Modeling axonal injury in vitro: injury and regeneration following acute neuritic trauma.

Traumatic injury to axons was modeled in vitro using sympathetic principal neurons from the rat superior cervical ganglion. Neurons were grown as a pure culture on collagen in parallel tracks, with cell somata confined to the center, and neurites occupying the periphery of the culture dish. Growing as fascicles on tracks, the neurites demonstrated periodic varicosities. Neuritic transection was reliably and reproducibly achieved with a motor driven rubber impactor injury device. During a period lasting at least 1 h, dieback involving the proximal neurites averaged 105 +/- 10 microm. This was f…

Neurite fasciculation mediated by complexes of axonin-1 and Ng cell adhesion molecule.

Neural cell adhesion molecules composed of immunoglobulin and fibronectin type III-like domains have been implicated in cell adhesion, neurite outgrowth, and fasciculation. Axonin-1 and Ng cell adhesion molecule (NgCAM), two molecules with predominantly axonal expression exhibit homophilic interactions across the extracellular space (axonin- 1/axonin-1 and NgCAM/NgCAM) and a heterophilic interaction (axonin-1-NgCAM) that occurs exclusively in the plane of the same membrane (cis-interaction). Using domain deletion mutants we localized the NgCAM homophilic binding in the Ig domains 1-4 whereas h

Tenascin is accumulated along developing peripheral nerves and allows neurite outgrowth in vitro.

The extracellular matrix protein, tenascin, appears in a restricted pattern during organ morphogenesis. Here we studied the expression of te … /tenascin substrates and sprouted rapidly after a lag of several hours. Outgrowth was inhibited and neurites were detached by anti-tenascin antibodies. On substrates coated with tenascin alone, neurite outgrowth was achieved from 3 day spinal cord explants. Whereas growth cones were well spread and rapidly moving, the neurites were poorly attached, straigh …

Down patients: extracellular preamyloid deposits precede neuritic degeneration and senile plaques.

Using anti-SP28 (a polyclonal antibody to a 28 residue synthetic peptide homologous to the NH2-terminal region of the Alzheimer amyloid beta-protein) to investigate the cerebral cortex of 6 Down patients aged 6-55 y, we found that, besides senile plaques and congophilic vessels, extracellular deposits unrelated to degenerating neurites, tangle-bearing neurons or congophilic vessels were labelled. These deposits were similar to the extracellular deposits previously observed in the cerebral cortex of Alzheimer patients and non-demented individuals. The material accumulated in the deposits did no…

Fasciculation of granule cell neurites is responsible for the perpendicular orientation of small inhibitory interneurons in mouse cerebellar microexplant cultures in vitro.

To study the cellular and molecular mechanisms involved in the perpendicular orientation of stellate and/or basket cells to the direction of fasciculating granule cell neurites, we have used cultures of microexplants from early postnatal mouse cerebellum that show this cellular behaviour in vitro. When these cultures were maintained in the presence of antibodies to the neural cell adhesion molecules L1 and N-CAM or immunoglobulin-like domains I, II and IV of N-CAM, the resulting decrease or increase in the fasciculation of granule cell neurites changed the perpendicular orientation and morphol…

Evidence that paired helical filaments originate from neurofilaments--electron microscope observations of neurites in senile plaques in the brain in Alzheimer's disease.

Neurites in senile plaques in the cerebral cortex from an autopsied case of Alzheimer's disease were examined by electron microscope. Swollen axons containing vastly increased numbers of neurofilaments (NF) together with paired helical filaments (PHF) were common. In some of these axons the continuity of NF with PHF was frequently discernible. In one, there were numerous figures of continuity in a bundle, indicating transition from NF to PHF. Moreover, some of these figures showed that two NF seemed to join to form one PHF. No straight 15 nm filament was noted anywhere in the sectioned plane o…

14-3-3 protein sigma isoform co-localizes with phosphorylated α-synuclein in Lewy bodies and Lewy neurites in patients with Lewy body disease.

α-Synuclein shares structural homology with 14-3-3 proteins. Seven 14-3-3 protein isoforms have been identified in mammals. Among them, the 14-3-3 sigma isoform was initially considered absent in the mammalian brain. However, we previously identified immunohistochemical association of 14-3-3 sigma with Pick bodies. Because 14-3-3 isoforms other than sigma isoform have been identified in Lewy bodies, we were prompted to look for this 14-3-3 sigma-like immunoreactivity (IR) in Lewy bodies in the brainstem, cerebral cortex, and Lewy neurites in seven patients with Lewy body disease. Unexpectedly,…

Human neural stem cells survive long term in the midbrain of dopamine-depleted monkeys after GDNF overexpression and project neurites toward an appropriate target.

