Epidermal growth factor receptor (EGFR) and squamous cell carcinoma of the skin: molecular bases for EGFR-targeted therapy.

Cutaneous squamous cell carcinoma (SCC) ranks second in the frequency of all skin tumors. Its incidence has risen significantly due to an increased sun exposure and the number of immunocompromised patients. It has a well-defined progression with known precursor lesions called actinic keratosis. The degree of cellular differentiation, tumor thickness, location, and other features has prognostic value. It has a better prognosis than mucosal SCC of the head and neck, also called head and neck squamous cell carcinoma (HNSCC). Ultraviolet light plays a fundamental role as an initiator and promoter

EML4-ALK in Non-small Cell Lung Cancer: Molecular Mechanisms and Targeted Therapies.

Lung cancer is a malignancy characterized by high global incidence and mortality rates, with approximately 85% of cases classified as non-small cell lung cancer (NSCLC). Advances in molecular biology and targeted therapeutic agents have ushered NSCLC into the era of precision medicine. The echinoderm microtubule-associated protein-like 4 (EML4) and anaplastic lymphoma kinase (ALK) fusion gene represents a pivotal target for personalized treatment in NSCLC. Although ALK inhibitors exhibit significant efficacy against EML4-ALK-positive tumors, addressing drug resistance remains a major challenge

Programmed cell death in acute kidney injury: From molecular mechanisms to targeted therapies.

Acute kidney injury (AKI) represents a formidable global health challenge characterized by high morbidity and mortality, yet effective pharmacological interventions remain elusive. Programmed cell death (PCD) pathways, orchestrating the depletion of renal tubular epithelial cells, are increasingly recognized as central drivers of AKI pathogenesis. Despite the rapid expansion of PCD-targeted agents in other therapeutic areas, their translational potential in AKI has not been fully exploited. This review provides a systematic synthesis of the molecular mechanisms governing diverse PCD modalities

Beyond Percutaneous Coronary Intervention - Targeted Molecular Therapies for the Next Era of Coronary Care.

Coronary artery disease (CAD) remains the leading global cause of cardiovascular morbidity and mortality, driven by dysregulated lipid metabolism, chronic vascular inflammation, thrombosis, and genetic susceptibility. Management has long relied on mechanical revascularization-percutaneous coronary intervention and coronary artery bypass grafting supported by antithrombotic therapy. While these restore perfusion and reduce ischemic events, they do not address atherogenesis at the molecular level and leave substantial residual risk, often with increased bleeding. Advances in genomics, proteomics

Neuropeptide Y receptors 1 and 2 as molecular targets in prostate and breast cancer therapy.

Recent advances have revealed the overexpression of Neuropeptide Y (NPY) receptors in multiple cancers, positioning them as attractive molecular targets for cancer diagnostics and therapeutics. Despite this, a comprehensive roadmap for the rational development of anticancer agents targeting NPY receptors remains lacking. Therefore, we present the characteristics of NPY receptor subtypes, their abundance, and the correlation of their expression in different cancer types. It was found that NPY receptor subtypes 1 and 2 were extensively studied, especially in connection with breast and prostate c

Rheumatoid Arthritis and Osteoporosis as Prototypes of Immunosenescence in Osteoimmunology: Molecular Pathways of Inflammaging and Targeted Therapies.

Immunosenescence refers to the set of immunoendocrinological alterations underlying the progressive decline in innate and adaptive immune function that occurs with aging. It is closely linked to the concept of inflammaging, a state of low-grade chronic systemic inflammation that contributes to age-related diseases. In the elderly, key features of diseases such as rheumatoid arthritis, particularly in its elderly onset form, and senile osteoporosis are characterized by a decline in sex hormones and the immunoregulatory IL-2; an increase in serum autoantibodies and pro-inflammatory mediators suc

Meisoindigo Acts as a Molecular Glue to Target PKMYT1 for Degradation in Chronic Myeloid Leukemia Therapy.

Meisoindigo (Mei) has been clinically utilized for the treatment of chronic myeloid leukemia (CML), yet the precise molecular targets by which it exerts effects remain unclear. Through activity-based protein profiling (ABPP), the protein kinase, membrane-associated tyrosine/threonine 1 (PKMYT1) is identified as a direct target of Mei. Specifically, Mei forms a selective and reversible covalent bond with the Cys301 residue of PKMYT1, triggering its K48-linked polyubiquitination and accelerating proteasomal degradation, which is mediated by the E3 ligase TRIM25. The study reveals that Mei acts a

Targeted genetic and molecular therapies in neurofibromatosis - A review of present therapeutic options and a glimpse into the future.

Neurofibromatosis type 1, the most common phakomatoses, can present with a host of signs and symptoms, usually involving the skin and the peripheral nervous system. It is characterized by a mutation in the neurofibromatosis type 1 gene on chromosome 17q11.2 that codes for the protein neurofibromin. Neurofibromin acts as a tumor suppressor gene by inhibiting rat sarcoma (Ras) activity and its deficiency leads to increased Ras activity, cellular proliferation and tumor formation. This review was conducted to analyze the various targeted therapies at the genetic and molecular level employed to ma

The molecular, cellular and clinical consequences of targeting the estrogen receptor following estrogen deprivation therapy.

During the past 20 years our understanding of the control of breast tumor development, growth and survival has changed dramatically. The once long forgotten application of high dose synthetic estrogen therapy as the first chemical therapy to treat any cancer has been resurrected, refined and reinvented as the new biology of estrogen-induced apoptosis. High dose estrogen therapy was cast aside once tamoxifen, from its origins as a failed "morning after pill", was reinvented as the first targeted therapy to treat any cancer. The current understanding of the mechanism of estrogen-induced apoptosi

Molecular imaging of immune and fibrosis targets to guide therapy for repair after myocardial infarction.

Heart injury after acute myocardial infarction requires adequate repair to prevent left ventricular remodelling and the development of chronic heart failure. The early immune response to myocardial injury and its tight interconnection to fibroblast activation have a crucial role in determining the outcome after myocardial infarction. Emerging therapies aim to exploit these targets, harnessing the subacute window after cardiac injury to prevent subsequent contractile dysfunction. Given the complexity of the tissue response to myocardial injury and the molecular specificity of targeted therapies

Systemic Mastocytosis: Molecular Pathophysiology, WHO Diagnostic Framework, and KIT-Directed Targeted Therapies.

BACKGROUND: Systemic mastocytosis (SM) is a clonal mast cell (MC) neoplasm driven by somatically acquired activating mutations in the KIT receptor tyrosine kinase (CD117), resulting in pathological accumulation of morphologically atypical MCs in extracutaneous organs. The KIT D816V substitution is detectable in over 95% of cases by high-sensitivity next-generation sequencing (NGS) or allele-specific PCR. This gain-of-function variant confers ligand-independent receptor autophosphorylation, leading to constitutive activation of downstream signaling cascades that promote MC progenitor survival,
