In many animal systems, females exhibit a localized immune response to insemination that helps defend against sexually transmitted disease. However, this response may also kill sperm, reducing a male's reproductive potential. If males could suppress this response, they may be able to increase their sperm's representation in the female's reproductive tract, thereby increasing their fitness. Here we address the hypothesis that, under conditions of sperm competition, males interfere with female immunity. To test our hypothesis, we manipulated levels of female mating frequency (single vs. multiply

Oculocutaneous albinism is an autosomal recessive genetic disorder. Several types of oculocutaneous albinism are caused by mutation in related genes. Oculocutaneous albinism 1 is associated with the tyrosinase gene. The human tyrosinase gene (TYR) encodes tyrosinase, a key enzyme in melanin biosynthesis. As exon 1 of the gene shows an MboI-RFLP within codon 192 in Caucasians, we studied allele frequencies of MboI 192 polymorphism in 200 chromosomes from 100 unrelated normal Korean individuals. As a result, only one allele system, the presence of the MboI 192 site, was detected in the Korean.

Oculocutaneous albinism (OCA) is a complex group of genetic disorders that have historically been defined by clinical and biochemical methods. Recent advances in the molecular biology of pigmentation have greatly increased our understanding of the complexity of this group of disorders. To date, two different types of OCA (OCA1 and OCA2) have been mapped to specific chromosomal regions. Mutations have been found in the tyrosinase locus associated with OCA1 and the human homologue to the murine pink-eyed dilution locus associated with OCA2. Analysis of these genes and their mutations will allow

The human tyrosinase gene is greater than 35 kb and is organized in four introns and five exons [Tomita et al. (1989) Biochem. Biophys. Res. Commun., 164, 990-996]. Using the full-length cDNA encoding human tyrosinase and its exon-specific fragments as hybridization probes, we show that overall structural organization of the tyrosinase gene is unchanged in three patients affected with tyrosinase-negative oculocutaneous albinism (OCA). Moreover, we are able to show the presence of additional DNA segments cross-hybridizable to exon 4 or exon 5 of the tyrosinase gene in the genome of three OCA pa

We examined tyrosinase activity in pigmented tissues from five human eyes. Tissue extract was prepared in cholate-phosphate buffer by homogenization, centrifugation, trypsin digestion and hydroxylapatite column chromatography. Tyrosinase activity was spectrophotometrically assayed as dopa (L-3,4-dihydroxylphenylalanine) oxidase activity. Tyrosinase activity was detected in the extracts of the ciliary body. Enzyme activity was inhibited by phenylthiourea but not by 3-iodo-tyrosine. We believe that our findings confirm the presence of tyrosinase activity in the human ciliary body.

When the Legionnaires' disease (LD) bacterium is grown on supplemented Mueller-Hinton agar, brown pigmentation of the medium occurs. Since this browning may result from tyrosinase-mediated formation of melanin, we supplemented yeast-extract agar with various aromatic precursors of melanin and inoculated it with eight strains of the LD bacterium. Browning occurred with growth of each LD strain of the bacterium on yeast-extract agar enriched with 2.5 mmol/L of L-phenylalanine or L-tyrosine but not without such enrichment. Equimolar D-phenylalanine or D-tryosine in yeast-extract agar did not enha

As people age and are exposed to the sunlight for a long time, the excessive production and accumulation of melanin in the skin can affect their appearance and health. In addition, some whitening active ingredients have limitations in terms of solubility, stability, and skin irritation, which prompted us to research more effective techniques to improve the skincare effects of active ingredients. 4-Butylresorcinol (BR) can achieve whitening by inhibiting the activity of tyrosinase (TYR) and dihydroxyindole carboxylate oxidase. However, the high irritancy of BR limits its application. Therefore,

Tyrosinase (TYR) and hydrogen sulfide (H₂S) are two notable biomarkers implicated in melanocyte physiology and melanoma progression. However, no fluorescent probe has been developed for their simultaneous detection in biological systems. Here, we present a dual-locked fluorescein-based probe TyNitro-1 capable of concurrently sensing TYR and H₂S, which incorporates a 3-hydroxyphenyl group for TYR recognition and a nitro moiety for H₂S recognition. Upon dual-analyte activation, the probe displays an obvious fluorescence enhancement at 517 nm. It offers an efficient response (≤120 min for TYR; ≤6

Oculocutaneous albinism (OCA) are genetically and clinically heterogeneous recessive disorders with at least 23 associated genes. Isolated OCA is characterized by hypopigmentation in the skin, hair, and eyes combined with ocular abnormalities. Hermansky Pudlak syndrome (HPS) and Chediak-Higaski syndrome are syndromic forms of OCA, distinguished by immunological and hematological symptoms in addition to hypopigmentation and ocular anomalies. Targeted clinical care is crucial for the patients and molecular genetic diagnosis is important for classification of patients. Current diagnostic yield is

BACKGROUND: Extremophilic fungi thrive in extreme environments, revealing life's origins and enhancing biodiversity while offering insights into evolutionary biology. This study investigates the adaptation mechanisms of haloalkaliphilic fungi Aspergillus salinarum, Cladosporium sphaerospermum, and Penicillium camemberti, isolated from Egyptian soils, adapted to life under extreme conditions of high salt (15%) and alkaline pH (10). These properties make them interesting for fundamental research and the exploration of biotechnological potential.
RESULTS: These fungi exhibited increased levels o

