Longitudinal evolution of the concentration of gangliosides GM3 and GD3 in human milk.

It has been reported that dietary gangliosides may have an important role in preventing infections and in brain development during early infancy. However, data related to the evolution of their concentration over the different stages of lactation are scarce. Liquid chromatography coupled with electrospray ionization high resolution mass spectrometer (LC/ESI-HR-MS) has been optimized to quantify the two major ganglioside classes, i.e., aNeu5Ac(2-8)aNeu5Ac(2-3)bDGalp(1-4)bDGlcp(1-1)Cer (GD3) and aNeu5Ac(2-3)bDGalp(1-4)bDGlcp(1-1)Cer (GM3) in human milk. Gangliosides were extracted using chlorofo

[Multicenter study on the effects of human milk fortification in premature infants].

OBJECTIVE: To study the effects of human milk fortification on growth and safety in premature infants in China during hospital stay. METHODS: A prospective controlled study was conducted in 4 tertiary hospitals in Beijing and Shanghai, premature infants born from Nov. 2009 to Mar. 2011 were included according to certain enrollment criteria. Premature infants were divided into two groups: fortified human milk group, i.e., FHM group who were fed with human milk over 50 percent during hospital stays, preterm formula was used for those whose mothers did not have enough milk, and preterm formula g

The possibility of vertical transmission of human papillomavirus through maternal milk.

Human papillomavirus (HPV) DNA has been detected in the oral cavity of infants and breast cancer tissue, suggesting its vertical transmission through maternal milk. We determined whether HPV is detected in maternal milk and is vertically transmitted by breast-feeding. Informed consent was obtained, and maternal milk samples (n=80) were analysed for high-risk HPV DNA. In 43 women, this DNA was measured in the uterine cervix. In women with positive samples, this DNA was measured in the oral cavities of their children. The domain including HPV E6 and E7 was amplified by polymerase chain reaction

Improving the use of human milk during and after the NICU stay.

The feeding of human milk (milk from the infant's own mother; excluding donor milk) during the newborn intensive care unit (NICU) stay reduces the risk of costly and handicapping morbidities in premature infants. The mechanisms by which human milk provides this protection are varied and synergistic, and appear to change over the course of the NICU stay. The fact that these mechanisms include specific human milk components that are not present in the milk of other mammals means that human milk from the infant's mother cannot be replaced by commercial infant or donor human milk, and the feeding

Milk folate secretion is not impaired during iron deficiency in humans.

The purpose of this study was to examine whether maternal iron and/or folate status influences human milk folate secretion and is responsible for growth faltering of Otomi infants in Capulhuac, Mexico. Breast-feeding mothers (n = 71) were randomized at 22 +/- 13 d (baseline) postpartum to receive a daily multivitamin supplement containing folic acid (400 microg) with and without iron (18 mg). Mothers provided blood and milk samples at baseline, and at 82 +/- 15 and 138 +/- 18 d postpartum. Iron supplementation significantly improved hematocrit and transferrin receptor concentrations but had no

Dietary phosphorus reduction by pretreatment of human breast milk with sevelamer.

Hyperphosphatemia leading to hyperparathyroidism and ultimately renal osteodystrophy is a well-known complication of chronic renal failure. A new hydrogel binder, sevelamer, has recently become available for use in hyperphosphatemic patients with renal failure. We had previously mixed the capsule with pumped breast milk and formula, but discovered that the hydrogel formed a viscous solution that infants were unable or unwilling to swallow. We therefore evaluated the phosphorus content of fresh and frozen breast milk before and after treating with different doses of sevelamer at different tempe

Insulin in human milk: postpartum changes and effect of gestational age.

OBJECTIVE: To determine if human milk insulin (HMI) concentrations are affected by gestational age and postnatal age. DESIGN AND SETTING: An observational study carried out in a level III neonatal intensive care unit. Insulin concentrations were determined in human milk of 90 parturient mothers who delivered between 30 and 41 weeks gestation. Samples were collected on days 3 and 10 after delivery. RESULTS: HMI concentrations for mothers of preterm infants were not significantly different from those of full term infants, on either day 3 or 10 post partum. When results for all 90 mothers were

Anti-endotoxin antibodies in human milk: correlation with infection of the newborn.

UNLABELLED: A longitudinal study was performed to investigate the content of human colostrum and milk of antibodies against endotoxins of Escherichia coli, Pseudomonas aeruginosa and Salmonella minnesota during the first 6 mo of lactation. The influence of the gestational age of the newborn and the prevalence of a systemic infection in the child on maternal antibody production were observed. Colostrum of mothers of term infants who had shown signs of systemic infection contained higher antibody concentrations compared to colostrum of mothers of healthy newborns. After the first week post partu

Antiinflammatory effects of human milk on chemically induced colitis in rats.

