Multisystem triglyceride storage disorder without ichthyosis in two siblings.

A four-year-old boy presented with hepatomegaly, vacuolized granulocytes (Jordans' anomaly) and slightly progressive myopathy as signs of multisystem triglyceride storage disease. The nine-year-old sister of the patient also showed Jordans' anomaly and early fatigability, but no overt weakness. Biochemical analysis revealed normal values for carnitines, carnitine palmityl transferase in serum and striated muscle, and beta-oxidation enzymes in striated muscles. Distribution of non-membrane-bound lipids in granulocytes, fibroblasts, smooth muscle cells and striated muscle was compatible with Cha

Characterization of vagal input to the rat esophageal muscle.

There is recent morphological evidence for an interaction of autonomic nerve fibers and extrinsic motor nerves of the rat esophagus. The aim of the present study was to investigate a possible functional role of this autonomic innervation of vagal motor fibers on rat esophageal smooth and striated muscle function in vitro. The entire esophagus with both Nn vagi, including the Nn recurrentes, was dissected and placed in an organ bath with oxygenated Krebs-Ringer buffer. Contractile activity was measured in longitudinal direction with a force transducer. Both Nn vagi were placed on a bipolar plat

Physiological and Pathological Relevance of Selective and Nonselective Ca2+ Channels in Skeletal and Cardiac Muscle.

Contraction of the striated muscle is fundamental for human existence. The action of voluntary skeletal muscle enables activities such as breathing, establishing body posture, and diverse body movements. Additionally, highly precise motion empowers communication, artistic expression, and other activities that define everyday human life. The involuntary contraction of striated muscle is the core function of the heart and is essential for blood flow. Several ion channels are important in the transduction of action potentials to cytosolic Ca2+ signals that enable muscle contraction; however, othe

Responses of feline esophagus to cervical vagal stimulation.

We used an in vivo feline model to study striated and smooth-muscle esophageal responses to cervical vagal stimulation (VS). Circular esophageal contractions were measured by intraluminal pressure sensors and longitudinal contractions by a force transducer. Responses to VS were recorded before and after giving succinylcholine, atropine, or both. Circular contractions characteristic of striated and smooth muscle were recorded from the mid- and distal esophagus, respectively. With increases in VS frequency, the striated muscle showed rapid, repetitive twitches progressing to tetany. These fast c

Malignant hyperthermia in a family. The ultrastructure of muscle biopsies of healthy members.

Twelve healthy relatives of two children who died of malignant hyperthermia--volunteered to give biopsies of striated muscle. Electron microscopy of their muscles revealed nonspecific myofibrillary degeneration and profound alterations of parts of muscle fibres. Furthermore, constructional faults were found in the architecture of the muscle. Previous investigations correlated with these abnormalities support the assumption, that inborn errors of metabolism may involve membranes of other tissues as well as striated muscle.

Effect of nebivolol on altered skeletal and cardiac muscles induced by dyslipidemia in rats: impact on oxidative and inflammatory machineries.

AIM: High cholesterol diet is greatly linked to deleterious health consequences. In this work we tried to explore direct effects of high cholesterol diet on striated (skeletal and cardiac) muscle tissues and the mechanisms by which nebivolol could improve such harmful effects. METHODS: The study included 24 healthy adult male albino rats weighing 200-220 grams that were assigned into four groups: control group, control drug group, high cholesterol diet fed groups; one untreated the other was treated with nebivolol. RESULTS: In the cholesterol fed group, we found decreased blood HDL and NO wi

On Structure-Function Relationships in the Female Human Urethra: A Finite Element Model Approach.

Remarkably little is known about urethral striated and smooth muscle and vascular plexus contributions to maintaining continence or initiating micturition. We therefore developed a 3-D, multiphysics, finite element model, based on sequential MR images from a 23-year-old nulliparous heathy woman, to examine the effect of contracting one or more individual muscle layers on the urethral closure pressure (UCP). The lofted urethra turned out to be both curved and asymmetric. The model results led us to reject the current hypothesis that the striated and smooth muscles contribute equally to UCP. Whi

Rescue of enzyme deficiency in embryonic diaphragm in a mouse model of metabolic myopathy: Pompe disease.

Several human genetic diseases that affect striated muscle have been modeled by creating knockout mouse strains. However, many of these are perinatal lethal mutations that result in death from respiratory distress within hours after birth. As the diaphragm muscle does not contract until birth, the sudden increase in diaphragm activity creates permanent injury to the muscle causing it to fail to meet respiratory demands. Therefore, the impact of these mutations remains hidden throughout embryonic development and early death prevents investigators from performing detailed studies of other striat

Presence of smooth muscle continuous with the rectal and vaginal walls in the deep perineal space prompts reconsideration of the deep transverse perineal muscle.