Microalgae are promising biorefineries because of their rapid growth and carbon fixation abilities. However, the functional applications of microalgal extracellular polysaccharides (EPS), particularly those from edible strains, remain underexplored, limiting their potential use in health-related and cosmetic industries. In this study, we investigated the EPS produced by the edible green alga Parachlorella sp. BX1.5 (bxEPS). The bxEPS was extracted from nitrogen-depleted BG11 (BG11-N) medium and identified as an acidic rhamnan (rhamnose: xylose: glucuronic acid = 55.1:35.4:9.5 mol%) without sul

This study employed the predicted data mining approach (PDMA) to guide the biotransformation of scutellarin methyl ester by Bacillus megaterium tyrosinase (BmTYR). While the enzymatic conversion yielded 6-hydroxyluteolin 7-O-methylglucuronide, a known derivative, this study highlights the ability of PDMA to predict and synthesise single products with high specificity. This approach presents a valuable alternative to the extraction of bioactive compounds from traditional medicinal herbs, which inherently contain a complex mixture of secondary metabolites. It facilitates the efficient production

Melanin is produced by melanocytes to protect human skin from harmful ultraviolet radiation. During skin cell renewal, melanin and dead skin cells are disposed of. However, prolonged exposure to ultraviolet rays or aging can disturb this cycle, leading to skin hyperpigmentation due to melanin accumulation. Tyrosinase is a crucial enzyme involved in melanin biosynthesis. Although various compounds, including tyrosine inhibitors, that counteract melanin accumulation have been reported, some, such as hydroquinone, are toxic and can cause vitiligo. Meanwhile, the skin is the largest organ and the

BACKGROUND: Tyrosinase, a copper-containing metalloenzyme with catalytic activity, is widely found in mammals. It is the key rate-limiting enzyme that catalyzes melanin synthesis. For humans, tyrosinase is beneficial to the darkening of eyes and hair. However, excessive deposition of melanin in the skin can lead to dull skin color and lead to pigmentation. Therefore, many skin-whitening compounds have been developed to decrease tyrosinase activity. This study aimed to identify a new tyrosinase inhibitory peptide through enzymatic hydrolysis, in vitro activity verification, molecular docking, a

BACKGROUND: Albinism is a heterogeneous condition in which patients present complete absence, reduction, or normal pigmentation in skin, hair and eyes in addition to ocular defects. One of the heterogeneous forms of albinism is observed in Hermansky-Pudlak syndrome (HPS) patients. HPS is characterized by albinism and hemorrhagic diathesis due to the absence of dense bodies in platelets.
METHODS: In this report, we describe a case of a pair of Puerto Rican siblings with albinism that were clinically diagnosed with HPS during childhood. Since they did not harbor the founder changes in the HPS1

Onosma riedliana Binzet & Orcan, a traditionally used plant species, has been explored for its therapeutic potential in this study. The work presented here is the first report on the phenolic profile and biological activity of this species. Three extracts of varying polarity were prepared, with the methanolic extract containing the highest phenolic content (97.62 ± 0.20 mgGAE/g). Key phenolic compounds identified included pinoresinol, hesperidin, 4-hydroxybenzoic acid, and p-coumaric acid. The methanolic extract exhibited exceptional antioxidant properties, rivaling Trolox as a positive contro

In human, Tyrosinase enzyme (TyH) is involved in the key steps of protective pigments biosynthesis (in skin, eyes and hair). The use of molecules targeting its binuclear copper active site represents a relevant strategy to regulate TyH activities. In this work, we targeted 2-Hydroxypyridine-N-oxide analogs (HOPNO, an established chelating group for the tyrosinase dicopper active site) with the aim to combine effects induced by combination with a reference inhibitor (kojic acid) or natural substrate (tyrosine). The HOPNO-MeOH (3) and the racemic amino acid HOPNO-AA compounds (11) were tested on

A resorufin-based dual-locked fluorescent probe (RHT) was introduced to image melanoma cells selectively. RHT was shown to function as an AND molecular logic gate as it emitted a signal only in the presence of both hydrogen sulfide (H2S) and tyrosinase (Tyr), which are known to be overexpressed in melanoma cells. In vitro cell culture studies revealed that RHT can be activated with endogenous H2S and Tyr and allows selective imaging of B16-F10 cancer cells under confocal microscopy. RHT marks the first ever example of a fluorescent probe that is sequentially activated by H2S and Tyr.

Although melanogenesis is a defense mechanism against ultraviolet (UV)-induced skin damage, abnormally excessive melanin production causes pigmentation disorders. Tyrosinase, as a key factor for melanin synthesis, plays an important role in inducing skin pigmentation. Therefore, the inhibition of tyrosinase is crucial in preventing skin pigmentation in the cosmetics and medicine fields. However, the majority of well-known tyrosinase inhibitors have been discontinued due to toxic effects on the skin or lack of selectivity and/or stability. In this study, we evaluated possible anti-melanogenic e
