Development and validation of an actin-1 endogenous internal control for multiplex RT-qPCR surveillance of mosquito- and sand fly-borne viruses.

Internal controls are essential to ensure the reliability of molecular assays, confirming the integrity of nucleic acid extraction and testing process. This is particularly important in entomovirological surveillance, which relies on molecular testing of field-collected arthropod vectors. A key challenge is identifying a control target conserved among diverse vector species. We designed a novel primer-probe set for hydrolysis probe-based real-time PCR, targeting the actin-1 gene and assessed its performance across 21 mosquito and 9 sand fly species in Israel. The assay performance was compared

Efficacy of ceiling-mounted mosquito nets for malaria vector control in a Peruvian Amazon riverine community: A stepped-wedge cluster randomized trial.

Malaria remains a major global health concern, especially in tropical regions such as the Peruvian Amazon. Installing ceiling-mounted mosquito nets in houses has been proposed as a strategy to reduce mosquito-human contact and, in turn, lower malaria transmission. We aimed to assess the effectiveness of ceiling-mounted mosquito nets in reducing Anopheles mosquito density in a high-transmission Amazonian setting. We conducted a stepped-wedge cluster-randomized trial from March to December 2024 in the Llanchama community of the San Juan Bautista district, Loreto, Peru. A total of 69 households w

Evaluation of AaDOP2 receptor antagonists reveals antidepressants and antipsychotics as novel lead molecules for control of the yellow fever mosquito, Aedes aegypti.

The yellow fever mosquito, Aedes aegypti, vectors disease-causing agents that adversely affect human health, most notably the viruses causing dengue and yellow fever. The efficacy of current mosquito control programs is challenged by the emergence of insecticide-resistant mosquito populations, suggesting an urgent need for the development of chemical insecticides with new mechanisms of action. One recently identified potential insecticide target is the A. aegypti D1-like dopamine receptor, AaDOP2. The focus of the present study was to evaluate AaDOP2 antagonism both in vitro and in vivo using …

Pou2f3 Modulates Mosquito Host-Seeking Behavior by Regulating Skin Microbiota and Their Production of Volatiles: Implications for Eco-Friendly Pest Control Strategies.

Skin microbiota-derived volatile metabolites are critical for mosquito host-seeking and disease transmission. Similar volatile organic compounds (VOCs)-mediated mechanisms may apply to agricultural pests (e.g., aphids, whiteflies). We demonstrate that Pou2f3, a skin-enriched transcription factor, regulates β-defensin1 expression, shaping microbiota composition and volatile production. Pou2f3-deficient mice exhibited increased Staphylococcus abundance and elevated 1-methoxy-2-propanol levels, enhancing attraction in Culex and Aedes mosquitoes. This VOC, alongside other microbiota-derived compou

Mutant allele formation and inheritance during Cas9/guide RNA-mediated gene drive in a population modification mosquito strain for human malaria control.

Gene-drive systems are under development for the population modification of anopheline vectors of human malaria parasites. The key to their success is the fixation of genes in target mosquito populations that encode molecules preventing parasite transmission. High-efficiency Cas9/guide RNA (gRNA)-based gene-drive systems can facilitate this objective. A potential challenge to these systems is the presence of naturally-occurring or drive system-induced sequence polymorphisms in the genomic target site that could impede Cas9/gRNA-mediated cleavage and negatively impact gene-drive dynamics and fi

A dietary risk assessment of the pyrethroid insecticide resmethrin associated with its use for West Nile Virus mosquito vector control in California.

An outbreak of human illnesses associated with West Nile Virus (WNV) occurred in New York City in 1999. Since then, it has gradually spread westwards, reaching northern California for the first time in 2005. WNV is transmitted by several mosquito species and birds serve as the main reservoir. Several control measures have been used, targeting both the aquatic larvae and the adult mosquitoes. In the latter case, roosting birds in trees are sprayed with pyrethroid insecticides because these are highly toxic to mosquitoes, but have low avian toxicity. A request was made to use a resmethrin-contai

Single immunization with genetically attenuated Pf∆mei2 (GA2) parasites by mosquito bite in controlled human malaria infection: a placebo-controlled randomized trial.

