The timing of enteric neural crest cell colonisation of the chick embryo cloaca.

Neural crest cell (NCC) migration and formation of the enteric nervous system (ENS) is an essential process in the development of the normal human gut. Abnormalities of the ENS lead to a number of neurochristopathies. In avian embryos, the cloaca acts as a common chamber into which gastrointestinal, urinary and genital tracts emerge. Previous studies have elucidated the specific timeframes at which NCCs reach the various regions of the developing chick gut but, to date, none have looked at NCC colonisation of the cloaca. The aim of our study was to investigate the exact timing of the appearanc…

Effects of retinoic acid on the neural crest-controlled organs of fetal rats.

Prenatal exposure of rat embryos to retinoic acid induces severe malformations involving various organs. The mechanisms of this embryopathy are known only in part. This study describes the malformations of the neural crest-derived organs in this model and shows that many of them fit into the pattern of disturbed neural crest organogenic control. Pregnant rats were exposed to either all-trans retinoic acid (125 mg/kg; n=17) or vehicle ( n=10) on E10. Fetuses were recovered on E21 and external and internal malformations were sought. The craniofacial area, the trachea, parathyroids, thymus, thyro…

Sox10 is required for the early development of the prospective neural crest in Xenopus embryos.

The Sox family of transcription factors has been implicated in the development of different tissues during embryogenesis. Several mutations in humans, mice, and zebrafish have shown that depletion of Sox10 activity produces defects in the development of neural crest derivatives, such as melanocytes, ganglia of the peripheral nervous system, and some specific cell types as glia. We have isolated the Xenopus homologue of the Sox10 gene. It is expressed in prospective neural crest and otic placode regions from the earliest stages of neural crest specification and in migrating cranial and trunk ne…

The role of the epicardium and neural crest as extracardiac contributors to coronary vascular development.

At species-specific times in embryonic development, the pro-epicardial organ appears as an outcropping of the mesothelial body wall, near th … to interstitial fibroblasts and adventitial and smooth muscle cells of the coronary arteries. Using neural crest tracings in mouse embryos (Wnt1-Cre-lacZ), we studied the patterning of cardiac neural crest cells during development. Participation of neural crest cells in the formation of the vascular media could not be excluded, although epicardium-derived cell …

The effects of mesencephalic neural crest cell extirpation on the development of chicken embryos.

The mesencephalic neural crest cells of Hamburger-Hamilton Stage 9-Stage 11 chick embryos were surgically extirpated unilaterally in 148 embryos and bilaterally in 8 embryos. Sham operations were performed unilaterally on 16 control embryos and bilaterally on one control embryo. Embryos were fixed at various time intervals after operation, studied macroscopically, and by light and scanning electron microscopy, and their development compared with that of 47 normal embryos. The extirpated mesencephalic region was repopulated by crest cells within 6-8 hours after operation. These 'new' crest cell…

Regulative response of the cranial neural tube after neural fold ablation: spatiotemporal nature of neural crest regeneration and up-regulation of Slug.

After unilateral ablation of the avian cranial neural folds, the remaining neuroepithelial cells are able to replace the missing neural crest population (Scherson et al., 1993). Here, we characterize the cellular and molecular nature of this regulative response by defining: (1) the time and location of neural crest cell production by the neuroepithelium; (2) rostrocaudal axial differences in the regulative response; and (3) the onset of expression of Slug, a transcription factor present in premigratory and migrating neural crest cells. Using DiI and HNK-1 antibody labeling techniques, we find …

Establishment and neural differentiation of neural crest-derived stem cells from human dental pulp in serum-free conditions.

The potential of obtaining cell cultures with neural crest resemblance (neural crest-derived stem cells [NCSCs]) from dental-related tissues, including human dental pulp cells (hDPCs), has been discussed in the literature. However, most reports include the use of serum-rich conditions and do not describe the potential for neural differentiation, slowing translation to the clinic. Therefore, we aimed to culture and characterize NCSCs from the human dental pulp in vitro and evaluate their ability to differentiate into neurons; we also investigated the effectiveness of the addition of BMP4 to enh…

Aberrant neural crest cells migration leads to melanocyte presence in the umbilical cord.

BACKGROUND: Systematic evaluation of the umbilical cord is an integral part of placental examination. Among the important parameters of this … of melanocytes and melanin pigment in the umbilical cord. We propose that an aberrant migration of neural crest cells into the umbilical cord, with subsequent differentiation into melanocytes, is the most likely pathogenesis.

In Vivo Differentiation of hESC-Derived Neural Crest Cells into Trabecular Meshwork Cells.

Primary open-angle glaucoma is a prevalent type of degenerative eye disease that results in lifelong blindness, and its critical pathogenic cause is trabecular meshwork (TM) dysfunction or decreased TM cellularity. Considering that TM develops from neural crest cells (NCCs), we investigate the potential of human embryonic stem cell (hESC)-derived NCCs transplantation for TM regeneration. We used a chemically defined method to induce the differentiation of NCCs and injected 1.0 × 106 hESC-derived NCCs combined with 100 μmol/L Y-27632 into the anterior chamber of rabbit. Intraocular pressure (IO…

Core microRNAs regulate neural crest delamination and condensation in the developing trigeminal ganglion.

