Reduction of oral mucositis by palifermin (rHuKGF): dose-effect of rHuKGF.

PURPOSE: The aim of the present study was to determine the dose effect of palifermin (recombinant human keratinocyte growth factor, rHuKGF) for reduction of the response of oral mucosa to fractionated radiotherapy in a mouse model. MATERIAL AND METHODS: Ulceration (confluent mucositis) of mouse tongue epithelium was analysed as the clinically relevant endpoint. Palifermin at doses from 1 - 30 mg/kg was administered before the onset (day -3), at the end of the first (day +4) or the second week of irradiation (day +11) with 5 x 3 Gy/week. Each protocol was terminated by graded radiation test (t…

Risk factors for ulcerative oral mucositis in cancer patients: unanswered questions.

A multitude of laboratory and clinical research studies of ulcerative oral mucositis induced by cytotoxic cancer therapies have been reported during the past decade. However, a comprehensive understanding of oral mucositis pathogenesis, together with a clear definition of risk factors for development and severity of the lesion, remain under investigation. The literature presents sometimes divergent data regarding these issues, which in turn restrict efforts to develop a unified approach for management of this morbid condition. The current review summarizes these controversies and highlights th…

Role of mucosal IgA antibodies as novel therapies to enhance mucosal barriers.

To prevent infection, the experience of the recent severe acute respiratory syndrome coronavirus 2 (SARS-CoV2) pandemic has led to recognition of the importance of not only vaccines but also the strengthening of mucosal barriers by secretory immunoglobulin A (IgA). Strong mucosal barrier provided by IgA is also possible to prevent allergies and chronic inflammatory conditions in the intestinal tract, since it can protect foreign enemies or antigens at the first line of defense before their invasion. Therefore, it is important to understand the role of IgA antibodies secreted by the mucosa of t

Humulene Inhibits Acute Gastric Mucosal Injury by Enhancing Mucosal Integrity.

This study was designed to determine whether α-humulene, a major constituent in many plants used in fragrances, has a protective role against gastric injury in vivo and in vitro. A rat model of hydrochloric acid (HCl)/ethanol-induced gastritis and human mast cells (HMC-1) were used to investigate the mucosal protective effect of α-humulene. α-Humulene significantly inhibited gastric lesions in HCl/ethanol-induced acute gastritis and decreased gastric acid secretion pyloric ligation-induced gastric ulcers in vivo. In addition, α-humulene reduced the amount of reactive oxygen species and malondi

Sepsis and mucositis-related biomarkers in adult hematologic patients with febrile neutropenia.

Febrile neutropenia (FN) is a common complication of chemotherapy and potentially fatal. It is often associated with mucositis. Timely recognition of complications enables early supportive treatment and potentially reduces mortality. Common diagnostic parameters are often nonspecific or have slow kinetics. This study examines the applicability of mucosa-related biomarkers in the recognition of complicated course among 78 adult hematologic patients with FN after intensive chemotherapy. Plasma concentrations of citrulline, intestinal fatty acid binding protein (I-FABP), zonula occludens-1 (ZO-1)…
