Evaluation of pre- and post-fusion Human metapneumovirus F proteins as subunit vaccine candidates in mice.

Human metapneumovirus (hMPV) is an important respiratory pathogen especially in young children and elderly subjects. Our objective was to assess the immunogenicity and protection conferred by predominant pre- and post-fusion (F) hMPV-F constructs in Balb/C mice. Immunizations without adjuvant were not immunogenic whereas alum-adjuvanted hMPV-F proteins, regardless of their conformations, generated comparable neutralizing antibody titers with undetectable pulmonary viral titers following viral challenge. In conclusion, we found no apparent advantage for mixtures of predominant pre-fusion F prot…

Streptococcus pneumoniae exposure is associated with human metapneumovirus seroconversion and increased susceptibility to in vitro HMPV infection.

It remains largely unknown which factors determine the clinical outcome of human metapneumovirus (HMPV) infections. The aim of the present study was to analyse whether exposure to bacterial pathogens can influence HMPV infections. From 57 children, serum samples and colonization data for Haemophilus influenzae, Moraxella catarrhalis, Staphylococcus aureus and Streptococcus pneumoniae were collected at 1.5, 6, 14 and 24 months of age. Seroconversion rates to HMPV were determined and related to bacterial carriage. Frequent nasopharyngeal carriage (≥2 times in the first 2 years of life) of S. pne…

Effects of N-linked glycosylation of the fusion protein on replication of human metapneumovirus in vitro and in mouse lungs.

The fusion (F) protein is an important membrane glycoprotein necessary for cellular entry and replication of human metapneumovirus (hMPV). Selective prevention of N-linked glycosylation may compromise the catalytic and fusion functions of the F protein. By using site-directed mutagenesis and reverse genetics, recombinant mutant viruses lacking one or two N-linked glycosylation sites in the F protein were constructed. M1, which lacked glycosylation at position 57 of the F protein, had slightly compromised replication, whereas M2 and M4, which lacked glycosylation at position(s) 172 or 57 and 17…

Rates of hospitalisation for influenza, respiratory syncytial virus and human metapneumovirus among infants and young children.

To inform the development of a national influenza immunisation programme and the potential role of antiviral drugs in young children, we studied 613 children aged 71 months or younger who attended Leicester Childrens' Hospital during winter 2001-2002. During periods of respiratory syncytial virus (RSV), influenza, and human metapneumovirus activity, an estimated 12.2% (95% CI: 11.4-13.1), 7.1% (6.3-7.9), and 2.5% (2.1-2.9), respectively, of all medical cases assessed in the hospital were associated with these infections. The respective rates of hospital assessments for RSV, influenza, and huma…

Mechanistic insights and emerging applications of human metapneumovirus (HMPV) treatment: From molecular design to translational immunity.

Human metapneumovirus (HMPV) is a significant cause of acute respiratory tract infections, particularly in children, the elderly, and immunocompromised individuals, with no approved vaccines or antivirals available. This review provides mechanistic insights into HMPV virology, immunopathogenesis, and emerging therapeutic and vaccine strategies, bridging molecular design to translational immunity. HMPV, a member of the Pneumoviridae family, encodes nine proteins, including key surface glycoproteins F, G, and SH, which mediate attachment, fusion, and immune evasion. The virus’s life cycle involv…

Mathematical modelling and analysis of human metapneumovirus transmission dynamics using neural network intelligence and optimal control.

Human metapneumovirus (hMPV) is a serious global health threat because it causes human respiratory diseases in people of all ages. The complicated dynamics of this virus transmission exist in complications of waning immunity and reinfection that are extremely problematic for mathematical epidemiology. The aim of this research is to develop an integrated approach that combines a fractional-order mathematical model of hMPV transmission with a computational framework that forecasts outbreaks and optimal control, which applies effective control measures and captures complex immunological memory ef…
