Metabolomics Suggests That Soil Inoculation with Arbuscular Mycorrhizal Fungi Decreased Free Amino Acid Content in Roots of Durum Wheat Grown under N-Limited, P-Rich Field Conditions.

Arbuscular mycorrhizal fungi (AMF) have a major impact on plant nutrition, defence against pathogens, a plant's reaction to stressful environments, soil fertility, and a plant's relationship with other microorganisms. Such effects imply a broad reprogramming of the plant's metabolic activity. However, little information is available regarding the role of AMF and their relation to other soil plant growth-promoting microorganisms in the plant metabolome, especially under realistic field conditions. In the present experiment, we evaluated the effects of inoculation with AMF, either alone or in co

Broad-scale shift in dominance of arbuscular mycorrhizal trees along the Japanese archipelago associated with tree diseases and climate warming.

Distribution shifts of mycorrhizal tree symbioses are predicted to alter the nutrient cycling in temperate forests worldwide. Yet, our understanding of the drivers and specific impacts of such shifts remains limited. Here, we assign mycorrhizal types to trees at the species and genus level in >13,000 forest plots to assess the dominance of arbuscular mycorrhizal trees based on basal area proportions along the Japanese archipelago in temperate East Asia. We show a significant increase in arbuscular mycorrhizal tree dominance within 15 years associated with climate warming and the spread of tree

Belowground Fungal Volatile Exposure Induces Systemic Metabolic Reprogramming in Poplar.

Although trees encounter diverse soil-associated fungi, it is unclear how they adjust their physiology in response to volatile organic compounds (VOCs) from fungi with contrasting ecological strategies before physical contact. Here, we tested whether VOCs emitted by fungi are sufficient to induce systemic, fungus-dependent metabolic responses in poplar. Populus × canescens roots were exposed to VOCs from a pathogen (Heterobasidion annosum), a saprotroph (Postia placenta), or an ectomycorrhizal mutualist (Laccaria bicolor) for 6 weeks in a contact-free pot-in-pot system. Untargeted LC-MS metabo

Microbial communities in the rhizosphere of three Mentha species: Links to soil properties and essential oil profiles.

Mentha species are widely cultivated aromatic plants valued for their essential oils and antimicrobial properties. However, despite their agricultural and pharmacological significance, limited information is available on how different Mentha species influence rhizosphere microbial communities and their relationships with soil physicochemical parameters and essential oil composition. In this study, we examined the rhizosphere microbiota of three closely related taxa - Mentha × villosa B10, M. spicata B17, and M. suaveolens J17 - cultivated under uniform field conditions. Rhizosphere and bulk so

Differential Responses of Tree Phyllosphere and Rhizosphere Microbiomes to Mycorrhizal Types and Planting Patterns in a Young Subtropical Forest Plantation.

Arbuscular mycorrhizal (AM) and ectomycorrhizal (EM) tree species are ubiquitous in subtropical forests and have distinctive root colonisation characteristics and leaf traits, resulting in differences in nutrient acquisition strategies and ecological functions. Here, we investigated the responses of bacteria, fungi and protists inhabiting tree phyllosphere and rhizosphere to tree mycorrhizal types (AM vs. EM) and planting patterns (single or double tree species planting with the same or different mycorrhizal type) in two seasons. Both leaf- and root-associated fungal richness and community com

Amazonian understory forests change phosphorus acquisition strategies under elevated CO2.

The potential for the Amazon forest to continue functioning as a carbon (C) sink strongly depends on soil nutrient availability, particularly phosphorus (P), and on plants' ability to adjust nutrient acquisition strategies. However, limited experimental evidence constrains the mechanistic representation of nutrient-carbon interactions in climate change models. Here, we conducted an experiment in a P-depleted Amazonian understory forest, increasing atmospheric CO₂ in-situ by approximately 300 ppm using open-top chambers. We show that elevated CO₂ (eCO₂) induced contrasting responses by roots al

Impact of agricultural systems on arbuscular mycorrhizal fungi community composition in robusta coffee roots in the Democratic Republic of congo.

Robusta coffee, grown by 25 million farmers across more than 50 countries, plays an important role in smallholder farmers' livelihoods and the economies of many low-income countries. Coffee establishes a mutualistic symbiosis with arbuscular mycorrhizal fungi (AMF); however, the impact of agricultural practices and soil characteristics on AMF diversity and community composition is not well understood. To address this, we characterised the AMF community composition of robusta coffee in part of its region of origin, the Democratic Republic of Congo. AMF diversity and community composition were c

Orchid mycorrhizal fungus from the Brazilian Cerrado exhibits biocontrol activity against Meloidogyne javanica.

