagi Desy, were studied on a model of acute toxic hepatitis induced in random bred young male albino rats (90-100 g). Two experimental models were used: acute geliotrine hepatitis and acute CCl4 hepatitis. Yantocine was added to preparations of damaged plasma membranes (PM) in ascending doses. The results indicate that the level of malonic dialdehyde in the rat liver PM depended on the drug dose in both models of hepatitis. Linear dependence was observed for doses of 1.0-5.0 gamma. The capacity of yantocine to decrea

Decreased lipid fluidity of the erythrocyte membrane in dogs with leishmaniasis-associated anaemia.

In both man and the dog, anaemia resulting from natural leishmaniasis is often severe and mainly associated with a shortened life span of erythrocytes. Lipid fluidity of erythrocyte membranes from 17 dogs with anaemia caused by visceral leishmaniasis was investigated by means of fluorescence polarization. Results were compared with those from three groups of control animals (10 healthy dogs, seven dogs with visceral leishmaniasis but no anaemia, and 10 dogs with anaemia unrelated to leishmaniasis). Fluorescence polarization values recorded for animals with leishmaniasis-associated anaemia were

The cell membrane of Mycoplasma penetrans: lipid composition and phospholipase A1 activity.

Analysis of Mycoplasma penetrans membrane lipids revealed that, in addition to large amounts of unesterified cholesterol, M. penetrans incorporated exogenous phospholipids, preferentially sphingomyelin, from the growth medium. The major phospholipids synthesized de novo by M. penetrans were phosphatidylglycerol (PG) and diphosphatidylglycerol (DPG). In vivo labeling of PG and DPG by growing the cells with radioactive palmitate or oleate, followed by snake venom phospholipase A2 treatment, enabled us to assess the positional distribution of fatty acids in these lipids. Saturated fatty acids wer

A general model for the interaction of foreign molecules with lipid membranes: drugs and anaesthetics.

A general microscopic interaction model is proposed to describe the changes in the physical properties of phospholipid bilayer membranes due to foreign molecules which, to different degrees, partition between the membrane phases and the aqueous environment. The model is a multi-state lattice model for the main phase transition of lipid bilayers and the foreign molecules are assumed to intercalate as interstitials in the lattice. By varying the model parameters, the diversity in the thermodynamic properties of the model is explored using computer-simulation techniques which faithfully take acco

Liver microsomal membrane fluidity and lipid characteristics in vitamin A-deficient rats.

Rat liver microsomes (microsomal fraction) were isolated from vitamin A-deficient and -sufficient rats and analysed for membrane lipid characteristics. Membrane fluidity was found to be significantly decreased in microsomes from the vitamin A-deficient rats, but not in liposomes prepared from lipid extracts. Microsomes from vitamin A-deficient animals showed a significant decrease in C18:2, omega 6 and an increase in C22:5, omega 6 fatty acids.

Effect of factors of favism on the protein and lipid components of rat erythrocyte membrane.

Erythrocytes prepared from riboflavin- and tocopherol-deficient (RT-) and from control rats were used to investigate the mechanism of oxidative hemolysis by the factors of favism. RT- erythrocytes have a defense system against the oxidative stress which is blocked either where regeneration of GSH occurs or the scavenging of the radicals from the membrane is prevented. The oxidative factors used were isouramil, divicine and diamide. When RT- erythrocytes were treated with isouramil, GSH decreased to undetectable levels and was not regenerated. Complete hemolysis occurred, but no oxidation of SH

Quantitative characterization of the lateral distribution of membrane proteins within the lipid bilayer.

The dependence of the lateral distribution of membrane proteins on the size, protein/lipoid molar ratio, and the magnitude of the interaction potentials has been investigated by computer modeling protein-lipid distributions with Monte Carlo calculations. These results have allowed us to develop a quantitative characterization of the distribution of membrane proteins and to correlate these distributions with experimental observables. The topological arrangement of protein domains, protein plus annular lipid domains, and free lipid domains is described in terms of radial distribution, pair conne