Transplanted multipotent human fetal neural stem cells (hfNSCs) significantly improved the function of parkinsonian monkeys in a prior study primarily by neuroprotection, with only 3%-5% of cells expressing a dopamine (DA) phenotype. In this paper, we sought to determine whether further manipulation of the neural microenvironment by overexpression of a developmentally critical molecule, glial cell-derived neurotrophic factor (GDNF), in the host striatum could enhance DA differentiation of hfNSCs injected into the substantia nigra and elicit growth of their axons to the GDNF-expressing target.

Alzheimer's disease-like dystrophic neurites characteristically associated with senile plaques are not found within other neurodegenerative diseases unless amyloid beta-protein deposition is present.

Swollen, bulbous-shaped (dystrophic) neurites are a common pathologic feature of Alzheimer's disease (AD) and represent one of the most abundant neuritic abnormalities within the brains of patients with this disease. In the present study, we sought to determine whether the dystrophic neurites which are observed in association with senile plaques are unique to AD or whether they are characteristic of a more generalized process of neuritic and/or neuronal degeneration which can be observed in other neurodegenerative diseases. To accomplish this, we examined post-mortem brain material from patien…

Role of olfactory receptor Olfr544 in neuronal cells: Insights into neurite outgrowth and CREB signaling.

Olfactory receptors (ORs) are expressed beyond the nasal epithelium and implicated in brain function, yet their roles remain unclear. This study investigated the function of the OR Olfr544, which has been reported to be highly expressed near amyloid plaques in Alzheimer's disease model mice, using the mouse neuroblastoma N2a cell line. We found that its agonist azelaic acid (AZA) promotes neurite outgrowth and activates the cAMP-PKA-CREB signaling cascade in an Olfr544-dependent manner. Transcriptome analysis of Olfr544-knockout cells revealed altered expression of genes involved in neuroactiv

Neuroprotective Effects of Two N-Benzylamides in Models of Oxidative Stress, Neurite Outgrowth and Neurodegeneration.

Oxidative stress is a central pathological mechanism in neurodegenerative diseases (NDDs), contributing to mitochondrial dysfunction, impaired proteostasis, and progressive neuronal loss. The sigma-1 receptor (S1R) is an endoplasmic reticulum (ER)-resident chaperone that coordinates cellular stress responses and has emerged as a promising therapeutic target for neuroprotection. In this study, we investigated the neuroprotective properties and mechanisms of the structurally related N-benzylamides, N-benzylcinnamide (NBCA) and N-benzylbenzamide (NBBA) using complementary in vitro, in vivo and in

A rare occurrence of lepra Type II reaction in pure neuritic leprosy: a case report.

BACKGROUND: Pure neuritic leprosy (PNL) is an uncommon clinical form of leprosy characterized by nerve involvement without skin lesions, making diagnosis challenging. Type II lepra reaction (erythema nodosum leprosum, ENL) is rarely associated with PNL. CASE PRESENTATION: We report a 39-year-old male from Harar, Ethiopia, who presented with a 10-month history of progressive numbness, tingling, and weakness in the left hand. Clinical examination revealed thickened and tender ulnar nerve, sensory loss, hypothenar atrophy, and clawing of the ring and little fingers. Nerve conduction studies confi

Diagnosis and therapeutic outcomes of biopsy-confirmed Pure Neuritic Leprosy: A two-decade experience.

BACKGROUND: Pure neuritic leprosy (PNL) is a distinct form of leprosy characterized by peripheral nerve involvement without skin lesions. Diagnosis is often delayed, and 'silent neuritis' contributes to progressive and irreversible nerve damage. Despite appropriate therapy, many patients develop long-term disability. This study describes the clinical profile, diagnostic patterns, therapeutic outcomes, and predictors of disability in biopsy-confirmed PNL. METHODS: We conducted a retrospective observational cohort study at a tertiary referral centre in South India, enrolling biopsy-confirmed PN

Methylone promotes neurite outgrowth and has long-lasting effects on fear extinction learning.

Post-traumatic stress disorder (PTSD) is a prevalent and debilitating disorder, and available treatments are limited. TSND-201 (methylone) is in clinical trials for PTSD, showing potential to have rapid, robust and long-lasting benefit without direct agonist/antagonist activity at 5HT2A. Alterations in structural neuroplasticity are a well-studied mechanism that may underlie both the pathophysiology and treatment of PTSD. Previous work showed that methylone rapidly induced neuroplasticity-related factors in PTSD-relevant brain areas. The current study was undertaken to determine whether methyl
