Menthol ameliorates thioacetamide-induced renal fibrosis in rats by upregulating SIRT1/Nrf2 and downregulating TGF-β1/Smad3 signalling pathways.

INTRODUCTION: Renal fibrosis is a significant factor in the progression of chronic kidney disease. This study examined how menthol affects thioacetamide (TA)-induced biochemical, molecular, and histopathological damage that leads to renal fibrosis and dysfunction. METHODS: Male Wistar rats were treated with TA (200 mg/kg, intraperitoneally) twice a week for four consecutive weeks, along with menthol (10 mg/kg, intraperitoneally) for the same duration. RESULTS: Menthol effectively reduced oxidative stress and inflammation in the kidneys of rats treated with TA. It also lowered the expression …

Comparison between in vitro toxicities of tobacco- and menthol-flavored electronic cigarette liquids on human middle ear epithelial cells.

Since electronic-cigarettes (e-cigarettes) are considered less toxic than conventional tobacco smoking, the use of e-cigarettes has increased, and the market for e-cigarette liquids (e-liquids) is continuously increasing. However, many studies showed that e-cigarettes may cause various harmful effects in lung, oral and heart. In this study, we investigated the effects of e-liquids on otitis media (OM) using human middle ear epithelial cells (HMEECs). Menthol-flavored e-liquid induced significant cell death in HMEECs (IC50: 1.45 ± 0.14%) and tobacco-flavored e-liquid led to increase in inflamma…

Understanding the impact of cooling and warming compounds on in-mouth aroma release from flavoured aqueous systems: the unexpected effect of menthol.

Menthol and capsaicin, known for their distinctive cooling and warming trigeminal sensations, respectively, are widely utilised as food flavour ingredients globally. Despite their well-documented effects on sensory perception, the impact of these compounds, individually and in combination, on in-mouth aroma release remains poorly understood. In this study, four flavoured aqueous model systems were formulated with either capsaicin, menthol, or in combination to compare against a control. Each model system contained five aroma compounds selected with diverse physicochemical properties: butyl for…

Ingesting menthol in a beverage may alter the sweating response during passive heat stress, but only when the beverage is cold.

Consuming cold fluids during heat stress can alter sweating responses; however, it remains unclear why chemical activation of cold-sensitive transient receptor potential melastatin 8 (TRPM8) with menthol only induces a cold sensation. Animal models propose that complete cold transduction with TRPM8 chemical stimulation must be combined with physical cooling to impact thermoeffector responses. To address this, 12 participants (5 females and 7 males; 22 ± 5 yr; 79.5 ± 15.0 kg; 1.8 ± 0.1 m) conducted four passive heating protocols in random order where, within trial, they consumed 3.2 mL/kg body …

Influence of topical application of capsaicin, menthol and local anesthetics on intraoral somatosensory sensitivity in healthy subjects: temporal and spatial aspects.

The aim of this study was to investigate temporal and spatial aspects of somatosensory changes after topical application of capsaicin, menthol and local anesthetics (LA) on the gingiva with the use of intraoral palpometers and thermal devices. Sixteen healthy volunteers (eight male, eight female) participated. Four topical preparations (capsaicin, menthol, LA and Vaseline as a control) were randomly applied to the gingiva around the first premolar in the upper jaw via individual oral templates, which allowed spatial mapping of somatosensory changes at and adjacent to the site of application. T…

Neuroprotective Efficacy of Menthol Against Ipomoea carnea-Induced Neurotoxicity in Rats: A Mechanistic Study on Oxidative Stress, Neuroinflammation, and Pyroptosis.

Ipomoea carnea, an invasive plant widespread in tropical and subtropical regions, contains the toxic alkaloids swainsonine and calystegines, which induce lysosomal dysfunction, neurodegeneration, and neuroinflammation. Menthol, a cyclic terpene alcohol from peppermint oil, exhibits antioxidant, anti-inflammatory, and neuroprotective properties. This study is the first to investigate the neuroprotective efficacy of menthol against neurotoxicity induced by I. carnea leaf extract in rats, with a novel focus on the pyroptosis pathway. Twenty-four male Sprague Dawley rats were divided into four gro…

Body Temperature and Discriminative Stimulus Effects of Nicotine and Menthol Aerosols in Male Rats: Role of α4β2 Nicotinic Acetylcholine Receptors.

Menthol, a prevalent additive in tobacco and electronic cigarette products, has been reported to interact with nicotine at both neuropharmacological and behavioral levels. Although menthol alone is not reinforcing, converging evidence suggests that it may modulate the pharmacological and behavioral effects of nicotine through nicotinic acetylcholine receptor (nAChR) mechanisms. Using an inhalation-based rodent model that mimics human exposure to electronic nicotine delivery systems, we examined how menthol aerosol influences nicotine-induced hypothermia, discriminative stimulus effects, and th…

Lower Successful Quit Rate of Menthol Tobacco Users in a Tobacco Cessation Program: An Explanatory Analysis in Search of Potential Mechanisms.

BACKGROUND: Menthol-flavored tobacco products are disproportionately used in low-income African American communities, a result of decades of targeted marketing and systemic inequities. Menthol use has been associated with lower quit rates, often compounded by factors such as lower trust in healthcare systems, reduced access to cessation programs, and other structural barriers. Despite this, few studies have systematically examined the explanatory mechanisms that might clarify why menthol-flavored tobacco is linked to poorer cessation outcomes among participants in tobacco cessation programs. …

Menthol as an effective inhibitor of quorum sensing and biofilm formation in Candida albicans and Candida glabrata by targeting the transcriptional repressor TUP1.

BACKGROUND: Menthol, a natural quorum sensing molecule, is derived from the Mentha species. Combating pathogenicity by inactivating quorum sensing is an emerging approach. Therefore, our objective was to investigate anti-quorum sensing and anti-biofilm potentials of menthol in Candida albicans and Candida glabrata. METHODS: The antifungal properties of menthol were evaluated using a broth microdilution assay and a time-kill assay, and its effects on quorum sensing-mediated virulence factors, cellular reactive oxygen species (ROS), and biofilm formation were tested by evaluating TUP1 expressio…

Irritant-evoked reflex tachyarrhythmia in spontaneously hypertensive rats is reduced by inhalation of TRPM8 agonists l-menthol and WS-12.

Inhalation of noxious irritants activates nociceptive sensory afferent nerves innervating the airways, inducing reflex regulation of autonom … individuals with cardiovascular disease (CVD). Previous studies have shown that the cooling mimic l-menthol, an agonist for the cold-sensitive transient receptor potential melastatin 8 (TRPM8), can alleviate nasal inflammatory symptoms and respiratory reflexes evoked by irritants. Here, we investigated the impact of inhalation of TRPM8 agonists l-menthol …
