Neuropathological features of cerebrovascular diseases.

Optimal blood flow through a patent cerebral circulation is critical for supply of oxygen and nutrients for brain function. The integrity of vascular elements within arterial vessels of any calibre can be compromised by various disease processes. Pathological changes in the walls of veins and the venous system may also alter the dynamics of cerebral perfusion. The consequences of both systemic vascular and cerebrovascular diseases range from acute focal changes to irreversible chronic restructuring of the brain parenchyma. Cerebral infarcts of different sizes may instigate a cascade of program

Neuropathological report of propionic acidemia.

Propionic acidemia (PA) is an autosomal recessive inheritable metabolic disease caused by mutations in the propionyl CoA carboxylase gene (PCC) that affects multiple systems of the human body. Here, we report neuropathological findings of a PA patient. The patient was a male infant who presented with increasing lethargy and poor feeding from four days postpartum. He gradually became comatose and died from complications after liver transplantation at three months old. The results of laboratory examination were consistent with PA, and genetic analysis revealed compound heterozygous mutations in

Neuropathological alterations in cocaine abuse.

Cocaine abuse represents a significant health issue worldwide. Besides cardiovascular complications, psychiatric and neurologic symptoms are the most common manifestations of cocaine toxicity. In cocaine abusers brain abnormalities have been shown with computed tomography (CT), magnetic resonance imaging (MRI), positron emission tomography (PET), or single photon emission computed tomography (SPECT). The spectrum of neuropathologic alterations seen in cocaine abusers is broad. The major findings consist of vascular complications such as stroke, subarachnoid and intracerebral hemorrhages and ce

[Neuropathological diagnosis of prion disease].

Neuropathological diagnosis of prion disease consists of sequence analysis of PRNP (prion protein gene), located on chromosome 20 and characterization and visualization of deposited proteinase K-resistant prion protein (PrP(Sc)). SNP at 129 locus (M/V) and Type 1 and Type 2 difference in Western blot analysis of PrP(Sc) from the postmortem brain influence the clinical and pathological presentations. Prion disease is classified into sporadic, hereditary and infectious subtypes, but PrP(Sc) from almost all the subtypes can transmit the disease to transgenic mice expressing human PRNP. Variant CJ

Neuropathologic findings in essential tremor.

Pathologic findings, including cerebellar changes and brainstem Lewy bodies, distinguished 10 essential tremor (ET) cases from 12 controls. Numbers of torpedoes (p = 0.009) and Bergmann glia (p = 0.046) were increased in cases. Six cases (60%) had Lewy bodies vs 2 controls (16.7%) (odds ratio 7.5, 95% CI 1.04 to 54.1; p = 0.035). Four of these six had an atypical distribution of brainstem Lewy bodies. ET may be pathologically heterogeneous.

Neuropathologic substrate of cerebral palsy.

Animal models have assisted in understanding the mechanisms of brain injury underlying cerebral palsy. Nevertheless, no such models replicate every aspect of the human disease. This review summarizes the classic and more recent studies of the neuropathology of human perinatal brain injury most commonly associated with cerebral palsy, for use by researchers and clinicians alike who need to analyze published animal models with respect to their fidelity to the human disorder. The neuropathology underlying cerebral palsy includes white-matter injury, known as periventricular leukomalacia, as well

The neuropathology of hyperthermic seizures in the rat.

PURPOSE: Single and repeated hyperthermic seizures were induced in rats beginning at age 22 days to determine the neuroanatomic consequences to the hippocampus and to compare these changes with those in the hippocampi of patients with temporal lobe epilepsy (TLE) experiencing febrile seizures. METHODS: Hyperthermic seizures were induced by placing rats in a bath of water at 45 degrees C for 4 min. Seizures were visually observed, and some animals also were monitored electroencephalographically. Neurodegeneration was examined with a silver stain, whereas granule cell sprouting was detected wit

[Neuropathological aspects of Alzheimer disease].

In the present review we describe the morphological features of Alzheimer's disease (AD) and compare these findings with those obtained in argyrophilic grain disease, a frequent but often unrecognized form of late-onset dementia. Macroscopically AD brains exhibit a marked atrophy of the medical temporal lobe, including the hippocampus, entorhinal cortex and amygdala. Neuronal loss, decreased synapse density and the intra- and extracellular deposition of abnormal proteins constitute the histological hallmark lesions of AD. The intraneural accumulation of the microtubule-associated protein tau i

Neuropathologic aspects of lentiviral infections.

Studies of lentiviral infections of various animals and man have shown that all may invade the CNS and induce pathological lesions. This is well established in infections with VV, CAEV, SIV, HIV-1, and FIV. Although VV and CAEV do not cause an overt immunodeficiency, they share several features pertinent for the establishment of neuropathologic lesions with those that induce immunodeficiency. This holds especially true for the initial steps and early CNS lesions. 1) Infection of the CNS is from the blood stream. Although a definite proof of how the different viruses cross the blood-brain barri

[A neuropathological study on respirator brain].