The deep transverse perineal muscle was considered a striated muscle within the deep perineal space; however, recent studies suggest that it consists of smooth muscle. We investigated the histological composition and spatial distribution of the deep perineal space and its anatomical relationships with surrounding structures to understand its role in the perineum. Perineal muscle shape and location from nine cadavers (five males; average age, 79 years) were macroscopically examined. Serial sections were histologically analyzed using Masson's trichrome, Elastica Masson, and immunostaining of smo

[Growth and differentiation of adult canine autologous skeletal myoblasts after transplanted into acute myocardial infarction region].

OBJECTIVE: To study the growth and differentiation of adult canine autologous skeletal myoblasts after being transplanted into acute myocardial infarction (AMI) region by intramyocardium injection (IMI) and intracoronary infusion (ICI). METHODS: Autologous skeletal myoblasts were procured by a modified method. AMI model of adult canine was obtained through left anterior descending branch ligation and was divided into 4 groups (n = 5 for each group). Autologous skeletal myoblasts (1.0 - 1.4 x 10(8) cells) were injected locally into AMI region or infused into infarction-related coronary artery.

Abnormalities in esophageal smooth muscle induced by mutations in collagen XIX.

Collagen XIX is a nonfibrillar collagen that localizes in restricted tissues at very low amounts. A previous study on Col19a1 null mice revealed that collagen XIX is involved in esophageal muscle physiology and morphogenesis. Here, we use histological analysis to show that mice with a Col19a1 mutant lacking the NC3 domain and seven collagen triplets display abnormal transition of smooth to striated muscle in the abdominal segment of esophagus, and a widened esophagus with age. With two newly prepared antibodies, we analyzed the expression of collagen XIX in the mouse esophagus and show that co

[The striated sphincter of the urethra. 1: Recall of knowledge on the striated sphincter of the urethra].

The first part of this report is devoted to the anatomy, nerve supply, and physiology of the striated sphincter, three subjects which greatly aroused a great deal of controversies. The conclusions of the authors are as follows. The striated sphincter of the urethra is a muscle of the perineum, arising from the cloacal sphincter. The deep transverse, bulbo-cavernous, anal sphincter, ischio-cavernous and superficial transverse muscles have the same embryological origin and together with the sphincter of the urethra form the accessory pelvic diaphragm. The embryological origin of the levator ani

Urethral pharmacological mechanisms incontinence and bladder emptying: An updated review.

BACKGROUND: The urethral wall consists of layers of striated muscle, circular and longitudinal smooth muscles, collagen fibers, and a vascular plexus. However, the relative contributions of these components to urethral pressure in humans remain poorly understood. The circular and longitudinal smooth muscle components can develop a spontaneous contractile activity, generating a basal tone. They can further contract or relax in response to excitatory or inhibitory stimuli. Animal studies suggest that smooth muscle activity in the mid-urethra plays a crucial role in determining maximal urethral c

Tissue Engineering Applied to Skeletal Muscle: Strategies and Perspectives.

Muscle tissue is formed by elongated and contractile cells with specific morphofunctional characteristics. Thus, it is divided into three basic types: smooth muscle tissue, cardiac striated muscle tissue and skeletal striated muscle tissue. The striated skeletal muscle tissue presents high plasticity, regeneration and growth capacity due to the presence of satellite cells, quiescent myoblasts that are activated in case of injury to the tissue and originate new muscle fibers when they differentiate. In more severe deficiencies or injuries there is a loss of their regenerative capacity, thus com

Role of male pelvic floor muscles and anterior fibromuscular stroma in males on α(1)-blocker treatment: a magnetic resonance imaging study.

Dynamic motion of the pelvic floor muscles during voiding was analyzed using real-time magnetic resonance imaging. To evaluate the contraction of the pelvic floor muscles, striated urethral sphincter distance, levator ani muscle thickness and anterior fibromuscular stroma distance were measured. The percent contraction of the striated urethral sphincter from before voiding to just before initiation of voiding was 14% in the normal group and 5% in the voiding dysfunction group. The percent contraction of the anterior fibromuscular stroma from before voiding to just before initiation of voiding

Role of puborectalis muscle in the genesis of urethral pressure.

PURPOSE: The internal (smooth muscle) and the external (rhabdosphincter striated muscle) urethral sphincters have important roles in the genesis of urethral closure pressure. The U-shaped pelvic floor puborectalis muscle is important in the closure of anal and vaginal orifices in humans. We defined the contribution of the puborectalis to urethral pressure. MATERIALS AND METHODS: A total of 11 female rabbits were anesthetized and prepared to measure urethral, vaginal and anal canal pressure using manometric methods. Pressure was recorded at rest, after administration of pharmacological agents