We examined the effects of a human milk diet on rats with chemical colitis induced with a 4% acetic acid enema. Colonic myeloperoxidase activity was used as a surrogate marker for neutrophil infiltration. Control rats fed rat chow had little colonic myeloperoxidase activity; geometric mean, 0.27 U/g of tissue. Rats with colitis fed rat chow had significantly increased colonic myeloperoxidase activity (geometric mean, 6.76 U/g, p < 0.01 versus no colitis), as did rats with colitis fed infant formula or Pedialyte (geometric mean, 6.92 and 8.13 U/g, respectively, both p < 0.01 versus no colitis).

[Colonic fermentation in humans: effect of the ingestion of meat associated to milk].

The amount of breath H2 produced for 6 h following the ingestion of 240 ml of milk was used as an index of the rate of colonic fermentation of undigested lactose in 8 lactase non-persistent Guatemalan adults. Treatments in separate days included milk alone, milk with 400 g cooked beef, lactose pre-hydrolyzed milk with the same amount of beef, beef alone and fasting for 6 h. Excess excretion of H2, calculated as the mean area under the curve were 178 +/- 31 ppm h with milk alone, 50 +/- 17 with milk plus beef, -1 +/- 26 with prehydrolyzed milk (p > 0.001). Peak increments of breath H2 followed

Need for riboflavin supplementation in small prematures fed with human milk.

It is the practice in Finland to feed small premature infants with human milk and with no riboflavin supplementation. In this study the riboflavin status was analyzed in 39 such premature infants, 19 with riboflavin supplementation (0.3 mg/day) and 20 without, in their mothers, and in breast-milk samples during a period of 12 wk after delivery. The mean gestational age of the infants was 30.1 wk and their birth weight 1,183 g. Stimulation of erythrocyte glutathione reductase by flavin-adenine-dinucleotide was used as the criterion for riboflavin status in the blood samples. At age 6 wk 47% of

[The degradation of lactose in the intestines of rats on human and cow's milk diets].

Female lactose adapted rats were kept for 10 days on human milk (HM), a milk diet adapted to human milk (MD1), and a milk diet rich in protein and phosphate (MD2), the lactose supply being always the same. In the caecum, colon and faeces, the pH value, the phosphorus content, the buffer capacity and the numbers of microorganisms with proteolytic activity (Bacteroides, Klebsiella, Proteus) were lower and the lactose concentration and the beta-galactosidase activity were higher on HM and MD1 than on MD2.

Identification of copper- and zinc-binding ligands in human and bovine milk.

The object of this study was to identify copper and zinc ligands detected during modified gel chromatography ( MGC ) of bovine and human milk ultrafiltrates. Isolation by anion-exchange chromatography and subsequent proton nuclear magnetic resonance (NMR) spectroscopy analysis demonstrated that the sole major low-molecular-weight ligand binding copper and zinc in bovine milk is citrate. Human milk apparently also contains citrate as a major metal-binding ligand but also contains amino acids, of which primarily glutamate was purified by anion-exchange chromatography. The amino acids bind copper

Multi-omics analysis of functional component networks in human, porcine, and ruminant milk: Insights for precise design of infant formula and milk replacers based on bovine milk.

This integrated multi-omics study (metabolomics, lipidomics, proteomics) compared the compositions of bovine, caprine, ovine, porcine, and human milk. Human milk is defined by a unique microenvironment where complex human milk oligosaccharides and immune regulating small molecules act together. Components like Lnnt and Lnfp iii form interactive networks, which help modulate both gut microbiota and the immune system. Porcine milk contributes to neural development and immune function. It is rich in polar phospholipids and polyunsaturated fatty acids such as docosahexaenoic acid, important for ne

Prenatal SARS-CoV-2 Infection Alters Human Milk-Derived Extracellular Vesicles.

Human milk-derived extracellular vesicles (HMEVs) are key components in breast milk, promoting infant health and development. Maternal conditions could affect HMEV cargo; however, the impact of SARS-CoV-2 infection on HMEVs remains unknown. This study investigated the influence of SARS-CoV-2 infection during pregnancy on postpartum HMEV molecules. The median duration from SARS-CoV-2 test positivity to milk collection was 3 months. After defatting and casein micelle disaggregation, HMEVs were isolated from milk samples of nine mothers with prenatal SARS-CoV-2 and six controls by sequential cent