Waitea circinata, an orchid-associated mycorrhizal fungus, was evaluated for the first time as a biological control agent against the root-knot nematode Meloidogyne javanica. The fungus was originally isolated from the roots of Epidendrum nocturnum, a rupicolous orchid to the Brazilian Cerrado. In vitro assays demonstrated that W. circinata parasitized nematode eggs, with parasitism rates of 13-15% and characteristic emergence of septate hyphae with right-angle branching. Microscopy revealed eggshell degradation, arrested embryogenesis, and juvenile deformation. A mycelial suspension (MSF) of

Microbial island biogeography: isolation shapes the life history characteristics but not diversity of root-symbiotic fungal communities.

Island biogeography theory is one of the most influential paradigms in ecology. That island characteristics, including remoteness, can profoundly modulate biological diversity has been borne out by studies of animals and plants. By contrast, the processes influencing microbial diversity in island systems remain largely undetermined. We sequenced arbuscular mycorrhizal (AM) fungal DNA from plant roots collected on 13 islands worldwide and compared AM fungal diversity on islands with existing data from mainland sites. AM fungal communities on islands (even those >6000 km from the closest mainlan

Mycorrhizal technology and phosphorus in the production of primary and secondary metabolites in cebil (Anadenanthera colubrina (Vell.) Brenan) seedlings.

BACKGROUND: The application of arbuscular mycorrhizal fungi (AMF) can increase the growth and concentration of primary and secondary metabolites in several plant species. Cebil (Anadenanthera colubrina), a medicinal plant, benefits from mycorrhizal association, but the influence of the symbiosis on the production of its bioactive compounds is unknown. In this study the effect of mycorrhizal inoculation and phosphorus (P) supply on the production of primary and secondary metabolites in cebil seedlings was determined. RESULTS: The production of proteins and carbohydrates in terms of both concen

Effect of arbuscular mycorrhizal colonization and two levels of compost supply on nutrient uptake and flowering of pelargonium plants.

Two challenges frequently encountered in the production of ornamental plants in organic horticulture are: (1) the rate of mineralization of phosphorus (P) and nitrogen (N) from organic fertilizers can be too slow to meet the high nutrient demand of young plants, and (2) the exclusive use of peat as a substrate for pot-based plant culture is discouraged in organic production systems. In this situation, the use of beneficial soil microorganisms in combination with high quality compost substrates can contribute to adequate plant growth and flower development. In this study, we examined possible a

Loss of the flagellar regulator FlhC changes the transcriptional response of plant-associated Acidovorax delafieldii strains to metabolites from Rhizophagus irregularis-colonized Lotus japonicus roots.

Acidovorax is a cosmopolitan bacterial genus comprising both beneficial and pathogenic plant-associated species. In plant-associated bacteria, flagella can facilitate colonization but also trigger plant immune responses driving mutation or loss of flagella in some strains. FlhC is an important transcriptional regulator of flagella assembly genes. Here, we investigated whether it modulates root colonization and the transcriptional response of plant-commensal Acidovorax strains to different plant cues. As model cues, we used root extracts from Lotus japonicus that forms symbioses with arbuscular

Role of phosphate solubilizing Burkholderia spp. for successful colonization and growth promotion of Lycopodium cernuum L. (Lycopodiaceae) in lateritic belt of Birbhum district of West Bengal, India.

Profuse growth of Lycpodium cernuum L. was found in phosphate deficient red lateritic soil of West Bengal, India. Interaction of vesicular-arbuscular mycorrhiza (VAM) with Lycopodium rhizoids were described earlier but association of PGPR with their rhizoids were not studied. Three potent phosphate solubilizing bacterial strains (P4, P9 and P10) associated with L. cernuum rhizoids were isolated and identified by 16S rDNA homologies on Ez-Taxon database as Burkholderia tropica, Burkholderia unamae and Burkholderia cepacia respectively. The Effect of Mycorrhization with Fungi of Different Efficiency on the Root Metabolome of Medicago lupulina Within Development.