A neuropathological study was conducted on 101 respirator brains. The patients comprised 62 men and 39 women, ranging in age from 11 to 84 years, the average being 49.4 years. Of these, 51 cases died of cerebrovascular disease, including 26 cases of subarachnoid hemorrhage. The average duration of mechanical supportive care was 99 hours. The mean weight of the brain at autopsy was 1,460 g. Characteristic pathological findings were brain edema, congestion, herniation and various subarachnoid hemorrhages. Brain consistency was soft despite adequate fixation. Histologically, the cytoplasm of neur

AIDS-myelopathy. A neuropathological study.

Vacuolar myelopathy belongs to the AIDS-associated diseases. It is characterized by vacuolation and infiltration of the long tracts of the spinal cord by macrophages. The clinical and morphological findings of 8 AIDS-patients with vacuolar myelopathy are reported here. The syndrome developed during the final stages of AIDS and was associated with HIV-encephalopathy in 5 cases. The vacuoles were mainly due to intramyelinic swelling and vacuolation. Vacuolated macrophages and axons contributed only to a minor degree. In one case only, HIV-antigens were detected immunohistochemically. The results

Neuropathologic findings in Reye syndrome.

Brain tissue from three patients with a clinical diagnosis of Reye syndrome was compared with tissue from three control patients. All Reye syndrome patients demonstrated cytotoxic cerebral edema, with swelling of astrocyte foot processes, which was not seen in controls. Myelin sheath splitting was seen both in controls and patients, but myelin blebs were uncommon in both. Mitochondrial changes also were not seen. Although cerebral edema is a nonspecific finding, it appears to be characteristic of Reye syndrome.

Neuropathologic correlates of leuko-araiosis.

We describe the pathologic findings in 17 persons with dementia, 12 of whom exhibited leuko-araiosis on computed tomographic scan. The presence of white matter pallor was confirmed on autopsy in 11 of these 12 cases, 9 with Alzheimer's disease and 2 with multi-infarct dementia. Two further patients, 1 with Alzheimer's disease and 1 with multi-infarct dementia, proved to have white matter changes on pathologic examination. White matter pallor coexisted with cerebral amyloid angiopathy in the brains of the patients with Alzheimer's disease. The presence of severe white matter pallor in patients

[Neuropathologic principles of senile dementia].

Several groups of senile dementias with differing causes and neurohistological substrates can be distinguished. Genetically determined Pick's disease produces severe neuronal atrophy; Alzheimer's disease is characterized by pathological neurofibrillary changes. Jakob-Creutzfeldt disease and Gerstmann-Sträussler-Scheinker disease are slow virus diseases induced by unconventional agents. These diseases, after an incubation time of several years, take the course of slowly progressive degenerative diseases of the central nervous system without inflammatory and specific immune reactions of the host

Möbius syndrome. Neuropathologic observations.

Neuropathologic findings in an infant with congenital right facial and bilateral lateral rectus palsy (Möbius syndrome) are presented. Multiple microscopic foci of necrosis were found in the lower pontine tegmentum, involving the region of the intramedullary course of 6th and 7th cranial nerves. The significance of pathologic findings in this infant and 14 additional published cases of Möbius syndrome is discussed.

A neuropathologic study of Whipple's disease.

A patient with Whipple's disease developed massive and irreversible CNS involvement after a drug-induced intestinal remission. The multifocal brain lesions were apparent grossly with the heaviest concentration being noted in rhinencephalon, diencephalon, and mesencephalon. This patient exhibited progressive neurologic deterioration, although viable organisms could not be identified in the brain at autopsy. Astrocytes, pericytes, and choroid plexus cells attempted to dispose of the organism, in additional to the traditionally implicated microglial and ependymal cell.

Disruption of BDNF signalling in neuropathologies.

The vital role of brain-derived neurotrophic factor (BDNF) in neuronal development, synaptic plasticity, and neuroprotection has been explored for decades. Therefore, the expression, processing, and signalling activities of this neurotrophin, which is reliant upon TrkB and p75NTR receptors, have been well characterised in both health and disease. This review summarises the latest findings on BDNF dysregulation in neuropathologies. Indeed, across diseases of both the central and peripheral nervous systems, BDNF signalling is frequently disrupted, contributing to neuronal dysfunction and degener

The Human Cerebellum as a Semaphore for Neuropathology.

The cerebellum has intricate connections either directly or indirectly with most levels of the brain. While many of these are functionally relevant, prominent neuropathologic changes that reflect the underlying anatomy arise from injury to a few major pathways. Identifying these pathologic sequelae can help to identify the cells involved in the underlying disease. The cerebellum has two distinct components: the aesthetically pleasing cortical dendritic folia and the deep nuclei. The uniform and monotonous cerebellar cortex contains molecular, Purkinje, and granular layers. Several deep nuclei