Malaria vaccines consisting of metabolically active Plasmodium falciparum (Pf) sporozoites can offer improved protection compared with currently deployed subunit vaccines. In a previous study, we demonstrated the superior protective efficacy of a three-dose regimen of late-arresting genetically attenuated parasites administered by mosquito bite (GA2-MB) compared with early-arresting counterparts (GA1-MB) against a homologous controlled human malaria infection. Encouraged by these results, we explored the potency of a single GA2-MB immunization in a placebo-controlled randomized trial. Primary

Pixel intensity of wing photos used to infer efficacy of mosquito control interventions against Anopheles gambiae caught during the RIMDAMAL II clinical trial.

Mosquito age-grading is important for evaluating mosquito control efforts and estimating pathogen transmission risk. We previously developed a simple, low-cost, and high-throughput method to age-grade mosquitoes by computing the pixel intensity (PI) of wing photos, which reflects wing scale loss over time. Here the technique was refined and used to understand wild Anopheles gambiae population structures from the RIMDAMAL II clinical trial. PI distributions from wing photos of wild An. gambiae caught during the trial reflected wild population structures assessed by gold standard techniques, whe…

Exploring the contributions of bed nets, cattle, insecticides and excitorepellency to malaria control: a deterministic model of mosquito host-seeking behaviour and mortality.

Domestic and personal protection measures against malaria exposure either divert host-seeking vectors to other hosts or kill those attempting to feed. Here, we explicitly model mosquito host-seeking processes in the context of local host availability and elucidate the impacts and mechanisms of pyrethroid-treated bed nets in Africa. It has been suggested that excitorepellent insecticides could increase exposure of unprotected humans by concentrating mosquito biting activity on this vulnerable group. This worst-case scenario is confirmed as a possibility where vector populations lack alternative

Integrating 3D video tracking with the standard WHO tunnel assay: a proof-of-concept to support improving insecticide-treated nets for mosquito control.

The World Health Organization (WHO) tunnel test is a standardised laboratory assay used to characterise the biological availability and potency of active ingredients on the surface of an insecticide-treated bed net (ITN) against host-seeking mosquitoes. However, the assay provides only endpoint measurements-the proportions of mosquitoes killed and blood-fed - and therefore offers no insight into how mosquitoes interact with the ITN. Therefore, complementary behavioural data would be highly valuable, for example to reveal the extent to which mosquitoes engage with the net, thereby helping to ex

RNA interference to combat the Asian tiger mosquito in Europe: A pathway from design of an innovative vector control tool to its application.

The Asian tiger mosquito Aedes albopictus is currently spreading across Europe, facilitated by climate change and global transportation. It is a vector of arboviruses causing human diseases such as chikungunya, dengue hemorrhagic fever and Zika fever. For the majority of these diseases, no vaccines or therapeutics are available. Options for the control of Ae. albopictus are limited by European regulations introduced to protect biodiversity by restricting or phasing out the use of pesticides, genetically modified organisms (GMOs) or products of genome editing. Alternative solutions are thus urg

Exploring Natural Alkaloids from Brazilian Biodiversity as Potential Inhibitors of the Aedes aegypti Juvenile Hormone Enzyme: A Computational Approach for Vector Mosquito Control.

This study explores the potential inhibitory activity of alkaloids, a class of natural compounds isolated from Brazilian biodiversity, against the mJHBP enzyme of the Aedes aegypti mosquito. This mosquito is a significant vector of diseases such as dengue, zika, and chikungunya. The interactions between the ligands and the enzyme at the molecular level were evaluated using computational techniques such as molecular docking, molecular dynamics (MD), and molecular mechanics with generalized Born surface area (MMGBSA) free energy calculation. The findings suggest that these compounds exhibit a hi

Towards green oviposition deterrents? Effectiveness of Syzygium lanceolatum (Myrtaceae) essential oil against six mosquito vectors and impact on four aquatic biological control agents.

Essential oils (EOs) from plants may be alternative sources of molecules toxic against mosquito vectors of public health relevance. Most of researches in this field focused on EOs as larvicides or ovicides, while limited efforts focused on the exploitation of EOs as oviposition deterrents. In the present study, the larvicidal and oviposition deterrent activity of Syzygium lanceolatum leaf EO was evaluated against six mosquito species, Anopheles stephensi, An. subpictus, Aedes aegypti, Ae. albopictus, Culex quinquefasciatus, and Cx. tritaeniorhynchus.
