Anthranilate N-methyltransferase, a branch-point enzyme of acridone biosynthesis.

Acridone alkaloids formed by acridone synthase in Ruta graveolens L. are composed of N-methylanthraniloyl CoA and malonyl CoAs. A 1095 bp cDNA from elicited Ruta cells was expressed in Escherichia coli, and shown to encode S-adenosyl-l-methionine-dependent anthranilate N-methyltransferase. SDS-PAGE of the purified enzyme revealed a mass of 40 +/- 2 kDa, corresponding to 40 059 Da for the translated polypeptide, whereas the catalytic activity was assigned to a homodimer. Alignments revealed closest relationships to catechol or caffeate O-methyltransferases at 56% and 55% identity (73% similarit…

Structural basis for inhibition of histamine N-methyltransferase by diverse drugs.

In mammals, histamine action is terminated through metabolic inactivation by histamine N-methyltransferase (HNMT) and diamine oxidase. In addition to three well-studied pharmacological functions, smooth muscle contraction, increased vascular permeability, and stimulation of gastric acid secretion, histamine plays important roles in neurotransmission, immunomodulation, and regulation of cell proliferation. The histamine receptor H1 antagonist diphenhydramine, the antimalarial drug amodiaquine, the antifolate drug metoprine, and the anticholinesterase drug tacrine (an early drug for Alzheimer's

Guanidinoacetate methyltransferase deficiency identified in adults and a child with mental retardation.

Our study describes the adult clinical and biochemical spectrum of guanidinoacetate methyltransferase (GAMT) deficiency, a recently discovered inborn error of metabolism. The majority of the previous reports dealt with pediatric patients, in contrast to the present study. A total of 180 institutionalized patients with a severe mental handicap were investigated for urine and plasma uric acid and creatinine. Patients with an increased urinary uric acid/creatinine ratio and/or decreased creatinine were subjected to the analysis of guanidinoacetate (GAA). Four patients (three related and one from

Characterization of Streptococcus pneumoniae TrmD, a tRNA methyltransferase essential for growth.

Down-regulation of expression of trmD, encoding the enzyme tRNA (guanosine-1)-methyltransferase, has shown that this gene is essential for growth of Streptococcus pneumoniae. The S. pneumoniae trmD gene has been isolated and expressed in Escherichia coli by using a His-tagged T7 expression vector. Recombinant protein has been purified, and its catalytic and physical properties have been characterized. The native enzyme displays a molecular mass of approximately 65,000 Da, suggesting that streptococcal TrmD is a dimer of two identical subunits. In fact, this characteristic can be extended to se

Hepatotoxic profile of catechol-O-methyltransferase inhibitors in Parkinson's disease.

Entacapone and tolcapone are selective catechol-O-methyltransferase (COMT) inhibitors developed recently as adjuncts to levodopa for the treatment of Parkinson's disease (PD). They extend the duration of action of levodopa. As a result, they increase 'on' time, decrease 'off' time and improve motor scores in patients with motor fluctuations. Both benefits and main side effects are related to increased dopaminergic activity. This paper reviews the use of those COMT inhibitors in PD with particular focus on the issue of hepatotoxicity. Neither tolcapone nor entacapone caused hepatotoxicity in pr

The ICF syndrome, a DNA methyltransferase 3B deficiency and immunodeficiency disease.

Only one human disease that involves Mendelian inheritance of immunodeficiency and aberrant DNA methylation has been identified. This is a rare chromosome breakage disease called the immunodeficiency, centromeric region instability, and facial anomalies syndrome (ICF). Its diagnostic characteristics are agammaglobulinemia with B cells as well as DNA rearrangements targeted to the centromere-adjacent heterochromatic region (qh) of chromosomes 1, 16, and sometimes 9 in mitogen-stimulated lymphocytes. These rearrangement-prone regions show DNA hypomethylation in all examined ICF cell populations.

Activity of phosphatidylethanolamine-N-methyltransferase in brain affected by Alzheimer's disease.