The mechanisms underlying the symbiotic efficiency of arbuscular mycorrhizal (AM) fungi are actively debated, but comparative metabolomic studies with fungi of contrasting efficiency are scarce. This study aimed to evaluate the influence of effective (Rhizophagus irregularis RCAM00320) and ineffective (Glomus sp. 129.1Te) AM fungal strains on the root metabolome of the responsive Medicago lupulina line MlS-1 at two vegetative and two reproductive stages. Using GC-MS, over 150 metabolites (amino acids, carboxylic and fatty acids, sugars, etc.) were annotated. Effective AM symbiosis was associat

From Sampling to Identification of Arbuscular Mycorrhizal Fungi Through Next Generation Sequencing.

In recent years, DNA sequencing technologies have advanced considerably with the rise of Next-Generation Sequencing (NGS) platforms, which have transformed microbial ecology research. These approaches enable the characterization of entire communities by using DNA traces to identify organisms taxonomically from a single sample. Arbuscular mycorrhizal fungi (AMF) are no exception and represent one of the most extensively studied groups of soil fungi. This chapter presents protocols for high-throughput, sequence-based analysis of AMF communities, covering the complete workflow from DNA extraction

New record of ectomycorrhizal association of the genus Nothojafnea in the Northern Hemisphere.

Nothojafnea is recognized as an ectomycorrhizal (ECM) fungal genus currently comprising only two species, both previously reported exclusively from the Southern Hemisphere. Although this genus has been taxonomically recognized, detailed descriptions of its ECM morphology and anatomical characteristics have remained limited and, in some cases, ambiguous. Simultaneously, molecular data for this genus deposited in the NCBI database are still quite limited, with a substantial proportion consisting of unpublished sequences. In this study, we report the first occurrence of Nothojafnea in Vietnam, ba

Effects of arbuscular mycorrhizal inoculation on the phytoremediation of PAH-contaminated soil: A meta-analysis.

Inoculation with arbuscular mycorrhizal (AM) fungi can accelerate the phytoremediation process by increasing plant biomass and improving soil physicochemical and biological characteristics. However, a quantitative, data-based conclusion is yet to be derived on the roles of AM fungi in remediating soils polluted by polycyclic aromatic hydrocarbons (PAHs), and the impact factors are unclear. To address these issues, we performed a meta-analysis of 45 articles to estimate the effects of AM inoculation on the phytoremediation of soils polluted by PAHs and to examine the influence of experimental c

An ectomycorrhizal fungus alters sensitivity to jasmonate, salicylate, gibberellin, and ethylene in host roots.

The phytohormones jasmonate, gibberellin, salicylate, and ethylene regulate an interconnected reprogramming network integrating root development with plant responses against microbes. The establishment of mutualistic ectomycorrhizal symbiosis requires the suppression of plant defense responses against fungi as well as the modification of root architecture and cortical cell wall properties. Here, we investigated the contribution of phytohormones and their crosstalk to the ontogenesis of ectomycorrhizae (ECM) between grey poplar (Populus tremula x alba) roots and the fungus Laccaria bicolor. The arbuscular mycorrhizal fungus Rhizophagus irregularis MUCL 41833 increases the phosphorus uptake and biomass of Medicago truncatula, a benzo[a]pyrene-tolerant plant species.

The accumulation of phosphorus (P) in plants increases their biomass and resistance/tolerance to organic pollutants. Both characteristics are mandatory for the utilization of plants in phytoremediation. Arbuscular mycorrhizal (AM) fungi improve plant P nutrition, and thus growth. However, only a few studies have focused on the dynamics of inorganic P (Pi) uptake in AM fungal-colonized plants in the presence of organic pollutants. Indeed, most of the results so far were obtained after harvesting the plants, thus by evaluating P concentration and content at a single time point. Here, we investig

Hijacking common mycorrhizal networks for herbivore-induced defence signal transfer between tomato plants.

Common mycorrhizal networks (CMNs) link multiple plants together. We hypothesized that CMNs can serve as an underground conduit for transferring herbivore-induced defence signals. We established CMN between two tomato plants in pots with mycorrhizal fungus Funneliformis mosseae, challenged a 'donor' plant with caterpillar Spodoptera litura, and investigated defence responses and insect resistance in neighbouring CMN-connected 'receiver' plants. After CMN establishment caterpillar infestation on 'donor' plant led to increased insect resistance and activities of putative defensive enzymes, induc