Cranial neural crest cells (NCCs) undergo dynamic processes during embryonic development, including delamination from the neural tube by epithelial-to-mesenchymal transition (EMT), migration to the periphery, condensation via mesenchymal-to-epithelial transition (MET), and differentiation into structures like the trigeminal ganglion. Here, we identify and characterize the function of a core set of miRNAs involved in these transitions during the formation of the trigeminal ganglion in the chick embryo. We further identify putative targets of miRNAs involved in neural crest EMT and MET. Notably,…

Adult leptomeningeal vestigial neural crest-derived multipotent cells promote vascular repair after stroke.

The neural crest (NC) is a transient embryonic structure composed of highly migratory multipotent stem cells that generate diverse cell types and orchestrate early neurovascular patterning. It has long been assumed that NC cells are exhausted after development once their progeny fully differentiate. However, through NC lineage tracing, single-cell and spatial transcriptomics, interactome modeling, and in vivo imaging, we identified a population of NC-derived multipotent (not lineage-restricted) cells persisting within the adult mouse leptomeninges. Following ischemic stroke, loss- and gain-of-…

Associations between birth defects with neural crest cell origins and pediatric embryonal tumors.

BACKGROUND: There are few assessments evaluating associations between birth defects with neural crest cell developmental origins (BDNCOs) and embryonal tumors, which are characterized by undifferentiated cells having a molecular profile similar to neural crest cells. The effect of BDNCOs on embryonal tumors was estimated to explore potential shared etiologic pathways and genetic origins. METHODS: With the use of a multistate, registry-linkage cohort study, BDNCO-embryonal tumor associations were evaluated by generating hazard ratios (HRs) and 95% confidence intervals (CIs) with Cox regression…

Glycosaminoglycans promote osteogenesis from human induced pluripotent stem cells via neural crest induction.

The stepwise development of bone is rigidly controlled from mesenchymal cells through osteoblasts. Dysregulation of this process causes various bone diseases, such as osteoporosis and osteogenesis imperfecta. Recently, it has been noted that the decrease in bone density due to aging occurs not only in the axial skeleton but also in the facial bone. To address this issue, we focused on neural crest-derived osteoblasts that form craniofacial bone, and evaluated several functional ingredients that have been reported to activate osteoblast function using mineralization ability as an index. Glucosa…

NB-UVB Induces Melanocytic Differentiation of Human Hair Follicle Neural Crest Stem Cells.

BACKGROUND: Phototherapy is an important method to treat vitiligo. However, it is unclear how phototherapy affects melanocyte precursors and skin neural crest stem cells. OBJECTIVE: To investigate the underlying mechanisms of narrow-band ultraviolet B (NB-UVB) induced melanocyte lineage differentiated from human scalp-derived neural crest stem cells (HS-NCSCs). METHODS: HS-NCSCs were expanded from scalp hair follicles. The c-Kit-/CD57- HS-NCSCs were isolated by cell sorting. Different doses of NB-UVB were used to irradiate these HS-NCSCs. Cell ultrastructure was examined by transmission elec…

Olig2 positive cells derived from hair follicle neural crest stem cells in rats.

Motor neuron disease (MND) is a kind of common clinical nervous system disease with typical characteristic of progressive motor neurons degeneration or death. Motor neuron derived from stem cells or motor neuron progenitor cells will be a good choice to be used for treatment of the disease. In this study, we used the combination of 5 small molecular including CHIR99021 (CHIR), SB431542 (SB), DMH1 (DMH), retinoic acid (RA) and Purmorphamine (Pur) to induce hair follicles neural crest stem cells (hfNCSCs) to motor neurons progenitors (MNPs). Valproic acid (VPA) was used to make MNPs proliferatio…

Mesenteric Neural Crest Cells Are the Embryological Basis of Skip Segment Hirschsprung's Disease.

BACKGROUND & AIMS: Defective rostrocaudal colonization of the gut by vagal neural crest cells (vNCCs) results in Hirschsprung's disease (HSCR), which is characterized by aganglionosis in variable lengths of the distal bowel. Skip segment Hirschsprung's disease (SSHD), referring to a ganglionated segment within an otherwise aganglionic intestine, contradicts HSCR pathogenesis and underscores a significant gap in our understanding of the development of the enteric nervous system. Here, we aimed to identify the embryonic origin of the ganglionic segments in SSHD. METHODS: Intestinal biopsy speci…

Neogenin-loss in neural crest cells results in persistent hyperplastic primary vitreous formation.

Neogenin is a transmembrane receptor critical for multiple cellular processes, including neurogenesis, astrogliogenesis, endochondral bone formation, and iron homeostasis. Here we present evidence that loss of neogenin contributes to pathogenesis of persistent hyperplastic primary vitreous (PHPV) formation, a genetic disorder accounting for ~ 5% of blindness in the USA. Selective loss of neogenin in neural crest cells (as observed in Wnt1-Cre; Neof/f mice), but not neural stem cells (as observed in GFAP-Cre and Nestin-Cre; Neof/f mice), resulted in a dysregulation of neural crest cell migratio…

Induction of Neural Crest Stem Cells From Bardet-Biedl Syndrome Patient Derived hiPSCs.

Neural crest cells arise in the embryo from the neural plate border and migrate throughout the body, giving rise to many different tissue types such as bones and cartilage of the face, smooth muscles, neurons, and melanocytes. While studied extensively in animal models, neural crest development and disease have been poorly described in humans due to the challenges in accessing embryonic tissues. In recent years, patient-derived human induced pluripotent stem cells (hiPSCs) have become easier to generate, and several streamlined protocols have enabled robust differentiation of hiPSCs to the neu…