To determine whether phospholipid abnormality in Alzheimer's disease is associated with modification of phosphatidylethanolamine-N-methyltransferase, the activity of the enzyme was analysed in the frontal and occipital cortex of the brain from patients with Alzheimer's disease and from aged-matched control. The optimum pH for phosphatidylethanolamine-N-methyltransferase in human brain was 9.0. The enzyme activity was stimulated by detergent TWEEN 20 but inhibited by Triton X-100. Neither magnesium dependence nor chemical methylation was found. A decrease in activity of phosphatidylethanolamine

Abnormal regulation of DNA methyltransferase expression during colorectal carcinogenesis.

Somatic changes in CpG dinucleotide methylation occur quite commonly in human cancer cell DNA. Relative to DNA from normal human colonic cells, DNA from human colorectal cancer cells typically displays regional CpG dinucleotide hypermethylation amid global CpG dinucleotide hypomethylation. The role of the maintenance DNA methyltransferase (DNMT1) in the acquisition of such abnormal CpG dinucleotide methylation changes in colorectal cancer cells remains controversial; in one study, 60-200-fold increases in DNMT1 mRNA expression were detected in colorectal polyps and cancers relative to normal c

Hydroxyindole-O-methyltransferase in Y-79 cells: regulation by serum.

Hydroxyindole-O-methyltransferase (HIOMT) catalyzes the last step in the synthesis of melatonin. In the present study, the regulation of HIOMT expression was examined in the human Y-79 retinoblastoma cell line. Cells were grown in suspension culture using medium supplemented with 10% fetal calf serum (FCS). HIOMT activity and mRNA were strongly reduced when FCS was substituted with 0.1% bovine serum albumin (BSA), and were restored by addition of FCS. The effect of FCS on HIOMT expression was relatively selective, because the abundance of mRNA encoding actin, G3PDH or interphotoreceptor retino

Changes in DNA methyltransferase induced by treatment with N-2-acetylaminofluorene.

We have compared the levels of DNA methyltransferases from rat liver and spleen in both sexes following a single injection of N-2-acetylaminofluorene (AAF). Enzyme extracts from treated animals were obtained at different intervals (2-34 days) after treatment. The extracts were assayed in the presence of chicken erythrocyte DNA and S-adenosyl-L-[Me-3H]methionine. A 55% increase in male rat-liver methyltransferase activity measured by Me-3H incorporation into DNA occurred on day 14. By contrast, female methyltransferase after a similar period revealed a 33% decrease in activity. Between days 21 …

The effect of vitamin A deficiency on testicular transfer RNA methyltransferase activity.

Testicular transfer RNA methyltransferase activity was examined in normal and vitamin A-deficient rats. The specific activity was reduced by 50% in the vitamin A-deficient rats. In addition, a 5-fold decrease in the extent of tRNA methylation was observed with enzyme preparations from deficient testes. Both the rate and extent of tRNA methylation returned to control levels in vitamin A-repleted rats. In contrast, retinoic acid repletion did not reverse the effect of vitamin A deficiency on testicular tRNA methyltransferase activity. The methylated nucleoside composition of tRNA methylated by e

Kmt2c and Kmt2d histone methyltransferase deficiencies compromise macrophage function.

Methylation of histone (H) 3 lysine (K) 4 (H3K4) has a well-established role in innate immune responses, but the contribution of H3K4 methyltransferases Kmt2c and Kmt2d in innate immunity is incompletely understood. Using conditional knockout mouse models, we investigated how Kmt2c- and Kmt2d-deficiencies affect innate immune cell function. Through functional, transcriptomic, and metabolic analyses, we delineate the consequences of disrupted epigenetic regulation on macrophage biology. Our findings reveal that loss of Kmt2c or Kmt2d in macrophages leads to impaired pro-inflammatory cytokine re…

Allosteric Inhibitors of Erythromycin Resistance Methyltransferase Reverse Antibiotic Resistance.

Methylation of a critical adenine residue of the 23S rRNA residing in the nascent exit tunnel of the bacterial ribosome, by Erythromycin resistant methyltransferases (Erms), reduces binding of macrolide, lincosamide, and streptogramin B (MLSB) class of antibiotics and confers high-level resistance. Using an allosteric site-directed focused virtual screening approach, nucleoside analogues were identified as the initial scaffold. 6-thioinosine, recognized as a significant initial hit molecule, was confirmed through in vitro screening to be a potent scaffold. Further, structure-based inhibitor ev…

[Therapeutic potential of histamine N-methyltransferase inhibition for the treatment of hypersomnia].

Histamine plays a crucial role in maintaining wakefulness. Decreased histamine levels in the cerebrospinal fluid have been reported in patients with hypersomnia, including narcolepsy and idiopathic hypersomnia. Histamine N-methyltransferase (HNMT) is a key enzyme responsible for histamine degradation in the brain. In this study, we examined the effects of pharmacological HNMT inhibition on animal models of hypersomnia disorders. Metoprine, an HNMT inhibitor, significantly increased brain histamine levels and produced robust wake-promoting effects and suppressed cataplexy in narcolepsy model mi

Histamine N-methyltransferase upregulation, cardiac hypertrophy, and heart failure.

BACKGROUND AND AIMS: Histamine signalling is critical in heart failure (HF). Histamine N-methyltransferase (HNMT) serves as the primary enzyme responsible for histamine clearance in heart, but its role in HF remains undefined. This study investigates HNMT-mediated metabolic-epigenetic crosstalk in HF and evaluates therapeutic strategies targeting this axis. METHODS: Cardiomyocyte-specific Hnmt-knockout and Hnmt-overexpression mice were generated. Transverse aortic constriction (TAC) was applied to induce HF in mice. RNA sequencing and targeted metabolomics were employed to explore mechanisms

Protein arginine methyltransferase 5 is essential for virulence in Toxoplasma gondii.

BACKGROUND: Protein arginine methyltransferase 5 (PRMT5) is a key enzyme responsible for catalyzing symmetric dimethylarginine (SDMA) modifications and plays crucial roles in epigenetic regulation, transcription, and cell cycle progression in eukaryotes. Although our previous study determined the expression and cellular localization of PRMT5 in tachyzoites and bradyzoites, and confirmed its type II PRMT activity, its functional significance in Toxoplasma gondii remains entirely uncharacterized. METHODS: This study aimed to explore the biological functions of PRMT5 in T. gondii. The prmt5 gene

Clinical relevance of thiopurine methyltransferase genotype testing for azathioprine in dermatology.

BACKGROUND: Polymorphisms in thiopurine methyltransferase (TPMT) are a predominant cause of azathioprine-induced leucopenia in Western countries. The exact role of these polymorphisms in the Indian population with dermatological disorders is uncertain. OBJECTIVES: To evaluate the frequency of genetic polymorphism of TPMT and its impact on the safety of azathioprine in dermatological disorders. METHODS: We included consecutive patients on azathioprine who were initiated for dermatological disorders from South India. Three TPMT polymorphisms (c.238G>C, c.460G>A and c.719A>G) were assessed. The histone methyltransferase MLL1 complex inhibits expression of fetal hemoglobin.

Increasing fetal-type hemoglobin expression in adult erythroid cells holds promise in the treatment of sickle cell disease (SCD) and β-thalassemia. We have identified the MLL1 complex as a critical regulator of fetal and embryonic hemoglobin repression. Knockdowns of MEN1 and KMT2A, encoding essential components of the complex, caused a significant downregulation of BCL11A expression and a substantial increase in γ- and ε-globin mRNA levels in HUDEP-2 cells. Significant binding of MEN1 and KMT2A was readily detected at the promoter and a critical enhancer of BCL11A in HUDEP-2 cells, suggesting

Furamidine, a methyltransferase inhibitor, is a potential anti-Babesia spp. chemotherapeutic.

Epigenetic inhibitors targeting histone methyltransferases (HMTs) have been proven to be promising for blood protozoan treatment. However, little is known about the effects of HMT inhibitors on Babesia parasites. Here, in vitro and in vivo drug tests were performed to evaluate the efficacy of several compounds targeting various HMTs for Babesia treatment. Their cytotoxicity to MDOK cells was also assessed. Among these compounds, furamidine demonstrated outstanding activity in vitro at the nanomolar level (IC50s of 0.03 ± 0.55, 0.02 ± 0.50, and0.02 ± 0.76 μM at 48, 72, and 96 h, respectively). …
